Com o objetivo de investigar a possível transmissão vertical em bovinos, colheu-se também sangue de fêmeas e de seus respectivos fetos, abatidos em frigorífico, provenientes da região sudoeste do Estado de Mato Grosso. O total de 1076 soros correspondente a: 932 de fêmeas bovinas leiteiras, 37 de cães, 67 de humanos, provenientes de 24 propriedades da região sudoeste do Estado de Mato Grosso, 20 soros de fêmeas bovinas e 20 de seus respectivos fetos. Os soros examinados por meio da Imunofluorescência Indireta (IFI) revelaram a presença de anticorpos contra Neospora caninum em 499 bovinos (53,54%), 25 caninos (67,57%) e sete humanos (10,45%). Dos exames sorológicos de animais de abatedouro, duas fêmeas e seus respectivos fetos apresentaram positividade, com infecção caracterizada no terço médio da gestação. Ainda do mesmo grupo, três fêmeas mostraram-se positivas, em diferentes titulações, sem transmissão de anticorpos para os fetos, sendo duas analisadas em período gestacional correspondente ao terço final, e uma inicial. Os resultados obtidos permitem inferir que a infecção pelo protozoário encontra-se amplamente disseminada na região estudada. / To determine the prevalence of anti-Neospora caninum antibodies, 932 samples of blood sera from female dairy cattle belonging to 24 farms in the southwest of the state of Mato Grosso were analyzed by means of the indirect immunofluorescent-antibody test (IFAT = 200). Serum samples from 37 dogs and 67 humans living in the same farms were tested for anti- N. caninum antibodies. From these, 499 (53,54%) of the cattle sera, 25 (67,57%) of the dog sera and seven from the human sera (10,45%) were positive for the infection. In order to determine the occurrence of vertical transmission of N. caninum in bovine, maternal and fetal sera obtained from 20 Killed pregnant heifers from the same region were analyzed. Antibodies were found in five heifers (25%) from which two fetuses (10%) showed positive results. Fetal antibodies were detected on mid gestation. These data suggest that neosporosis is present among dairy cattle in the studied geographic region.

Development of diagnostic tests for the detection of Neospora caninum infected cattle

