Resumo: As abelhas Apis mellifera são insetos ecologicamente e economicamente importantes. Elas asseguram a polinização de diversas plantas contribuindo para a manutenção da biodiversidade. Seu valor econômico resulta não somente de seus produtos diretos mas também pela ativa polinização que exercem em culturas. A polinização feita por animais é importante para a reprodução sexuada de muitas culturas e para a maioria das plantas nativas, as quais também podem ser importantes como fonte de calorias e micronutrientes para os seres humanos. Além do mais, o declínio no número de polinizadores pode acarretar uma redução concomitante no número de espécies vegetais. A abelha A. mellifera, destaca-se como polinizador economicamente mais valioso para culturas em todo o mundo. Por outro lado, a agricultura moderna cada vez mais depende do uso de produtos químicos para controlar plantas daninhas, fungos e insetos-praga para assegurar a produtividade. Abelhas melíferas podem entrar em contato com tais agentes químicos devido suas atividades de coleta de água, resinas vegetais, pólen e néctar. A intoxicação resultante desta exposição pode ser letal, o que é facilmente identificável, ou causar efeitos na fisiologia e no comportamento do inseto. Tais efeitos, ocasionados por doses subletais, são difíceis de serem mensurados, como paralisia, desorientação ou mudanças comportamentais; porém, podem comprometer toda a estrutura social da colônia. Visando um melhor conhecimento dos efeitos de doses subletais de inseticidas em abelhas, estudou-se a ação dos ingredientes ativos Acetamiprido, Tiametoxam e Fipronil na sobrevivência e nos comportamentos de reflexo de extensão da probóscide e da atividade locomotora em operárias de A. mellifera. Their economic value derives not only from their direct products but also from their pollinating activity in crop plants. Animal pollination is important to sexual reproduction of many crops and the majority of wild plants, which can also be important for providing calories and micronutrients for humans. Furthermore, the decline of pollinating species can lead to a parallel decline in number of plant species. A. mellifera stands out as the most economically valuable crop pollinator in the world. Modern agriculture increasingly depends on the use of chemicals substances to control weeds, fungi and insect pests to ensure high yields. Honey bees may frequently become exposed to such chemicals as a consequence of their foraging activities collecting water, natural resins, pollen and nectar. Intoxication resulting from this exposure can be lethal, which is easily identifiable, or cause effects on the physiology and insect behavior. These effects, caused by sublethal doses are difficult to measure (such as paralysis, disorientation or behavioral changes), and can compromise the entire social structure of the colony. To improve the knowledge about the effects of insecticides sublethal doses effects in honey bees, we studied the action of the active ingredients Acetamiprid, Thiamethoxam and Fipronil on survival and behavior of the proboscis extension reflex (PER) and locomotor activity in workers of A. mellifera. Twenty-four hours after topic application, the LD50 values of Acetamiprid, Thiamethoxam and Fipronil were 9,3 μg/bee, 17 ng/bee and 1,9 ng/bee, respectively. We also estimated a LT50 with values of 1,4 hours, 3,8 hours and 19,8 hours to Acetamiprid, Thiamethoxam and Fipronil respectively. Este trabalho está centrado na análise dos fenômenos socioespaciais que envolvem a criminalidade violenta no meio urbano. Busca-se investigar a relação entre o aumento da violência letal e os elevados índices de desigualdade social nos últimos 30 anos. O trabalho trata respectivamente das causas da violência urbana, das relações do crime e da violência com o espaço, além da participação dos jovens nas ações violentas e no trabalho do tráfico de drogas como fator decisivo na elevação das taxas de letalidade juvenil. Selecionamos o bairro Santa Tereza, por tratar-se de uma região conflituosa no município de Porto Alegre - RS, como objeto de estudo de caso, visto que concentra características de uma sociabilidade violenta. Investigamos as relações de pertencimento nas comunidades e o medo dos lugares em virtude do aumento da criminalidade nos centros urbanos. Da mesma forma, consideramos as causas do aumento da violência como sendo um dos fatores de repulsão dos espaços de uso comum e promotor de novas territorialidades a partir do esgarçamento do tecido sócio espacial. / This work focuses on the analysis of socio-spatial phenomena involving violent crime in urban areas. The aim is to investigate the relationship between the increase in lethal violence to high levels of social inequality in the last 30 years. The work deals respectively of the causes of urban violence, crime and violence relations with the space on the participation of young people in violent actions and the work of drug trafficking as a decisive factor in the rise of juvenile mortality rates. We selected Santa Tereza district, because it is a conflictive region in the city of Porto Alegre - RS, as a case study object as it focuses characteristics of a violent sociability. We investigated the relationships of belonging in communities and the fear of places due to the increase of crime in urban centers. Similarly we consider the causes of increasing violence as one of repulsion factors of spaces for common use and promoter of new territoriality from the fraying of the socio-spatial fabric.

