Während die güteorientierte Bewirtschaftung von Stauräumen in der Vergangenheit hauptsächlich bei der langfristigen Bewirtschaftung eine Rolle spielte, gewinnt sie in jüngerer Zeit immer mehr auch bei der kurzfristigen Abflusssteuerung als Vorentlastung und während der Hochwasserereignisse an Bedeutung. Dabei soll vor allem die Entlastung aus Horizonten mit geringerer Wasserqualität ermöglicht werden. Der vorliegende Beitrag beschäftigt sich mit den diesbezüglichen Möglichkeiten bei Nutzung vorhandener Entnahmeeinrichtungen und den Anforderungen an bauliche Ergänzungen. Es werden Grundsätze für die Planung, die Berechnung, den Betrieb und die Überwachung aus hydraulischer Sicht besprochen.

Der vorliegende Bericht stellt Ergebnisse des zweijährigen Projektes 'Veränderte Pflanzenschutzmittel- und Nährstoffausträge bei bodenschonender Bewirtschaftung - Risikoanalyse und -vorsorge' vor. Auf landwirtschaftlichen Flächen in der Lommatzscher Pflege wurden mit einer Beregnungsanlage Starkniederschläge (worst-case-Szenario) simuliert. Die Untersuchungen zu Wasser- und Agrochemikalientransport fanden auf Flächen statt, die konventionell mit Pflug, konservierend flach bearbeitet bzw. in Direktsaat bestellt wurden. Die Auswirkungen auf vertikale und horizontale Stofftransporte, insbesondere von Nitrat-Stickstoff und dem Totalherbizid Durano mit dem Wirkstoff Glyphosat wurden untersucht.

Symptomatic Repetitive Right Ventricular Outflow Tract Ventricular Tachycardia in Pregnancy and Postpartum

Idiopathic ventricular tachycardias, which occur in patients without structural heart disease, are a common entity, representing up to 10% of all ventricular tachycardias evaluated by cardiac electrophysiology services. Pregnancy can increase the incidence of various cardiac arrhythmias. Factors that can potentially promote arrhythmias in pregnancy include the effects of hormones, changes in autonomic tone, hemodynamic perturbations, hypokalemia, and underlying heart disease. Ventricular arrhythmias in pregnancy are repetitive monomorphic ventricular premature complexes and couplets that frequently originate at the right ventricular outflow tract. New onset symptomatic repetitive right ventricular outflow tract ventricular tachycardia during pregnancy has been inadequately reported in the literature. We present a case of symptomatic repetitive right ventricular outflow tract tachycardia that started during pregnancy and continued in the postpartum period, requiring curative treatment with electrophysiology study and radiofrequency ablation.

Monte Carlo Simulations of Polarimetric and Light Variability From Corotating Interaction Regions in Hot Stellar Winds

We use a 3D Monte Carlo radiative transfer code to study the polarimetric and photometric variability from stationary corotating interaction regions (CIR) in the wind of massive stars. Our CIRs are approximated by Archimedean spirals of higher (or lower) density formed in a spherical wind originating from the star and we also made allowance for a bright Gaussian spot at the base of the CIR. Comparing results from our code to previous analytical calculations in the optically thin case, we find differences which we attribute mainly to a better estimation of the total unpolarized flux reaching the observer. In the optically thick case, the differences with the analytical calculations are much larger, as multiple scattering introduces additional complexities including occultation effects. The addition of a Gaussian spot does not alter the shape of the polarization curve significantly but does create a small excess in polarization. On the other hand, the effect can be larger on the light curve and can become dominant over the resulting CIR, depending on the spot parameters and density of the wind.

