Assim sendo, foi elaborado um plano de ação educacional, como projeto piloto, visando à formação continuada em gestão escolar para as 148 escolas estaduais e 2 técnicos da jurisdição da Superintendência Regional de Ensino (SRE) Metropolitana A da Secretaria de Estado de Educação de Minas Gerais (SEEMG). Espera-se que este estudo possa contribuir para o fortalecimento de uma gestão escolar integrada, estratégica e participativa das escolas estaduais de Minas Gerais, buscando a melhoria da qualidade da educação. / This work, developed in the Juiz de Fora’ Federal University Program of PostGraduation in Evaluation and Management of Public Education, aims to analyse how school managers use the strategic management tool provided by Ministry of Education (MEC), through the School Development Plan (PDE Escola). This study case analysed four urban state school located in Belo Horizonte and Sabará (both in Minas Gerais). The research has as guiding question: how the PDE Escola has been used as strategic management tool for school units? The examination and description of implementation and appropriation process of PDE Escola by school managers in this four cases, show the need of a continuous training for school managers with focus on strategic, participatory and integrated management, considering also the Political Pedagogical Program materialization most suitable to school’s reality. Thus was elaborated an educational action plan as a pilot project for a continuous formation in school management for 148 state schools and 2 experts of Metropolitan A Regional Educational Superintendence (SRE) of Minas Gerais State Secretariat for Education. This study expects to contribute to strengthening of an integrated, strategic and participatory school management in Minas Gerais’ state schools and, with this, promote an improvement in educational quality.

Dessa forma, buscamos compreender como se dá a implementação do programa no âmbito da escola, tendo como foco o papel da liderança no processo. A pesquisa de cunho qualitativo envolveu entrevistas com diretores escolares e a análise dos dados encontrados se deu com base na produção recente de autores do campo da gestão escolar, como Dourado (2001), Fonseca (2003a, 2003b), Libâneo (2004), Lück (2008), Mendonça (2000), Paro (1998), Teixeira (2010). Os resultados apontam que a escola precisa apropriar-se de uma cultura de planejamento tendo como articulador central o gestor, devendo esse exercer uma forte liderança e ser capaz de dialogar com todos os segmentos da comunidade escolar de forma democrática, em prol de uma educação de qualidade. / We intend in this work study the Strategic Planning in the Education applied to the Educational Management as instrument of managerial innovation, having as a starting point the analysis of a Program of the Department of the Education (Ministry of Education and Culture - MEC) which comes being implemented in the public schools of the country since 1997, the Plan of Development of the School – PDE School (Plano de Desenvolvimento da Escola – PDE Escola). Into a new educational context and considering the new public politics for the area after the launching in 2007 of the Plan of Goals of the Federal Government "Commitment Everybody by the Education"( Plano de Metas do Governo Federal “Compromisso Todos pela Educação”) and of the Plan of Development of the Education (Plano de Desenvolvimento da Educação - PDE), we have done an approach concerns to the implementation of that action under the point of view of the director, in six municipal public schools of Juiz de Fora which presented low Development index of the basic education (Índice de Desenvolvimento da Educação Básica - Ideb) in the year of 2007. In this way, we tried to understand how happens the implementation of the program in the scope of the school, having like the focus the paper of the leadership in the process. The research of qualitative character involved interviews with school directors and the data analysis found was happened on the basis of recent production of authors from the field of the school management as Dourado (2001), Fonseca (2003a, 2003b), Libâneo (2004), Lück (1986), Mendonça (2000), Paro (1998), Teixeira (2010). The results shows the school necessity to appropriate of a culture of planning having as a central articulator the gestor and he must exercise a strong leadership and to be capable of talk with all of the segments of the school community in a democratic form, in behalf of an education of quality.

