Bebida láctea fermentada com cultura probiótica adicionada de semente de chia (Salvia hispanica L.)

O objetivo deste trabalho foi desenvolver uma bebida láctea fermentada preparada com 60% de leite, 32% de soro lácteo e 8% de açúcar, utilizando micro-organismos probióticos. Foram desenvolvidas 3 formulações, com 1% (A), 3% (B) e 5% de semente de chia (Salvia hispanica L.). Foram realizadas análises físico-químicas na formulação padrão e nas formulações A, B e C durante 0, 7 e 14 dias; análise sensorial nas formulações A, B, e C nos tempos 0, 7 e 14 dias e avaliação microbiológica. O teor de umidade nas 3 formulações apresentou diferença significativa aos 0 e 7 dias de estocagem (p < 0,05). Os teores de cinzas, proteínas, lipídeos e a viscosidade da bebida aumentaram significativamente à medida que se aumentou a proporção de chia na bebida. Foi observada uma redução gradativa do pH e aumento da acidez titulável em função do tempo de estocagem da bebida. A bebida láctea obteve boa aceitação sensorial, mas foi observada uma diminuição nas notas da escala hedônica e intenção de consumo durante o período de estocagem. A contagem de bactérias probióticas teve redução de aproximadamente 1 ciclo logarítmico por semana. A formulação preferida no teste de ordenação de preferência foi a formulação A. / The aim of this study was to develop a fermented dairy beverage made with 60% milk, 32% whey and 8% sugar, using probiotic microorganisms. Three formulations, with 1% (A), 3% (B) and 5% chia seed (Salvia hispanica L.) have been developed. Physico-chemical analyzes on standard formulation and the formulations A, B and C for 0, 7 and 14 days were made; sensory analysis in formulations A, B, and C for times 0, 7 and 14 days and microbiological evaluation The moisture content of the three formulations showed a significant difference at 0 and 7 days of storage (p <0.05). Contents of ash, protein, lipids and viscosity increased significantly as it increased the proportion of chia. A gradual reduction of pH and increase in acidity due to the storage time of the beverage was observed. The product has obtained good acceptability, but a decrease was observed in the global acceptability and intention of consumption during the storage period. The probiotic bacteria count was reduced by about 1 log cycle per week. The preferred formulation of the ranking preference test was the formulation A.

O objetivo desse estudo foi avaliar o efeito do Lactobacillus reuteri aplicado localmente ou sistemicamente como coadjuvante ao tratamento periodontal não cirúrgico de raspagem e alisamento radicular (RAR), na periodontite experimentalmente induzida em animais submetidos à quimioterapia com 5-fluorouracil (5-FU). A indução da periodontite experimental (PE) foi realizada com a colocação de um fio de algodão ao redor dos molares inferiores esquerdos de 108 ratos, que permaneceu por 7 dias. Todos os animais receberam o quimioterápico no momento da indução da PE (60mg/kg) e 48 horas após (40mg/kg) e foram separados aleatoriamente em 6 grupos com 18 animais cada um, com os seguintes tratamentos: 5FU: animais tratados com 5-FU; 5FU/RAR: animais tratados com 5-FU, que receberam tratamento com RAR seguido de 1 aplicação de solução fisiológica salina (SS; 0,16mL); Grupo 5FU/SSL: animais tratados com 5-FU, que receberam tratamento com RAR e 4 aplicações locais de SS (0,16mL); 5FU/SSS: animais tratados com 5-FU, que receberam tratamento com RAR e 4 gavagens com SS (0,16mL); 5FU/PL: animais tratados com 5-FU, que receberam tratamento com RAR e 4 aplicações locais de PRO (0,16 mL); 5FU/PS: animais tratados com 5-FU, que receberam tratamento com RAR e 4 gavagens com PRO (0,16mL). 6 animais de cada grupo foram submetidos à eutanásia aos 7, 15 e 30 dias após os tratamentos. A área da furca dos molares foi submetida às análises histopatológica, histométrica e dos padrões de imunomarcação para TRAP, PCNA, RANKL, OPG, OCN e TGF-β1. Os dados obtidos foram submetidos à análise estatística (α=5%). O grupo 5FU/PS apresentou melhora no processo inflamatório em todos os períodos. Houve maior perda óssea alveolar (PO) nos espécimes do grupo 5FU/SSS comparado ao grupo 5FU/SSL (p˂0,01) no período de 7 dias de avaliação. No período de 15 dias os espécimes dos grupos 5FU/SSL e 5FU/SSS demonstraram maior PO comparado aos grupos 5FU (p˂0,05) e 5FU/PS (p˂0,05). Aos 30 dias a PO mostrou-se maior nos espécimes do grupo 5FU/PL comparado aos grupos 5FU/RAR (p˂0,05), 5FU/SSS (p˂0,05) e 5FU/PS (p˂0,05). Na análise intragrupo houve menor PO no grupo 5FU/RAR e 5FU/PS aos 30 dias em relação aos 7 dias (p˂0,05). Na imunomarcação de células TRAP-positivas não foi evidenciada diferença significante entre os grupos e períodos; houve maior imunomarcação