The protozoan parasite Neospora caninum is among the most frequently diagnosed infectious causes of bovine abortion thus causing significant economic losses, production inefficiency and welfare concern to the cattle sector worldwide. The control of bovine neosporosis relies on management techniques within which the identification of infected animals by serological testing plays a key role. However, the reliable diagnosis of the disease is hindered by the complex host-parasite interactions and the intrinsic limitations of the serological diagnostic tools currently available; as a result, some infected animals may not be detected. At the herd, regional and national levels, this can potentially undermine efforts for the control of the disease. The work presented in this thesis was undertaken to further investigate avenues to improve the diagnosis of N. caninum infection in cattle. It has been shown previously that recombinant antigens expressed by the rapidly proliferating tachyzoite stage and the slowly multiplying bradyzoite stage of N. caninum can be successfully employed for the detection of specific antibody responses during acute and persistent infections respectively. Following the establishment of persistent infections, sustained by the bradyzoite stage, the antibody levels against the tachyzoite stage may decline below the detection limits of currently available diagnostic tests which are exclusively based on antigens expressed by the tachyzoite. Consequently, the use of bradyzoite antigens for the development of serological diagnostic tests, may enhance the identification of infected animals. Novel antigens putatively expressed by the quiescent bradyzoite stage of N. caninum have been identified, expressed as recombinant proteins and assayed for the detection of specific antibodies. The recognition of recombinant tNcSRS12A-B and tNcSRS44- A by specific antibodies in sera from persistently N. caninum-infected cattle suggested that these proteins could be used for the detection of persistently infected animals. Indirect ELISAs based on previously characterised N. caninum antigens, such as the tachyzoite surface protein rNcSRS2, the immunodominant dense granule protein rNcGRA7 and the bradyzoite specific surface antigens rNcSAG4, rNcBSR4 and rNcSRS9, as well as a commercial test using whole tachyzoite lysate as antigenic preparation, were evaluated within a cross-sectional study to estimate the seroprevalence of bovine neosporosis in British dairy cattle. Moderate, but not high, agreement was found amongst the tests based on whole tachyzoite lysate, rNcSRS2 and rNcGRA7, and amongst the bradyzoite-specific antigen-based iELISAs. In contrast, only slight agreement was observed when each test detecting antibody responses indicative of acute infection (whole tachyzoite lysate, rNcSRS2 and rNcGRA7) was compared with each test detecting antibody responses indicative of persistent infection (bradyzoite-specific antigen-based iELISAs). Most N. caninum antibody-positive cattle samples showed detectable antibodies only against either antigens predominantly expressed by the tachyzoite or bradyzoite antigens thus suggesting that the exclusive use of one type of test may result in the misclassification of a proportion of animals, which test negative despite harbouring the parasite. This may result in the underestimation of the seroprevalence. Consequently, the combination of multiple tests in parallel, both tachyzoite and bradyzoite antigen-based, would improve the diagnosis of bovine neosporosis. Molecular tools for the genetic discrimination of different N. caninum isolates were also investigated. A novel multilocus fragment typing (MLFT) tool based on twelve highly polymorphic microsatellite markers was developed and applied to the analysis of DNA samples obtained from laboratory-maintained N. caninum isolates and tissues collected from bovine foetuses aborted due to N. caninum. The locus-specific nested PCRs associated with automated fragment analysis by capillary electrophoresis enabled to evaluate the markers in terms of typeability and discriminatory power. Overall, the typing tool was characterised by good typeability and discriminatory power and enabled to provide information on the genetic diversity amongst the laboratory-maintained and clinical N. caninum isolates studied. The MLFT tool may help to investigate the likely source of infection within abortion outbreaks and aid the study of the association between the genetic heterogeneity of N. caninum and the diverse biological features in vitro and in vivo. Furthermore, the loci characterised by the highest discriminatory power and typeability may be used alongside already established microsatellite markers for the development of an improved typing tool which could be proposed at the inter-laboratory level. Finally, current perceptions and common veterinary practice related to the diagnosis and control of bovine neosporosis were studied by developing a questionnaire for cattle practitioners in the United Kingdom. The survey highlighted the awareness of the limitations of current serological techniques and the demand for additional tools in terms of diagnostics and vaccines to tackle the economic losses and animal welfare implications related to N. caninum in cattle.

Caracterização molecular da actina do Apicomplexa Neospora caninum / Molecular characterization of the actin from the Apicomplexan Neospora caninum

Neospora caninum é um protozoário pertencente ao filo Apicomplexa que atinge, dentre diversos hospedeiros intermediários, principalmente bovinos e tem emergido como um importante causador de problemas reprodutivos e abortos em rebanhos de corte e leiteiro. Organismos do filo Apicomplexa são parasitas intracelulares obrigatórios que, para locomoverem-se e realizarem a invasão das células hospedeiras, utilizam um mecanismo próprio de locomoção ativa impulsionada pelo motor actina/miosina denominado motilidade por deslizamento (gliding motility), cujo complexo motor localiza-se entre a membrana plasmática e o complexo de membrana interno do parasita. A investigação a respeito do funcionamento desse mecanismo de locomoção e invasão vem sendo realizada principalmente em Toxoplasma gondii e Plasmodium spp., entretanto não há nenhuma publicação envolvendo actina em N. caninum. Esse trabalho envolveu a clonagem e expressão da sequência NcAct201-310 e deu início a caracterização da actina de N. caninum (NcAct). A sequência NcAct foi obtida em banco de dados ToxoDB, e uma comparação por alinhamento entre as actinas de Apicomplexas relacionados revelou que NcAct é idêntica à TgACT1 (100% identidade). Com outras espécies, a NcAct tem maior identidade/similaridade com a actina de Eimeria tenella (97%/99%), seguida da actina de Plasmodium falciparum PfACT1 (93%/97%), da actina de Babesia bovis (86%/94%) e PfACT2 (80%/92%). Quando localizada com anticorpo anti-?-actina C4, NcAct apresenta-se em duas bandas de 43 e 45 kDa em gel de acrilamida 1D e em nove isoformas em gel de acrilamida 2D. Todas as identidades das bandas e spots foram confirmados por espectrometria de massas (MS/MS). Além disso, NcAct localiza-se, em sua maioria, na região periférica do taquizoíta de N. caninum e sua distribuição é alterada após incubação dos taquizoítas com 5 ?M de jasplakinolida (JAS) ou 2 ?M de citocalasina D (CytD). Pathogenic and antigenic characterization of <I>Neospora hughesi</I>

