Characteristics of Soil Heterogeneity and Effectiveness of Crop Reflectance on Detection of Corn (Zea mays L.) Nitrogen Status

Spatial variations in soil properties can directly affect Nitrogen status of corn (Zea mays L.) and decrease efficiency of uniform fertilizer N applications. The objective of this study was to assess the spatial variations of soil properties and measure the effect on corn Nitrogen status through canopy reflectance. Field trials were conducted in 2010 and 2011 on a producer’s field west of Yazoo City, MS that contained high in field variability. Soil physical and chemical properties all exhibited moderate to high spatial dependency during both years of this study. Vegetative indices were derived from canopy reflectance values and indices utilizing the red-edge were the strongest and most consistent descriptors of tissue N percent and whole plant N uptake. The Canopy Chlorophyll Content Index (I) shows the greatest potential of assessing variations of corn Nitrogen status among the indices tested.

Whole Cell Bacterial Biosensor for Glutamine and Applications to Plants and Microbes

Glutamine (Gln) is a critical intermediate in nitrogen metabolism in all organisms. Here, a whole cell biosensor (GlnLux) for Gln was constructed by transforming a bacterial Gln auxotroph with a constitutive lux reporter. The biosensor was optimized for sensitivity, linearity, efficiency, specificity and robustness to permit detection of Gln in vitro and in vivo. The optimized GlnLux biosensor achieved nanomolar sensitivity with Gln standards. Extracts from only 1 mg of maize (Zea mays L.) leaf tissue were sufficient for Gln detection by GlnLux. Measurements of Gln in leaf extracts by GlnLux correlated with quantification by high performance liquid chromatography (Spearman r = 0.95). GlnLux permitted indirect in planta imaging of Gln using a CCD camera, enabling identification of plants that had been fertilized with nitrogen. Imaging using GlnLux also resolved predicted spatial differences in leaf Gln concentration. In a second application, it was demonstrated that GlnLux embedded into agar permits non-destructive screening of co-inoculated bacterial colonies for biological nitrogen fixation (BNF). GlnLux agar was able to distinguish a Bradyrhizobium japonicum wild type strain (nif+) from a mutant strain defective in nitrogenase (nif-) following ≥8 h of co-incubation. The technology was used to screen a bacterial endophyte diversity library cultured from Zea mays (L.) seeds for biological nitrogen fixation. / OMAFRA

Hierarchy of factors impacting grape berry mass at different scales and its direct and indirect effects on grape and wine composition / Hiérarchisation des facteurs impactant la masse de la baie de raisin à différentes échelles et leurs effets directs et indirects sur la composition du raisin et du vin

La masse de la baie est le résultat de l’effet intégré de plusieurs facteurs. La recherche a été dessinée afin d’étudier l’effet simultané des facteurs majeurs influençant la masse et la composition de la baie, de les hiérarchiser selon leur degré d’impact à des échelles différentes, de séparer leur effet direct et indirect sur la composition du raisin et de comparer le profil de vins élaborés à partir de petites et grosses baies. L’étude a été conduite sur deux sites expérimentaux, localisés dans les régions de Saint-Emilion (France) et Alcamo (Italie), pendant les années 2014 et 2015. Sur le premier site, les vignes sont plantées sur deux types de sols, tandis que sur le deuxième, deux traitements hydriques étaient appliqués. A l’échelle intra-parcellaire, l’état hydrique de la vigne représente le facteur le plus important, tandis que l’effet du nombre de pépins par baie n’est pas significatif. Des résultats opposés sont obtenus lorsque les relations sont étudiées à l’échelle de la grappe et de la plante. A large échelle, les facteurs impactent directement et indirectement la composition du raisin et les petites baies produisent des moûts et des vins plus concentrés. A l’inverse, à l’échelle de la grappe et de la plante, la masse de la baie n’influence pas la composition du raisin. Seule la concentration en anthocyanes est significativement liée à la masse à toutes les échelles. Cette relation est particulièrement évidente sous conditions hydriques limitantes. Un déficit hydrique augmente le ratio pellicule/pulpe, indépendamment de la masse de la baie. Petites et grosses baies d’une parcelle ayant une condition hydrique homogène, tendent à avoir un profil similaire. / Final berry mass is the result of the integrated effect of several factors. They also influence berry composition. The present work was designed to study the simultaneous effect of major factors influencing berry mass and composition, to hierarchize their impact at different scales, to distinguish their direct and indirect effect on berry composition and to compare the profile of wines made from large and small berries. The study was carried out simultaneously on two vineyards located in the Saint Emil ion (France) and Alcamo (Sicily) areas, during 2014 and 2015. On the first site, vines were planted on two soil types, while on the second site two different irrigation treatments were applied. Depending on the scale, some factors homogeneously impacted the berry mass and composition. At the intra-parcel scale, vine water status represented the most impacting factor, while berry seed number did not have significant effect. Opposite results were obtained when the investigation was carried out at the intra-bunch and intra-plant scales. At large scale, factors impacted directly and indirectly berry compounds and grape juices and wines produced from smaller berries were more concentrated. Neither at intra-bunch, nor at intra-plant scales, berry size effect on juice composition was significant. Only anthocyanin concentration was related to berry size at all scales. This fact was particularly obvious in berries produced under limited water conditions. Water deficit increased the skin to flesh ratio, independently of berry size. This means that small and large berries, produced from a single parcel with homogenous water uptake conditions, tend to have similar enological profiles.

