Michaela Klusáková DEPARTMENT: The History and History Didactics Department SUPERVISOR: PhDr. Jiří Hnilica, Ph.D. ABSTRACT The main goal of the thesis The "Religious" Remigration during the First Czechoslovak Republic Era is to provide a comprehensive view on the issue, focusing on specific places of return, which are the towns of Velký dvůr u Zábřehu, Sulejovice, Čížkovice, Encovany, Vikýřovice, Suchdol nad Odrou a Žitný ostrov na Slovensku. The thesis is divided into three thematic chapters. The first chapter focuses on the general development of remigration in chronological order, the second chapter addresses to specific locations where the descendants of the White Mountain refugees had settled. The third chapter discusses general issue of the First Czechoslovak Republic "religious" remigration. The core of the study lies in the analysis of the White Mountain emigrants descendants return to Czechoslovakia. While processing this thesis we used direct methods of research of primary written and pictorial sources. We have conducted their criticism of interpretation and synthesis. of thesis titled Insurance intermediaries and supervision over their activities. In my thesis I deal with legislation status and activities of insurance intermediaries. The thesis is divided into three chapters. The first one is devoted to general insurance and the insurance law. I emphasize the importance of the sector and necessity its legal regulation. I mention the roots and development of insurance legislation since 1989. The core of my work is the second chapter, which describes the importance of insurance intermediaries, their function and relationship to other distribution paths. I am talking about the history of the business and its legal regulation and focusing on the development of regulatory activities of insurance brokers since 1989. View of the earlier legislation is important because the anchor institutions and concepts that are still part of the applicable law. The most important part of my theses is a evaluation od the existing legislation, the Act on Insurance Intermediaries, Insurance Act and the Insurance Contract Act in relation to insurance intermediaries. Cette thèse décrit différentes stratégies pour co-immobiliser au sein d'un film sol-gel une déhydrogénase, le cofacteur NAD+/NADH et un système pour régénérer électrochimiquement ce cofacteur. L'immobilisation de la déshydrogénase dans la matrice sol-gel a été obtenue en utilisant un polyélectrolyte positivement chargé comme additif dans le sol de départ. Le film peut être déposé par les procédés d'évaporation ou d'électrogénération, permettant alors la fonctionnalisation d'électrodes d'or macroporeuses. La diaphorase a également pu être co-encapsulée pour la régénération du cofacteur NAD+. L'immobilisation du cofacteur a ensuite été obtenue par couplage chimique du NAD+ avec le groupement époxy du glycidoxypropylsilane avant formation du film. Plusieurs stratégies d'immobilisation du médiateur électrochimique ont alors été étudiées avec succès. Les espèces de type ferrocène ou des complexes d'osmium(III) peuvent être incorporées dans la matrice sol-gel par encapsulation de polymères portant ces médiateurs (Fc-PEI et polymère d'osmium) ou par co-condensation avec un ferrocène fonctionnalisé par un groupement silane. Finalement d'autres stratégies ont été étudiées basées sur la fonctionnalisation de nanotubes de carbone par un traitement micro-onde, par électropolymérisation du vert de méthylène, ou par recouvrement par un polymère de type acrylate portant des complexes d'osmium(III). L'électrogénération d'une couche mince sol-gel servant à immobiliser les protéines et le cofacteur à la surface des nanotubes fonctionnalisés par le polymère d'osmium(III) a alors permis d'observer une réponse électrocatalytique de stabilité remarquable. / In this thesis, the research work was focused on designing functional layers based on silica sol-gel thin films to co-immobilize dehydrogenase, cofactor and electron mediator to get the most highly active systems. Immobilization of dehydrogenase in an active form in a sol-gel matrix was obtained by using a positively-charged polyelectrolyte as additive in the starting sol. The optimal sol can be deposited by evaporation or by electrodeposition and was successfully deposited in macroporous electrodes. The immobilization of the cofactor was investigated by simple entrapment, adsorption to carbon nanotube (CNTs), encapsulation of NAD+ chemically attached to dextran(NAD-dextran), and by in-situ coupling with glycidoxypropyltrimethoxysilane (GPS). The last approach allowed stable immobilization of the cofactor. Several mediators (Fc-PEI, Fc-Silane or Osmium polymer) were successfully co-immobilized with dehydrogenase and cofactor in the sol-gel matrix deposited by drop-coating. However, such co-immobilization did not operate in the electrogenerated sol-gel films. The strategies based on CNTs for mediator immobilization were finally developed. They include (1) micro-wave treatment (MWCNTs-µW), (2) electrochemical deposition of poly(methylene green) (MWCNTs-PMG), and (3) wrapping by osmium(III) polymer (MWCNTs-Os). MWCNTs-Os has been the only system that was successfully combined with sol-gel electrodeposition for co-immobilization of dehydrogenase and cofactor.

Based on the formulated objectives and hypotheses, using a combination of theoretical knowledge and self-evaluation of data on the administrative district of municipality with extended powers Nove Mesto nad Metuji, analyzes segregation as a result of suburbanization processes. The conclusion brings the selected possibilities of solving the problem of segregation and recommendations.

