Sorghum hot water extract : influence of grain physico-chemical characteristics

Traditionally, barley is the preferred cereal for traditional lager brewing. The use of sorghum for the production of European-style lager beers has recently been recognised. The use of sorghum as brewing adjunct could be a major socio-economic advantage in the developing countries in Africa. Limitations for its use, however, include its low amylolytic potential, high gelatinization temperatures, and the presence of tannins. Adjuncts are often combined with cereal malt during the brewing process to provide extra sources of fermentable carbohydrates. As with all cereals, the functional properties of sorghum grains are influenced by their physico-chemical characteristics. It is therefore critical to understand the structure, chemistry and functionality of the sorghum cultivar(s) considered for use as brewing adjuncts. Hot water extract describes the quality of the wort of an adjunct and depicts the amount of starch that was solubilised during mashing. The determination of hot water extract is expensive, laborious and time-consuming. The provision of a possible predictive marker(s) for sorghum hot water extract that is less complicated to determine, could be of great economical value to the brewer. Hot water extract was determined for 43 sorghum cultivars and then compared to various physico-chemical characteristics. Sorghum endosperm texture was visually determined. Suspensions of whole sorghum flour were pasted using a Rapid Visco Analyser (RVA) with an extended heating cycle. Significant negative correlations were obtained between extract content and pasting temperature and time in corneous endosperm samples. There was also a significant positive relationship between tannin-free sorghums and peak viscosity in pasted samples. Protein contents of 10 different sorghums were compared to their hot water extracts, where there was a significant negative relationship between these characteristics. Protein content could be used successfully as a predictive marker for extract. No significant relationship could be established between sorghum hot water extract and starch content. Tannin-containing sorghum cultivars gave significantly lower extracts and had higher malt diastatic power (DP) than non-tannin cultivars. There was no significant relationship between the DP and extract content of non-tannin sorghums. When only non-tannin sorghum cultivars that pasted were subjected to principal component analysis, it seemed that a positive relationship existed between peak viscosity and extract content. Low protein sorghum cultivars with no tannins and corneous endosperm would be suitable for use as brewing adjuncts. Sensory quality of deep fat fried potato chips manufactured from potatoes with different physico-chemical characteristics

Studies generally agree on the critical influence of physico-chemical characteristics of raw potatoes on the physico-chemical and sensory characteristics of potato chips. However, the actual magnitude of differences in sensory quality and consumer acceptance of potato chips as a result of differences in the physico-chemical characteristics of raw potatoes are scarcely reported in most studies. This research was conducted to determine the effect of different physico-chemical characteristics of potato samples on the sensory quality and acceptance of potato chips. The potato samples which represented typical variations in physico-chemical characteristics that a potato chip manufacturer experiences were sourced over a six month period. The samples comprised of four potato varieties (A, B, C, D) sourced from five regions (V, W, X, Y, Z). The samples were coded AV, BW1, BW2, CW, CX, DY, DW and DZ. BW1 and BW2 were of the same variety and grown in the same region but harvested at different time periods. The sourcing of potatoes and potato chips were done at the manufacturer’s processing line during regular production. The potatoes were analyzed for physico-chemical characteristics generally reported to influence potato chip sensory quality, i.e. specific gravity (solids contents), starch content and reducing sugars content. After processing, the physico-chemical characteristics i.e. moisture content, colour and oil content of the respective unflavoured potato chip samples were analysed. The sensory qualities of both unflavoured and flavoured (sour cream and onion) potato chips of the respective potato samples were also determined. There were significant differences (p<0.05) in the specific gravity/solids content of the potato samples. The specific gravity and solids content ranged from 1.073 to 1.098g/cm3 and 19.2 to 24.9% respectively. Reducing sugars were not detected in any of the potato samples at a minimum detection level of 0.05 %. The potato chip samples differed significantly (p<0.05) in moisture content, oil content and colour (a * and b* values, chroma, hue angle) with ranges of 1.2 to 1.9 % (moisture), 33.1 to 40.8 % d.b (oil content), 