CLA causes significant economic losses in commercial goat herds; the best procedure to avoid spread of this disease is elimination of infected animals. Secreted antigens from T1 strain bacteria grown in brain heart infusion (BHI) broth were tested in an indirect ELISA system to determine if this system could be used to identify specific immunoglobulins against C. pseudotuberculosis. The electrophoretic profile of the BHI antigen was analyzed, as well as the recognition pattern by CLA infected sheep sera. The ELISA results were compared to Multiplex PCR assay and IFN-gamma production. The ELISA was able to discriminate between negative and positive animals, with a sensitivity of 89% and a specificity of 99%, and a large difference between positive and negative optical density values. When this assay was compared with other immunological and molecular methods, it was found only two discrepant results among thirty-two samples. We concluded that an ELISA using secreted antigens from C. pseudotuberculosis T1 strain growth in BHI broth culture would be a reliable tool for the serodiagnosis of caseous ymphadenitis in sheep. / Salvador

Entretanto, diversos fatores na criação desses animais ainda necessitam melhorar, principalmente na área sanitária, na qual muitas doenças acometem o rebanho, dentre elas, a Linfadenite Caseosa, doença causada por Corynebacterium pseudotuberculosis, uma bactéria que apresenta sua toxicidade através de genes de virulência, assim como pela produção de biofilme, que protege a bactéria de ambientes hostis. Dessa forma, estudos sobre esse patógeno são necessários, a exemplo do sequenciamento genômico, na busca de conhecer genes para análises de biomoléculas e com isso compreender melhor a sua antigenicidade e imunogenicidade com o objetivo de melhorar a especificidade e sensibilidade de testes sorológicos, utilizados como prevenção da doença no rebanho. Além disso, é essencial entender o mecanismo de patogenicidade desta bactéria no hospedeiro e suas formas de defesa no sistema imunológico do animal. Assim, no presente estudo, foram caracterizados os genomas de quatro cepas de C. pseudotuberculosis, comparando as cepas produtoras (OVI2C, CAPJ4) e não produtoras (OVI03, CAP3W) de biofilme. As cepas foram extraídas para produção de antígenos somáticos e de superfícies. A partir de estudos prévios com immunoblotting, foram escolhidas as cepas de origem caprina (CAPJ4 e CAP3W) para o estudo in vivo com dezoito caprinos da raça Canindé, divididos em três grupos experimentais, um grupo controle e dois grupos infectados com cepas de C. pseudotuberculosis, para avaliar a patogenia clinicamente, realizando hemograma, além de outras análises sorológicas através do ensaio de ELISA e Western Blotting. Também foi realizada a cultura de células sanguíneas para o estudo da imunidade celular com IFN- e cultivo de células mononucleares do sangue periférico para observar a modulação da expressão de proteínas do sistema GTPases, Rab 5 e Rab 7. Após esses estudos, observou-se que os genes contidos no genoma das cepas de C. pseudotuberculosis possuem similaridade, mostrando que eles são altamente conservados. A extração das bactérias resultou em antígenos de superfície F3, que apresentaram reatividade no immunoblotting frente aos soros de ovinos experimentalmente infectados. De todos os antígenos, os de origem caprina, apresentaram melhor antigenicidade. No experimento in vivo, após a infecção dos animais, foram realizadas outras colheitas de sangue e foi possível observar que a cepa produtora de biofilme é mais resistente, causando danos maiores aos animais. Para as condições de infecção experimental, os valores do hemograma estão dentro dos parâmetros. Na análise sorológica os caprinos infectados experimentalmente responderam à exposição primária, o que contribui para o aumento da imunoglobulina IgG. A produção de IFN-  também foi observada. Foi verificado que as cepas de C. pseudotuberculosis produtora e não produtora de biofilme podem alterar a expressão de proteínas Rab 5 e Rab 7 em macrófagos derivados de células mononucleares do sangue periférico cultivados in vitro. Esses resultados forneceram informações importantes para a produção de testes diagnósticos, com maior acurácia. / The consumption of sheep and goat products is frequent and increases the values of world economy, so it is necessary to produce large scale and quality to meet the demand of population. However, several factors in the breeding of these animals still have to improve, especially in sanitary area, where many diseases affect the cattle, among them, Caseous Lymphadenitis, caused by Corynebacterium pseudotuberculosis, a bacterium that presents its toxicity through virulence genes, also by production of biofilm that protects the bacteria from hostile environments. Thus, studies of this pathogen are necessary, such as genomic sequencing, in search to know antigenic genes for biomolecule analysis and with this to understand better its antigenicity and immunogenicity, to be possible improve the specificity and sensitivity of serological tests, that are used as prevention of disease in the herd. Besides, it is essential to understand the mechanism of pathogenicity of this bacterium in host and its forms of defense against the animals immune system. At the present study, the genomes of four strains of C. pseudotuberculosis were characterized, comparing the biofilm producing (OVI2C, CAPJ4) and non-producing (OVI03, CAP3W) strains. These strains were extracted for production of somatic and surfaces antigens. From previous studies with immunoblottingting, strains of