Internal combustion engine durability monitor : Identifying and analysing engine parameters affecting knock and lambda / Livslängdsövervakning av förbränningsmotor : Identifiering och analys av motorparametrar som påverkar knack och lambda

This study has been performed at Powertrain Engineering Sweden AB (PES), a fully owned subsidiary of Volvo Cars Group, which is constantly working to develop and improve internal combustion engines. As part of this work, durability tests are performed to analyse the impact of wear on the engines. At present, there is a strong focus on visual inspections after the engines have undergone durability tests. PES wants to develop a method where collected data from these tests can be used to explain how the phenomenon of knocking and the control of lambda changes over time. The study analyses one specific durability test and investigates the methodology of data analysis by using the open-source software platform Sympathy for Data, with an add-on developed by Volvo Cars Group, for data management, visualisation and analysis. To execute the analysis, engine parameters that affect these systems as well as parameters suitable to use as response variables are identified through literature studies of internal combustion engine fundamentalsas well as internal material, and knowledge acquired at the company. The result is presented in the form of an analysis generated by the node for partial least squares regression (PLSR) which is pre-programmed in Sympathy for Data as well as the images and graphs obtained as output. For knock, the signal for the final ignition angle was found to be suitable to use as the response variable in the PLSR. A suitable response variable for lambda was more difficult to identify, this is why both signals for the measured lambda and lambda adaptation are analysed. Studies of the internal material and knowledge highlighted the fact that several engine subsystems are highly dependent on each other and that even deeper research would be necessary to fully understand the process and identify the primary cause for the variations observed in the generated models. However, partial least squares regression was performed using parameters derived from literature reviews as input (predictors) in order produce regression models to explain the variance in sought response. Well-fitting models could be created with a varying number of latent variables needed for the different responses. The output obtained from the PLSR enables further studies of the specific cases as well as the methodology itself, hence, increase the use of data analysis with the help of the software used in the department for durability testing at PES. / Denna studie är utförd hos Powertrain Engineering Sweden AB (PES), vilka är ett helägt dotterbolag till Volvo Cars Group, som arbetar med att ta fram och förbättra förbränningsmotorer. En del i detta arbete är att genomföra långtidstest för att analysera hur motorernas egenskaper ändras vid förslitning över tid. I nuläget ligger stort fokus på visuella inspektioner efter att motorerna genomgått långtidstester. PES önskar utveckla en metod där redan insamlad data som registrerats i dessa tester kan förklara hur fenomenet knack och regleringen för lambda förändras över tid. Studien är genomförd i form av en fallstudie av ett specifikt långtidstest där den öppna programvaran Sympathy for Data, tillsammans med det av Volvo Cars Group utvecklade tillägget, används för datahantering, visualisering och analys. Studien undersöker också metodiken för dataanalys med nämnd programvara. För att genomföra detta identifieras motorparametrar som påverkar de undersökta systemen samt parametrar som lämpar sig att användas som responsvariabler i en regressionsmodell. Dessa parametrar togs fram genom litteraturstudier om de fundamentala delarna i en förbränningsmotor samt från företaget förvärvad intern kunskap kring systemen. Resultatet presenteras i form av en analys genomförd med den, i Sympathy for Data, förprogrammerade noden för partial least squares regression(PLSR) samt de bilder och grafer som erhålls. För knack visade det sig att den slutliga tändningsvinkeln var lämplig att använda som respons i PLSR-modellen. En lämplig responsvariabel för lambda var mer svåridentifierad, detta förklarar varför signalerna för uppmätt lambda och lambda adaption analyseras. Inläsning av internt material och grundläggande information om förbränningsmotorer visade att delsystem i ottomotorn är beroende och påverkas av varandra vilket innebär att mer ingående studier i dessa delsystem är nödvändigt för att förstå hela processen och hitta grundorsakerna till variationerna som påvisas för responssignalerna. Vidare utfördes PLSR med de parametrar som härletts från litteraturstudier som indatasignaler (prediktorer) för att skapa en regressionsmodell som förklarar variansen i sökta responssignaler. Beroende av responssignal krävdes varierande antal latenta variabler för att uppnå en tillräckligt precis modell. Resultatet från PLSR möjliggör vidare forskning inom området och metoden som använts och har på så sätt möjliggjort för fortsatt utveckling. Detta i sin tur kan öka användandet av dataanalys med hjälp av den programvara som används vid avdelningen för långtidstest hos PES.

