The aim of this diploma thesis is to design a mountain hotel in the village Rokytnice nad Jizernou. The object is placed on a sloping plot located in an attractive touristic and skiing area of mountain range Krkonoše. The mountain hotel has 4 floors and a basement. In the First floor there is a restaurant with a terrace, kitchen and administrative facilities of hotel. In the second and third floor we can find guest rooms and room for the hotel staff. In the basement there is wellness, technical facilities and garage compound. Construction of building is made of the reinforced concrete monolithic transverse frames with beam filling of ceramic block.

The thesis deals with the design of a sustainable urban structure, based on a medieval city, in the 21st century. The main idea of the proposal is the creation of a new part of the town Náměšť nad Oslavou with the aim of connecting it with the existing urban structure and at the same time extending it to the missing elements of the developing city. The newly built area is designed with an emphasis on the construction of new residential housing with civil facilities services, while designing and creating public spaces and private parts of courtyards and villas. The aim of the thesis was to design a growing territory so that it adapts to the needs of today's society and at the same time became part of the existing city. From an urban point of view, based on the theme of the thesis, the aim is to combine elements of a medieval city with elements of a city built in the 21st century. The concept is based on a planned network of streets with multifunctional buildings designed for trade and production in the ground floor and housing on other floors, which originated in the medieval city. The emblem of the medieval city was the emotional perception of the connection of strict lines and strange shapes into one whole, and therefore buildings were designed in a new part of the city for living with almost clean geometric lines, while the objects of civil facilities are organic shapes.

The assignment of diploma thesis was the design of a sustainable urban structure based on a medieval city in Náměšť nad Oslavou. As part of the work, I tried to find the best solution for this solved area, which is isolated by a railway corridor and suggest a new city district based on the principle of a medieval city, which will be linked to the existing city. The proposed development of city blocks is built on a system of platforms that level the terrain within the blocks and enable optimal parking spaces and the use of commercials. This development is complemented by several blocks with villa houses and a city SPA complex. The proposal supports the construction of cycle paths that connect parts of the solved area with the Rathan recreation pond. The design also includes a large park with a lookout tower and a sports ground on the city horizon ner the SPA complex. The accent was also placed on the public area near the new railway station, which serves to gather people for cultural and entertainment opportunities. This public space should not compete with an existing Masaryk square, which fulfills a representative and administration function.