goat origin (CAPJ4 and CAP3W) were chosen for in vivo study with 18 Canindé goats, divided into three experimental groups, one control group and two groups infected with strains of C. pseudotuberculosis, to evaluate clinically the pathogenesis and through hemogram, in addition to other serological tests as ELISA assay and Western Blotting. Also was performed blood cell culture for the study of cellular immunity with IFN-, as culture of peripheral blood mononuclear cells to observe the modulation of expression of GTPases, Rab 5 and Rab 7 system proteins. After these studies, it was observed that the genes contained in the genome of the strains of C. pseudotuberculosis are genetically homogeneous, showing that they are highly conserved. The extraction of bacteria resulted in F3 surface antigens, that showed reactivity in immunoblottingting against sera from experimentally infected sheep. Of all antigens, those of goat origin had better antigenicity. At the in vivo experiment, after the infection of animals, other blood samples were collected and it was possible to observe that the biofilm producing strain is more resistant, causing greater damages to animals. For the conditions of experimental infection, blood count values are between normal limits. In the serological analysis, the experimentally infected goats responded to primary exposure, which contributes to increase of immunoglobulin IgG. The production of IFN- was also observed. In conclusion, it was observed that strains of C. pseudotuberculosis producing and not producing biofilm can modify the expression of Rab proteins in macrophages derived from mononuclear cells of peripheral blood cultivated in vitro. These results provided important information for production of diagnostic tests, with greater accuracy.

In Brazil the clinical prevalence CLA is 30%. The control measures are vaccination and diagnosis of infected animals. Currently, commercial vaccines are not completely effective, the same occur with the methods of diagnosis, because the medical signs are not immediately detectable. By sequencing and additional proteomic analysis, several targets were identified, including cp1002_0369 gene, which codifies a secreted protein which is probably described as a potentially antigenic phosphoesterase, that can subsequently be used in the development of recombinant vaccines and development of diagnostic tests. This protein was used as recombinant and it was tested as subunit vaccine and as a DNA vaccine and also to develop a diagnostic test based on ELISA. For this, the cp1002_0369 gene was cloned in the eukaryotic expression vector pTARGET and in a vector of expression in prokaryotes pAE. The recombinant protein CP0369 was purified and evaluated as a recombinant vaccine associated with both of the adjuvants xanthan and aluminum hydroxide and then measured in an indirect ELISA with sheep serum. The immunoprotective potential was assessed in a murine model by challenge with 104 CFU of strain Mic-6 of C. pseudotuberculosis in 9 vaccinal groups. The vaccine formulation that increased the survival rate of animals after challenge was the group containing the recombinant protein (rCP0369 + aluminum hydroxide). The ELISA for diagnostic of CL was evaluated by ROC analysis of 182 sera and showed a sensitivity and specificity of 90% and 75.6%, respectively. The format of ELISA assay (ELISA-rCP0369) can be used in the diagnosis of LC sheep herds with a good sensitivity index. However, new vaccine strategies employing the CP0369 protein will be evaluated to improve the effectiveness of the vaccine. / A linfadenite Caseosa (LC) é uma doença que acomete principalmente pequenos ruminantes sendo causada pela bactéria Corynebacterium pseudotuberculosis. No Brasil a prevalência clínica da LC é de 30%. As medidas de controle são a vacinação e o diagnóstico dos animais infectados. As vacinas disponíveis comercialmente não são totalmente eficazes, o mesmo ocorre com os métodos de diagnóstico, pois os sinais clínicos não são perceptíveis de imediato. Através de sequenciamento e da análise proteômica complementar foram identificados vários alvos, dentre eles o gene cp1002_0369, que codifica para uma proteína secretada sendo esta descrita provavelmente como uma fosfoesterase, potencialmente antigênica, a qual pode vir a ser utilizada no desenvolvimento de vacinas recombinantes e no desenvolvimento de testes de diagnóstico. Esta proteína foi utilizada na forma recombinante como vacina de subunidade e como vacina de DNA e também para o desenvolvimento de um teste de diagnóstico baseado em ELISA. Para isso o gene cp1002_0369 foi clonado no vetor de expressão em eucariotos pTARGET e de expressão em procariotos pAE. A proteína CP0369 recombinante foi purificada e avaliada como vacina recombinante associada aos adjuvantes xantana e hidróxido de alumínio e em um ELISA indireto com soros de ovinos. O potencial imunoprotetor foi avaliado em modelo murino através de desafio com a 104 UFC da cepa Mic6 de C. pseudotuberculosis em 9 grupos vacinais. A formulação vacinal que aumentou a taxa de sobrevida dos animais após o desafio foi o grupo contendo a proteína recombinante (rCP0369 + hidróxido de alumínio). O ELISA para diagnóstico de LC foi avaliado através da análise ROC em um total de 182 soros revelou uma sensibilidade e a especificidade de 90% e 75,6%, respectivamente. O formato de ELISA (ELISA-rCP0369) pode ser utilizado no diagnóstico de LC em rebanhos ovinos com bom índice de sensibilidade. No entanto, novas estratégias vacinas empregando a proteína CP0369 serão avaliadas para melhorar a eficácia da vacina.

