Cytoplasmic and Mitochondrial NADPH-Coupled Redox Systems in the Regulation of Aging

The reduced form of nicotinamide adenine dinucleotide phosphate (NADPH) protects against redox stress by providing reducing equivalents to antioxidants such as glutathione and thioredoxin. NADPH levels decline with aging in several tissues, but whether this is a major driving force for the aging process has not been well established. Global or neural overexpression of several cytoplasmic enzymes that synthesize NADPH have been shown to extend lifespan in model organisms such as Drosophila suggesting a positive relationship between cytoplasmic NADPH levels and longevity. Mitochondrial NADPH plays an important role in the protection against redox stress and cell death and mitochondrial NADPH-utilizing thioredoxin reductase 2 levels correlate with species longevity in cells from rodents and primates. Mitochondrial NADPH shuttles allow for some NADPH flux between the cytoplasm and mitochondria. Since a decline of nicotinamide adenine dinucleotide (NAD + ) is linked with aging and because NADP + is exclusively synthesized from NAD + by cytoplasmic and mitochondrial NAD + kinases, a decline in the cytoplasmic or mitochondrial NADPH pool may also contribute to the aging process. Therefore pro-longevity therapies should aim to maintain the levels of both NAD + and NADPH in aging tissues.

This thesis defines and applies a methodology for analyzing the possibility of extending the operating sector of DP shuttle tankers for offloading operations in Spread Moored FPSO Platforms. Extending the operating sector is beneficial to increase operations\' availability and to reduce DP power demand under certain environmental conditions. This study is important, since several emergency disconnections have occurred during offloading in Santos Basin because the shuttle tanker was pushed out of the green sector by environmental resultants that pointed to the West. However, this proposal has to be carefully analyzed in order to guarantee the operations\' safety and not to increase the risks of collision between FPSO and shuttle tanker, of oil pollution and of personnel safety. The methodology consists in five basic steps: Preliminary Risk Analysis to assess the potential hazards associated with the new sector; evaluation of the uptime gain through static analysis; evaluation of DP power demand inside the original and extended sector; real time simulations to evaluate the operation in a realistic environment; and, finally, field tests to validate the proposal. This thesis presents the contextualization of the problem, a bibliographical research, theoretical concepts, the detailed methodology and results of each step. The results show that the average uptime gain is significant both in Campos and Santos Basins - up to 9% and 13% respectively - and that the additional risks created by the sector extension are well mitigated if some recommendations are put into place. The conclusion of this thesis is that extending the operating sector is not only beneficial but also safe. / Esta dissertação define e aplica uma metodologia para analisar a possibilidade de extensão do setor operacional de navios aliviadores DP para operações de offloading em plataformas FPSO ancoradas em Spread-Mooring. Esta proposta apresenta como vantagens o aumento da disponibilidade das operações e redução na demanda de energia do navio DP em certas condições ambientais. O estudo é importante tendo em vista que várias desconexões de emergência já ocorreram durante operações de alívio na Bacia de Santos, porque o navio-tanque foi empurrado para fora do setor verde por resultantes ambientais que apontavam para oeste. No entanto, a proposta deve ser cuidadosamente analisada para garantir que o novo setor não aumenta os riscos de colisão, de poluição ambiental e de segurança às pessoas. A metodologia consiste em cinco etapas básicas: Análise Preliminar de Riscos (APR); avaliação do ganho de disponibilidade da operação; avaliação da demanda de energia do sistema DP nos setores original e estendido; simulações de manobra em tempo real; e, finalmente, testes em campo para validação da proposta. Esta dissertação apresenta a contextualização do problema, pesquisa bibliográfica, conceitos teóricos, a metodologia detalhada e os resultados de cada etapa. NADPH oxidase-dependent reactive oxygen species stimulate the differentiation of endocrine progenitors in murine pancreas.