Regulation von oxidativem Stress durch biomechanische Kräfte und fettreiche Ernährung im Herz-KreislaufsystemGöttsch, Claudia 27 February 2007 (has links)
Erkrankungen des Herz-Kreislaufsystems sind trotz erheblicher Fortschritte in Diagnostik und Therapie noch immer die häufigste Todesursache in Deutschland. Neben bekannte Risikofaktoren wie Hypercholesterinämie, Hyperlipoproteinämie, Diabetes mellitus, Adipositas, Bewegungsmangel, Stress und hohem Alter wird eine pathophysiologisch erhöhte Bildung reaktiver Sauerstoffspezies (ROS) als Ursache für deren Entstehung diskutiert. NAD(P)H-Oxidasen, von denen 7 Isoformen der katalytischen Nox-Untereinheiten bekannt sind, stellen dabei die Hauptquelle für vaskuläre Superoxidanionen und oxidativen Stress dar. In dieser Arbeit konnte die vorrangige Bedeutung eines intrazellulär lokalisierten Nox4-haltigen NAD(P)H-Oxidase-Komplexes für die konstitutive Radikalbildung in primären humanen Endothelzellen nachgewiesen werden. Weiterhin konnte gezeigt werden, dass durch chronische Applikation der biomechanischen Kräfte Schubspannung und Dehnung oxidativer Stress in humanen Endothelzellen in vitro vermindert werden kann. Die Herabregulation der Superoxidanionen-Bildung sowie die vermehrte Freisetzung von NO durch chronische Applikation biomechanischer Kräfte trägt zur positiven Balance von NO/Superoxidanionen und zum vasoprotektiven Potential physiologischer Schubspannung bzw. Dehnung bei. Durch Nox4-Promotordeletionsanalysen und Mutationsstudien konnte der Transkriptionsfaktor AP-1 als entscheidend für die schubspannungsabhängige Herabregulation von Nox4 identifiziert werden. Durch Stimulation von Endothelzellen bzw. murinen Gefäßringen mit oxidiertem LDL konnte dagegen die vaskuläre ROS-Bildung in vitro und ex vivo induziert werden. Zur weiteren Aufklärung des Mechanismus der LDL-induzierten ROS-Bildung in vivo und des Einflusses von NAD(P)H-Oxidasen wurden C57BL/6 (Wildtyp)- und Nox2-/--Mäuse 10 Wochen lang mit einer fettreichen Diät (Western diet) gefüttert und anschließend der Einfluss dieser Fütterung auf die NAD(P)H-Oxidase-Expression und ROS-Bildung analysiert. In der Aorta thoracalis beider Mausstämme zeigte sich durch das fettreiche Futter ein signifikanter Anstieg der NAD(P)H-Oxidase-Aktivität im Vergleich zum Standardfutter. Durch Western diet-Fütterung wurde die Nox4-mRNA-Expression in der A. thoracalis von Nox2-/--Mäuse und die p22phox-mRNA-Expression in beiden Mausstämmen induziert. Die Analyse weiterer Organe (Herz, Niere) zeigte keine Induktion von NAD(P)H-Oxidase-Untereinheiten durch Western diet-Fütterung. Zusammenfassend sprechen die Ergebnisse der vorliegenden Arbeit für eine entscheidende Rolle der Nox4-haltigen NAD(P)H-Oxidase bei der vaskulären Radikalbildung in vitro und in vivo. / Cardiovascular diseases are the most common causes of death in Germany. Beside the known risk factors hypercholesteremia, hyperlipoproteinemia, diabetes mellitus, obesity, sedentary lifestyle, stress and high age, a pathophysiologically increased formation of reactive oxygen species (ROS) are discussed as cause of development of cardiovascular diseases. Nicotine adenine dinucleotide phosphate (NADPH) oxidase complexes have been identified as main source of oxidative stress and vascular superoxide anions. There are 7 known isoforms of the catalytic Nox subunit of the NADPH oxidase. In this dissertation it was shown that NADPH oxidase subunit Nox4 is the major Nox isoform in human endothelial cells. Nox4 could be localized in the perinuclear space. Overexpression of Nox4 enhanced endothelial superoxide anion formation. Furthermore, a reduction of oxidative stress could be demonstrated by chronic application of the biomechanical forces laminar shear stress and cyclic strain in endothelial cells in vitro. The observed downregulation of superoxide anion formation and upregulation of NO formation by application of biomechanical forces contribute to the positive balance between NO and superoxide anion and the vasoprotective potential of physiological shear stress and cyclic strain. Molecular cloning and functional analysis of the human Nox4 promoter revealed that an AP-1 binding site is essential for downregulation of Nox4 by laminar shear stress. On the other hand stimulation of endothelial cells and murine vessels with oxidized lipids caused an upregulation of vascular ROS production in vitro and ex vivo. In order to examine the mechanism of LDL induced ROS formation and the influence of NADPH oxidase, C57BL/6 (wild-type) and Nox2-/- mice were feed with a diet high in fat and sugar (Western-type diet) for 10 weeks. After feeding, the influence of diet on the expression of NADPH oxidase and ROS production was analyzed in the A. thoracalis. Both mice strains showed a significant upregulation of aortic ROS production in comparison to normal chow. The mRNA expression of aortic Nox4 was induced in Nox2-/- mice. Furthermore, the aortic p22phox mRNA expression was upregulated in both mice strains. The analysis of other organs (heart, kidney) showed no influence of the Western-type diet. In conclusion, the results demonstrate a major role of a Nox4 containing NADPH oxidase in the vascular radical formation in vitro and in vivo.