As moscas-das-frutas são consideradas importantes pragas da fruticultura mundial, causando perdas significativas à produção e ao livre trânsito de frutas. Com a preferência por alimentos com baixos níveis de agrotóxicos, pesquisas estão buscando alternativas ecológica, social e econômica viáveis para o controle destas pragas. A Técnica do Inseto estéril (TIE) é uma técnica onde os insetos são criados, esterilizados e liberados em áreas infestadas, reduzindo a fertilidade da população selvagem, além de ser um método não poluente e não agressivo ao ambiente. Com o desenvolvimento de novos estudos da metodologia da TIE, foi introduzida uma nova linhagem mutante a Sensibilidade a Letal Temperatura (tsl) Viena-8, cujas fêmeas são sensíveis à alta temperatura, podendo ser eliminadas já na fase embrionária, no início da criação massal, agregando mais benefícios a este método de controle e/ou supressão de praga. Com diferentes testes de incubação de ovos da linhagem tsl, com temperaturas e tempos combinados e variados, comprovou-se a extrema sensibilidade às mudanças de temperatura. A temperatura ideal para a incubação dos ovos dessa linhagem está entre 24ºC a 26ºC pelo período de 24 a 48 horas. Deve-se tomar o máximo de cuidado com a elevação da temperatura acima de 26ºC durante a manutenção e multiplicação dessa linhagem em condições de laboratório ou massalmente. Acima de 26ºC, há sérios riscos de comprometer a qualidade dos insetos produzidos. O curto período de 6 horas a uma temperatura de 34ºC, já é suficiente para alterar negativamente a proporção de fêmeas de ovos tratados termicamente / The fruit flies are considered important pests of the world horticulture, causing significant loses to the production and the free traffic of fruits. With the preference for foods with low levels of pesticides, researches are looking for alternatives ecological, social and economical viable for the control of these pests. The Sterile Insect Technique (SIT) is a technique where the insects are reared, sterilized and release in infested areas, reducing the fertility of the wild population, and is a method no pollutant and no aggressive to the environment. With the development of new studies of the methodology of SIT, a new mutation, Temperature Sensitive Lethal (tsl) Vienna 8, whose females are sensitive to the high temperature, could already be eliminated in the embryonic phase, in the beginning of the mass-rearing, resulting in more benefits to the control/suppression this pests. Different tests of incubation with eggs of tsl strain, with variation of time and temperature was done and proved a very sensitive to temperature changes. The ideal temperature for the incubation of the eggs is among 24ºC to 26ºC by the period from 24 to 48 hours. The maximum of care should be taken with the elevation of the temperature above 26ºC during the maintenance and multiplication in laboratory conditions or mass-rearing. Above this temperature it can commit the quality of the produced insects. The short period of 6 hours with temperature to 34ºC, is enough to change negatively the proportion of females of eggs in the thermal treatment

An Exploratory Analysis of Media Reporting of Police Involved Shootings in Florida

The focus of this study is on media reporting of police involved shootings in Florida. Given that knowledge of killings committed by law enforcement are frequently restricted to what people get from news sources, it is important to investigate the way these messages are being communicated. An exploratory analysis of 199 articles and transcripts covering 86 cases relevant to deadly use of force by police officers as reported from 2013 to 2015 provided the primary data source. The analysis engaged a critical examination of media content and goodness of fit models to identify commonalities used by the media when reporting police involved shootings. The goal is to determine the potential impact of race, age and gender of a victim with regard to the media’s portrayal of events that result in lethal use of force by police. It is expected that exposure of the discoveries in this study will influence future law enforcement reporting systems for a more transparent relationship between police, news and the community. The demographics characteristics race, sex and age that were assessed in this study were not significantly associated with the length of media coverage on police involved shootings. However, further analysis using different modeling approaches are required for a better understanding as to whether a relationship exists.

