Cultivos celulares podem ser utilizados para isolamento viral, caracterização de novas amostras virais e produção de imunobiológicos. Podem ser empregados cultivos celulares primários, secundários ou de linha. Estes últimos podem ser obtidos pela transformação física, química ou biológica de cultivos primários ou secundários. Para verificar a interação entre células transformadas com o Ag T do vírus símio 40 (SV40) e os Lentivírus de Pequenos Ruminantes (SRLV), amostras brasileiras de SRLV foram utilizadas para inoculação em cultivos celulares de membrana sinovial ovina transformados pelo Ag T e comparadas à amostras inoculadas em cultivos não transformados. Inicialmente, as células transformadas pelo Ag T foram caracterizadas e observou-se um aumento na cinética de crescimento e alterações no cariótipo, provavelmente induzidas pela presença do Ag T. Este foi detectado por PCR no núcleo e no citoplasma das células transformadas e sua expressão, confirmada através de RT-PCR. A fim de avaliar a permissividade e a possível seleção de populações virais em células transformadas, dois isolados, um de maedi-visna dos ovinos e um de artrite-encefalite caprina, foram inoculados em células MSO e TMSOpSV1. Através da análise de sequências dos genes gag e env e LTR obtidos por PCR, procurou-se avaliar a variabilidade viral em função do tipo de célula utilizada. Analisando-se as seqüências obtidas pelo método de Maximum Likelihood, embora tenha sido observada variabilidade viral, esta não estava associada ao tipo celular utilizado para inoculação viral.

Vliv maedi-visna viru na reprodukční ukazatele ve vybraném chovu plemene šumavská ovce

The aim of the thesis was to evaluate the influence of maedi-visna on reproductive indicators in selected Šumava sheep breeding. Serological test ELISA was accomplished in the evaluated herd, and its results were used for comparing reproductive rates of healthy and infected ewes. Maedi-visna disease was proved in 16 out of 193 samples (8, 29 %). Statistically significant influence of maedi-visna was shown in indicators such as age of infected ewes and number of reared lambs. For other indicators (number of live birth lambs, lambs weight in 100 days, occurrence of sterility, mortality and stillborn lambs) higher success rate was observed in maedi-visna negative ewes, however these findings were not statistically significant. Maedi-visna is incurable disease and most important tool how to avoid this disease is preventive protection of the sheep breedings.

Foram avaliadas 400 amostras de soro sangüíneo de ovinos de oito propriedades de corte, com criação em sistema semi-intensivo, de diferentes municípios da região. Nenhuma das amostras de soro foi reagente na prova de IDGA. Com base nos resultados obtidos, foi verificada uma discordância com estudos realizados em outros estados brasileiros, nos quais a prevalência da doença vem aumentando progressivamente. / The present study aimed to verify the prevalence of the ovine progressive pneumonia in Botucatu region by means of imunodiffusion test in agar-gel. Serum samples of 400 sheep from eight specific farms for meat, with type of semi-intensive breding of different areas. All the samples tested were negative to Maedi-Visna. With base in the obtained results, is verified a discordance when compared with other studies of other Brazilians states, where the prevalence of the disease comes increasing progressively.

Study on the effects of a natural Maedi visna virus infection on sheep productivity