Laser-Induced Breakdown Spectroscopy: Simultaneous Multi-Elemental Analysis and Geological Applications

Under high irradiation, a fourth state of matter named plasma can be obtained. Plasmas emit electromagnetic radiation that can be recorded in the form of spectra for spectroscopic elemental analysis. With the advent of lasers in the 1960s, spectroscopists realized that lasers could be used simultaneously as a source of energy and excitation to create plasmas. The use of a laser to ignite a plasma subsequently led to laser-induced breakdown spectroscopy (LIBS), an optical emission spectroscopy capable of analyzing samples in various states (solids, liquids, gases) with minimal sample preparation, rapid feedback, and endowed with in situ capability. In this dissertation, studies of LIBS for multi-elemental analysis and geological applications are reported. LIBS was applied to cosmetic powders for elemental analysis, screening and classification based on the raw material used. Principal component analysis (PCA) and internal standardization were used. The intensity ratios of Mg/Si and Fe/Si observed in talcum powder show that these two ratios could be used as indicators of the potential presence of asbestos. The feasibility of LIBS for the analysis of gasification slags was investigated and results compared with those of inductively-coupled plasma−optical emission spectrometry (ICP-OES). The limits of detection for Al, Ca, Fe, Si and V were determined. The matrix effect was studied using an internal standard and PLS-R. Apart from V, prediction results were closed to those of ICP-OES with accuracy within 10%. Elemental characterization of outcrop geological samples from the Marcellus Shale Formation was also carried out. The matrix effect was substantially reduced. The limits of detection obtained for Si, Al, Ti, Mg, Ca and C were determined. The relative errors of LIBS measurements are in the range of 1.7 to 12.6%. Gate delay and laser pulse energy, have been investigated in view of quantitative analysis of variation of trace elements in a high-pressure environment. Optimization of these parameters permits obtaining underwater plasma emission of calcium with quantitative results on the order of 30 ppm within a certain limit of increased pressure. Monitoring the variation of the trace elements can predict changes in the chemical composition in carbon sequestration reservoir.

Accuracy and Reproducibility of Laboratory Diffuse Reflectance Measurements with Portable VNIR and MIR Spectrometers for Predictive Soil Organic Carbon Modeling

Soil spectroscopy in the visible-to-near infrared (VNIR) and mid-infrared (MIR) is a cost-effective method to determine the soil organic carbon content (SOC) based on predictive spectral models calibrated to analytical-determined SOC reference data. The degree to which uncertainty in reference data and spectral measurements contributes to the estimated accuracy of VNIR and MIR predictions, however, is rarely addressed and remains unclear, in particular for current handheld MIR spectrometers. We thus evaluated the reproducibility of both the spectral reflectance measurements with portable VNIR and MIR spectrometers and the analytical dry combustion SOC reference method, with the aim to assess how varying spectral inputs and reference values impact the calibration and validation of predictive VNIR and MIR models. Soil reflectance spectra and SOC were measured in triplicate, the latter by different laboratories, for a set of 75 finely ground soil samples covering a wide range of parent materials and SOC contents. Predictive partial least-squares regression (PLSR) models were evaluated in a repeated, nested cross-validation approach with systematically varied spectral inputs and reference data, respectively. We found that SOC predictions from both VNIR and MIR spectra were equally highly reproducible on average and similar to the dry combustion method, but MIR spectra were more robust to calibration sample variation. The contributions of spectral variation (ΔRMSE < 0.4 g·kg−1) and reference SOC uncertainty (ΔRMSE < 0.3 g·kg−1) to spectral modeling errors were small compared to the difference between the VNIR and MIR spectral ranges (ΔRMSE ~1.4 g·kg−1 in favor of MIR). For reference SOC, uncertainty was limited to the case of biased reference data appearing in either the calibration or validation. Given better predictive accuracy, comparable spectral reproducibility and greater robustness against calibration sample selection, the portable MIR spectrometer was considered overall superior to the VNIR instrument for SOC analysis. Our results further indicate that random errors in SOC reference values are effectively compensated for during model calibration, while biased SOC calibration data propagates errors into model predictions. Reference data uncertainty is thus more likely to negatively impact the estimated validation accuracy in soil spectroscopy studies where archived data, e.g., from soil spectral libraries, are used for model building, but it should be negligible otherwise.

