Efeitos da quantização em sistemas de controle em rede

Este trabalho investiga a influência da quantização em sistemas de controle em rede. São tratados problemas de estabilidade e estabilização de sistemas lineares de tempo discreto envolvendo quantização finita nas entradas da planta controlada, considerando dois tipos de quantizadores: os uniformes e os logarítmicos. Como consequência da quantização finita, ocorrem também efeitos de saturação e zonamorta dos sinais de entrada. Tais comportamentos não-lineares são considerados explicitamente na análise. Para plantas instáveis, o objetivo é estimar a região onde os estados estarão confinados em regime permanente. Esta região, denominada atrator dos estados, é estimada por meio de um conjunto elipsoidal. Ao mesmo tempo, determina-se um conjunto elipsoidal de condições iniciais admissíveis, para o qual se garante a convergência das trajetórias para o atrator em tempo finito. Primeiramente, esses conjuntos são determinados para o caso de um controlador dado e, posteriormente, sintetiza-se um controlador que minimiza o atrator. Em se tratando de plantas estáveis, investiga-se como o desempenho dinâmico é afetado pela quantização. Para tanto, utiliza-se como critério o coeficiente de decaimento exponencial que é garantido para o sistema. Nesta parte, excluem-se os comportamentos na região de saturação e na região da zona-morta. Primeiramente, o coeficiente de decaimento garantido é estimado para um sistema com controlador dado. Neste caso, faz-se uma análise de degradação de desempenho induzida pela quantização com relação ao comportamento do sistema em malha fechada sem quantização. Posteriormente, sintetiza-se um controlador que minimiza este coeficiente na presença da quantização. Na obtenção dos resultados, utilizam-se condições de setor respeitadas pelas não linearidades e formulam-se os problemas na forma de inequações matriciais que podem ser resolvidas a partir de problemas de otimização baseados em LMIs. / This work investigates the in uence of quantization over networked control systems. At rst, we tackle stability and stabilization problems of discrete-time linear systems involving nite quantization on the input of the controlled plant, considering two kinds of quantizers: uniform and logarithmic. As a consequence of the nite quantization, saturation and dead-zone e ects on the input signals are also present. These non-linear behaviors are explictly considered in the analysis. For unstable plants, the objective is to estimate the region where the states will be ultimately bounded. This region, which we call the attractor of the states, is estimated through an ellipsoidal set. Simultaneously, we determine an ellipsoidal set of admissible initial conditions, for which the trajectories will converge to the attractor in nite time. At rst, the sets are determined for the case where the controller is given and, in the sequel, a controller that minimizes the attractor is designed. When dealing with stable plants, we investigate how the dynamic performance is a ected by the quantization. To do that, we use as criterion the exponential decay rate which is guaranteed for the system. At this point, we exclude the behaviour in the saturation and deadzone regions. At rst, the guaranteed decay rate is estimated for a system where the controller is given. In this case, we analyze the deterioration of the performance in uenced by the quantization, compared to the behavior of the closed-loop system without quantization. In the sequel, a controller that minimizes that rate in the presence of quantization is designed. To obtain the results, we use sector conditions which are respected by the nonlinearities and we state the problems as matrix inequalities which can be solved using LMI-based optimization problems.

Nanocristaux optiquement non linéaires pour des applications en imagerie biologique : synthèse et caractérisations d'iodate de fer en microémulsions

Le développement de nanomatériaux à propriétés optiques et fonctionnalisés pour un marquage spécifique est en plein essor dans le domaine de l'imagerie biologique. Parmi les agents de contraste exogènes déjà utilisés, les marqueurs fluorescents tels que les nanocristaux semi-conducteurs (CdSe/ZnS,...) et les molécules organiques naturelles (GFP,...) ou synthétiques (fluorescéine,...) souffrent respectivement de clignotements (blinking) et de photo-blanchiment (bleaching) c'est-à-dire d'une faible tenue au rayonnement lumineux incident. Récemment, la microscopie de Génération de Second Harmonique (GSH) à partir de structures non-centrosymétriques de certains matériaux ou molécules optiquement non linéaires (ONL), s'est révélée un outil particulièrement prometteur. Les inconvénients du clignotement et du photo-blanchiment sont en effet absents pour le processus non linéaire de GSH. De plus, le principe de fonctionnement des marqueurs ONL repose sur un processus non résonant, contrairement aux marqueurs fluorescents, ce qui est un avantage décisif pour le choix de la longueur d'onde d'excitation des nanosondes. Pour des illuminations dans le proche infrarouge, cela permet de limiter l'énergie déposée dans le milieu biologique, d'augmenter la profondeur d'imagerie et enfin de bien séparer spectralement les signaux des marqueurs ONL de l'auto-fluorescence naturelle de certains échantillons. Notre objectif, dans ce contexte, était la synthèse et la caractérisation de nouvelles nanosondes ONL de forme sphérique et de taille inférieure à 100nm. Le matériau de structure cristalline non centrosymétrique retenu est l'iodate de fer (Fe(IO3)3) car ses éléments chimiques sont peu toxiques et que ses propriétés paramagnétiques peuvent également donner un contraste en imagerie par résonance magnétique (IRM) ce qui est potentiellement intéressant par rapport à d'autres cristaux ONL tels que ZnO, KNbO3, BaTiO3 et KTP. D'un point de vue synthèse, les microémulsions inverses sont bien référencées dans la littérature pour leur rôle de gabarit permettant un bon contrôle de la taille et de la morphologie des nanomatériaux obtenus par co-précipitation. Dans ce travail, les nano-réacteurs ont été préparés à partir des systèmes AOT/alcane/eau et Triton/1-hexanol/cyclohexane/eau. De manière très originale et pratique, le développement d'un banc optique de diffusion Hyper-Rayleigh (HRS) a permis de suivre in-situ et en temps réel les cinétiques de cristallisation des nanoparticules de Fe(IO3)3 en fonction de conditions expérimentales variables. Les mécanismes de croissance et de cristallisation des nano-bâtonnets de Fe(IO3)3 ont été élucidés en combinant d'autres techniques physico-chimiques usuelles comme la diffraction des rayons X, la diffusion dynamique de la lumière et la microscopie électronique en transmission. Nous avons démontré que la température et la nature du tensioactif influencent les forces d'interaction à l'interface organique-inorganique ce qui permet, pour certaines conditions expérimentales, de réduire la taille et la polydispersité des nanocristaux en fin de processus. Toutefois, avant d'envisager l'utilisation de ces derniers en tant que marqueurs optiques spécifiques, il est nécessaire d'encapsuler ces nanocristaux en raison de la faible stabilité du composé aux pH physiologiques. Les premiers essais de stabilisation en microémulsions par une couche de silice ont permis d'obtenir des nanoparticules de taille ~ 10 nm avec une forte réponse ONL. La caractérisation complète et la fonctionnalisation de ces nanostructures ainsi qu'une optimisation des interactions à l'interface particules - films de tensioactifs constituent les perspectives de ce travail.