de células PCNA-positivas aos 7 dias no grupo 5FU/RAR comparado ao 5FU/SSS. Prevaleceu baixo padrão de imunomarcação de TGF-β1 e OCN nos grupos 5FU, 5FU/RAR, 5FU/SSL e 5FU/SSS em todos os períodos. O grupo 5FU/PS apresentou aos 7 dias um moderado padrão de imunomarcação e aos 15 e 30 dias um alto padrão de imunomarcação de TGF-β1 e OCN; o grupo 5FU/PL apresentou um moderado padrão de imunomarcação de TGF-β1 e OCN aos 7,15 e 30 dias. Na análise de RANKL prevaleceu um alto padrão de imunomarcação nos grupos 5FU, 5FU/RAR, 5FU/SSL, 5FU/SSS em todos os períodos e o grupo 5FU/PS apresentou aos 7 e 15 dias um moderado padrão de imunomarcação e aos 30 dias predominou um padrão de imunomarcação que variou do moderado ao baixo. Prevaleceu um padrão baixo de imunomarcação em todos os grupos experimentais e períodos, com exceção dos grupos 5FU/PL e 5FU/PS aos 30 dias (moderado padrão de imunomarcação) na análise de OPG. Diante dos resultados obtidos e com a metodologia empregada pode ser concluído que o uso sistêmico do Lactobacillus reuteri promoveu redução da inflamação e beneficiou o processo de reparação dos tecidos periodontais, porém não foi capaz de reduzir a PO na região de furca, demonstrando ser uma terapia periodontal coadjuvante promissora em animais submetidos à quimioterapia com 5-FU. / Probiotics (PROs) have been intensively evaluated to prevent or treat oral cavity diseases, associated with a change in the composition and microbial activity of the biofilm and response of the host. The aim of this study was to evaluate the effect of Lactobacillus reuteri applied locally or systemically as a coadjuvant to the non-surgical periodontal treatment of scaling and root planing (SRP) in experimentally induced periodontitis in animals treated with 5-fluorouracil (5-FU). Induction of experimental periodontitis (EP) was performed by placing a cotton thread around the left lower molars of 108 rats, which were kept there for 7 days. All animals underwent chemotherapy twice. They were 60mg/kg at the time they were subjected to ligature induction and again another 40 mg/kg, 48 hours later. They were randomly divided into 6 groups with 18 animals each and they received the following treatments: 5FU: treated animals with 5-FU; 5FU/SRP: 5-FU treated animals receiving SRP treatment followed by physiological saline (SS) solution; 5FU/SSL group: animals treated with 5-FU, who received SRP treatment and local SS applications; 5FU/SSS: animals treated with 5-FU, who received SRP treatment and systemic treatment with SS; 5FU/PL: animals treated with 5-FU, who received SRP treatment and local applications of PRO; 5FU/PS: animals treated with 5-FU, who received SRP treatment and systemic treatment with PRO (0.16ml x 4 days). Six animals from each group were submitted to euthanasia at 7, 15 and 30 days after treatments. The area of the molar furcation was submitted to histopathological, histometric and immuno-labeling analysis for TRAP, PCNA, RANKL, OPG, OCN and TGF-β1. The data were submitted to statistical analysis (α = 5%). There was greater bone loss (BL) in the 5FU/SSS group compared to the 5FU/SSL group (p˂0.01) in the 7-day evaluation period. In the 15-day period, specimens from the 5FU/SSL and 5FU/SSS groups showed a higher BL compared to the 5FU (p˂0.05) and 5FU/PS (p˂0.05) groups. At 30 days the BL was higher in the 5FU/PL group compared to the 5FU/SRP (p˂0.05), 5FU/SSS (p˂0.05) and 5FU/PS groups (p˂0.05). In the intragroup analysis there was a lower BL in the 5FU/SRP group and 5FU/PS at 30 days compared to 7 days (p˂0.05).In the immunostaining of TRAP-positive cells no significant difference between groups and periods was evidenced; there was greater immunolabeling of PCNA-positive cells at 7 days in the 5FU/SRP group compared to 5FU/SSS. Low prevalence of TGF-β1 and OCN immunostaining in the 5FU, 5FU/RAR, 5FU/SSL and 5FU/SSS groups prevailed at all periods. The 5FU/PS group presented at 7 days a moderate pattern of immunostaining and at 15 and 30 days a high standard of immunoblotting of TGF-β1 and OCN; the 5FU/PL group presented a moderate pattern of TGF-β1 and OCN immunoregulation at 7, 15 and 30 days; for RANKL a high standard of immunostaining in the 5FU, 5FU/SRP, 5FU/SSL, 5FU/SSS groups prevailed at all periods, 5FU/PL prevailed at 7 days a high standard of immunostaining and a moderate immunostaining pattern at 15 and 30 days and 5FU/PS showed a moderate immunostaining pattern at 7 and 15 days, and at 30 days a predominance of the immunoblot pattern varied from moderate to low; for OPG a low pattern of immunoblotting prevailed in all experimental groups and periods, except for 5FU/PL and 5FU/PS at 30 days that presented a moderate pattern of immunostaining. Based on the results obtained in this study and the methodology used to work on it, it was concluded that the use of Lactobacillus reuteri promoted reduction of inflamation and benefited the process of repairing periodontal tissues, but it was not able to reduce the BL in the furca region, proving to be a therapy promising adjuvant periodontal in animals submitted to 5-FU chemotherapy. / Capes