Multi-omic data integration study of immune system alterations in the development of minimal hepatic encephalopathy in patients with liver cirrhosis

[ES] El objetivo principal de este trabajo fue conocer las alteraciones inmunológicas asociadas a la inflamación periférica que desencadenan deterioro cognitivo en los pacientes cirróticos encefalopatía hepática mínima (EHM). Estos cambios pueden ser monitorizados como cascadas de señalización a lo largo de los tipos celulares del sistema inmune. Como estudio preliminar, se analizaron los cambios en la expresión génica (transcriptómica), los metabolitos de plasma (metabolómica) y un panel de citoquinas extracelulares en muestras de sangre de pacientes cirróticos con y sin EHM. Los resultados del análisis transcriptómico apoyaron la hipótesis de alternancias en las poblaciones celulares de linfocitos Th1/Th2 y Th17 como principales impulsores de la EHM. El análisis clúster de las moléculas del suero dio como resultado 6 grupos de compuestos químicamente similares. También se ha realizado un análisis de integración multiómica para detectar las relaciones entre los componentes intra y extracelulares que podrían contribuir a la inducción del deterioro cognitivo. Los resultados de este análisis integrativo sugirieron una relación entre las citocinas CCL20, CX3CL1, CXCL13, IL-15, IL-22 e IL-6 con la alteración de la quimiotaxis, así como un vínculo entre los fosfolípidos insaturados de cadena larga y el aumento del transporte de ácidos grasos y la producción de prostaglandinas. Estudios previos sugieren que un cambio en la inflamación periférica, orquestado principalmente por las células T CD4+, es un factor crítico que desencadena el deterioro cognitivo en EHM. La segunda parte de la tesis se centró en la comprensión de las rutas genéticas y los mecanismos por los que las alteraciones en los linfocitos CD4+ pueden contribuir a la inflamación periférica en EHM. Se analizaron los niveles de expresión de genes, factores de transcripción y miARNs en este subtipo de linfocitos mediante secuenciación de alto rendimiento (RNA-seq y miRNA-seq). El análisis individual de cada grupo de datos mostró diferencias de expresión de ARNm y miARN, así como las vías biológicas alteradas en los linfocitos CD4+ comparando pacientes cirróticos con y sin EHM. Encontramos alteraciones en 167 ARNm y 20 rutas biológicas en los pacientes con EHM, incluyendo los receptores tipo Toll, la señalización de la IL-17 y las vías del metabolismo de histidina y triptófano. Trece miRNAs y 7 factores de transcripción presentaron alteraciones en los pacientes con EHM. Utilizando bases de datos para determinar sus genes diana, encontramos una modulación por el aumento de miR-494-39, miR-656-3p y miR-130b-3p de la expresión de TNFAIP3 (proteína A20) y ZFP36 (proteína TTP) aumentaría los niveles de citoquinas proinflamatorias como IL-17 y TNF¿. Finalmente, estudiamos el repertorio de receptores de células T (TCR) de pacientes control, y de pacientes cirróticos con y sin EHM, a partir del conjunto de datos de RNA-seq procedentes de células T CD4+ aisladas previamente. Dado que los experimentos de RNA-seq contienen genes del TCR en una fracción de los datos, se puede analizar el repertorio sin necesidad de generar datos adicionales. Tras el alineamiento de las lecturas con la base de datos de los genes VDJ realizada por la herramienta MiXCR, recuperamos entre 498-1114 cadenas TCR beta distintas por paciente. Los resultados mostraron un bajo número de clones públicos (convergencia clonal), una alta diversidad (expansión clonal) y una elevada similitud en la arquitectura de la secuencia dentro de los repertorios, independientemente del estado inmunitario de los 3 grupos de pacientes. Además, detectamos una sobrerrepresentación significativa de los TCRs relacionados con la enfermedad celíaca y la enfermedad inflamatoria intestinal en los repertorios de los pacientes con EHM. / [CA] L'objectiu principal d'aquest treball va ser conèixer les alteracions immunològiques associades a la inflamació perifèrica que desencadenen deteriorament cognitiu en els pacients cirròtics amb encefalopatia hepàtica mínima (EHM). Aquests canvis poden ser monitoritzats com cascades de senyalització al llarg dels tipus cel·lulars del sistema immune. Com a estudi preliminar, es van analitzar els canvis en l'expressió gènica (transcriptòmica), els metabòlits de plasma (metabolòmica) i un conjunt de citocines extracel·lulars en mostres de sang de pacients cirròtics amb i sense EHM. Els resultats de l'anàlisi transcriptòmica van recolzar la hipòtesi d'alternances en les poblacions