The goals are to identify the main actors participating in the process, understand their behaviors and the justifications that they give for their activities at the time of formulation of policy. The search was developed through semi-structured interviews with sectors of civil society organizations, such as corporate entities based in São Paulo state and trade unions representing workers in education. In addition to these, were also interviewed governmental actors, representatives of the Education and Cultures Ministry (MEC) and the Espírito Santo’s Department of Education (SEDU). Through this search, could be concluded that the MEC selected partners for the formulation of the PDE considering short-term’s needs, as political and ideological conflicts, including inner conflicts in the Workers’ Party (PT), the convenience of find programs and projects ready to render the scope of its plan, as well as the concession of public spaces for a sector, making an clear agreement to their educational policies.

Homogenised models of Smooth Muscle and Endothelial Cells.

Numerous macroscale models of arteries have been developed, comprised of populations of discrete coupled Endothelial Cells (EC) and Smooth Muscle Cells (SMC) cells, an example of which is the model of Shaikh et al. (2012), which simulates the complex biochemical processes responsible for the observed propagating waves of Ca2+ observed in experiments. In a 'homogenised' model however, the length scale of each cell is assumed infinitely small while the population of cells are assumed infinitely large, so that the microscopic spatial dynamics of individual cells are unaccounted for. We wish to show in our study, our hypothesis that the homogenised modelling approach for a particular system can be used to replicate observations of the discrete modelling approach for the same system. We may do this by deriving a homogenised model based on Goldbeter et al. (1990), the simplest possible physiological system, and comparing its results with those of the discrete Shaikh et al. (2012), which have already been validated with experimental findings. We will then analyse the mathematical dynamics of our homogenised model to gain a better understanding of how its system parameters influence the behaviour of its solutions. All our homogenised models are essentially formulated as partial differential equations (PDE), specifically they are of type reaction diffusion PDEs. Therefore before we begin developing the homogenised Goldbeter et al. (1990), we will first analyse the Brusselator PDE with the goal that it will help us to understand reaction diffusion systems better. The Brusselator is a suitable preliminary study as it shares two common properties with reaction diffusion equations: oscillatory solutions and a diffusion term.

Numerical Methods for Nonlinear Equations in Option Pricing

This thesis explores numerical methods for solving nonlinear partial differential equations (PDEs) that arise in option pricing problems. The goal is to develop or identify robust and efficient techniques that converge to the financially relevant solution for both one and two factor problems. To illustrate the underlying concepts, two nonlinear models are examined in detail: uncertain volatility and passport options. For any nonlinear model, implicit timestepping techniques lead to a set of discrete nonlinear equations which must be solved at each timestep. Several iterative methods for solving these equations are tested. In the cases of uncertain volatility and passport options, it is shown that the frozen coefficient method outperforms two different Newton-type methods. Further, it is proven that the frozen coefficient method is guaranteed to converge for a wide class of one factor problems. A major issue when solving nonlinear PDEs is the possibility of multiple solutions. In a financial context, convergence to the viscosity solution is desired. Conditions under which the one factor uncertain volatility equations are guaranteed to converge to the viscosity solution are derived. Unfortunately, the techniques used do not apply to passport options, primarily because a positive coefficient discretization is shown to not always be achievable. For both uncertain volatility and passport options, much work has already been done for one factor problems. In this thesis, extensions are made for two factor problems. The importance of treating derivative estimates consistently between the discretization and an optimization procedure is discussed. For option pricing problems in general, non-smooth data can cause convergence difficulties for classical timestepping techniques. In particular, quadratic convergence may not be achieved. Techniques for restoring quadratic convergence for linear problems are examined. Via numerical examples, these techniques are also shown to improve the stability of the nonlinear uncertain volatility and passport option problems. Finally, two applications are briefly explored. The first application involves static hedging to reduce the bid-ask spread implied by uncertain volatility pricing. While static hedging has been carried out previously for one factor models, examples for two factor models are provided. The second application uses passport option theory to examine trader compensation strategies. By changing the payoff, it is shown how the expected distribution of trading account balances can be modified to reflect trader or bank preferences.