0.39 to 3.69 (a *), 15.88 to 21.26 (b*), 15.93 to 21.37 (chroma) and 79.92 to 88.96 o (hue angle). Descriptive sensory evaluation showed that 6 and 8 of the attributes identified in the unflavoured and flavoured potato chip samples, respectively, differed significantly (p<0.05). Consumer sensory evaluation indicated that there was no clear preference of one chip sample over the others despite of differences observed through descriptive sensory analysis and physico-chemical characteristics. All the chip samples were generally liked. To the manufacturer this research could be regarded as of a quality control nature since the potato chip samples used in this study were of chipping varieties actually used by the manufacturer. Aproveito este espaço para contar um pouco da minha trajetória com as abelhas, mas também para falar sobre as atividades desenvolvidas e possibilidades surgidas no decorrer do curso de Doutorado, informações estas que normalmente passam despercebidas. Nascido em Salvador, tive meu primeiro contato com as abelhas com pouco mais de 12 anos por iniciativa de meu pai, que nesta época já era um entusiasta pela atividade e possuía conhecimentos práticos. Alguns anos após, fizemos nosso primeiro curso de iniciação em apicultura para consolidar as nossas experiências práticas e as informações obtidas em literatura. Terminei por cursar a graduação em Engenharia Agronômica na Escola de Agronomia da UFBA, atualmente Centro de Ciências Agrárias, Ambientais e Biológicas da Universidade Federal do Recôncavo da Bahia. Foi um período de crescimento em que me envolvi com atividades de iniciação científica, em outras áreas que não apicultura. Já no mestrado, me foi dada a oportunidade de trabalhar com um grupo de abelhas até então novo para mim, os meliponíneos, e de ter como orientador o Prof. Dr. Carlos Alfredo Carvalho, pessoa que me direcionou para o futuro curso de doutorado na USP, dando continuidade à linha de pesquisa iniciada no mestrado. Em 2004 iniciei o meu doutoramento na ESALQ/USP sob orientação do Prof. Dr. Luis Carlos Marchini. Fazendo um balanço desta última etapa, além do desenvolvimento das atividades regulares do programa tive a oportunidade de me envolver com a formulação de projetos de pesquisa para agências de fomento, de participar de eventos nacionais e internacionais, de publicar artigos científicos e de consolidar parcerias de trabalho no Brasil e no exterior. Em síntese, este período de doutorado que se encerra (2004 - 2008) pode ser representado em números por: quatro cursos realizados como formação complementar, quatro premiações em eventos científicos, nove trabalhos publicados em periódicos (alguns submetidos antes de ingressar no doutorado), quatro livros, três capítulos de livros, três cartilhas, dois textos em revista de divulgação, quatro trabalhos completos publicados em anais de congressos, 40 resumos, 17 trabalhos apresentados e participação em 12 eventos técnico-científicos; além do envolvimento em atividades de monitoria junto a disciplinas de graduação, treinamento este fundamental para uma futura atividade de docência. Acredito que todas estas amizades feitas, parcerias consolidadas, atividades desenvolvidas e produções decorrentes do doutorado contribuirão de forma substancial e decisiva para esta nova etapa que se inicia em minha vida. / The stingless bees honey is a product presenting a growing market demand but, in spite of its consumption as food and even medicinal use, there are few studies to define quality standards for its commercialization. This work was carried out to determine the physico-chemical characteristics and microbiological quality of honey samples produced by meliponid species from Bahia State, Brazil, contributing to the establishment of quality control standards of this product. Forty-seven samples collected from December 2004 to May 2006 were analyzed, being determined their physico-chemical characteristics and microbiological quality, regarding to the standard counting of moulds and yeasts, and presence of microorganisms of the coliform group. Concerning the physico-chemical characteristics, the obtained medium values, excepting moisture content, fulfill the quality criteria established by the current Brazilian Legislation. Considering the number of samples individually disqualified, adjustments in reducing sugars limits should be made for best attend to the requirements of meliponid honey, as well as the use of the diastasic activity in the Melipona honeys as quality criterion. In the microbiologic analyses, 53.2% of the honey samples presented standard counting for moulds and yeasts above the maximum value allowed by the Brazilian Legislation. For microorganisms of the coliform group only one sample was positive for coliform at 35ºC. The presence of these microorganisms, mainly moulds and yeasts, even in samples aseptically harvested indicates the need of identification of this microbiota and its possible natural occurrence in the honey produced by this group of bees.