cel·lulars de limfòcits Th1/Th2 i Th17 com a principals impulsors de la EHM. L'anàlisi clúster de les molècules del sèrum va donar com a resultat 6 grups de compostos químicament similars. A més, detectàrem una sobrerepresentació significativa dels TCRs relacionats amb la malaltia celíaca i la malaltia inflamatòria intestinal en els repertoris dels pacients amb EHM. / [EN] The main objective of this work was to understand the immunological alterations associated with the peripheral inflammation that trigger minimal hepatic encephalopathy (MHE) in patients with cirrhosis. These changes can be monitored through the signaling cascades of different immune system cell types. In this work, in a preliminary study, changes in gene expression (transcriptomics), plasma metabolites (metabolomics), and a panel of extracellular cytokines were analyzed in blood samples from patients with cirrhosis with and without MHE. Transcriptomics analysis supported the hypothesis that alternations in the Th1/Th2 and Th17 lymphocyte cell populations are the major drivers of MHE. Cluster analysis of serum molecules highlighted 6 groups of chemically similar compounds. We also developed a multi-omic integration analysis pipeline to detect covariation between intra- and extracellular components that could contribute to the induction of cognitive impairment. Results of this integrative analysis suggested a relationship between cytokines CCL20, CX3CL1, CXCL13, IL-15, IL-22, and IL-6 and altered chemotaxis, as well as a link between long-chain unsaturated phospholipids and increased fatty acid transport and prostaglandin production. A shift in peripheral inflammation in patients with MHE, mainly orchestrated by CD4+ T cells, had been proposed in previous studies as a critical factor that triggers cognitive impairment. The second part of this thesis focused on understanding the pathways and mechanisms by which alterations in CD4+ lymphocytes may contribute to peripheral inflammation in MHE. Thus, the expression levels of genes, transcription factors, and miRNAs were analyzed in this lymphocyte subtype by high throughput sequencing (RNA-seq and miRNA-seq). Separate analysis of each dataset showed mRNA and miRNA expression differences and altered biological pathways in CD4+ lymphocytes when compared to patients with cirrhosis with and without MHE. We found alterations in 167 mRNAs and 20 pathways in patients with MHE, including toll-like receptors, IL-17 signaling, histidine, and tryptophan metabolism pathways. In addition, 13 miRNAs and 7 transcription factors presented alterations in patients with MHE. We used public databases to determine the target genes of these regulatory molecules and found that increased miR-494-39, miR-656-3p, and miR-130b-3p expression may modulate TNFAIP3 (A20) and ZFP36 (TTP) to increase levels of pro-inflammatory cytokines such as IL-17 and TNF¿. Finally, we present a case study of the T-cell receptor (TCR) repertoire profiles of control patients and patients with cirrhosis with and without MHE obtained from the bulk RNA-seq dataset previously generated from isolated CD4+ T cells. Given that RNA-seq experiments contain the TCR genes in a fraction of the data, the receptor repertoire analysis without the need to generate additional data is possible. After read alignment to the VDJ genes was performed with the MiXCR tool, we successfully recovered 498-1,114 distinct TCR beta chains per patient. Results showed fewer public clones (clonal convergence), higher diversity (clonal expansion), and elevated sequence architecture similarity within repertoires, independently of the immune status of the 3 groups of patients. Additionally, we detected significant overrepresentation of celiac disease and inflammatory bowel disease related TCRs in MHE patient repertoires. To the best of our knowledge, this is one of the few studies to have shown a step-by-step pipeline for the analysis of immune repertoires using whole transcriptome RNA-seq reads as source data. In conclusion, our work identified potentially relevant molecular mechanisms of the changes in the immune system associated with the onset of MHE in patients with cirrhosis. Future work with a large sample cohort will be required to validate these results in terms of biomarker determination and the development of new, more effective treatments for MHE. / Rubio Martínez-Abarca, MT. An Evaluation of Technological, Organizational and Environmental Determinants of Emerging Technologies Adoption Driving SMEs’ Competitive Advantage