Structural and functional investigation of the trabecular outflow pathway

Primary open-angle glaucoma (POAG) is a leading cause of blindness in the world. A primary risk factor for POAG is elevated intraocular pressure (IOP), caused by increased aqueous humor outflow resistance. Currently, lowering the IOP is the only effective way of treating glaucoma; however, the cause of increased outflow resistance remains unclear. This thesis will present a series of studies which investigated structures of the trabecular outflow pathway, including Schlemm’s canal endothelium, juxtacanalicular tissue, and trabecular beams, and their roles in regulating aqueous outflow resistance. The studies were conducted in both human and animal models using ex vivo ocular perfusion as well as in vitro microfluidic systems. In the first study, we investigated the effects of Y27632, a derivative of Rho-kinase inhibitor that is being developed as next generation glaucoma drug with unclear IOP lowering mechanism, on aqueous humor outflow dynamics and associated morphological changes in normal human eyes and laser-induced ocular hypertensive monkey eyes. In the second study, we developed and validated a novel three-dimensional microfluidic system using lymphatic microvascular endothelial cells. The microfluidic system can be used to study Schlemm’s canal endothelial cell dynamics and aqueous humor transport mechanism in the future. In the last study, we characterized the morphological structure, distribution, and thickness of the endothelial glycocalyx in the aqueous humor outflow pathway of human and bovine eyes. Together these studies will help define new directions for therapy that will help control IOP and preserve vision throughout a normal life span.

The Role of MCTP2 in Health and Disease

Indiana University-Purdue University Indianapolis (IUPUI) / MCTP2 (multiple C2 domain transmembrane containing protein 2) encodes a protein with poorly understood roles in lipid metabolism and lipid droplet biogenesis. Genetic studies previously identified variations in MCTP2 in conjunction with left ventricular outflow tract obstructive forms of congenital heart disease (CHD). This dissertation research aimed to delineate the biomedical significance of Mctp2 by investigating its expression and consequences of its genetic deletion in mouse models. Temporal and spatial expression of Mctp2 was investigated by RT-PCR and in-situ hybridization. A novel isoform, designated as isoform 2 in mice, results from alternative pre-mRNA splicing. Similar levels of Mctp2 isoforms 1 and 2 are present in embryonic tissues, whereas isoform 1 is preferentially expressed in adult tissues with high lipid metabolism. During mouse embryonic development, in-situ hybridization suggests expression of Mctp2 at the gut tube, liver bud and near the pharyngeal arches from E8.5 – E10.5. Given association of MCTP2 with CHD, the biological significance of Mctp2 was addressed using gene trap (GT) and conditional mouse models. Survival of Mctp2 GT mice was dependent on the genetic background strain, suggesting a role for strain-specific modifiers. Conditional knockout of Mctp2 in cardiac progenitor cells displayed no effect on survival. The role of Mctp2 in cardiac development remains to be delineated. The role of Mctp2 in cardiac function was addressed in both mouse models. Initial findings suggest Mctp2 allele dosage effects on the development of heart failure. GT mice lacking one, or both, copies of Mctp2 display cardiac systolic dysfunction, with upregulation of heart failure markers at 50 weeks of age in heterozygotes and increases in cardiac fibrosis in homozygotes. Systemic conditional deletion of Mctp2 did not show heart failure phenotypes using the strain protective from lethality. However, cardiac specific deletion of Mctp2 using the Nkx2.5-Cre driver, a line that is sensitized for cardiac dysfunction, led to decreased ejection fraction and fractional shortening in mice with conditional deletion of both copies of Mctp2 as well as Mctp2 dosage dependent penetrance of cardiac dilation. These studies of knockout mice suggest a role for Mctp2 in maintenance of cardiac function and possible genetic interaction with Nkx2.5. / 2022-02-02

Stanovení tenzidů v odpadních vodách / Determination of surfactants in waste waters

Surfactants belong to surface-active compounds that have ability to restrain the surface tension; this ability is exploited to eliminate impurities. This study is focused on determination of surfactants in waste water to which these compounds can be transported from various cleaning and washing articles. In theoretical part are listed basic classifications of surfactants, their properties and requests of Czech legislative for their content in waste water. Furthermore here are described chosen analytical procedures for determination of anion-active, cation-active and non-ionic surfactants in waste water. The conclusion of the work is evaluation of measured results of surfactants concentration in inflow and outflow of waste water from waste-treatment plant in University of Veterinary and Pharmaceutical Sciences Brno and waste water from neutralizating station in FCH BUT. To determination of anionic surfactants were used two methods, arbitration method with usage of methylene blue and mobile analytics method which is based on chemical reaction between target compound and chemical agent. This reaction leads to formation of coloured compound able to spectrofotometric determination.