Características fisico-químicas e celulares na secreção láctea de caprinos da raça Saanen durante o processo de secagem da glândula mamária / Physico-chemical and cellular characteristics on milk secretion of Saanen goats during the mammary gland drying off

Para avaliar a influência do processo de secagem da glândula mamária sobre as características físico-químicas e celulares da secreção láctea foram avaliadas 216 amostras provenientes de 27 glândulas mamárias de 15 cabras da raça Saanen consideradas sadias, sendo que essas mamas foram separadas de acordo com o resultado do exame microbiológico da secreção láctea, caracterizando dois grupos: com e sem isolamento bacteriano, composto por 17 e 10 mamas, respectivamente. A partir do dia da interrupção da ordenha e em mais sete momentos (1, 3, 5, 7, 10, 15 e 30 dias após o início da secagem) foi feita a inspeção e a palpação das glândulas mamárias e posterior avaliação do aspecto da secreção láctea. Imediatamente após o exame clínico foram colhidas amostras de leite para a realização do exame microbiológico e das determinações das características físico-químicas e celulares (pH, eletrocondutividade, cloreto, lactose, proteínas, gordura, sólidos totais, índice cloreto/lactose, California Mastitis Test (CMT) e contagem de células somáticas). As glândulas mamárias apresentaram sua maior distensão no terceiro dia após a interrupção da ordenha, mesmo período em que foi possível observar o aparecimento de edema na base dos tetos e em toda a glândula, desaparecendo após o quinto dia, quando teve início a involução do úbere e a freqüência de mamas distendidas começou a declinar, sendo que ao final do processo 37,04% das mamas observadas eram classificadas como não distendidas. A secreção láctea inicial com aspecto de leite foi substituída a princípio por uma de coloração creme clara e mais tardiamente por uma de aspecto aquoso, sendo que ao final do processo as duas últimas eram mais observadas que a com aspecto de leite. Apenas o pH, a eletrocondutividade, os teores de cloretos, lactose, proteínas e índice cloreto/lactose sofreram uma significativa influência do processo de secagem, sendo que os valores do pH, eletrocondutividade, cloretos, proteínas e índice cloreto/lactose aumentaram e os valores de lactose declinaram. O CMT também sofreu uma influência do processo de secagem, mais especificamente as respostas \"traços\" e \"duas cruzes\". Estas, de maneira geral, aumentaram no decorrer da secagem, sendo que no último momento de colheita ocorreu uma queda na freqüência da resposta \"traços\" e a freqüência da resposta \"duas cruzes\" teve uma pequena diminuição no meio do processo. Ainda foi observado que, apesar de existirem diferenças estatísticas significativas entre valores das características físico-químicas e celulares das amostras de secreção láctea com e sem isolamento bacteriano, as diferenças são isoladas e limitadas a algumas características estudadas, impossibilitando afirmar que o processo de secagem dos dois grupos difere de maneira significativa. / To evaluate the influence of the mammary gland drying off on the physiochemical and cellular characteristics in the milk secretion they were appraised 216 coming samples of 27 mammary glands of 15 Saanen goats considered healthy, those mammas were separate in agreement with the result of the microbiological test of the milky secretion characterizing two groups: one with 17 mammas without bacterial isolation and other with 10 mammas that presented positive bacterial isolation. Starting from the day of the interruption of the milking and in more seven moments (1, 3, 5, 7, 10, 15 and 30 days after the beginning of the drying off) it was made the inspection and palpation of the mammary glands with subsequent evaluation of the aspect of the milky secretion. Immediately after the exam they were picked samples of milk for the accomplishment of the microbiological test and of the determinations of the physiochemical and cellular characteristics (pH, electrical conductance, chloride, lactose, proteins, fat, total solids, chloride/lactose ratio, California Mastitis Test (CMT) and somatic cell count). The mammary glands presented its largest distention in the third day after the milking interruption, same period in that was possible to observe the edema emergence in the teats base and in the whole gland, that disappeared after the fifth day, when involution of the udder begins and the frequency of distended mammas began to refuse, and at the end of the process 37,04% of the observed mammas they were classified as no distended. The initial milky secretion with aspect of milk was substituted at first for one of coloration cream clear and, so late, for one of aqueous aspect, and in the end of the process those two last they were more observed that the secretion with aspect of milk. Just the pH, electrical conductance, chloride, lactose, proteins and chloride/lactose ratio suffered a significant influence of the drying off, and the values of the pH, electrical conductance, chloride, proteins and chloride/lactose ratio increased and the lactose values refused. CMT also suffered an influence of the drying off, more specifically the answers \"lines\" and \"two crosses.\" These, in a general way, increased in elapsing of the drying off, and in the last moment studied it happened a fall in the frequency of the answer \"lines\" and the frequency of the answer \"two crosses\" had a small decrease in the middle of the process. It was still observed that in spite of significant statistical differences exist among values of the physiochemical and cellular characteristics of the samples of milky secretion with isolation and without bacterial isolation, the differences are punctual and limited to some studied characteristics, disabling to affirm that the process of drying of the two groups differs in a significant way.