Embora sua origem ainda não seja completamente conhecida, existe uma crescente evidência baseada em estudos recentes associando predisposição genética, composição da microbiota intestinal e sistema imunológico como fatores precursores para a iniciação e progressão dessas patologias. O uso de determinadas cepas probióticas vem sendo apontado como uma forma promissora de se reduzir o risco de doenças inflamatórias intestinais, mais precisamente da colite ulcerativa. Diversos mecanismos têm sido propostos para explicar os benefícios dos probióticos, indicando que algumas cepas bacterianas são capazes de modular positivamente a microbiota intestinal e o sistema imune, além de produzirem metabólitos com capacidade antiinflamatória. Como os efeitos e os mecanismos de ação envolvidos são cepa-específicos, é de extrema importância a realização de novos estudos para compreender as interações entre microbiota intestinal e hospedeiro, contribuindo para o entendimento do potencial terapêutico dos probióticos nas doenças relacionadas ao desequilíbrio da microbiota intestinal. O presente trabalho se propõe a investigar o efeito da ingestão diária de um produto à base de extrato aquoso de soja, fermentado com Enterococcus faecium CRL 183 e Lactobacillus helveticus 416 e com adição de Bifidobacterium longum ATCC 15707, na colite induzida quimicamente em ratos. A colite foi induzida, pela administração de dextran sulfato de sódio (DSS) a 4%, que foi dissolvido na água fornecida diariamente aos animais e administrado durante um período de sete dias. Os animais foram distribuídos aleatoriamente em cinco grupos (n=10): Grupo C: Animais sadios que não receberam os produtos em estudo. ... / Abstract: Inflammatory bowel disease (IBD), including Crohn's disease (CD) and ulcerative colitis (UC), comprise a group of disorders and their main characteristic is an inflammation of the intestinal mucosa. Although its origin is not yet fully known, there is growing evidence linking genetics predisposition, composition of the intestinal microbiota and immune system factors as precursors for the initiation and progression of these pathologies. The use of certain probiotic microorganisms has been considered as promising way to reduce the risk of inflammatory bowel disease, specifically ulcerative colitis. Several mechanisms have been proposed to explain the benefits of probiotics, indicating that some bacterial strains are able to positively modulate the intestinal microbiota and the immune system, and produce metabolites with anti-inflammatory capacity. As the effects and the mechanisms involved are strain-specific, further studies are extremely important to understand the interactions between gut microbiota and host, contributing to the knowledge on therapeutic potential of probiotics in diseases related to dysbiosis. This current paper aims to bring together the various results and information based on scientific evidence that are related to probiotics and the inflammatory bowel disease, with emphasis on the possible mechanisms involved in this action. Furthermore, the effects of daily ingestion of a product based on an aqueous extract of soybean, fermented by Enterococcus faecium CRL 183 and Lactobacillus helveticus 416 with the addition of Bifidobacterium longum ATCC 15707, were also investigated on chemically induced colitis in rats. Možnosti využití potravních doplňků v prevenci a péči o zdraví telat / Possibilities of use of food supplements in prevention and health care calves

The basis for a successful cattle production is the proper care taking of pregnant cows as well as a sufficient care taking of the new-born calves. During the last years, a big attention has been given to this area because the higher farming quality the higher economic benefits and the lower losses in meat and milk yield will be respectively. For this reason, an influence of different feed supplements (Lactovita, Biopolym, Homeopathics) on haematological and biochemical parameters as well as an effect on weight gain and amounts of microorganisms in new-born calves has been studied. The specific aim of this thesis is to evaluate an influence of these supplements on incidences of diarrhea, microbial activity and to assess its effect on selected haematological and biochemical blood parameters. Three experimental and one control group (ten calves per