Estudio experimental de migración de finos en minería de caving

Ingeniero Civil de Minas / A medida que las reservas mineras superficiales se agotan y las minas subterráneas se profundizan, la minería de Caving aflora como una mejor opción. Dentro de los desafíos de la minería de Caving se encuentra estudiar el flujo gravitacional, el cual influye en la recuperación de las reservas del yacimiento. En particular, la migración de finos incide en el flujo diluyendo reservas, afectando la recuperación y la seguridad de la operación. Esta incidencia se ve acentuada si una gran proporción de las partículas ha fragmentado de tamaño fino en la columna de extracción. Dados estos argumentos y problemáticas que presentan las partículas finas, se vuelve necesario estudiar en mayor profundidad la migración de finos en minería de Caving. El objetivo de este trabajo es realizar experimentos que permitan estudiar la migración de finos en condiciones similares a las encontradas en minas de Caving y comparar los resultados con aquellos obtenidos en simulaciones. Para esto se llevan a cabo dos experimentos en un modelo físico, uno de extracción uniforme y otro de extracción aislada, que en una escala de 1:200 emulan una mina de Block/Panel Caving en general. Para el diseño del modelo físico se considera el análisis de similitud y los factores de escala. Para la realización de los experimentos el modelo es cargado con 240 [cm] de partículas gruesas (tamaño medio 4.45 [mm]) y por sobre las partículas gruesas son ubicadas partículas finas (tamaño medio 0.14 [mm]). Por otro lado se realizan seis simulaciones en REBOP 3.1 (Rapid Emulator Based On PFC3D) emulando los dos experimentos, tres imitando al experimento de extracción aislada y otras tres imitando al de extracción uniforme. Las simulaciones se diferencian en la variación de la granulometría de las partículas gruesas siendo igual o menor en promedio y desviación estándar a la del modelo físico. Los resultados experimentales indican que el punto de entrada de la dilución (PED) para los experimentos de extracción uniforme y extracción aislada es de 94.5% y 60.4% respectivamente (si no se considera la masa de la zona estancada ISZ para el experimento de extracción aislada corresponde a 78%). Además, la migración final que alcanza el experimento de extracción aislada es del orden de 3.5 veces mayor al experimento de extracción uniforme. Las simulaciones con resultados más cercanos a los experimentos registran un PED muy similar al modelo físico, aunque para esto deben ser calibradas. En general las simulaciones son capaces de representar resultados cuantitativos del modelo físico, sin embargo, es posible encontrar diferencias entre las imágenes del modelo físico y de las simulaciones. Finalmente se concluye que la migración de finos sí es influenciada por la estrategia de extracción, siendo este efecto mayor en una estrategia de extracción aislada. Esto se debe a que una estrategia de extracción aislada genera mayor deformación por corte en las partículas lo que permite la migración. Por otro lado es posible concluir que el software REBOP es capaz de replicar los experimentos una vez calibrado, siendo la calibración una etapa fundamental del proceso de simulación. A modo de recomendación, para facilitar la toma de imágenes en el modelo físico, se sugiere utilizarlo hasta bajas alturas y evitar el uso excesivo barras.