Esta bactéria faz parte do grupo CMNR (Corynebacterium, Mycobacterium, Nocardia e Rhodococcus), que demonstra grande importância veterinária e médica, uma vez que estes micro-organismos comumente infectam animais, podendo infectar o homem, causando perdas econômicas pela ineficácia ou alto custo de terapias existentes. Um exemplo seria a linfadenite caseosa (LC) causada por C. pseudotuberculosis, que afeta particularmente a pecuária de caprinos e ovinos, com a condenação da carcaça e redução da produção de lã e de carne. A transmissão da doença e contágio dos animais é direta, muitas vezes através da alimentação e ingestão de água em local contaminado por animais doentes. Essa doença possui incidência na pecuária mundial, principalmente de caprinos e ovinos, havendo registros de ocorrência no Brasil, Europa, Oriente Médio, Austrália, Nova Zelândia, África do Sul, Canadá e Estados Unidos e mesmo com todos os avanços tecnológicos, ainda não há métodos de prevenção totalmente eficazes, como vacinas e medicamentos, tampouco para o tratamento de animais infectados, que geralmente são de custo elevado, por longos períodos e sem a garantia de cura ou de isenção de reincidência da LC. Deste modo, técnicas mais rápidas e fáceis para detecção e diagnóstico da doença, bem como para seu tratamento, se tornam imprescindíveis, evitando não só a disseminação da doença, mas também suas consequentes perdas econômicas. Atualmente, devido ao uso indiscriminado de antibióticos para o tratamento de infecções de origem bacteriana, bem como à constante exposição destes micro-organismos a essas drogas em ambientes hospitalares, observa-se o desenvolvimento de micro-organismos resistentes às terapias disponíveis, sendo um desafio mundial a descoberta e elaboração de tratamentos eficazes para a prevenção, controle e eliminação destes patógenos, como alternativa aos já existentes. Visando terapias alternativas e direcionadas para infecção por C. pseudotuberculosis, as proteínas tioredoxina, tioredoxina redutase e diphtheria toxin repressor foram estudadas no presente trabalho, a fim de melhor compreender este micro-organismo. / Corynebacterium pseudotuberculosis (C. pseudotuberculosis), is a gram-positive, facultative anaerobe, pleomorphic, non-sporulating bacterium with two biotopes or biovars, being the biovar equi capable of infecting horses, whereas the biotype called ovis infects small ruminants. It is part of the CMNR group (Corynebacterium, Mycobacterium, Nocardia and Rhodococcus), which demonstrates great veterinary and medical importance, since these common microorganisms infect animals and can infect humans, causing economic losses due to the inefficiency or high cost of existing therapies. An example is a caseous lymphadenitis (LC) caused by C. pseudotuberculosis, which particularly affects the goats and sheep livestock, with carcass condemnation and reduction of wool and meat production. The transmission of disease and the contagion of animals is direct, often through feeding and drinking water in places contaminated by sick animals. This disease has an incidence in the world livestock, mainly of goats and sheep, with occurrence records in Brazil, Europe, the Middle East, Australia, New Zealand, South Africa, Canada and the United States and even with all technological advances, still there are no totally effective prevention methods, such as vaccines and medications, nor for the treatment of infected animals, which are usually of a high cost, for long periods and without guarantee of cure or exemption from recurrence of LC. In this way, faster and easier techniques for the detection of the diagnosis of this disease as well as for its treatment become essential, avoiding not only a spread of the disease, but also its consequent economic losses. Currently, the use of indiscriminate antibiotics for the treatment of infections of bacterial origin, as well as the constant exposure of these microorganisms to these drugs in hospital environments, shows the development of microorganisms resistant to the available therapies, being one world-wide challenge the elaboration of effective treatments for the prevention, control and elimination of these pathogens, as an alternative to the existing ones. Aiming alternative therapies for infection by C. pseudotuberculosis, proteins such as thioredoxin, thioredoxin reductase, diphtheria toxin repressor, were studied in the present work, for a better comprehension of this microorganism.

