NMR als Mittel zur Beobachtung der gelösten Eisen-Konzentration im Porenraum von Sedimenten

In der vorliegenden Arbeit wurde die Methode der magnetischen Kernspinresonanz (NMR) eingesetzt, um beim Schadstoffabbau stattfindende Prozesse und geochemische Reaktionen zerstörungs- und beprobungsfrei zu untersuchen. Dies ist möglich, da die gelösten Elektronenakzeptoren Sauerstoff und Eisen paramagnetisch sind und somit einen Ein uss auf die NMRRelaxationszeiten ausüben. Der lineare Zusammenhang zwischen der gelösten Sauerstoff- beziehungsweise Eisen-Konzentration und den NMR-Relaxationsraten 1/T1 und 1/T2 wurde quantifiziert. Weiterhin wurde der bereits bekannte Einfluss der Matrixoberflächen von porösen Medien auf die Relaxation von Wasser nachgewiesen. Die paramagnetischen Zentren auf Sandoberflächen führen ebenfalls zu einer Verkürzung der Relaxationszeiten. Es wurde gezeigt, dass die kleinsten Korngrößen der verwendeten Sande den größten Einfluss auf die Oberflächenrelaxation haben. Wird die Oberflächenrelaxation berücksichtigt, ist auch in porösen Medien die ermittelte lineare Abhängigkeit der Relaxationszeiten von der Ionenkonzentration anwendbar, um den Gehalt an gelösten paramagnetischen Ionen aus Relaxationsmessungen zu ermitteln. Beispielhaft wurde der Anstieg der Eisen(III)-Konzentration in der Porenlösung von natürlichen Sanden infolge der Auflösung eisenhaltiger Mineralien von den Oberflächen zeitlich und räumlich detailliert betrachtet. Eine durchgeführte Modellierung zeigte, dass das Reaktionssystem zu Beginn der Reaktion von der Diffusion dominiert wird, am Ende dann die Reaktionsgeschwindigkeit der bestimmende Parameter ist. Die beim biologischen Schadstoffabbau auftretenden Redoxprozesse des Eisens wurden durch rein chemische Reaktionen unter Verwendung von Oxidations- und Reduktionsmitteln simuliert. Die zeitlich und räumlich detaillierte Beobachtung des Anstiegs beziehungsweise des Abfalls der gelösten Eisen(III)-Konzentration in der (Poren-)Lösung war mit NMR-Relaxometrie trotz der Schnelligkeit der Reaktionen möglich. Mit Hilfe der anschliessenden Modellierung wurde der wichtige Einfluss des pH-Wertes auf den genauen Ablauf der Reaktionen deutlich gemacht. Nur in sehr sauren pH-Bereichen (pH < 3) liegen die Eisen(III)-Ionen in Lösung vor. Weiterhin wurde der Einfluss der Mikroorganismen selbst auf die NMR-Relaxations- und Diffusionsmessungen untersucht. Im Rahmen dieser Arbeit wurde an Medien mit Lactobacillus und Penicillium eine Verschiebungen in den Relaxationszeitverteilungen hin zu kleineren Relaxationszeiten gemessen. Dies basiert auf der bereits bekannten Verringerung der Mobilität der Spins innerhalb der Biomasse. Für Bakterien von Geobacter metallireducens konnte erstmals der Verbrauch von Eisen(III)-Ionen durch Reduktion während des Wachstum anhand der ansteigenden T2-Relaxationszeit gezeigt werden.

Methemoglobin Formation via Nitric Oxide and Comparison of Methemoglobin, Deoxyhemoglobin, and Ferrous Nitrosyl Hemoglobin as Potential MRI Contrast Agents