Reaction-Diffusion kinetics of Protein DNA Interactions

Transcription factors need to rapidly find one specific binding site among millions of nonspecific sites on the chromosomal DNA. In this thesis I use various aspects of reaction-diffusion theory to investigate the interaction between proteins and DNA and to explain the searching, finding and binding to specific operator sites. Using molecular dynamics methods we calculate the free energy profile for the model protein LacI as it leaves a nonspecific stretch of DNA and as it slides along DNA. Based on the free energy profiles we estimate the microscopic dissociation rate constant, kdmicro ~1.45×104s-1, and the 1D diffusion coefficient, D1 ~ 0.05-0.29 μm2s-1 (2-40μs to slide 1 basepair (bp)). At a non-atomistic level of detail we estimate the number of microscopic rebindings before a macroscopic dissociation occurs which leads to the  macroscopic residence time, τDmacro ~ 48±12ms resulting in a in vitro sliding length estimate of 135-345bp. When we fit the DNA interaction parameters for in vivo conditions to recent single molecule in vivo experiments we conclude that neither hopping nor intersegment transfer contribute to the target search for the LacI dimer, that it appears to bind the specific Osym operator site as soon as it slides into it, and that the sliding length is around 40bp in the cell. The estimated in vivo D1 ~ 0.025 μm2s-1 is higher than expected from estimates of D1 based on viscosity and the atomistic simulations. Surprisingly, we were also forced to conclude that the nonspecific association for the LacI dimer appeared reaction limited which is in conflict with the free energy profile. This inconsistency is resolved by allowing for steric effects. Using reaction-diffusion theory and simulations we show that an apparent reaction limited association can be diffusion limited if geometry and steric effects are taken into account. Furthermore, the simulations show that a protein binds ~2 times faster to a DNA molecule with a helical reactive patch than to a stripe patch running along the length of the DNA. This facilitated binding has a direct impact on the search time especially in the presence of other DNA binding proteins.

Pharmaceutical tablets have been the most dominant form for drug delivery and they need to be strong enough to withstand external stresses due to packaging and loading conditions before use. The strength of the produced tablets, which is characterised by their compressibility and compactibility, is usually deter-mined through a physical prototype. This process is sometimes quite expensive and time consuming. Therefore, simulating this process before hand can over-come this problem. A technique for shape modelling of pharmaceutical tablets based on the use of Partial Differential Equations is presented in this thesis. The volume and the sur-face area of the generated parametric tablet in various shapes have been es-timated numerically. This work also presents an extended formulation of the PDE method to a higher dimensional space by increasing the number of pa-rameters responsible for describing the surface in order to generate a solid tab-let. The shape and size of the generated solid tablets can be changed by ex-ploiting the analytic expressions relating the coefficients associated with the PDE method. The solution of the axisymmetric boundary value problem for a finite cylinder subject to a uniform axial load has been utilised in order to model a displace-ment component of a compressed PDE-based representation of a flat-faced round tablet. The simulation results, which are analysed using the Heckel model, show that the developed model is capable of predicting the compressibility of pharmaceutical powders since it fits the experimental data accurately. The opti-mal design of pharmaceutical tablets with particular volume and maximum strength has been obtained using an automatic design optimisation which is performed by combining the PDE method and a standard method for numerical optimisation.

Identification and Characterization of PDE8 Inhibitors Using a Fission Yeast Based High-throughput Screening Platform

Hoffman / In this thesis, I describe the development of a screening platform for detecting PDE8A inhibitors using the cAMP-dependent glucose sensing pathway of the fission yeast Schizosaccharomyces pombe, which led us to discover several PDE8A selective inhibitors. In this system, the only PDE of the fission yeast is replaced with mammalian PDE8A1 in strains that have been engineered such that PDE inhibition is required to allow cell growth. Using this system, I screened 56 compounds obtained from PDE4 and PDE7 high throughput screens (HTSs) and identified a PDE4-PDE8 dual specificity inhibitor. Using this as a positive control, I developed a robust high-throughput screen (HTS) for PDE8A inhibitors and screened 240,267 compounds at the Harvard Medical School ICCB Screening Facility. Approximately 0.2 % of the screened compounds were potential PDE8A inhibitors with 0.03% displaying significant potency. Secondary assays of 367 of the most effective compounds against strains expressing PDE8A (both full length and catalytic domain), PDE4A and PDE7A or PDE7B led to the selection of structurally diverse compounds for further testing. To profile the selectivity of twenty-eight of these compounds, dose response assays were conducted using 16 yeast strains that express different PDE isoforms (representing all PDE families with the exception of the PDE6 family). These assays identified compounds with different patterns of inhibition, including structurally-distinct PDE8A-specific inhibitors. By evaluating the effects of these compounds for steroid production in mouse Leydig cells, biologically active compounds that can elevate steroid production were identified. / Thesis (PhD) — Boston College, 2011. / Submitted to: Boston College.