This research evaluates the technological, organizational, and environmental determinants of emerging technologies adoption represented by Artificial Intelligence (AI) and Internet of Things (IoT) driving SMEs’ competitive advantage within a resource-based view (RBV) theoretical approach supported by the technological-organizational-environmental (TOE)-framework setting. Current literature on SMEs competitive advantage as outcome of emerging technologies in the technological, organisational, and environmental contexts presents models focused on these contexts individual components. There are no models in the literature to represent the TOE framework as an integrated structure with gradual levels of complexity, allowing for incremental evaluation of the business context in support of decision making towards emerging technologies adoption supporting the firm competitive advantage. This research gap is addressed with the introduction of a new concept, the IT resource-based renewal, underpinned by the RBV, and supported by the TOE framework for providing a holistic understanding of the SMEs strategic renewal decision through information technology. This is achieved through a complex measurement model with four level constructs, leading into a parsimonious structural model that evaluates the relationships between IT resource-based renewal, and emerging technologies adoption driving SMEs competitive advantage. The model confirms the positive association between the IT resource-based renewal and emerging technologies adoption, and between the IT resource-based renewal and SME competitive advantage for the SMEs managers model, with the SME owners model outcomes are found not being supportive towards emerging technologies adoption driving SME competitive advantage. As methodology, PLS-SEM is used for its capabilities of assessing complex paths among model variables. Analysis is done on three models, one for the full sample, with two subsequent ones for owners and managers, respectively, as SME decision makers, with data collected using a web-based survey in Canada, the UK, and the US, that has provided 510 usable answers. This research has a theoretical contribution represented by the introduction of the IT resource-based renewal concept, that integrates the RBV perspective and the TOE framework for supporting organization’s decision on emerging technologies adoption driving SMEs competitive advantage. As practical implications, this thesis provides SMEs with a reference framework on adopting emerging technologies, offering SME managers and owners a comprehensive model of hierarchical factors contributing to SMEs competitive advantage acquired as outcome of AI and IoT adoption. This research makes an original contribution to the enterprise management, information systems adoption, and SME competitive advantage literature, with an empirical approach that verifies a model of emerging technologies adoption determinants driving SMEs competitive advantage.

A multivariate approach to characterization of drug-like molecules, proteins and the interactions between them

En sjukdom kan många gånger härledas till en kaskadereaktion mellan proteiner, co-faktorer och substrat. Denna kaskadreaktion blir många gånger målet för att behandla sjukdomen med läkemedel. För att designa nya läkemedelsmoleyler används vanligen datorbaserade verktyg. Denna design av läkemedelsmolekyler drar stor nytta av att målproteinet är känt och då framförallt dess tredimensionella (3D) struktur. Är 3D-strukturen känd kan man utföra så kallad struktur- och datorbaserad molekyldesign, 3D-geometrin (f.f.a. för inbindningsplatsen) blir en vägledning för designen av en ny molekyl. Många faktorer avgör interaktionen mellan en molekyl och bindningsplatsen, till exempel fysikalisk-kemiska egenskaper hos molekylen och bindningsplatsen, flexibiliteten i molekylen och målproteinet, och det omgivande lösningsmedlet. För att strukturbaserad molekyldesign ska fungera väl måste två viktiga steg utföras: i) 3D anpassning av molekyler till bindningsplatsen i ett målprotein (s.k. dockning) och ii) prediktion av molekylers affinitet för bindningsplatsen. Huvudsyftena med arbetet i denna avhandling var som följer: i) skapa modeler för att prediktera affiniteten mellan en molekyl och bindningsplatsen i ett målprotein; ii) förfina molekyl-protein-geometrin som skapas vid 3D-anpassning mellan en molekyl och bindningsplatsen i ett målprotein (s.k. dockning); iii) karaktärisera proteiner och framför allt deras sekundärstruktur; iv) bedöma effekten av olika matematiska beskrivningar av lösningsmedlet för förfining av 3D molekyl-protein-geometrin skapad vid dockning och prediktion av molekylers affinitet