This thesis is aimed at the Czech ZOO´s education activities, concept of self-service worksheets for Ohrada Zoo and evaluation of these worksheets. Three tests (pre-test and two post-tests) and two groups of students were used for the evaluation. First group (experimental) passed the education activities and the tests and the second one (control group) just passed the tests. The self-service worksheets ?Our Fauna? create a practical output of the thesis. As a result of the evaluation we can say that the education activities provided in the Zoo had a pozitive and permanent influence on the students knowledge, irrespectively to their academic performance.

This thesis examines labour market and unemployment not only in chosen region but also at the country level with special focus on people over 50 years of age. Current growing share of older people at labour market is caused especially by demographic trends and by rising of retirement age. From this reason its necessary to diagnose and solve the topic and area of unemployment of elderly persons. In the first part of thesis basic concepts and theoretical basis are defined. We give emphasis on theoretical information regarding of unemployment, its types and rate and employment politics too. The second part of thesis focuses on unemployments evolution and trend in Czech republic, actual state of the labour market and its analysis from the older population point of view. Following part of work characterizes trend of unemployment in Kralupy nad Vltavou and villages in close neighborhood that are part of same administrative district and evaluates overall situation of local labour market. By confirming or declining of stated hypothesis at the end of thesis we aim to define main and root causes of unemployment of chosen age group.

The main task of this thesis is to summarize the diplomatics material of guild corporations at Havlíčkův Brod area into integrated form. Bacause of broad topic, the selection was only focused on the textile craft occurring in this region and especially in deeds. In the introducion the work presents history of the guilds from their beginning to the final downfall - focusing on textile production. It summarizes the genaral situation of the surviving monuments. The edition which is created from deeds issued by these corporations and also by publishers for mentioned guilds - this is the main core of the thesis. The deeds are arranged chronologically according particular guilds and towns. They are completed by diplomatic analysis of internal and external signs and by the final study, which summarizes all the research.

The house of canons regular in Roudnice nad Labem was founded 1333 by John IV of Dražice after his return from Avignon. It was a very important and richly supported monastery, which is demonstrated besides other things by the younger fragment of necrologium, dated 1458-1464 (State Regional Archive in Třeboň, fond Velkostatek Třeboň, registratura IA 3K β 28e). The thesis deals with the Bohemian illuminated manuscripts from the 2nd third of the 14th century from the library from Roudnice, which is quite unique between all the Bohemian houses of regular canons for the high percentage of illuminated manuscripts. The Missale of John of Dražice ( KNM XIII B 9) is an excellent example of some high quality manuscript in the time around 1340 in Bohemia. The ornamental initials show parallels to the recent Western European style. One of them, VD initial on the folio 57r, is filled by the coat-of-arms of the Dražice family, gold wine sprig with three leafs in the red field. But the most interesting is the canon leaf (65r), a classical representative of Central European linear style. The leaf was glued to its place. The same praxis we see in the slightly younger Missale of Chotěšov Monastery (NK XIV C 3). For a long time, 5' cap has been thought to be privilege only for eukaryotic organisms in form of 7-methylguanosine cap at the end of mRNA. This was changed only a few years ago. By using methods liquid chromatography and mass spectrometry a new molecule associated with RNA of Escherichia coli has been found. This molecule turned out to be nicotinamide adenine dinucleotide (NAD+ ) attached to 5' end of some small regulatory RNAs (sRNA). Later it has been shown, that RNA polymerase can attach NAD+ at 5' of RNA ab initio, meaning that RNA polymerase can utilize NAD+ as a substrate for transcription initiation. To some extent substrate for transcription initiation is chosen based on promoter sequence. Crucial requirement is presence of adenine at +1 position of DNA coding strand. This thesis focuses on promoter sequence requirements for transcription initiation with NAD+ . As a template for transcription four promoters with different modifications and their chimeras are used: RNA1, Pveg, lac UV5 and rrnB P1. Also, I tried to compare RNA polymerase from E. coli and B. subtilis in terms of transcription initiation substrate usage. The thesis deals with the issue of employment and unemployment, including its socio- economic aspects in the Ústí nad Labem region compared to the Czech Republic between years 2007 and 2015. The theoretical part describes the nature of unemployment (breakdowns, causes, impacts, unemployment solution) including the impact of macroeconomic indicators, fiscal and monetary policies, with particular respect to active employment policy. The practical part includes the analysis of the Ústí nad Labem Region itself and its employment and unemployment, taking into account the benchmark of the Czech Republic with a polemic on the effectiveness of employment policy and its settings. The thesis implements the SWOT analysis of the Ústí nad Labem Region, which describes weaknesses and strengths and identifies individual opportunities and threats. The data were gained from the Czech Statistical Office and the Ministry of Labour and Social Affairs.

Supervision of financial activity Abstract The activity of entities operating on the financial market is one of the most regulated ones. Economists generally agree that services provided by entities in the financial system are so vital for the functioning of the economy that the overgrown regulatory rules are well- founded. In response to the global financial crisis, there has been a significant development in the regulation and supervision of the financial system, in particular the expansion of the prudential rules of major institutions and the emergence of new areas of macro-prudential supervision and resolution supervision. These are largely the subject of the submitted diploma thesis. The aim of the thesis is to analyze the supervision of the financial system of the Czech Republic in the perspective of institutional and functional classification and in context with their development after the global financial crisis. The diploma thesis The preparation of an educational pathway Hluboka upon Vltava - Zoo Ohrada deals with compiling of the terrain in the neighbourhood of the Zoo Ohrada, subsequent creating of designs and texts for individual information boards. The Educational pathway is called, "On the dam of the Municky pond". The first part of the diploma thesis describes physical-geographical characteristics of the defined area and the history of building educational pathways in the Czech Republic, their types and classification. One chapter is devoted to building educational pathways and possibilities of their financing. The content of the second part of the diploma thesis is a design of the educational pathway itself, texts about individual stages, suggestions for an arrangement of information on information boards including visual documentation, their marking and realization. Scope of the boards is varied, they provide a lot of information about flora and fauna that can be found in this area and also information about natural and environmental regularities and relationships between organisms. A part of the diploma thesis is also a methodical guide with plenty of games, suggestions and activities to make the course of the educational pathway more interesting. The appendix of the diploma thesis contains fotodocumentation, maps and a set of worksheets wit a key.