Structural Studies of Bacteriophage PRR1 and HIV-1 protease

Viruses are a diverse genera of organisms adapted to thrive in many different hosts from prokaryotic to eukaryotic. We present here the structure of bacteriophage PRR1 virus-like particle (VLP), belonging to Leviviridae family. Our structure reveals calcium ions in the VLP. Metal ions are rare in the VLP among the Leviviridae and the calcium ions were found to affect VLP stability. Gene expression in Leviviridae is controlled by a specific interaction between the viral coat protein that assembles to create the VLP, and the genomic RNA. This interaction has been thoroughly studied for the levivirus MS2 but other structural data are scarce. We have solved the structure of PRR1 VLP in complex with its RNA operator stem-loop. Binding of the stem-loop in PRR1 shows similarities to MS2 but also a different arrangement of the nucleotides, in the area of the loop that we could interpret, compared to MS2. The structures of PRR1 increase our knowledge about translational control in Leviviridae and add new information about particle stability within this family. The other virus we investigated is the more infamous human pathogen, the HIV. Because of the high mutation rate of HIV new drugs are needed on a continuous basis. We describe here the structure of two new protease inhibitors bound to the HIV-1 protease and compare them with two previously published inhibitors. <p>This study provides educators with an opportunity to share their commitments and biases, their beliefs about what these learners need and what, as educators, they have to offer them. The study focuses specifically on what curriculum educators believe should be offered to the youth in these centres. It is important to note that the focus of the thesis is on the educational debates and challenges within SYCECs, and not on the overall goals and role to be played by these institutions in rehabilitating youth. This piece of work acknowledges the systemic issues that impact on learning and development of youth, but foregrounds what learners in SYCECs should be taught and why. This is its main contribution.</p>

The Synaptic Role of Neuronal Calcium Sensor 1 in Dentate Gyrus Plasticity, Curiosity and Spatial Memory

Only 200 years ago, virtually nothing was known about the biological workings of the mind. Today, there is a deep (though far from complete) understanding of the cellular and molecular mechanisms underlying the encoding of memory, arguably the most fundamental aspect of a cognitive being. In this thesis, I describe experiments that help complete this understanding and identify the very first molecules underlying curiosity. By using an inducible rtTA2-M2 double transgenic system to selectively overexpress the calcium sensor Ncs1 in the adult murine dentate gyrus, I created an animal with facilitated long-term potentiation, enhanced rapid acquisition of spatial memory and greater curiosity. These phenotypes are reversed by direct infusion of a small membrane-permeant interfering peptide designed to block complex formation between NCS-1 and Dopamine type-2 receptors (D2 receptors). Pharmacological antagonism of D2 receptors also attenuates plasticity in wild-type mice and direct antagonism of D2 receptors in the dentate of cannulized wild-type mice prevents spatial memory formation. Conversely, application of a dominant negative NCS-1 peptide reduces synaptic transmission in the dentate gyrus and impairs spatial fear learning. Far less understood than the mechanisms governing learning and memory, are the mechanisms used by the brain to generate curiosity. Strikingly, Ncs1 overexpressing mice also demonstrate increased exploratory behaviours in a variety of novel, non-fearful environments. But they do not explore novel fearful environments any more than their littermate controls. I argue that the specificity of this phenotype represents an effect on curiosity, thereby identifying NCS-1 and D2 receptors as the first known regulators of this primordial mental state. I propose that the generation of curiosity is a fundamental feature of the nervous system and is upstream of learning and cognition. As such, molecular cascades involved in curiosity likely also play roles in mental illnesses. To investigate this theory, I generated an NCS-1 point mutant mouse line. NCS-1P144S/P144S mice show endophenotypes of schizophrenia and depression, supporting the link between curiosity and mental illness. I integrate my findings with the current literature and propose a means to investigate the role of NCS-1 in humans with mental illnesses.