för proteiners bindningsfickor. Ett övergripande syfte var att använda kemometriska metoder för modellering och dataanalys på de ovan nämnda punkterna. För att sammanfatta så presenterar denna avhandling metoder och resultat som är användbara för strukturbaserad molekyldesign. De rapporterade resultaten visar att det är möjligt att skapa kemometriska modeler för prediktion av molekylers affinitet för bindningsplatsen i ett protein och att dessa presterade lika bra som andra vanliga metoder. Dessutom kunde kemometriska modeller skapas för att beskriva effekten av hur inställningarna för olika parametrar i dockningsprogram påverkade den 3D molekyl-protein-geometrin som dockingsprogram skapade. Vidare kunde kemometriska modeller andvändas för att öka förståelsen för deskriptorer som beskrev sekundärstrukturen i proteiner. Förfining av molekyl-protein-geometrin skapad genom dockning gav liknande och ickesignifikanta resultat oberoende av vilken matematisk modell för lösningsmedlet som användes, förutom för ett fåtal (sex av 30) fall. Däremot visade det sig att användandet av en förfinad geometri var värdefullt för prediktion av molekylers affinitet för bindningsplatsen i ett protein. Förbättringen av prediktion av affintitet var markant då en Poisson-Boltzmann beskrivning av lösningsmedlet användes; jämfört med prediktionerna gjorda med ett dockningsprogram förbättrades korrelationen mellan beräknad affintiet och uppmätt affinitet med 0,7 (R2). / A disease is often associated with a cascade reaction pathway involving proteins, co-factors and substrates. Hence to treat the disease, elements of this pathway are often targeted using a therapeutic agent, a drug. Designing new drug molecules for use as therapeutic agents involves the application of methods collectively known as computer-aided molecular design, CAMD. When the three dimensional (3D) geometry of a macromolecular target (usually a protein) is known, structure-based CAMD is undertaken and structural information of the target guides the design of new molecules and their interactions with the binding sites in targeted proteins. Many factors influence the interactions between the designed molecules and the binding sites of the target proteins, such as the physico-chemical properties of the molecule and the binding site, the flexibility of the protein and the ligand, and the surrounding solvent. In order for structure-based CAMD to be successful, two important aspects must be considered that take the abovementioned factors into account. These are; i) 3D fitting of molecules to the binding site of the target protein (like fitting pieces of a jigsaw puzzle), and ii) predicting the affinity of molecules to the protein binding site. The main objectives of the work underlying this thesis were: to create models for predicting the affinity between a molecule and a protein binding site; to refine the geometry of the molecule-protein complex derived by or in 3D fitting (also known as docking); to characterize the proteins and their secondary structure; and to evaluate the effects of different generalized-Born (GB) and Poisson-Boltzmann (PB) implicit solvent models on the refinement of the molecule-protein complex geometry created in the docking and the prediction of the molecule-to-protein binding site affinity. A further objective was to apply chemometric methodologies for modeling and data analysis to all of the above. To summarize, this thesis presents methodologies and results applicable to structure-based CAMD. Results show that predictive chemometric models for molecule-to-protein binding site affinity could be created that yield comparable results to similar, commonly used methods. In addition, chemometric models could be created to model the effects of software settings on the molecule-protein complex geometry using software for molecule-to-binding site docking. Furthermore, the use of chemometric models provided a more profound understanding of protein secondary structure descriptors. Refining the geometry of molecule-protein complexes created through molecule-to-binding site docking gave similar results for all investigated implicit solvent models, but the geometry was significantly improved in only a few examined cases (six of 30). However, using the geometry-refined molecule-protein complexes was highly valuable for the prediction of molecule-to-binding site affinity. Indeed, using the PB solvent model it yielded improvements of 0.7 in correlation coefficients (R2) for binding affinity parameters of a set of Factor Xa protein drug molecules, relative to those obtained using the fitting software.

