Τα έντομα και άλλοι ποικιλόθερμοι οργανισμοί έχουν αναπτύξει διάφορους μοριακούς
μηχανισμούς για την προστασία τους από ακραίες θερμοκρασίες και άλλες στρεσογόνες
καταστάσεις που συμβαίνουν κατά τη διάρκεια της ζωής τους. Ένας από τους σημαντικότερους
μηχανισμούς είναι η παραγωγή των θερμοεπαγόμενων πρωτεϊνών οι οποίες, ως μοριακοί συνοδοί,
προστατεύουν τα κύτταρα από τη μετουσίωση και συσσωμάτωση των πρωτεϊνών τους. Πολλές από
τις θερμοεπαγόμενες πρωτεΐνες συνθέτονται και σε φυσιολογικές συνθήκες και παίζουν
σημαντικούς ρόλους στην ορθή αναδίπλωση, μεταφορά και αποικοδόμηση των πρωτεϊνών του
κυττάρου.
Το εργαστήριο μας ενδιαφέρεται για το ρόλο των θερμοεπαγόμενων πρωτεϊνών και τη ρύθμιση
των γονιδίων τους στη Μεσογειακή μύγα. Τέσσερα θερμοεπαγόμενα γονίδια, hsp90, hsp70, hsp27
και hsp26, έχουν κλωνοποιηθεί και χαρακτηριστεί στο εργαστήριό μας. Στόχοι αυτής της
μεταπτυχιακής εργασίας είναι α) η λειτουργική ανάλυση της 5’ ανοδικής περιοχής του γονιδίου
hsp90 της Μεσογειακής μύγας και β) η έκφραση του γονιδίου σε συνθήκες ψυχρού στρες.
Για να μελετήσουμε την ρύθμιση του γονιδίου hsp90, δημιουργήσαμε διαγονιδιακά στελέχη που
έφεραν το γονίδιο της λουσιφεράσης (ως γονίδιο αναφοράς) υπό τον έλεγχο διαφορετικών 5’
περιοχών του γονιδίου hsp90. Η ανάλυση των στελεχών αυτών έδειξε ότι η ευρύτερη περιοχή του
υποκινητή που εκτίνεται έως και το -3536 περιλαμβάνει όλα τα απαραίτητα στοιχεία για τη
θερμοεπαγόμενη έκφραση του γονιδίου.
Για να μελετήσουμε εάν η χαμηλή θερμοκρασία (0 oC) έχει κάποια επίδραση στη βιωσιμότητα
του εντόμου και στην έκφραση του hsp90, ομάδες αρσενικών ενήλικων εντόμων επωάστηκαν για
διαφόρους χρόνους στους 0 oC και ακολούθως μελετήθηκε η επιβίωσή τους και η έκφραση του
θερμοεπαγόμενου γονιδίου με RT-PCR και qRT-PCR. Τα αποτελέσματά μας υποδηλώνουν ότι οι
χαμηλές θερμοκρασίες αυξάνουν την έκφραση του hsp90 σχεδόν στα ίδια επίπεδα με εκείνα του
θερμικού στρες. Επίσης η βιωσιμότητα των εντόμων βρέθηκε πολύ υψηλή στους 0 oC. / Insects and other poikilotherms have multiple molecular mechanisms for their protection from
extreme environmental and other stressful conditions which happens during their life. One of these
mechanisms is the production of heat shock proteins. These proteins, being molecular chaperones,
protect the cells from protein denaturation and aggregation. Some of these proteins are produced
under normal conditions, and they play important role in proper protein folding, transport and
degradation.
We are interested in the role of heat shock proteins and the regulation of their genes in
Mediterranean fruit fly (medfly). Four heat shock genes have been cloned and characterized in our
laboratory. These are hsp90, hsp70, hsp27 and hsp26. The aim of this master thesis is: a) to study the
normal and heat-induced regulation of the medfly’s hsp90 gene and b) to study the expression of this
gene under cold shock conditions.
For the first aim we created transgenic flies which they had the lusiferase gene (as a reference
gene) under the control of different 5’ upstream regions of hsp90 gene. The analysis of these
transgenic strains showed that the region between the first transcription base pair and -3536 base pair
is crucial for the expression of the gene under normal and heat shock conditions.
In order to examine whether the low temperature (0 oC) has any effect in the medfly’s viability
and the expression of hsp90 gene, male groups were exposed at different intervals at 0 oC and then it
was examined the viability and the expression of the gene. Our results suggest that the low
temperature increases the expression of hsp90 nearly to the same levels of the thermal stress and this
temperature does not affect the viability of the insect.

