Mikroskopie und Spektroskopie an Farbstoffmolekülen in photonischen Kristallen

The modification of the radiation pattern and radiative lifetime of dye molecules in three-dimensional colloidal photonic crystals is studied, using high-resolution optical microscopy and spectroscopy. The experimental observations are compared to corresponding calculations of the local optical density of states. It can be concluded, that the fluorescence of the molecules is redistributed spectrally and spatially within the photonic crystal. / Mit Hilfe hochauflösender optischer Mikroskopie und Spektroskopie werden Änderungen der Abstrahlcharakteristik und strahlenden Lebensdauer von Farbstoffmolekülen in dreidimensionalen kolloidalen photonischen Kristallen untersucht. Die experimentellen Beobachtungen werden mit entsprechenden Berechnungen der lokalen optischen Zustandsdichte verglichen. Hieraus kann auf eine spektrale und räumliche Umverteilung der Fluoreszenz der Moleküle im photonischen Kristall geschlossen werden.

Orts- und zeitaufgelöste optische Spektroskopie an Silizium-Nanokristallen

Gegenstand der Dissertation sind Untersuchungen zur Photolumineszenz von Silizium-Nanokristallen. Den Schwerpunkt bilden dabei die Messungen an isolierten Partikeln mittels konfokaler Mikroskopie und optischer Spektroskopie. Von einzelnen Silizium-Partikeln konnten relativ schmale, strukturierte Photolumineszenzbanden detektiert werden, die die Aussagen des Quantum-Confinement-Modells bestätigen. Ein weiteres Merkmal der Photolumineszenz von einzelnen Halbleiter-Nanopartikeln ist das so genannte Blinken. Die Erstellung von Blinkstatistiken unter verschiedenen Anregungsbedingungen ermöglichte es, die zum Blinken führenden photophysikalischen Prozesse genauer zu charakterisieren. Es wird unter anderem gezeigt, dass das reversible Bleichen der Lumineszenz von Silizium-Nanokristall-Ensemblen und porösem Silizium auf ein instationäres Blinkverhalten zurückzuführen ist. Abschließend werden Bezüge zu den astrophysikalischen Beobachtungen von der Extended Red Emission hergestellt und verschiedene Tunnel- und Random-Walk-Modelle zur Beschreibung der photophysikalischen Prozesse diskutiert.

Gegenstand dieser Diplomarbeit ist die Untersuchung der Dynamik in dünnen Polymerfilmen anhand von einzelnen Molekülen. Zu diesem Zweck wurden dünne Filme (kleiner 100nm) hergestellt und mittels Einzelmoleküldetektion und zeitaufgelöster Einzelphotonenzählung analysiert, was eine orts- und zeitaufgelöste Untersuchung einzelner Farbstoffmoleküle ermöglicht. Ziel war es, festzustellen ob, und auf welchem Weg, die Dynamik der Umgebung in Fluktuationen der Fluoreszenzlebensdauer einzelner Moleküle sichtbar wird. Neben der Evaluierung der Untersuchungsmethoden wurden in dieser Arbeit zwei Arten von Sensormolekülen - DiD und Malachit Grün - näher untersucht. / The subject of this diploma thesis is the analysis of dynamics in thin polymer films using single molecules. Thin polymer films (less than 100nm) were produced and analysed by Single Molecule Detection (SMD) and Time Correlated Single Photon Counting (TCSPC). This allows a spatial and time resolved investigation of the single dye molecule. The aim was to ascertain if, and in which way, the dynamics of the environment are reflected by fluctuations of the fluorescence lifetime of the single molecule. In addition to evaluating the investigation methods two kinds of molecules - DiD and Malachite Green - were also analysed.

In the diploma thesis the charge transfer reaction on thin layer N,N',N'',N'''-tetramethyl-tetra-3,4-pyridinoporphyrazinocobalt mediator is studied. The mediator is deposited on electrode surface formed by basal plane of highly ordered pyrolytic graphite. The modified electrode, which displays electrocatalytic activity to oxidation of propylene, has been characterized by cyclic voltammetry, backscattering spectroscopy and atomic force microscopy.