each) were set up in the chosen company. The experimental groups had a modified feeding ratio between the first and the fourth week of life the calves received either colostrum + 5ml "PVB" homeopathic or colostrum + 5 ml "Biopolym" prebiotics or colostrum + 1 pill of "Lactovita", while the control group received colostrum without supplement. The first day after birth, a blood sample and a microbial smear sample were taken. Second sample collection was carried out in the fourth week of life. The calves were weighed every week. Obtained data were processed graphically and in tables and were statistically evaluated. The influence of mentioned feed additives on weight gain and diarrhea occurrence was evaluated from the obtained results. The most of haematological and biochemical parameters have approximately the same values compared to those found by other authors. There were no significant differences in monitored parameters in the experimental groups compared to the control group. Furthermore, no influence was found on blood parameters. Only in case of zinc a significant difference was found between treated groups and control (p=0,0012). Changes between the treated groups (independently on control) were observed in haematocrit, total protein content, cholesterol, zinc and copper. Non-significant differences between the tested groups and control were found in haemoglobin, erythrocytes, leukocytes, glycaemia, urea, LF, GMT, phosphorus, calcium and magnesium. No impact of tested supplements was found on microbial composition. A positive effect of Lactovita, Biopolym and Homeopathic compared to the control group was found in case of weight gain. The obtained results show some positive trends which should be a subject for further research, where total amount of calves per groups is increased to reduce variability.

Možnosti využití doplňkových a nekonvenčních postupů v prevenci a péči o zdraví telat / Possibilities of the application of supplemental and nonconventional methods in prevention and health care in calves

Diarrheal diseases of calves are one of the most serious risks of rearing calves and cause economic losses. For this reason, the objective of this work was to assess the influence of means of support in operating conditions for the incidence of diarrheal disease in calves in the early post-weaning stages. A company was chosen for this work in which both the company employees and an external veterinarian were willing to collaborate and provide not only information about the breed in question, but also biological material. This company was JINOS - AGRO Veselí nad Lužnicí. The calves were observed in the period of April - June 2009. The calves were housed in separate boxes the second day after birth until the average age of 56 days. After storage, they were divided into 5 groups: 4 based on the chosen means of support, and a single control group. Faeces samples were collected from the calves and subsequently evaluated. The calves to whom were administered means of support were then evaluated to assess the effect thereof on the incidence of diarrhea, the duration of diarrhea, the method of treatment, and the duration of treatment. The results were summarized and analyzed using a statistical program (STATISTICA 7.0.). There was no statistically significant difference found between the individual groups and the experimental group. An evident and statistically significant difference was found only in the case of a difference between the drugs of the 2 experimental groups, specifically between Lactovita and Homeopatika. Based on this observation, it can be stated that there was no evidence supporting the effectiveness of administered means of support for the incidence of diarrheal disease in calves in the early post-weaning stages.

Healthy and well fed calves are the main prerequisite for high performances. The aim is to evaluate the concept of nutrition of calves and assess the dynamics of selected haematological and biochemical parameters of blood in the sample of agricultural holdings, depending on the influence of selected dietary supplements. The experiment was conducted in the agricultural cooperative Krásna hora nad Vltavou a.s. on the farm Perovice, where they breed Holstein cattle. There were three groups of calves - one control and two experimental. The test groups received dietary supplements to promote active immunity by the instruction. In the first group Lactovita dietary supplements, the latter group food supplement called Biopolym. The first blood sampling was performed from ages 5 to 7 days and a second collection has been made 3 weeks later. Findings and data analysis were summarized and evaluated. The experiment was conducted from August to November 2013.