A cohort study was conducted in order to measure the effect of the chronic indurative lymphocytic mastitis caused by the South African strain ofMaedi visna virus (SA-OMVV) on the pre-weaning growth of lambs born of naturally infected and uninfected ewes kept under similar conditions. 50 naturally infected ewes and 40 controls from an MVV-free source were purchased and kept separately. All ewes were of the same breed - the Dorper¬and 3 to 4 years old. From the adaptation period, through mating, pregnancy and lactation periods they were monitored for MVV antibodies and managed under similar conditions. The lambs were weighed at birth and thereafter every two weeks until the age of 90 days, when they were weaned. The ewes were slaughtered, their udders examined histologically and the lesions were assessed by counting typical lymphocytic follicles. Although the observed values indicated a correlation between the number of follicles in the udder and the reduction in the growth rate of the lambs, this was not statistically significant. Similarly, despite higher counts of lymphoid follicles in the udder of sero-positive ewes as compared to sero-negatives and the observed lower ewe productivity indexes (EPI) in infected ewes, no statistically significant differences were found in the EPI of ewes in different follicle categories. Carried out 64 questionnaires, examined 327 sera from 246 goats and sheep. The worms, foot pad dermatitis and lymphadenitis were more health problems reported by producers. The sanitary practices adopted, in general, are little used in the properties of goats and sheep. The semi-intensive system (67.7%) is the most used and 71% of producers buy animals from other places. The frequency of seropositive sheep B. ovis was 28% accounting for 71.9% of farms with sheep positive for B. ovis in the study. 25% of farms surveyed had one or more seropositive goats and sheep lentiviruses. Of the 138 animals tested positive for leptospirosis serovar Autumnalis was the most frequent, being found in 76.19% 38.88% in goats and sheep. In the study of case-control analysis of risk factors, herd size and purchase of animals showed a strong association. The caseous lymphadenitis has a high incidence in herds serological (92.2%). It can be concluded that the animals are seropositive for exposure to infectious diseases and inadequate management observed in most producers can spread the disease spread and put other animals at risk. / O objetivo deste trabalho foi caracterizar os sistemas de criações de caprinos e ovinos participantes das exposições agropecuárias, assim como estudar a ocorrência das infecções por Brucella ovis em ovinos e lentivírus, linfadenite caseosa e leptospirose em caprinos e ovinos e seus fatores de risco. Foram realizados 64 questionários, sendo examinados 327 e 246 soros de caprinos e ovinos, respectivamente. A verminose, linfadenite e pododermatite foram os problemas sanitários mais relatados pelos produtores. As práticas sanitárias adotadas, de maneira geral, são pouco utilizadas nas propriedades de caprinos e ovinos. O sistema semi-intensivo (67,7%) é o mais utilizado e 71% dos produtores compram animais de outros lugares. A frequência de ovinos soropositivos para B. ovis foi 28% representado em 71,9% das propriedades com criação de ovinos. 25% das propriedades investigadas apresentaram um ou mais caprinos e ovinos soropositivos para os lentivírus. Dos 138 animais soropositivos para leptospirose, o sorovar Autumnalis foi o mais frequente, sendo encontrado em 76,19% dos caprinos e 38,88% dos ovinos. No estudo de caso-controle para análise de fatores de risco, tamanho do rebanho e compra de animais apresentaram associação. A linfadenite caseosa possui uma elevada soropositividade nos rebanhos (92,2%). Pode concluir que os animais de exposição estão soropositivos para algumas doenças infecciosas e o manejo inadequado observado na maioria dos produtores, pode disseminar a propagação de doenças e colocar outros animais em risco.

Maedi-Visna virus : the development of serum and whole blood immunodiagnostic assays.

This thesis describes the development of serum and whole blood immunodiagnostic assays for Maedi-Visna virus (MVV). All previously described recombinant MVV ELISA assays utilised either the core p25 or transmembrane (TM) proteins alone, or combined, but as individual proteins. The p25 and TM genes of MVV were cloned individually into the pGEX-2T expression vector. Both proteins were expressed as a combined fusion protein in frame with glutathione S-transferase (GST). The purified recombinant antigens (GST-TM and GST-TM-p25) were used to develop a MVV ELISA. Sera from 46 positive and 46 negative sheep were tested using the GST-TM and GST-TM-p25 ELISAs and a commercial p25 EIA kit. A two-graph receiver operating characteristic (TG-ROC) analysis program was used to interpret the data. The GST-TM-p25 ELISA was more sensitive than the commercial assay which is based on the p25 antigen alone and more specific than the GST-TM ELISA. The GST-TM-p25 ELISA showed a sensitivity and specificity of 100%. The human AIDS lentivirus transmembrane (TM) glycoprotein portion of the envelope viral protein has been identified as the antigen most consistently recognised by antibodies. There is suggestive evidence that the same applies to MVV as the GST-TM fusion protein, expressed in E. coli, has comparable sensitivity to the GST-TM-p25 fusion protein, but lacks specificity. However, due to the hydrophobic nature of the MVV TM protein, purification of the expressed fusion protein required lengthy purification protocols. This was despite the fact that only a truncated version of the TM protein was expressed. This prompted investigating an alternative expression system that could possibly circumvent the above mentioned problems. The yeast Pichia pastoris is known to be suitable for the high-level expression of heterologous proteins which are secreted into the culture supernatant. These features made P. pastoris an attractive host for the expression of the hydrophobic TM protein of MVV. However, limited success was achieved as only low expression levels were obtained and detection and quantification was only accomplished by means of ELISA. Evaluation of the diagnostic performance of the P. pastoris expressed MVV TM-polypeptide was performed using a panel of 36 confirmed negative and positive sera, and evaluated using a TG-ROC analysis programme, which yielded an equal Se and Sp of 83%. The use of a novel rapid immunoassay system, which allows the detection of circulating antibodies in whole blood, has been investigated for use as a MVV diagnostic assay. The central feature of this immunoassay lies in a monoclonal antibody against a glycophorin epitope present on all sheep erythrocytes. A Fab'-peptide conjugate was constructed by coupling a synthetic peptide, corresponding to a sequence from MVV TM protein, to the hinge region of the Fab' fragment of the antisheep erythrocyte antibody. Within the limited number of 10 seronegative and 10 seropositive samples the autologous red blood cell agglutination assay had a sensitivity of 90% and a specificity of 80%. Despite the limitations and difficulties encountered, the use of such rapid whole blood immunodiagnostic assays for MVV holds promise. / Thesis (Ph.D.)-University of Natal, Durban, 1997.