Analyse du fonctionnement des performances des associations blé dur-pois d'hiver et blé dur-féverole d'hiver pour la conception d'itinéraires techniques adaptés à différents objectifs de production en systèmes bas-intrants / Analysis of the functioning and efficiency of durum wheat - winter pea and durum wheat - winter faba bean intercrops in order to design cropping systems

Les associations d'espèces sont définies comme la culture simultanée d'au moins deux espèces sur la même parcelle pendant une période significative de leur croissance. Ce système permettrait d'améliorer l'utilisation des ressources du milieu (eau, azote, lumière…) et ainsi d'augmenter le rendement et la qualité des grains par rapport aux cultures monospécifiques. L'objectif de notre travail était d'analyser le fonctionnement et d'évaluer la performance des associations blé dur - pois d'hiver et blé dur - féverole d'hiver pour aider à la conception d'itinéraires techniques adaptés à différents objectifs de production. Pour cela nous avons testé, au cours de trois années d'expérimentations, différentes combinaisons de variétés de blé dur, disponibilités en azote, structures de couverts et densités de plantes. Nos résultats ont confirmé l'intérêt de ces systèmes pour améliorer le rendement et la teneur en protéines du blé dur comparativement aux cultures monospécifiques mais également pour la réduction des ravageurs, maladies et de l'enherbement dans certaines conditions. Ces systèmes sont ainsi particulièrement bien adaptés aux situations à faible disponibilité en azote en raison de la complémentarité entre céréale et légumineuse pour l'utilisation de l'azote (minéral du sol et fixation symbiotique) mais aussi pour la captation de l'énergie lumineuse. In fine, ce travail a permis de proposer des prototypes d'itinéraires techniques d'associations adaptés à différents objectifs de production, grâce notamment à l'analyse dynamique des compétitions et complémentarités entre espèces au sein du couvert et en particulier de l'élaboration du rendement du blé dur en association. / Intercropping is the simultaneous growing of two or more species in the same field for a significant period. Such systems are known to use available resources (water, light, nitrogen…) more efficiently than their corresponding sole crops and consequently to increase yield and grain protein concentration particularly in low N input systems. The aim of our work was to analyse the functioning of durum wheat - winter pea and durum wheat - winter faba bean intercrops. A 3-year field experiment was carried out with different fertilizer-N levels, wheat cultivars and plant densities. The intercrop efficiency was studied in order to further design low inputs cropping systems for specific objectives. Our results confirm that intercropping is more suited to low-N-input systems because it takes advantage of complementary N sources (soil mineral N and N2 fixation) and light use between species of the intercrop. Thus, intercrops were more efficient than sole crops for yield, they increased durum wheat grain protein concentration and in certain cases reduced weeds, pests and diseases. Finally, our work indicates that intercrops can be optimized for specific objectives by the choice of crop species, cultivars, N fertilization and plant densities in order to maximize resource capture and minimize competition. Indeed, intercrop efficiency is determined by the complementary resource use between the two species as well as the relative strength of the intra- and interspecific dynamic competitive interactions.
