Formulation de nanosystèmes et évaluation de leur potentiel pour la délivrance cutanée de molécules actives / Formulation of nanosystems and evaluation of their potential for delivery of active molecules to the skin

Ces travaux visent à déterminer l‘intérêt de deux types de nanosystèmes (NS) coeur-couronne dans des applications dermatologiques ou cosmétiques. Les nanocapsules lipidiques (LNC) sont obtenues par une méthode déjà décrite dans la littérature. Leur formule est modifiée pour incorporer un actif cosmétique d‘intérêt. Les nanocapsules d‘alginate (ANC) sont développées au moyen de plans d‘expériences. Elles sont composées d‘un coeur huileux et d‘une coque d‘alginate de calcium gélifiée obtenue par gélification ionique de surface d‘une nanoémulsion. Des méthodes basées sur le phénomène de fluorescence nous permettent de mettre en évidence l‘endocytose des ANC par les kératinocytes. Leur contenu est rapidement libéré dans le cytoplasme. Une étude sur différents modèles ex vivo montre que les deux nanosystèmes permettent aux molécules encapsulées d‘atteindre les couches vivantes de l‘épiderme. ANC et LNC sont stables plusieurs mois dispersées dans des formes galéniques semi-solides. Ces deux NS sont donc adaptés à la délivrance de molécules actives dans la peau. / Two types of core-shell nanosystems have been evaluated for dermatological and cosmetic applications. Lipid nanocapsules (LNC) are obtained by a method that has already been described in the literature. Their composition is adapted for incorporation of a specific cosmetic ingredient. Alginate nanocapsules (ANC) are developed with the aid of experimental design. They consist of a triglyceride core with a rigid calcium alginate shell obtained by ionic gelation of the surface of a nanoemulsion. By incorporating fluorophores into these nanosystems, they can be studied by advanced spectral fluorescence imaging methods. We were thus able to show that ANC are first internalized into keratinocytes by endocytosis, and once inside the cells, their contents are rapidly released into the cytoplasm. A study of different ex vivo skin model systems has shown that both nanosystems enable active substances to reach the living epidermis. When incorporated into gels similar to those used as galenic forms for topical administration, LNC and ANC remain stable for months. They can thus be used as vectors for delivering active substances to the skin.

Small-scale Technologies for Enhanced Diagnostics and Therapeutics

<p>Miniaturization of technologies to milli-, micro- and nanoscale offers numerous advantages for diagnostic and therapeutic biomedical applications. In comparison to their macro-scale counterparts, these small-scale systems are more portable, less invasive and less costly. They can facilitate rapid, sensitive and high throughput detection of abnormalities, help track disease progression, reduce sample consumption and improve therapeutic efficacy of drug delivery while decreasing systemic toxicity. Thus, there is clearly a need for creating innovative milli-, micro- and nanoscale tools that can uncover new possibilities in detection and treatment of various types of diseases. The overall objective of this dissertation was to develop novel small-scale technologies that could help enhance diagnostic and/or therapeutic outcomes in patients with cancer, opioid addiction and inflammatory bowel disease. First, we developed an echogenically stable nanodroplet ultrasound contrast agent with potential applications in extravascular molecular imaging of tumors and targeted cancer therapies. Then, we created a polymer blend microsphere system that could be integrated in prescription opioid tablets to develop an abuse-deterrent formulation against smoking. Finally, we designed a release system for localized delivery of aminosalicylates from magnetically actuated millirobots in the colon to improve therapeutic outcomes in patients suffering from inflammatory bowel disease. Overall, the technologies we developed could serve as a basis for designing diagnostic and therapeutic tools that are superior to currently existing platforms.</p>

A presen?a de PTX levou a um aumento significativo no di?metro das part?culas em todos os sistemas avaliados, exceto no NE. Al?m disso, o aumento da concentra??o do f?rmaco (0,01% para 0,025%) produziu um aumento do di?metro para a prepara??o de CLN. However, the low solubility of PTX in aqueous medium (partition coefficient log of 3.96) represents a barrier for intravenous administration. The conventional PTX formulation contains a high concentration of Cremophor-EL? (polyethoxylated castor oil), which is associated with significant toxicities restricting its clinical use. The encapsulation of the PTX in drug delivery systems could improve the uptake and increase its therapeutic efficacy. In this study, three different lipid nanosystems containing PTX, solid lipid nanoparticle (SLN), nanoemulsion (NE), and nanostructured lipid carrier (NLC) were prepared, and the physicochemical properties and in vitro cytotoxic activity were evaluated. Concerning the mean diameter, NLC blank showed diameter values approximately 2 and 1.7-fold lower than those obtained for SLN and NE, respectively. The presence of PTX leads to a significant increase in the particle diameter in all systems evaluated, except NE. In addition, increases in drug concentration (0.01% to 0.025%) produced an enhanced diameter for NLC preparation. All formulations containing PTX showed PI higher than 0.3, except for NE-PTX at drug concentration equal to 0.01% (w/v). Negative zeta potential values were observed in all formulations evaluated. NLC-PTX was the system more stable after storage for 30 days at 4 oC. The cytotoxicity studies on breast cancer cell lines (MDA-MB-231 and MCF-7) demonstrated cytotoxic activity more pronounced for NCL-PTX than for free PTX for both tumor cell lines. Thus, the results showed that NCL-PTX seems to be a potential tool for the treatment of breast cancer.