Sledování buněčných populací z regresivních zubních primordií během ontogeneze / Tracing the fate of cell populations from regressive tooth primordia during ontogenesis

(v anglickém jazyce) Development of tooth primordia in mice is an important model for study of odontogenesis. Several dental rudiments develop during the mouse embryogenesis. These structures develop in functional teeth in their phylogenetically older relatives. Similarly, we can initiate growth of teeth from these germs in some mutant mice. In my diploma thesis we have focused on the importance of rudimentary structures with odontogenic potential in postnatal individuals. As a model of development, we have chosen a cell population originating from rudimentary primordia MS (mesial segment) that develops in diastema of the lower jaw during the embryonic day 12.5. Using the inducible Cre-lox technology we have marked the cells which are part of the signal domain of primordia at this time. As a marker of these cells we have used gene Shh. We have found out that these cells persist prenataly and also postnatally. Further we have isolated this cell area and we have tested it using a variety of methods. We have shown that in the cells of postnatal individual are expressed markers of stem cells (Sox2, Bmi1, Gli1) and also genes for major enamel matrix structural proteins: ameloblastin and amelogenin. Diploma thesis Iron Functionalized Nanoparticles of Titanium Dioxide Lenka Volfová 2017, ABSTRACT Iron-functionalized TiO2 were obtained by hydrolysis of aqueous solutions of titanyl sulfate with addition of ferric nitrate with ammonium hydroxide and the reaction filtered and washed with hydrogen peroxide. The colloid solutions thus prepared were lyophilized and the products were subsequently annealed at three different temperatures of 650 řC, 800 řC and 950 řC. The prepared doped materials were characterized by powder X-ray diffractometry, electron microscopy, infrared spectroscopy, Mössbauer spectroscopy, UV/VIS spectroscopy, thermogravimetric analysis and differential thermal analysis, and measurement of the specific surface area. Photocatalytic activity was determined by measuring of the decomposition of kinetics of 4-chlorophenol in an aqueous solution in the ultraviolet and visible area. For comparison of activity in the UV area and in the visible area were used a previously prepared highly photoactive specimen and standard TiO2 from Kronos, respectively. Metastabilní slitina Ti-15Mo připravená práškovou metalurgií / Metastable alloy Ti-15Mo prepared by powder metallurgy

This diploma thesis focused on manufacturing and characterization of Ti-15Mo metastable beta-Ti alloy prepared by cryogenic milling and spark plasma sintering. Initial powder was prepared by gas atomization and consequently deformed by cryogenic milling (milled powder). Both initial and milled powders were compacted by spark plasma sintering (SPS) at temperatures from 750 řC to 850 řC. Dependence of microstructure and mechanical properties on the parameters of preparation was studied. During cryo-milling, powder particles significantly changed shape from ball-shaped to disc-shaped. Particles were not refined by milling, but severely plastically deformed. SEM observations showed that all prepared samples contain duplex alpha + beta structure. Volume fraction of alpha phase is significantly higher in the sintered milled powder due to increased beta- transus temperature caused by contamination by oxygen and also due to easier alpha phase precipitation caused by refined microstructure. Maximum microhardness of 350 HV was achieved for both types of sintered powders. High microhardness of sintered initial powder can be attributed to formation of omega phase during cooling, while sintered milled powder is strengthened by refined microstructure and small alpha phase precipitates. Hydrogen processing is becoming increasingly important not only in the production of electricity but also during its accumulation. One of the energy storage options are liquid organic hydrogen carriers (LOHC). The main drawback of LOHC is the need for a large amount of thermal energy to release molecular hydrogen. We can bypass this issue using heterogeneous catalysis by transferring hydrogen from LOHC to acetone and using the produced 2-propanol (IPA) in the fuel cell. This innovative strategy of getting electri- cal energy from LOHC can be potentially energetically neutral. In this work, we studied highly ordered Co3O4(111) model catalysts for IPA oxidation in the as-prepared state and enhanced with platinum (Pt) nanoparticles. Catalysts were prepared by Physical Vapour Deposition (PVD) and further investigated by means of Low Energy Electron Diffrac- tion (LEED), X-ray Photoelectron Spectroscopy (XPS), Scanning Tunneling Microscopy (STM) and Temperature Programmed Desorption (TPD). The nucleation process of Pt on the as-prepared Co3O4(111) surface was studied by depositing low amounts 0.04 and 0.13 monolayer (ML) of Pt, that create clusters as small as 2 or 3 atoms with no need for a special nucleation site. Solid tumors are complex structures comprising besides the cancer cells vasculature, extracellular matrix (ECM), soluble molecules and a plethora of various other cell types. These components form a so-called tumour microenvironment. From the numerous cell types that are part of tumor microenvironment, tumor infiltrating lymphocytes (TILs) play a major role in patient prognosis. Their presence is also of major importance with regard to new biological therapies based on immune checkpoint inhibitors. Crucial role of TILs is also reflected by the new approaches in cancer diagnostics namely by Immunoscore method (currently used in clinical settings). Immunoscore is based on localization and quantification of CD3+ and CD8+ TILs in thin histological sections of tumor tissue. The question remains to which extent the information obtained from 2D slices reflects the situation in tumor microenvironment considering its spatial heterogeneity. The development of new methodological approaches allowing evaluation of histological information in 3D is the key to answer this question.