Carried out 64 questionnaires, examined 327 sera from 246 goats and sheep. The worms, foot pad dermatitis and lymphadenitis were more health problems reported by producers. The sanitary practices adopted, in general, are little used in the properties of goats and sheep. The semi-intensive system (67.7%) is the most used and 71% of producers buy animals from other places. The frequency of seropositive sheep B. ovis was 28% accounting for 71.9% of farms with sheep positive for B. ovis in the study. 25% of farms surveyed had one or more seropositive goats and sheep lentiviruses. Of the 138 animals tested positive for leptospirosis serovar Autumnalis was the most frequent, being found in 76.19% 38.88% in goats and sheep. In the study of case-control analysis of risk factors, herd size and purchase of animals showed a strong association. The caseous lymphadenitis has a high incidence in herds serological (92.2%). It can be concluded that the animals are seropositive for exposure to infectious diseases and inadequate management observed in most producers can spread the disease spread and put other animals at risk. / O objetivo deste trabalho foi caracterizar os sistemas de criações de caprinos e ovinos participantes das exposições agropecuárias, assim como estudar a ocorrência das infecções por Brucella ovis em ovinos e lentivírus, linfadenite caseosa e leptospirose em caprinos e ovinos e seus fatores de risco. Foram realizados 64 questionários, sendo examinados 327 e 246 soros de caprinos e ovinos, respectivamente. A verminose, linfadenite e pododermatite foram os problemas sanitários mais relatados pelos produtores. As práticas sanitárias adotadas, de maneira geral, são pouco utilizadas nas propriedades de caprinos e ovinos. O sistema semi-intensivo (67,7%) é o mais utilizado e 71% dos produtores compram animais de outros lugares. A frequência de ovinos soropositivos para B. ovis foi 28% representado em 71,9% das propriedades com criação de ovinos. 25% das propriedades investigadas apresentaram um ou mais caprinos e ovinos soropositivos para os lentivírus. Dos 138 animais soropositivos para leptospirose, o sorovar Autumnalis foi o mais frequente, sendo encontrado em 76,19% dos caprinos e 38,88% dos ovinos. No estudo de caso-controle para análise de fatores de risco, tamanho do rebanho e compra de animais apresentaram associação. A linfadenite caseosa possui uma elevada soropositividade nos rebanhos (92,2%). Pode concluir que os animais de exposição estão soropositivos para algumas doenças infecciosas e o manejo inadequado observado na maioria dos produtores, pode disseminar a propagação de doenças e colocar outros animais em risco.

Etude fonctionnelle du génome de Corynebacterium pseudotuberculosis par différentes stratégies protéomiques / Functional genome analysis of Corynebacterium pseudotuberculosis using various proteomic approaches

Corynebacterium pseudotuberculosis est une bactérie pathogène intracellulaire facultative, divisée en deux biovars : C. pseudotuberculosis ovis, agent de la lymphadénite caséeuse (petits ruminants), et C. pseudotuberculosis equi, responsable de lymphangites ulcéreuses (chevaux), mammites (bovins) et d’œdèmes cutanés (buffles). La génomique fonctionnelle a pour but d'élucider le rôle que joue chaque gène dans l'organisme, ainsi que l'interaction de ces gènes dans un réseau biologique. Au cours de ce travail de thèse, différentes stratégies protéomiques ont été adoptées pour l’étude fonctionnelle du génome de C. pseudotuberculosis : i) l’analyse du protéome extracellulaire des souches 1002_ovis et C231_ovis a permis la caractérisation totale de 60 protéines différentes de C. pseudotuberculosis dont 18 protéines sont régulées différemment ;ii) le protéome de la souche 1002_ovis été analysé en réponse au stress nitrosant révélant 58 protéines différentiellement régulées et impliquées dans différents processus biologiques susceptibles de favoriser la survie à ce stress ; iii) les souches 1002_ovis et 258_equi ont été utilisées pour induire des infections expérimentales sur modèle souris. L’analyse de leur protéome extracellulaire avant et après passage en série sur souris a permis d’identifier 250 protéines différentiellement régulées touchant diverses fonctions. Pour conclure, ce travail a permis de générer une base de données de protéines et de valider la fonctionnalité de différents gènes jusqu’alors prédits in silico seulement et des informations importantes sur les facteu / Corynebacterium pseudotuberculosis is a facultative intracellular pathogenic bacterium, subdivided