Στην παρούσα διπλωματική υλοποιήθηκε η σχεδίαση και υλοποίηση ασφαλούς υπηρεσίας με χρήση ελλειπτικής κρυπτογραφίας. Τα κρυπτογραφικά συστήματα που βασίζονται στις ελλειπτικές καμπύλες αποτελούν ένα πολύ σημαντικό κομμάτι της κρυπτογραφίας δημόσιου κλειδιού και τα τελευταία χρόνια όλο και περισσότεροι επιστήμονες ασχολούνται με τη μελέτη τους. Το πλεονέκτημα των συστημάτων αυτών σε σχέση με τα συμβατικά κρυπτογραφικά συστήματα είναι ότι χρησιμοποιούν μικρότερες παραμέτρους και κλειδιά, προσφέροντας τα ίδια επίπεδα ασφάλειας. Σχετικά με το πρόβλημα της προστασίας ευαίσθητων δεδομένων σκληρού δίσκου ή άλλου αποθηκευτικού μέσου διευθυνσιοδοτούμενου κατά τομείς (sector-adressed storage media), χρησιμοποιείται η τεχνική της κρυπτογράφησης δίσκου (disk encryption). Ορισμένα από τα υπεύθυνα για την υλοποίηση της κρυπτογράφησης λογισμικά (disk encryption software) χρησιμοποιούν την μέθοδο κρυπτογράφησης σε πραγματικό χρόνο (on-the-fly/real-time encryption). Ο όρος on-the-fly έγκειται στο γεγονός ότι τα αρχεία γίνονται προσβάσιμα αμέσως μόλις προσφερθεί το κλειδί κρυπτογράφησης (encryption key) όλο το διαμέρισμα (volume) «προσαρτάται» (mounted) σαν να ήταν ένας φυσικός δίσκος κάνοντας τα αρχεία να «φαίνονται» αποκρυπτογραφημένα.
Στην πλαίσιο αυτό τοποθετείται ο σκοπός του ερευνητικού μέρους της παρούσας εργασίας, που εντοπίζεται το ερώτημα της προσαρμογής βιβλιοθηκών που υλοποιούν κρυπτογραφία ελλειπτικών καμπυλών σε λογισμικό ικανό να κρυπτογραφεί «on the fly» φακέλους αρχείων και κατ΄ επέκταση σκληρούς δίσκους / -

Applying Mine Tailing and Fly Ash as Construction Materials for a Sustainable DevelopmentFeng, Qingming January 2015 (has links)
Geopolymerization has been considered as a new technology to replace the ordinary Portland cement in construction industry. It provides an option to manage the industry waste and byproducts like fly ash, mine tailings. At the same time, the CO₂ emissions can be reduced about 80% compared to that of ordinary Portland cement. The present research includes three main parts. First part is applying mine tailings as construction materials using geopolymerization method. The study is focused on efficiently activating mine tailings, reducing alkali consumption, decreasing curing time and improving compressive strength. We investigate the activation temperature effects, the impacts of additives and effects of forming pressures. The results show that a 40 MPa unconfined compressive strength (UCS) can be achieved with the geopolymerization samples after mine tailings are activated by sodium hydroxide at 170°C for 1 hour with the addition of calcium hydroxide and alkali dissolved aluminium oxide, further compressed with a 10 MPa forming pressure and finally cured at 90°C for 3 days. To elucidate the mechanism for the contribution of additives to geopolymerization, microscopic and spectroscopic techniques including scanning electron microscopy/ energy-dispersive X-ray spectroscopy (SEM/EDX), X-ray diffraction (XRD), and Fourier transform infrared (FTIR) spectroscopy are used to investigate the micro/nanostructure and the elemental and phase composition of geopolymerization specimens. The stress-strain behavior was also characterized. The results shows that the mechanical behavior is similar with that of concrete and the dynamic modulus is 22 GPa, which is comparable with that of concrete. The Young's modulus of geopolymer product was also calculated and the value is in the range of 2.9 to 9.3 GPa. The findings of the present work provide a novel method for the geopolymerization of mine tailings as construction materials. Second section is applying fly ash as a high strength water-resistant construction material. Through the present investigation, a procedure has been studied. The experiment results indicate that the concentration of NaOH, water content, and curing condition can significantly affect the mechanical property of geopolymer matrix. At the same time, the chemical composition, especially the Si/Al ratio and calcium content, is also an important factor during geopolymerization. XRD results show that the amorphous feature can be observed for both high and low calcium fly ash. It is the key of the success of geopolymerizaton due to its high reactivity. XRD, FTIR and SEM tests were performed to study how experiment conditions and the properties of fly ash affect geopolymerization. The obtained compressive strength of the geopolymerization product can reach above 100 MPa. The stress-strain behavior was also characterized. The results shows that the dynamic modulus is 36.5 GPa. The product obtained from the present work shows very high water resistance without losing any compressive strength even after a one month soaking time. Third part is applying the mixture of class C fly ash and mine tailings as construction materials. Through the present investigation, a protocol has been set up. The experiment results of the present work also help set up the working conditions such as activation temperature and time, the concentration of NaOH, the addition of Ca(OH)₂, forming pressure, mine tailing to class C fly ash weight ratio, curing temperature and curing time. To elucidate the mechanism for the contribution of additives to geopolymerization, microscopic and spectroscopic techniques such as SEM/EDX, X-ray diffraction and FTIR spectroscopy were used to investigate the micro/nanostructure and the elemental and phase composition of geopolymerization composite. The obtained compressive strength of the geopolymerization product can reach above 60 MPa. The stress-strain behavior of the geopolymer matrix of the mixture of mine tailing and fly ash were also characterized and the results show that the mechanical behavior is similar to that of concrete with a 24 GPa dynamic modulus. The Young's modulus of geopolymer product was also calculated and the value is in the range of 4.0 to 13.5 GPa. The findings of the present work provide a novel method for the geopolymerization of the mixture of mine tailings and class C fly ash as construction materials, such as bricks for construction and road pavement.