In the contemporary context, in order to achieve superior performance, it reinforces the company's need to have, and make good use, of scarce, valuable, non-substitutable and inimitable resources. These resources include brands, customer base, knowledge, ability and competence of the work teams, corporate culture, partnerships and operational processes established, among other intangible assets, usually arising from a long and risky development process. Mergers and acquisitions (M & A) arise, then, as an important strategic action, being an alternative means to obtain and accelerate the accumulation of these resources within the companies. That is the subject of this work, which discusses the importance of existing and intangible assets disclosed, previous to the M & A transactions, their classification into various types, measurement, and impact on the resulting firm's financial performance in long term. The overall objective of this thesis was to analyze how this performance, after a minimum period of 36 months of the event, is related to the existence, level of disclosure and the nature of intangible assets in the organizations involved. One hundred-eighteen (118) companies were investigated in fifty-nine (59) cases of M & A occurred in France between 1997 and 2007; the study reflects a multi-method research, pluralistic, on qualitative and quantitative aspects. Intangible assets disclosure indicators were built by applying the content analysis technique to financial and accounting reports provided by the companies prior to the events, as well as financial indicators (proxies) for the existence of intangibles were calculated. These indicators were initially confronted with each other and later their explanatory power in relation to financial ratios of growth and profitability (for the corporation and its shareholders), which are the analyzed dimensions of financial performance. Many methods for statistical analysis were used in the multivariate data analysis (correlations and factor analysis, multiple regressions) and in the structural equation modeling (SEM), via Partial Least Squares (PLS). A total of twelve models, with statistics significance, were established to express the relationship among the constructs examined. Best results were achieved in the models developed with variables of semantic origin, in detriment of those with financial indicators only. The results obtained in this thesis leads to deduce that, in this study, there are positive relationships between the existence and the disclosure of intangible assets by firms involved in the operations of M & A and subsequent financial performance, measured by the corporate profitability and the growth of the resulting organization. This suggests that the strategic choice for business growth via M & A operations is favorable to the accumulation of intangible assets in the firms, in search for better results. / A alocação de recursos e a constante busca por diferenciais competitivos, visando melhores resultados, são grandes desafios empresariais. No contexto contemporâneo, para obter desempenho superior, reforça-se a necessidade de a empresa dispor, e fazer bom uso, de recursos raros, valiosos, não-substituíveis e de difícil imitação. Dentre estes recursos, destacam-se aspectos como as marcas, a base de clientes, o conhecimento, a capacidade e competência das equipes de trabalho, a cultura corporativa, as parcerias e os processos operacionais estabelecidos, dentre outros ativos intangíveis, geralmente decorrentes de longos e arriscados processos de desenvolvimento. As fusões e aquisições (F&A) surgem, então, como movimentos estratégicos importantes, sendo meio alternativo para obter e acelerar a acumulação destes recursos nas empresas. É essa a temática deste trabalho, que discorre sobre a importância dos ativos intangíveis existentes e divulgados previamente às operações de F&A de empresas, sobre a classificação dos seus diversos tipos, a sua mensuração e o seu impacto sobre o desempenho financeiro da firma resultante, no longo prazo. O objetivo geral desta tese foi analisar como este desempenho, após prazo mínimo de 36 meses do evento, está relacionado à existência, ao nível de divulgação e à natureza dos ativos intangíveis das organizações envolvidas. Foram investigadas 118 empresas, em 59 casos de F&A ocorridos na França entre 1997 e 2007, em uma pesquisa multi-métodos, pluralística, nas vertentes qualitativa e quantitativa. Foram construídos indicadores de divulgação (disclosure) de ativos intangíveis, mediante aplicação da técnica de análise de conteúdos aos relatórios contábil-financeiros disponibilizados pelas empresas antes do evento, e calculados indicadores financeiros (proxies) para a existência de intangíveis. Estes indicadores foram inicialmente confrontados entre si e posteriormente quanto ao seu poder explicativo em relação aos índices financeiros de crescimento e de lucratividade (empresarial e para os acionistas), que são as dimensões analisadas do desempenho financeiro. Utilizaram-se métodos de análise estatística de dados multivariados (análises de correlações, fatoriais, regressões múltiplas) e modelagem em equações estruturais, via Partial Least Squares (SEM- PLS). Foram estabelecidos, no total, doze modelos com significância estatística para expressar o relacionamento entre os construtos examinados. Alcançaram-se melhores resultados nos modelos desenvolvidos com variáveis de origem semântica, em detrimento daqueles que utilizaram indicadores exclusivamente financeiros. Os resultados obtidos nesta tese permitiram deduzir que há relações positivas entre a existência e a divulgação de ativos intangíveis pelas firmas envolvidas nas operações de F&A estudadas e o posterior desempenho financeiro, mensurado pela lucratividade empresarial e pelo crescimento, da organização resultante. Isto sugere que a opção estratégica por crescimento empresarial via operações de F&A é favorável ao acúmulo de recursos intangíveis nas firmas, na busca por melhores resultados.