into two biovars: C. pseudotuberculosis ovis, agent of caseous lymphadenitis (small ruminants), and C. pseudotuberculosis equi, which causes ulcerative lymphangitis (equines), mastitis (bovines), and oedematous skin disease (buffalos). Functional genomics aims to elucidate the role that each gene plays in organism, as well as the interactions of these genes into a biological network. In this PhD work, various proteomic approaches were used to evaluate the functional genome of C. pseudotuberculosis: i) the analysis of the exoproteome of strains 1002_ovis and C231_ovis enabled the characterization of 60 proteins of C. pseudotuberculosis of which 18 were differentially regulated; ii) the proteome of the strain 1002_ovis was analyzed after a nitrosative stress revealing 58 proteins differentially regulated when compared to the control conditionsThese 58 proteins are involved in different biological processes which may favor the survival of this pathogen under exposition to nitrosative stress; iii) the strains 1002_ovis and 258_equi were used in a murine model of experimental infection. A comparative analysis of their extracellular proteomes before and after passage in mice revealed that 250 proteins ibnvolved in various functions were differentially regulated in C. pseudotuberculosis. Étude prospective des causes de mortalité chez l'espèce caprine avec emphase sur la lymphadénite caséeuse

En Amérique du Nord, et particulièrement au Canada, il y a très peu de données sur l’incidence des causes de mortalité chez l’espèce caprine. Le premier objectif de cette étude était de déterminer les principales causes de mortalité chez les chèvres au Québec. Depuis 2006, avec l’arrêt de la vaccination contre la lymphadénite caséeuse, les éleveurs de caprins laitiers et de boucherie du Québec ont rapporté une recrudescence des abcès chez leur bétail. Le second but de cette étude était de déterminer l’importance de la lymphadénite caséeuse dans le dépérissement et la mortalité des chèvres du Québec. Cent-cinquante-deux chèvres provenant de 13 élevages différents ont été soumises pour nécropsie et la cause de mortalité, de même que la présence d’abcès (s’il y a lieu), leur localisation et leur cause furent compilés. Les mortalités proportionnelles étaient, par ordre décroissant : l’entérotoxémie de type D (n= 26; 17,1%), la pneumonie (n= 21; 13,8%), la paratuberculose (n= 16; 10,5%), listériose encéphalitique (n= 10; 6,6%), la toxémie de gestation (n= 8; 5,3%), l’arthrite-encéphalite caprine (n= 7; 4,6%) et la lymphadénite caséeuse (n= 6; 3,9%). La lymphadénite caséeuse a été diagnostiquée chez 24,3% des chèvres soumises, mais sans être une cause majeure de dépérissement ou de mortalité. Les abcès étaient internes dans 54,1% des cas. Au total, la paratuberculose a été diagnostiquée chez 29 chèvres (16 en étant décédées) et fut considérée comme une cause majeure de dépérissement, d’émaciation et de mortalité. Le développement et l’implantation de mesures préventives contre cette maladie seraient donc à envisager dans le futur. / In North America, and especially in Canada, the incidence and the causes of death in goats are not well documented. The first objective of this study was to determine the main causes of mortality in goats in Quebec. Dairy and meat goat farmers had reported an increase in abscesses in their herd since vaccination for caseous lymphadenitis stopped being available (2006) in Quebec. The second objective of this study was to determine the importance of caseous lymphadenits in wasting and mortality in caprine herds from Quebec. One hundred and fifty-two goats from 13 different herds from Quebec were submitted for necropsy and the cause of mortality, as well as the presence, location and cause of abscesses (if present) were recorded. Proportional mortalities were, in decreasing order: type D enterotoxemia (n= 26; 17,1%), pneumonia (n= 21; 13,8%), paratuberculosis (n= 16; 10,5%), encephalitic listeriosis (n= 10; 6,6%), pregnancy toxemia (n= 8; 5,3%), caprine arthritis-encephalitis (n= 7; 4,6%) and caseous lymphadenitis (n= 6; 3,9%). Caseous lymphadenitis was diagnosed in 24.3% of the submitted goats, but was not a major cause of debilitation or mortality. Abscesses were localized internally in 54.1% of the cases. Overall, paratuberculosis was diagnosed in 29 goats (16 of which died of the condition) and was considered a major cause of wasting, emaciation and mortality. Development and implementation of preventive measures to control this disease in goat herds would certainly be relevant in the future.