O presente trabalho se propõe a investigar o efeito da ingestão diária de um produto à base de extrato aquoso de soja, fermentado com Enterococcus faecium CRL 183 e Lactobacillus helveticus 416 e com adição de Bifidobacterium longum ATCC 15707, na colite induzida quimicamente em ratos. A colite foi induzida, pela administração de dextran sulfato de sódio (DSS) a 4%, que foi dissolvido na água fornecida diariamente aos animais e administrado durante um período de sete dias. Os animais foram distribuídos aleatoriamente em cinco grupos (n=10): Grupo C: Animais sadios que não receberam os produtos em estudo. ... / Inflammatory bowel disease (IBD), including Crohn's disease (CD) and ulcerative colitis (UC), comprise a group of disorders and their main characteristic is an inflammation of the intestinal mucosa. Although its origin is not yet fully known, there is growing evidence linking genetics predisposition, composition of the intestinal microbiota and immune system factors as precursors for the initiation and progression of these pathologies. The use of certain probiotic microorganisms has been considered as promising way to reduce the risk of inflammatory bowel disease, specifically ulcerative colitis. Several mechanisms have been proposed to explain the benefits of probiotics, indicating that some bacterial strains are able to positively modulate the intestinal microbiota and the immune system, and produce metabolites with anti-inflammatory capacity. As the effects and the mechanisms involved are strain-specific, further studies are extremely important to understand the interactions between gut microbiota and host, contributing to the knowledge on therapeutic potential of probiotics in diseases related to dysbiosis. This current paper aims to bring together the various results and information based on scientific evidence that are related to probiotics and the inflammatory bowel disease, with emphasis on the possible mechanisms involved in this action. Furthermore, the effects of daily ingestion of a product based on an aqueous extract of soybean, fermented by Enterococcus faecium CRL 183 and Lactobacillus helveticus 416 with the addition of Bifidobacterium longum ATCC 15707, were also investigated on chemically induced colitis in rats. Por isso, o setor de alimentos investe cada vez mais no desenvolvimento de alimentos funcionais e entre esses, se destacam as bactérias probióticas e ingredientes prebióticos. O objetivo desta pesquisa foi avaliar os efeitos do consumo de probiótico (Lactobacillus acidophilus CRL 1014) e prebiótico (extrato de yacon) sobre a microbiota intestinal de animais induzidos quimicamente ao câncer de cólon. Foram utilizados 175 ratos machos Wistar divididos aleatoriamente em cinco tratamentos: Tratamento I-GCN: animais controle negativo (animais sadios que consumiram apenas a ração comercial); Tratamento II-GCP: animais controle positivo (induzidos quimicamente ao câncer de cólon e que consumiram apenas a ração comercial); Tratamento III-GPRO: (induzidos quimicamente ao câncer de cólon e que consumiram ração comercial + L. acidophilus CRL 1014); Tratamento IV-GPRE: (induzidos quimicamente ao câncer de cólon e que consumiram ração comercial + yacon); Tratamento V-GS: (induzidos quimicamente ao câncer de cólon e que consumiram ração comercial + L. acidophilus CRL 1014+ yacon). Os animais induzidos foram inoculados semanalmente com 1,2 dimetilhidrazina durante 2 semanas, sendo 2 induções semanais na concentração de 40mg/kg de peso. O L. acidophilus CRL 1014, na concentração de 109UFC/mL foi administrado aos animais na quantidade de 3 ml/kg de peso/dia, já o extrato aquoso de yacon foi administrado na quantidade de 10% de FOS/ração. Therefore, the food industry invests in the development of functional foods, such as probiotic bacteria and prebiotic ingredients. This research was evaluate the effects of consumption of probiotic (Lactobacillus acidophilus CRL 1014) and prebiotic (yacon extract) on the gut microbiota on 1.2 dimethylhydrazine (DMH) induced colon carcinogenesis in Wistar rats. The animals were randomly allocated into five groups (n=35): Treatment I-GCN: negative control animals (healthy animals + commercial feed); Treatment II-GCP: positive control animals (DMH induced + commercial feed); Treatment III-GPRO: (DMH induced + commercial feed + L. acidophilus CRL 1014); Treatment IV-GPRE: (DMH induced + commercial feed + yacon); Treatment V-GS: (DMH induced + commercial feed + L. acidophilus CRL 1014+ yacon). The induced animals were inoculated with DMH twice weekly for 2 weeks at a concentration of 40mg/kg weight. The probiotic, L. acidophilus CRL 1014 was administered in 109CFU/mL (3 ml/kg body weight) and the yacon extract (10% FOS/ feed),once daily. Microbiological analysis of Enterococcus spp., Lactobacillus spp., Bifidobacterium spp., Enterobacteriaceae, Clostridium spp., Bacteroides spp., total aerobes and facultative anaerobes were performed by plate count. Analysis of PCR-DGGE for the lactobacilli community, short-chain fatty acids (SCFA) production, ammonium ions and fecal pH were performed. The viable counts showed that the Enterococcus spp., at the end of the experimental protocol, the treated groups showed higher than controls count. The counts of Enterobacteriaceae and Bacteroides spp., were higher to GCP. There was an increase of Lactobacillus spp. for GPRO and Bifidobacterium ssp. for GPRE and GS.