Gadolinium-based contrast agents (GBCAs) are in widespread use to enhance magnetic resonance angiography images for evaluating vascular pathology. However, there are safety concerns and limitations regarding the use of GBCAs. It has been shown that the magnetic resonance imaging (MRI) signal intensity (T1-weighted images) in some of the brain's tissues is higher for patients who had multiple exposures to GBCAs compared to patients who had never had exposure to GBCAs. This implies that GBCAs are not sufficiently removed from body such that GBCAs may potentially have long-term effects on the human body. These potential safety concerns have led to an increased interest in alternative contrast agents. Methemoglobin (metHb) and oxygen-free hemoglobin (HHb) are two forms of hemoglobin with paramagnetic properties. It has been shown that the T1-weighted signal intensity of blood is changed during MRI scans for metHb and HHb, leading to enhanced contrast of MRI images. The ability of metHb and HHb to change the signal intensity has led to the idea that they can be used as effective contrast agents. MetHb can be made by exposing oxyhemoglobin (oxyHb) to nitric oxide (NO) and HHb can be made by removing the oxygen from hemoglobin using nitrogen (N2). In this study, a new gas delivery system was developed to make metHb and HHb. The new gas delivery system was developed to have greater experimental control compared to the PermSelect hollow-fiber module that was used in preliminary studies to make metHb. The same system can be used to make HHb. Initial experiments showed significant amounts of undesired nitrite (NO2-) formation during metHb formation due to the presence of contaminants in the NO gas source. To minimize this problem, flow of NO from the gas source was bubbled in a sodium hydroxide solution in order to reduce the NO2- concentration. Following metHb formation, continuous delivery of NO also led to the formation of ferrous nitrosyl hemoglobin (HbIINO). MRI studies showed that HbIINO can also increase the signal intensity of an MRI image. It is unknown as to whether metHb, HHb, or HbIINO would be a stronger and more appropriate contrast agent and to what extent the T1-weighted signal is affected by the concentration. This study evaluated T1-weighted images of blood samples over a range of metHb and HHb concentrations, as well as HbIINO concentrations. Comparison of T1 values showed that metHb is the strongest contrast agent and that HHb is a relatively weak contrast agent. This study showed for the first time that HbIINO can provide a contrast effect, although not as strong as metHb but stronger than HHb. With metHb providing a viable contrast between 10-20%, metHb has the potential to be a safe and effective contrast agent since it can be naturally converted back to hemoglobin.

An evaluation of the impact of the Non-ferrous Metals Crime Combating Committee on copper cable theft

Data was collected via in-depth interviews with awaiting trial detainees charged with copper cable theft at the Pollsmoor Correctional Centre in the Western Cape; members of the Mpumalanga and Gauteng Provincial Non-Ferrous Metals Crime Combating Committee; members of Business Against Crime South Africa; and investigators attached to Combined Private Investigations who investigate copper theft nationally. The in-depth interviews provided a comprehensive understanding of participant experiences relating to the impact of the Non-Ferrous Metals Crime Combating Committee on the combating of copper cable theft in South Africa. Moreover, the researcher performed a thorough literature study of the phenomenon locally and internationally. Research findings indicate shortcomings in the Non-Ferrous Metals Crime Combating Committee's effectiveness, limiting its impact on preventing copper cable theft. Based on these findings, the study recommends that the Non-Ferrous Metals Crime Combating Committee be replaced with a specialised South African Police Service Non-Ferrous Metals Theft Unit. Consequently, this study puts forward a set of recommendations proposing a systematic pro-active plan to address and minimise copper cable theft in South Africa. Simulation Monte-Carlo de la radiolyse du dosimètre de Fricke par des neutrons rapides / Monte-Carlo simulation of fast neutron radiolysis in the Fricke dosimeter

Monte-Carlo calculations are used to simulate the stochastic effects of fast neutron-induced chemical changes in the radiolysis of the ferrous sulfate (Fricke) dosimeter. To study the dependence of the yield of ferric ions, G(Fe[superscript 3+]), on fast neutron energy, we have simulated, at 25 [degree centigrade], the oxidation of ferrous ions in aerated aqueous 0.4 M H[subscript 2]SO[subscript 4] (pH 0.46) solutions when subjected to ~0.5-10 MeV incident neutrons, as a function of time up to ~50 s. The radiation effects due to fast neutrons are estimated on the basis of track segment (or"escape") yields calculated for the first four recoil protons with appropriate weighting according to the energy deposited by each of these protons. For example, a 0.8-MeV neutron generates recoil protons of 0.505, 0.186, 0.069, and 0.025 MeV, with linear energy transfer (LET) values of ~41, 69, 82, and 62 keV/[micro]m, respectively. In doing so, we consider that further recoils make only a negligible contribution to radiation processes. Our results show that the radiolysis of dilute aqueous solutions by fast neutrons produces smaller radical yields and larger molecular yields (relative to the corresponding yields for the radiolysis of water by [superscript 60]Co [gamma]-rays or fast electrons) due to the high LET associated to fast neutrons. The effect of recoil ions of oxygen, which is also taken into account in the calculations, is shown to decrease G(Fe[superscript 3+]) by about 10%. Our calculated values of G(Fe[superscript 3+]) are found to increase slightly with increasing neutron energy over the energy range covered in this study, in good agreement with available experimental data. We have also simulated the effect of temperature on the G(Fe[superscript 3+]) values in the fast neutron radiolysis of the Fricke dosimeter from 25 to 300 [degree centigrade]. Our results show an increase of G(Fe[superscript 3+]) with increasing temperature, which is readily explained by an increase in the yields of free radicals and a decrease in those of molecular products. For 0.8-MeV incident neutrons (the only case for which experimental data are available in the literature), there is a ~23% increase in G(Fe[superscript 3+]) on going from 25 to 300 [degree centigrade]. Although these results are in reasonable agreement with experiment, more experimental data, in particular for different incident neutron energies, would be needed to test more rigorously our Fe[superscript 3+] ion yield results at elevated temperatures.