<I>Neospora hughesi</I> is a recently described cause of equine protozoal myeloencephalitis (EPM). In the present study, we examined the susceptibility of BALB/c gamma-interferon gene knockout (gamma-INFKO), BALB/c, CD-1, and C57BL/6 strains of mice and gerbils to infection with tachyzoites of the Nh-A1 strain of <I>N. hughesi</I>. Only the gamma-IFNKO mice developed severe clinical disease following infection with <I>N. hughesi</I>. The most severe lesions were in the hearts of these mice. Two dogs fed the brains of mice, shown to contain <I>N. hughesi</I> tissue stages by cell culture and g-IFNKO mouse bioassay, did not shed <I>N. hughesi</I> oocysts over a 23 day observation period. We report important differences between the nucleotide and deduced amino acid sequences of the dense granule proteins GRA6 and GRA7 of <I>N. hughesi and N. caninum</I>. The newly defined proteins of <I>N. hughesi</I> are referred to as NhGRA6 and NhGRA7. From analysis of the sequences we found that there is a 14.8% difference in deduced amino acid sequence between NhGRA7 and NcGRA7, and a 4% difference between NhGRA6 and NcGRA6 in areas that could be compared. This thesis supports the identification of <I>N. hughesi</I> as a separate species from <I>N. caninum</I> and describes novel methods of distinguishing between the two. / Master of Science

Aborto bovino por Neospora caninum: fatores associados ao resultado positivo da imunoistoquímica

Um total de 237 fetos bovinos abortados foram enviados ao Setor de Patologia Veterinária da Universidade Federal do Rio Grande do Sul entre junho de 1999 e maio de 2004. Tecidos destes fetos foram analisados através da histologia e imunoistoquímica (IHQ) anti-Neospora caninum. Destes, 72 apresentaram lesões histológicas sugestivas de infecção por protozoário, caraterizadas pela presença de miosite não-supurativa, miocardite não-supurativa, pneumonia intersticial mononuclear associada a focos de necrose e encefalite necrosante não-supurativa multifocal. O cérebro apresentava consistência líquida ou semi-líquida em 61,1% dos fetos, dificultando a obtenção de tecido para a análise histológica e IHQ. A IHQ revelou a presença de taquizoítos e raros cistos de N. caninum no cérebro de 33/47 (70,2%) dos fetos que apresentam lesões histológicas sugestivas de infecção por protozoário. No modelo final da análise multivariada, três variáveis estiveram associadas significativamente com resultado positivo na IHQ nos fetos abortados: consistência do cérebro (P=0,031), lesão no cérebro (P=0,028) e idade (P=0,189). As chances de um resultado positivo na IHQ de N. caninum foram 4,35 vezes maiores (OR=4,35; 95% CI:1,1320-2,8116) quando o feto apresentava cérebro com consistência considerada normal e 4,94 vezes maiores (OR=4,94; 95%CI: 0,1646-3,0313) na presença de lesão sugestiva de infecção por protozoário caracterizada por encefalite de intensidade variada. A probabilidade de resultado positivo na IHQ foi inversamente proporcional à idade fetal, cada mês adicional de gestação diminuíram as chances de um feto apresentar resultado positivo na imunoistoquímica por 0,41 (OR=0,41, 95%CI: 1,5982- 0,1436).