Etude des propriétés électroniques et vibrationnelles de nano-objets métalliques et hybrides par spectroscopie femtoseconde / Study of electronic and vibrational properties of metallic and hybrid nano-objects using femtosecond spectroscopy

Ce travail a porté sur l’étude expérimentale de la dynamique électronique et vibrationnelle de nano-objets métalliques et hybrides par spectroscopie pompe-sonde femtoseconde. L’étude de la dynamique des échanges d’énergie électrons-réseau dans des systèmes métalliques bidimensionnels nous a permis de mettre en évidence une accélération du transfert d’énergie entre électrons et phonons due au confinement. Cette accélération est gouvernée par la plus petite dimension des nano-objets étudiés (nano-triangles d’argent 2D) lorsque celle-ci devient inférieure à environ 10 nm. Nous avons aussi étudié la dynamique vibrationnelle de nanoparticules métalliques, bimétalliques et métal diélectrique. Nous nous sommes intéressés aux modes de vibration longitudinaux et radiaux de bipyramides d’or en fonction de leurs dimensions, et à leurs évolutions consécutives à un dépôt d’argent. Une forte sensibilité des périodes et des amplitudes de vibration à de faibles dépôts a été mise en évidence. L’étude de l’évolution des modes de vibration de nano-objets de type coeur-coquille (argent-silice et or-silice) nous a permis d’obtenir des informations sur la qualité du contact mécanique à l’interface métal-diélectrique. Enfin, nous avons étudié les interactions électroniques dans des nano-systèmes hybrides métal/semi-conducteur (ZnO-Ag), et plus particulièrement les transferts de charge et les échanges d’énergie entre les deux composants.Nous avons montré la forte influence de l’environnement sur la dynamique électronique du ZnO et mis en évidence un transfert d’électron, photo-induit par une impulsion infrarouge, de la particule métallique vers la bande de conduction du semi-conducteur. / Electronic and vibrational dynamics have been studied in metallic and hybrid nano-object using femtosecond timeresolved spectroscopy. The study of electron-lattice energy exchanges in two-dimensional metallic systems showed an acceleration of the energy transfer between electrons and phonons due to confinement. This acceleration is governed by the smallest dimension of the nano-objects (2D-silver nano triangles ) when it becomes smaller than 10 nm. We also studied the vibrational dynamics of metallic nanoparticles, bimetallic and metal-dielectric. We investigated the longitudinal and radial modes of vibration of gold bipyramids which depend on their size, and studied their evolution under silver deposition. A high sensitivity of periods and amplitudes for small deposition were demonstrated. Our work on the evolution of acoustic vibrations of core-shell nano-objects (silver-silica and goldsilica) allowed us to obtain information on the quality of mechanical contact at the metal-dielectric interface. Finally, we studied the electronic interactions in hybrid metal / semiconductor (ZnO-Ag) nano-systems, and especially the charge transfer and energy exchanges between the two components. We showed a strong influence of the environment on the electron dynamics of ZnO and proved the existence of an electron transfer, photoinduced by an infrared pulse, from the metal particle to the semiconductor conduction band.

RATIONAL DESIGN OF VERTICAL SILICON NANONEEDLES FOR OCULAR DRUG DELIVERY AND INTRACELLULAR RECORDING

<p>The use of silicon nanoneedles provides a unique and versatile biointerface for a range of biomedical applications. In this work, we propose a rational design for vertical Si nanoneedles that are printed on a polymer substrate for ocular drug delivery, intracellular recording, and intra-organoid sensing. To enable minimally invasive and long-term sustained delivery of ocular drugs, we integrate vertical Si nanoneedles with a tear-soluble contact lens for ocular drug delivery. We demonstrate the effectiveness of this platform in treating corneal neovascularization in an in vivo rabbit model, surpassing the current gold standard surgical therapy. This platform has the potential to revolutionize the management of various chronic ocular diseases without causing significant side effects.</p> <p>To enable intracellular recording, we present a unique platform consisting of vertical Si nanoneedles coated with a thin, transparent network of Au-Ag nanowires. This platform is held in place and enclosed by a soft, transparent elastomer, providing simultaneous intracellular recording and live imaging with applications in neuroscience, cardiology, muscle physiology, and drug screening. To demonstrate the utility of this platform, we monitored electrical potentials from cardiomyocyte cells and cardiovascular organoids. Additionally, we propose an intra-organoid sensing platform with vertical Si nanoneedles transfer printed into a soft scaffold. This platform can be adjusted and tailored for various organoids and tumor tissues of interest, or used to deliver bioactive molecules of interest into organoids in response to external stimuli.</p> <p>Our proposed designs of vertical Si nanoneedles based platforms demonstrate their significant potential for a broad range of biomedical applications, including ocular drug delivery, intracellular recording, and intraorganoid sensing. These platforms have the potential to revolutionize current approaches and pave the way for future developments in biomedical research and clinical applications, offering new possibilities for the diagnosis and treatment of a wide range of diseases.</p>