Data was collected from literature sources as well as from security professionals combating copper theft. There are five primary objectives in this research: 1. To explore and to describe the extent and the impact of copper cable theft. 2. To gain insight into the profile and the modus operandi of the offender. 3. To evaluate current intervention measures used to combat the copper cable theft. 4. To describe the general factors limiting the success of combating copper cable theft. 5. To recommend probable intervention measures with which to combat copper cable theft. Semi-structured interviews were conducted with responsible security officials of victim stakeholder groups in Gauteng. Effects of iron and omega-3 supplementation on the immune system of iron deficient children in South Africa : a randomised controlled trial / Linda Malan

Background Iron deficiency (ID) is the world‟s most prevalent micronutrient deficiency and predominantly affects developing countries, also South Africa. In areas with low fish consumption and high n-6 PUFA vegetable oil intake, there is a risk for having inadequate n-3 PUFA status. Both iron and n-3 PUFA play important roles in the immune response, and supplementation is a strategy to alleviate deficiencies. However, little is known about potential interactive effects between concurrent iron and n-3 PUFA supplementation on the immune system. This is also important in the context that iron supplementation may be unsafe and may increase morbidity and mortality. Aim The overall aim of this thesis was to assess the effects of iron and docosahexaenoic (DHA)/eicosapentaenoic acid (EPA) supplementation, alone and in combination, on the immune system of ID children. More specifically, these effects were investigated on the occurrence and duration of illness and school-absenteeism due to illness, peripheral blood mononuclear cell (PBMC), red blood cell (RBC) and plasma total phospholipid fatty acid composition, iron status, fatty acid-derived immune modulators and targeted PBMC gene expression. Furthermore, association of PBMC, RBC and plasma total phospholipid fatty acid composition with allergic disease, were also examined. Design In a 2-by-2 factorial, randomised, double-blind, placebo-controlled trial, South African children (n = 321, aged 6–11 y) were randomly assigned to receive oral supplements of either 1) iron (50 mg as ferrous sulphate) plus placebo; 2) DHA/EPA (420/80 mg) plus placebo; 3) iron plus DHA/EPA (420/80 mg); or 4) placebo plus placebo for 8.5 mo, four times per week. Absenteeism and illness symptoms were recorded and biochemical parameters for compliance as well as parameters fundamental to immune function were assessed at baseline and endpoint. Furthermore, in a cross-sectional design, associations of allergic disease with baseline fatty acid composition of PBMC, RBC and plasma were examined. Results The combination of iron and DHA/EPA significantly attenuated respiratory illness caused by iron supplementation. DHA/EPA supplementation alone improved respiratory symptoms at school, but increased headache-related absenteeism. DHA/EPA and iron supplementation individually tended to increase and decrease anti-inflammatory DHA and EPA-derived mediators, respectively. Furthermore the anti-inflammatory DHA-derived immune mediator, 17HDHA was higher in the DHA/EPA plus placebo and iron plus DHA/EPA groups than in the iron plus placebo group. Also, the pro-inflammatory arachidonic acid (AA)-derived modulators (5- and 15-hydroxyeicosapentaenoic acid) were significantly lower in the iron plus DHA/EPA group compared to the placebo plus placebo groups. In the study population, 27.2% of the children had allergic disease and AA in PBMC phospholipids was significantly lower in the allergic children than in the non-allergic children. In RBC phospholipids dihomo-gamma-linolenic acid (DGLA) and the ratio of DGLA: linoleic acid (LA) correlated negatively and the n-6:n-3 PUFA ratio positively with total immunoglobulin E (tIgE). Furthermore, trans-C18:1n-9, tended to be higher in the allergic group. Conclusion DHA/EPA prevented respiratory illness caused by iron supplementation and although DHA/EPA on its own reduced respiratory morbidity when the children were present at school, surprisingly it increased the likelihood of being absent with headache and fever. The biochemical findings compliment the clinical results and support previous observations about DHA/EPA supplementation to reduce inflammation, but add to the current knowledge base that a relatively high oral dose of non-haem iron modulates circulating lipid-derived immune modulators and related gene expression. Furthermore, when supplementing with iron and DHA/EPA combined, in this ID population with low fish intake, the anti-inflammatory effect of DHA/EPA is maintained concurrently with attenuation of respiratory morbidity. This finding support the notion that excess iron (probably as non-transferrin bound iron) becomes available for pathogens and is probably why we found that iron increased respiratory infectious morbidity. The improved clinical outcome with combined supplementation seems to be related to increased lipid-mediator synthesis gene expression and the availability of DHA/EPA, leading to a more pro-resolving profile and enhanced immune competence. Overall these results give better insight into immune function and infectious morbidity in relation to n-3 PUFA and iron status and treatment, as well as the possible association of fatty acid status with allergic disease in young South-African school children. / PhD (Nutrition), North-West University, Potchefstroom Campus, 2015