The Diploma thesis "Křivoklát Altarpiece" tries to process the monographical issues of a late gothic altarpiece retable. The altar has survived as a part of stylistically uniform area of Křivoklát's chapel. Author primarily studies this work of an outstanding quality in the context of the second half of the 15th century of the Central European artistic product. The work is compared with the artistic works of the same type. In addition to that the author tries to clarify iconographic issues of the altarpiece which are often mentioned in specialized literature. The Diploma thesis mainly focuses on the altar's sculpture decoration with the goal to clarify its stylistic origins and the time of its creation. Křivoklát Altarpiece represents one of the most important works coming out of Prague's Jagellonian court.

Tato práce se zabývá vývojem efektivních technik pro konečné automaty a jejich aplikace. Zejména se věnujeme konečným automatům použitých pří detekci útoků v síťovém provozu a automatům v rozhodovacích procedurách a verifikaci. V první části práce navrhujeme techniky přibližné redukce nedeterministických automatů, které snižují spotřebu zdrojů v hardwarově akcelerovaném zkoumání obsahu paketů. Druhá část práce je je věnována automatům v rozhodovacích procedurách, zejména slabé monadické logice druhého řádů k následníků (WSkS) a teorie nad řetězci. Navrhujeme novou rozhodovací proceduru pro WS2S založenou na automatových termech, umožňující efektivně prořezávat stavový prostor. Dále studujeme techniky předzpracování WSkS formulí za účelem snížení velikosti konstruovaných automatů. Automaty jsme také aplikovali v rozhodovací proceduře teorie nad řetězci pro efektivní reprezentaci důkazového stromu. V poslední části práce potom navrhujeme optimalizace rank-based komplementace Buchiho automatů, které snižuje počet generovaných stavů během konstrukce komplementu.

La adaptación de la levadura Saccharomyces cerevisiae a condiciones de alta osmolaridad está mediada por la vía de HOG ((high-osmolarity glycerol). La activación de esta vía induce una serie de respuestas que van a permitir la supervivencia celular en respuesta a estrés. La regulación génica constituye una respuesta clave para dicha supervivencia. Se han descrito cinco factores de transcripción regulados por Hog1 en respuesta a estrés osmótico. Sin embargo, éstos no pueden explicar la totalidad de los genes regulados por la MAPK Hog1. En el presente trabajo describimos cómo el complejo transcripcional formado por las proteínas Rtg1 y Rtg3 regula, a través de la quinasa Hog1, la expresión de un conjunto específico de genes. Hog1 fosforila Rtg1 y Rtg3, aunque ninguna de estas fosforilaciones son esenciales para regulación transcripcional en respuesta a estrés. Este trabajo también muestra cómo la deleción de proteínas RTG provoca osmosensibilidad celular, lo que indica que la integridad de la vía de RTG es esencial para la supervivencia celular frente a un estrés osmótico. / In Saccharomyces cerevisiae the adaptation to high osmolarity is mediated by the HOG (high-osmolarity glycerol) pathway, which elicits different cellular responses required for cell survival upon osmostress. Regulation of gene expression is a major adaptative response required for cell survival in response to osmotic stress. At least five transcription factors have been reported to be controlled by the Hog1 MAPK. However, they cannot account for the regulation of all of the genes under the control of the Hog1 MAPK. Here we show that the Rtg1/3 transcriptional complex regulates the expression of specific genes upon osmostress in a Hog1-dependent manner. Hog1 phosphorylates both Rtg1 and Rtg3 proteins. However, none of these phosphorylations are essential for the transcriptional regulation upon osmostress. Here we also show that the deletion of RTG proteins leads to osmosensitivity at high osmolarity, suggesting that the RTG-pathway integrity is essential for cell survival upon stress.