Identifying genetic interactions of the spindle checkpoint in Caenorhabditis elegans.

Faithful segregation of chromosomes is ensured by the spindle checkpoint. If a kinetochore does not correctly attach to a microtubule the spindle checkpoint stops cell cycle progression until all chromosomes are attached to microtubules or tension is experienced while pulling the chromosomes. The C. elegans gene, san-1, is required for spindle checkpoint function and anoxia survival. To further understand the role of san-1 in the spindle checkpoint, an RNAi screen was conducted to identify genetic interactions with san-1. The kinetochore gene hcp-1 identified in this screen, was known to have a genetic interaction with hcp-2. Interestingly, san-1(ok1580);hcp-2(ok1757) had embryonic and larval lethal phenotypes, but the phenotypes observed are less severe compared to the phenotypes of san-1(ok1580);hcp-1(RNAi) animals. Both san-1(ok1580);hcp-1(RNAi) and san-1(ok1580);hcp-2(RNAi) produce eggs that may hatch; but san-1(ok1580):hcp-1(RNAi) larvae do not survive to adulthood due to defects caused by aberrant chromosome segregations during development. Y54G9A.6 encodes the C. elegans homolog of bub-3, and has spindle checkpoint function. In C.elegans, bub-3 has genetic interactions with san-1 and mdf-2. An RNAi screen for genetic interactions with bub-3 identified that F31F6.3 may potentially have a genetic interaction with bub-3. This work provided genetic evidence that hcp-1, hcp-2 and F31F6.2 interact with spindle checkpoint genes.

In this thesis, the role of Legal Autonomous Weapons Systems (LAWS) in the German institutional-political landscape is examined. The effort guiding the thesis is to uncover and understand how LAWS have been constructed in the German institutional-political discourse from 2017-2021. The field of LAWS is comparatively under-researched due to their recent emergence and highly contested spread as a weapon of choice. By trying to answer how securitization is taking place in German political-institutional debates surrounding LAWS, the aim is to better understand the different discursive elements of securitization in the specific German context. Building on the elements of defining the securitizing actor and referent subject, evaluating which discourse elements contribute and limit a securitization of LAWS and how suggested policy measures are tied to other discourses allows for a multileveled understanding of the issue at hand. What is clear is the need to further expand research in this field with the goal of contributing to the larger body of literature within Peace and Conflict Studies, detangling present discourses and suggesting potential policy paths. Finally, the conclusion drawn in this thesis suggests that both a technological discourse and discourse of threat of other actors contribute toward securitization of LAWS while a liberal economic discourse limits the securitization of LAWS.

Whole genome doubling confers unique genetic vulnerabilities on tumor cells

Whole genome doubling (WGD) generates genetically unstable tetraploid cells that fuel tumorigenesis. Cells that undergo WGD must acquire adaptive characteristics to accommodate their tetraploid state, and these adaptations may confer unique vulnerabilities that can be exploited therapeutically. We analyzed the genomes of ~9,700 primary human cancer samples to uncover genetic alterations that are specifically enriched in WGD+ cancer cells. Through integrating our genetic analysis with gene essentiality data acquired from Project Achilles, we identified gene dependencies in WGD+ cells. Moreover, we identified genes that are essential for the viability of WGD+ cancer cells, but non-essential to non-transformed diploid cells. We demonstrated that the gene encoding for the mitotic kinesin KIF18A is dispensable for mitosis in diploid cells, but becomes critical for accurate chromosome segregation and viability in WGD+ cells, making it an attractive drug target. Collectively, this work revealed new strategies to specifically target WGD+ cancer cells, namely targeting the gene KIF18A, while sparing the normal diploid cells from which they arise. / 2022-06-04T00:00:00Z

The thesis deals with the issue of the families taking care of an incurably ill children with the particular focus on the support of the parents by the general public. The thesis is divided into two parts. The theoretical part describes the current situation of the families with an incurably ill children in the Czech Republic and their needs and problems as well as the support provided by the society. In addition the theoretical part is focused on the sources of the support by the general public. The practical part contains a qualitative analysis which aims to investigate whether the respondents would provide the support to the families taking care of an incurably ill children or if the respondents would expect to get the support in a difficult life situation.