In RBC phospholipids dihomo-gamma-linolenic acid (DGLA) and the ratio of DGLA: linoleic acid (LA) correlated negatively and the n-6:n-3 PUFA ratio positively with total immunoglobulin E (tIgE). Furthermore, trans-C18:1n-9, tended to be higher in the allergic group. Conclusion DHA/EPA prevented respiratory illness caused by iron supplementation and although DHA/EPA on its own reduced respiratory morbidity when the children were present at school, surprisingly it increased the likelihood of being absent with headache and fever. The biochemical findings compliment the clinical results and support previous observations about DHA/EPA supplementation to reduce inflammation, but add to the current knowledge base that a relatively high oral dose of non-haem iron modulates circulating lipid-derived immune modulators and related gene expression. Furthermore, when supplementing with iron and DHA/EPA combined, in this ID population with low fish intake, the anti-inflammatory effect of DHA/EPA is maintained concurrently with attenuation of respiratory morbidity. This finding support the notion that excess iron (probably as non-transferrin bound iron) becomes available for pathogens and is probably why we found that iron increased respiratory infectious morbidity. The improved clinical outcome with combined supplementation seems to be related to increased lipid-mediator synthesis gene expression and the availability of DHA/EPA, leading to a more pro-resolving profile and enhanced immune competence. Removing Reds from the Old Red Scar: Maintaining and Industrial Peace in the East Tennessee Copper Basin from the Great War through the Second World War

This study considers industrial society and development in the East Tennessee Copper Basin from the 1890s through World War II; its main focus will be on the primary industrial concern, Tennessee Copper Company (TCC 1899), owned by the Lewisohn Group, New York. The study differs from other Appalachian scholarship in its assessment of New South industries generally overlooked. Wars and increased reliance on organic chemicals tied the basin to defense needs and agricultural advance. Locals understood the basin held expanding economic opportunities superior to those in the surrounding mountains and saw themselves as participants in the nation’s industrial and economic progress, and a vital part of its defense. The study upends earlier scholarship contending local industrial concerns acted proactively to challenges from farmers harmed by industrial pollution; investigation shows firms hesitated to initiate new production processes and manipulated local elections. Partisan developments woven amid all this underscore errors in assuming ancient regional affinity for Republicans. Confederate heritage gave Democrats an historic advantage that fractured before New Deal progressivism and expanding basin Republican power. Markets forced basin firms to merge and embrace technological change affecting working people’s relationships, forcing workers to improve skills or settle for low-skill jobs. Excepting TCC managers and supervisory staff, provincialism ruled; suspicions and competitiveness among workers grew as most miners lived a few scattered villages and most managers and craftsmen settled in the basin’s “Twin-cities” district. Early union efforts collapsed before union mismanagement, rational management and a company union based upon Sam Lewisohn’s ideals. Management managed to wrest control of its industrial relations despite the effects of Depression and the New Deal’s empowerment of workers. Workers’ infighting, reflecting neighborhood demographics and ideological differences, benefitted TCC; it convinced locals TCC could best protect industrial peace. The submissive AFL union installed fit of ownership’s nationally recognized program for industrial relations reliant on federal power. After competition crippled local industry, locals continued their reliance on government: to investigate the medical consequences of extraction work and coordinate environmental restoration. Recent regional anti-government populism makes the basin’s peculiar historic reliance on federal help engaging.