Audit a hodnocení IS bank / Audit and Assessment of IS in banks

Abstract (english) Objectives The main objective of this work is to design methods and proceadures enhancing effectiveness and efficiency of IT audit in banks with the accent given to their use by the supervisory authorities. Another objective of the work (and an essential starting point at the same time) is a summary and assesment of methods and proceadures developed and implemented into the CNB practice with regard to banking supervision in the area of information systems. Objectives Achievement From the methodological point of view the esential starting point of the work was represented by above mentioned objectives that were used for elaboration of a set of questions. Questions enabled to set up the hypotheses. (Another more particular hypotheses were defined in order to design the particular solutions in chapter 5.) Futhermore, the critical factors (problems) were defined in the process of the questions analyses. Subsequently, the solutions were specified. The solutions confirmed the hypotheses which reflected the achievement of the objectives. Description, categorisation, analyses, screening, modelling, comparative analyses and sample testing were used to achieve the objectives. In particular, the solutions that were elaborated, making use of methods described above, enhance effectiveness and efficiency of IT audit in banks. Moreover, the CNB's proceadures and methods were introduced and assesed within the work. Scientific Contribution The work brings an evidence of correlation between the quality of IT audit in banks and their economical performance. With this regard the work contributes with original conclusions, benchmarks and proceadures that may be used by banks, supervisory authorities and IT auditors. These conclusions are achieved by description, categorisation, analyses, modelling and screening research highlighting the role of the rentability, the productivity, the risks, the inovations and the economical value of information. Furthermore, the IT audit and IT supervision in banks are specified. They are also compared and contrast to the other audit cathegories. The work presents important peaces of evidence regarding the role of IT audit in this context. This is made by description, cathegorisation and analyses. Another contribution represents proceadures and methods developed and implemented (to the large extend by author) in the field od IT banking supervision in the Czech Republic. This delivers valuable outputs for foreign supervision authorities, banks and auditors. The work lead to original solutions of critical factors. These solutions are to use by IT audit and IT supervision (and also in audit work generally). The solutions make use of ceartain atributes of Capability Maturity Model (CMM) and were elaborated in the proces of decsription, cathegorisation, screening research, comparative analyses, hypotheses seting and testing. The solutions enhances acuracy and objectiveness of assesment done by IT auditors. The solutions lead to better comparativeness of audit outputs on both national and international level, give better preconditions for risk assesment and capital adequacy evaluation within BASEL II and enhance the information value of audit ouptuts. The structure (content) of the work reflects the above mentioned articles that give a brief description of the main four parts (chapters) of the work.

The thesis thoroughly examines the issue of response to juvenile delinquency and potential solutions of their criminal liability in the European context. The thesis describes the historical development of approach to juvenile delinquency, as well as basic models of youth justice. The important part of the thesis focuses on comparative analyses of selected foreign legislation, namely the legal order of Germany, Austria and Switzerland. In depth are analyzed penalty systems of those countries and their mutual comparison. Further part is dedicated to evaluation of selected sanctions in terms of their effectiveness based on knowledge about recidivism of juvenile offenders. In relation to effectiveness of sanctions the thesis deals with question how to influence different types of juvenile offenders from the perspective of developmental criminology, especially chronic offenders. Author pays attention to the phenomenon of chronic juvenile offenders, formulates risk factors used to identify and predict the life-course delinquency. This Master's thesis focuses on the spatial mobility of secondary school students and the way they spend their leisure time. Rychnov nad Kněžnou town and its surrounding is chosen for the empirical study. The topic of the thesis connects several disciplines especially geography, sociology and psychology in the theoretical part. Special attention is given to the Time Geography and the explanation of the concept of leisure time in the teen-agers's life. The research on the students from different types of secondary schools in Rychnov nad Kněžnou is the main part of the thesis where the method of questionnaire and diary is used. The objective of the survey is to explore daily activities of teen-agers in the time and space. Questions are focused especially on the way the respondents spend their leisure time. Stated activities are compared with the supply of leisure services in the region. The attention in the survey is also paid to the missing leisure services.

Cellular targets of Pseudomonas aeruginosa toxin Exoenzyme S

<p><i>Pseudomonas aeruginosa</i> is an opportunistic pathogen that can cause life-threatening infections in immunocompromised patients. It uses a type III secretion dependent mechanism to translocate toxic effector proteins directly into the eukaryotic cell. The enzymatic activity of two of these toxins, Exoenzyme S (ExoS) and Exoenzyme T (ExoT), have been studied in this thesis. ExoS is a bi-functional toxin known to contain a C-terminal ADP-ribosyltransferase activity, which has been shown to modify members of the Ras family in vitro. The N-terminal of ExoS contains a GTPase Activating Protein (GAP) domain, which shows specificity towards Rho proteins in vitro. ExoT shows high homology (76%) towards ExoS and has also been reported to contain ADP-ribosyltransferase activity <i>in vitro</i>. To study the biological effect of the two toxins, we inserted ExoS or ExoT into eukaryotic cells using the heterologous type III secretion system of <i>Yersinia pseudotuberculosis</i>. We found that Ras was ADP-ribosylated <i>in vivo</i> and this modification altered the ratio of GTP/GDP bound directly to Ras. We also found that ExoS could ADP-ribosylate several members of the Ras superfamily <i>in vivo</i>, modulating the activity of those proteins. In contrast, ExoT showed no ADP-ribosylation activity towards any of the GTPases tested. This suggests that ExoS is the major ADP-ribosyltransferase modulating small GTPase function encoded by <i>P. aeruginosa</i>. Furthermore, we have demonstrated that the GAP activity of ExoS abolishes the activation of RhoA, Cdc42 and Rap1 <i>in vivo</i>, and that ExoT shows GAP activity towards RhoA <i>in vitro</i>. </p><p>The ADP-ribosyltransferase activity of ExoS is dependent on the eukaryotic protein 14-3-3. 14-3-3 proteins interact with ExoS in a phospho-independent manner. We identified the amino acids <sup>424</sup>DALDL<sup>428</sup> on ExoS to be necessary for the specific interaction between ExoS and 14-3-3. Deletion of these five amino acids abolishes the ADP-ribosylation of Ras and hence the cytotoxic effect of P. aeruginosa on cells. Thus the 14-3-3 binding motif on ExoS appears to be critical for both the ADP-ribosylation activity and the cytotoxic action of ExoS <i>in vivo</i>.</p>
