Koherencí řízený holografický mikroskop ve výzkumu životního cyklu buňky / Coherence-controlled holographic microscope in cell's life cycle research

The goal of this diploma thesis was using of a coherence-controlled holographic microscope in cell’s life research. A brief history of interference microscopy and it’s applications in biology is described. Also other microscopy techniques routinely used for transparent objects imaging are mentioned and the biology of cell’s life cycle briefly explained. Characteristics describing the shape of a cell were proposed and tested with respect to identification of particular phases of its life cycle. The method of dynamic phase differences was modified in order to distinguish the internal motion of cell’s mass from the movement of the whole cell. Selected characteristics were used to evaluate observations carried out with the holographic microscope and the possibilities of their further applications were depicted. In conclusion, obtained findings were summarized and modifications of microscope construction as well as data-processing software were suggested.

This work is aimed at deposition of fibers, which are able to penetrate deeper in the human respiratory airways and cause health hazards. Opening chapters are dedicated to classification of aerosols, human lung anatomy basics and methods for measurement of micrometer-sized aerosols. An experiment was carried out to investigate the deposition of fibers. Fibers were delivered into the silicon cast of the human airways and data of deposition fraction and efficiency were acquired. A new method was established to acquire the data. This method works on a principle of image analysis. Results of the new method were compared with a standard method, which follows a methodology NIOSH 7400.

Měření rezonančních vlastností plazmonických nanostruktur v transmisním a reflexním režimu spektroskopie dalekých polí / Measurement of resonant properties of plasmonic nanostructures in transmission and reflection mode of far-field spectroscopy

Far-field optical spectroscopy allows for measurement of plasmonic resonances on metallic nanostructures of various shapes and sizes when illuminated by continuous light. Employing dark-field microscopy makes it possible to measure scattering on a localised plasmon excited on a single, isolated nanostructure. For the purpose of these measurements an apparatus based on commercial dual microscope system Nanonics has been put together. Experiments carried out during work on this thesis have shown a great sensitivity of the apparatus in its present form, allowing to measure a plasmonic resonance of a single nanoparticle. Proposed improvements of the apparatus should lead to even greater sensitivity and precision of measurements in the near future.

Charakterizace magnetických nanostruktur pomocí mikroskopie magnetických sil / Characterization of magnetic nanostructures by magnetic force microscopy

The thesis deals with magnetic force microscopy of soft magnetic nanostructures, mainly NiFe nanowires and thin-film elements such as discs. The thesis covers almost all aspects related to this technique - i.e. from preparation of magnetic probes and magnetic nanowires, through the measurement itself to micromagnetic simulations of the investigated samples. We observed the cores of magnetic vortices, tiny objects, both with commercial and our home-coated probes. Even domain walls in nanowires 50 nm in diameter were captured with this technique. We prepared functional probes with various magnetic coatings: hard magnetic Co, CoCr and soft NiFe. Hard probes give better signal, whereas the soft ones are more suitable for the measurement of soft magnetic structures as they do not influence significantly the imaged sample. Our probes are at least comparable with the standard commercial probes. The simulations are in most cases in a good agreement with the measurement and the theory. Further, we present our preliminary results of the probe-sample interaction modelling, which can be exploited for the simulation of magnetic force microscopy image even in the case of probe induced perturbations of the sample.

The first part contains a brief introduction of the company NTS Prometal Machining s.r.o. and brief description of the product. The following section is an analysis of existing manufacturing technology. The practical part is focusing on the proposal of rationalization versions of the production process. After evaluation of all possible options of rationalization there has been developed a new technological process with a respect to time and cost savings. Finally there was carried out economical evaluation and recommendation for further production of this component.

Analýza mazaného kontaktu poddajných těles / Analysis of lubricated compliant contact

This diploma thesis deals with the proposal of methodology for film thickness eva- luation within compliant contacts. With respect to characteristics of such contact pairs, like variable film thickness, high surface roughness, or poor conductivity and reflectivity, the usage of conventional experimental methods is particularly complicated. In present study, an optical method based on the principle of fluo- rescent microscopy was employed in present thesis. An evaluation algorithm in- volving background normalization and calibration of fluorescent intensity to film thickness was created in a form of experimental software. The proposed algori- thm was validated by using elastohydrodynamic contact formed between ceramic ball and glass disc. The measured film thickness was compared with theoretical prediction, while very good agreement of obtained data was observed. Further, a series of experiments with compliant samples was conducted, while the central film thickness was evaluated as a function of mean speed, applied lubricant, ap- plied load and slide-to-roll ratio. The last part of the thesis is focused on results analysis and discussion considering the previously published literature.