The expression of renin-angiotensin system (RAS) components has been detected in development tissue and organs, implicating their regulatory role in developmental processes. On the other hand, reactive oxygen species (ROS) are responsive to RAS signaling pathways and act as second messengers to promote differentiation through redox modification of transcriptional factors essential for differentiation. As a major source of ROS, NADPH oxidase has been shown to participate in the progenitor differentiation in a variety of cells and tissues. Despite this finding, the role of NADPH oxidase-dependent ROS in regulating pancreatic endocrine cell differentiation remains ambiguous. Against this background, the study was aimed at elucidating the roles of RAS components and NADPH oxidase-derived ROS during differentiation of pancreatic endocrine cells using mouse pancreatic rudiments and streptozotocin-treated neonatal rats. / Results showed that angiotensin II type 2 receptor (AT₂R), a major component of the classical RAS, was localized within the nuclei of endocrine progenitors in the cultured pancreatic rudiments; following the differentiation of endocrine progenitors into insulin producing cells, it translocated to cytoplasm. Blockade of AT₂R impeded the expression of Ngn3 and insulin as well as proliferation of β-cells. In addition, the dynamic changes of ROS levels were found in mouse pancreata at different embryonic days, concomitant with induction of endocrine cell differentiation induced by modest exogenous ROS in pancreatic rudiment cultures. Moreover, scavenger of ROS diminished the expression of islet cell markers for differentiation and maturation. NOX4 and its associated subunit p22phox, which are the member of NADPH oxidase, exhibited similar changes of expression to that of ROS levels during pancreas development and persisted in the endocrine lineage; they were located in NGN3⁺ cells at E15.5 during the burst of NGN3 expression and then distributed in insulin⁺ cell at E17.5, the latter being the phase that has a decline in NGN3 expression with an increase of insulin. Furthermore, administration of NADPH oxidase inhibitor, diphenylene iodonium (DPI) attenuated the differentiation of endocrine progenitors in rudiment cultures, while exogenous ROS reversed this effect. / On the other hand, studies performed in streptozotocin-induced neonatal rats showed that β cell regeneration was negatively affected by DPI treatments; consistently, impaired blood glucose control, disturbed islet architecture and deficient serum insulin were observed in DPI-treated groups. In addition, DPI treatments blunted NGN3 expression, but not Ki67-labeling beta-cells, suggesting that differentiation beyond proliferation of β-cells was accountable in response to ROS stimulation. Administration of DPI also suppressed the levels of SOX9, a transcriptional regulator of NGN3, in pancreatic progenitor cells, as evidenced by both in vivo and in vitro studies. Interestingly, over-expression of SOX9 could restore the repression of NGN3 induced by DPI. This diploma thesis deals with the development of an HPLC method for the determination of selected compounds participating in enzymatic reaction leading to the formation of gallic acid. The analysed reaction mixture contains the following reagents: shikimic acid, NADP+ and shikimatedehydrogenase (SDH) extracted from parsley; the presumed product of the reaction is gallic acid. Two chromatographic methods for the determination of the above mentioned compounds were developed using C18 HPLC column and porous graphitic carbon Hypercarb column. Molecular absorption spectrometric detection in the UV range was used in all measurements. Separation on the C18 column was found particularly suitable for analysing the composition of the end products of the reaction. Because of the NADP+ and shikimic acid peak overlap it is necessary to observe absorbance at 212 and 260 nm. Shikimic acid and NADP+ can be quantified due to the fact that shikimic acid does not absorb at 260 nm while NADP+ absorb radiation at both wavelengths. Separation via Hypercarb column was found particularly suitable for analysing the process of the reaction; additional products or intermediates can be seen in chromatograms compared to the C18 method. Regulation of Stomata Opening in the Crassulacean Acid Metabolism Plant Kalanchoe Laxiflora

Stomata are small pores that are located on the surface of epidermal leaves, and they can regulate the uptake of CO2 and prevent water lose by opening and closing the pores. Stomata of plants can be regulated by external condition such as CO2, biotic and abiotic stresses and internal factors. CAM (crassulacean acid metabolism) plants adapt to hot and dry environments by closing stomata during the day and opening stomata during the cool night. However, it is still unclear how CAM plants open their stomata during the night and close them during the day. In this study, a number of factors were evaluated for their potential roles in promoting stomatal opening in the model CAM plant Kalanchoe laxiflora. Citrate is an important organic acid and it accumulates during the night in CAM plants. It is shown in this study that citrate promoted stomatal opening in detached leaf epidermis of Kalanchoe laxiflora. Further, the cytokinin zeatin is also shown to stimulate stomatal opening in detached leave of Kalanchoe laxiflora. Melatonin is an important regulator of circadian rhythms in mammals and has been implicated in regulation of plant abiotic stress responses. Melatonin was detected in the leaves of Kalanchoe laxiflora. It promoted stomatal opening in detached epidermis of Kalanchoe laxiflora. Together, these results suggest that stomata of Kalanchoe laxiflora respond to citrate and malate which are the main organic acids accumulate during nighttime and also to some signaling molecules (zeatin, melatonin, and serotonin) by opening stomata during dark period.