Relações estrutura-retenção de flavonóides por cromatografia a líquido em membranas imobilizadas artificialmente / Structure retention relationships of flavonoids by liquid chromatography using immobilized artificial membranes

Para um composto químico exercer seu efeito bioativo é necessário que ele atravesse várias barreiras biológicas até alcançar seu sitio de ação. Propriedades farmacocinéticas insatisfatórias (como absorção, distribuição, metabolismo e excreção) são reconhecidamente as principais causas na descontinuidade de pesquisas na busca por novos fármacos. Neste trabalho, modelos biofísicos foram utilizados para o estudo de absorção de uma série de flavonóides naturais com atividade tripanossomicida. O coeficiente cromatográfico de partição, kw, foi determinado através da cromatografia líquida de alta eficiência em fase reversa, RP-HPLC, utilizando-se de colunas cromatográficas empacotadas com constituintes básicos da membrana biológica (fosfatidilcolina e colesterol). Os resultados obtidos demonstraram que nas colunas compostas por fosfatidilcolina a retenção de flavonóides hidroxilados é determinada por interações secundárias, além da partição, e no caso da coluna de colesterol, a partição é o principal mecanismo que rege a retenção. Uma série de descritores físico-químicos foi gerada pelos campos moleculares de interações (MIFs) entre os flavonóides naturais e algumas sondas químicas virtuais, utilizando o programa GRID. Os descritores físico-químicos gerados foram correlacionados com os log kw por análise dos mínimos múltiplos parciais (PLS), utilizando o programa VolSurf, com a finalidade de gerar um modelo quantitativo entre estrutura e propriedade (QSPR) para esta classe de compostos. O modelo produzido por este estudo, ao utilizar os dados de partição em colesterol, log kwCol, apresentou elevada consistência interna, com bom poder de correlação (R2 = 0, 97) e predição (Q2 = 0,86) para a partição destas moléculas / In order to a chemical compound exert its bioactive effect it is necessary that it crosses some biological barriers until reaching its site of action. Unfavorable pharmacokinetics properties (absorption, distribution, metabolism and excretion) are admittedly one of the main causes in the discontinuity of research in the search for new drugs. In this work, biophysics models were used for the study of absorption of a series of natural flavonoids with trypanocide activity. The chromatographic retention indices (log kw) were determined on immobilized artificial membranes columns (IAM.PC.DD, IAM.PC.DD2, Cholesteryl 10-Undecetonoato) obtained by the extrapolation method. The results demonstrated that in the composed columns for fosfatidilcolina the retention of hydroxil flavonoids is determined by secondary interactions, beyond the partition. In the case of the retention for the cholesterol column, the partition is the main mechanism that drives the retention. A series of physico-chemical descriptors were generated by the molecular interaction fields (MIF) between the flavonoids and some virtual chemical probes, using the program GRID. The descriptors were correlated with log kw by the partial least squares regression (PLS), using the VolSurf program, with the purpose to generate a quantitative model between the structure and the retention (QSRR) for this compounds class. The model produced for this study, when using the data of partition in cholesterol, log kwCol, presented high internal consistency, with good correlation power (R2 = 0, 97) and prediction (Q2 = 0,86) for the partition of these molecules