The work presents a method of dynamic testing of thin hard coatings on a newly constructed prototype of impact wear tester. It provides a comparison with standard methods of layer testing and presents new experimental results of dynamic impact wear test of the coatings. Finally, the thesis covers preparation techniques of thin film systems deposited on various substrates and their characterization using optical and electron microscopy.

Povrchová topografie a mechanické vlastnosti tenkých vrstev na bázi tetravinylsilanu / Surface topography and mechanical properties of thin films on tetravinylsilane basis

Proposed diploma thesis is focused on preparation and characterization of the plasma polymer thin films based on tetravinylsilane monomer (TVS). Plasma enhanced chemical vapour deposition (PECVD) method involving pulse and continual plasma discharge modes were used for thin film deposition on silicon wafer pieces. Reactive plasma composition was containing pure TVS or mixtures of TVS and argon or oxygen gas. Atomic force microscopy was used for surface topography and roughness characterization. Cyclic nanoindentation was involved to measurements to determine the Young’s modulus and hardness of prepared films and scratch test was performed to evaluate the degree of adhesion. Special attention was drawn to the characterization of films with a Young’s modulus below 10 GPa. Tip geometry of indenter influence on scratch test was also commented. Surface and mechanical properties of thin films in relation to the deposition conditions were correlated to the obtained results and final analysis of deposition conditions influence is proposed.

Studium krystalické struktury polyhydroxybutyrátu a nukleační aktivity vybraných typů aditiv / SStudy of crystalline structure of polyhydroxybutyrate and nucleating activity of selected additives

This diploma thesis deals with study of crystalline structure of polyhydroxybutyrate (PHB), which contains different types of additives for studying of their nucleation activity and which were prepared by mixing. It is about boronitrid (BN), sacharin, hydroxapatit, plasticizer Tegmer a tree types of talc. Crystal structure was analysed by differential scanning calorimetry and x-ray diffraction, supramolecular structure was observed by optical microscopy (polarized and confocal laser scanning). Nucleating activity was evaluated by isothermal and non-isothermal crystallization made on calorimeter and heated table of optical microscope. There is not influence of additives on crystallographic structure, but additives affects number and size of spherulites including crystal domains defects, which can have impact on final mechanical properties. BN and talcs react as nucleating agents, other additives during low and high cooling speeds (vc) inhibit nucleation and in middle cooling speeds are without effect. Nucleating activity is not evaluated by numerically, because decrease of crystallization temperature together with vc is not linear. Results of direct methods are based on picture analysis, which is great benefit for understanding of crystal behaviour of PHB.

Využití vybraných fluorescenčních technik ke studiu kvasinek a jejich metabolitů / Use of selected fluorescence techniques to study of yeasts and yeast metabolites

The scope of thesis was the optimization of methods for the study of yeast and their metabolites using flow cytometry and fluorescence microscopy. Red yeasts are characterized by overproduction of carotenoids and lipids, which are used in food, pharmaceutical and feed industries. Currently, intensive research is being carried on to find appropriate microbiological alternatives for synthesis of these substances. Present thesis is focused on selected yeast genera: Rhodotorula, Sporobolomyces, Cystofilobasidium and strain Phaffia rhodozyma. Yeasts were cultivated on different nutrient media, in which glucose was used as a nutritional source, and also on glycerol and whey as waste material. In two strains - Cystofilobasidium macerans and Rhodotorula mucilaginosa growth characteristics were determined on a synthetic glucose production medium. All studied strains were able to use waste substrates as a source of nutrients. Some of the strains displayed increased production of carotenoids, and, additionally, in some cases also relatively high production of lipids. In classical cultivation in lipid and glucose medium supplemented with vitamins the best production characteristics displayed Rhodotorula glutinisstrain. In glycerol medium the highest amount of carotenoids and lipidic substances produced Sporobolomyces shibatanus strain. Strain Sporobolomyces roseus showed the best production characteristics on whey as the main source of carbon. The results show use of whey and glycerol seems like appropriate option for potential carbon source to cultivate carotenogenic yeasts and production of carotenoids and selected lipidic substances as products with higher added value. Further optimization of nutrient medium on the given substrates is needed for higher production of selected metabolites. Fluorescence microscopy and flow cytometry have proved to be suitable options for determination of the observed metabolites in the cells, their amount and viability.