Sistema de recria e peso ao abate nas características de carcaça e qualidade da carne de cabritos mestiços

Os cabritos foram aleitados artificialmente em baldes com 1,5 L.dia-1 de leite natural de cabra e a partir do 10º dia de vida passaram a receber a dieta experimental (28,1% PB, 2,8% EE, 46,0% FDN, 25,5% FDA e 67,2% NDT) à vontade contendo 700 g kg-1 de concentrado farelado e 300 g kg-1 de feno de Coast cross, em matéria natural. Nas carcaças foram avaliadas as seguintes características: rendimentos de carcaça quente (RCQ) e frio (RCF), perda por resfriamento, área do olho de lombo, grau de acabamento e proporções dos cortes cárneos. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The objective of the present study was to evaluate the influence of growth systems and slaughter weight on carcass characteristics, as well as physico-chemical characteristics and sensory of the meat 99 crossbred kids (½ Anglo Nubiana x ½ Pardo Alpina), being 50 whole males and 49 females distributed in a completely randomized design with factorial scheme 2x2x3+1, being two sexes, submitted to two growth systems (GS): suckled up to 60 days (SL) and suckled to slaughter (CL), three weights slaughter (WS): 20, 30 and 40 kg and more additional treatment: weaned to slaughter at 60 days. The goats were artificially suckled buckets with 1,5 L.d-1 natural goat milk and from the 10th day of life, they began to receive the experimental diet (28.1% CP, 2.8% EE, 46.0% NDF, 25.5% ADF and 67.2% TDN) ad libitum containing 700 g kg-1 of concentrate ground and 300 g kg-1 of Coast cross hay, in natural matter. Na carne foram avaliadas as características físico-químicas e sensoriais: umidade (UM), proteína bruta (PB), extrato etéreo (EE), cinzas (CZ), pH, cor, capacidade de retenção de água (CRA), perda de peso por cozimento (PPC), força de cisalhamento (FC), aroma, sabor, maciez, suculência, mastigabilidade, além do perfil de ácidos graxos e índices: ácido linoleico conjugado (CLA), ω3 (ômega 3), ω6 (ômega 6), ácidos graxos saturados (AGS), ácidos graxos insaturados (AGI), ácidos graxos monoinsaturados (AGMI), ácidos graxos poliinsaturados (AGPI), ácidos graxos de cadeia curta (AGCC), ácidos graxos de cadeia média (AGCM), ácidos graxos de cadeia longa (AGCL), razões ω6/ω3, AGMI/AGS, AGPI/AGS, ácidos graxos hipocolesterolêmicos e hipercolesterolêmicos (h/H) e índices de aterogenicidade (IA) e trombogenicidade (IT). Os dados coletados foram analisados por meio do software estatístico SAS versão 9.4 pela aplicação do PROC GLM, ao nível de 5% de significância. O SR CL em relação ao SL, proporcionou aumentos no RCQ (48,7 vs 47,5%) e RCF (47,7 vs 46,5%). O PA 40 kg apresentou RCQ e RCF de 49,6 e 48,6% superiores aos PA 20 e 30 kg, os quais apresentaram valores de 46,8 e 45,5% e 47,9 e 47,1%, respectivamente. Para os cortes de desenvolvimento precoce (paleta e perna), o PA 20 kg apresentou resultados superiores em relação aos 40 kg. Porém, apresentou uma diminuição em relação aos cortes de desenvolvimento tardio (pescoço e costela) quando comparados ao PA 40 kg. O lombo não foi influenciado pelo SR e PA. Cabritos machos apresentaram RCQ superior ao das fêmeas (49,14 vs 48,21%), seguido de maiores proporções dos cortes pescoço e paleta. O SR CL em relação ao SL aumentou EE (2,12 vs 1,74%) e diminuiu a PB (17,74 vs 18,10%) da carne. O SR SL apresentou menor AGS (35,24 vs 38,82%), IA (0,45 vs 0,69) e IT (1,11 vs 1,34) em relação ao CL. O PA 20 kg apresentou maior UM e menor CZ em relação ao PA 40 kg e menor CRA em relação ao PA 30 kg. O PA 20 kg apresentou aumento nos AGPI e ω3 quando comparado ao PA 40 kg. Cabritos machos apresentaram resultados superiores para proteína bruta, pH, AGPI e ω3 em relação às fêmeas. Não houve influência do SR na análise sensorial, porém PA 20 kg apresentou maior maciez, suculência e mastigabilidade em relação ao PA 40 kg. Recomenda-se que os cabritos mestiços devem ser abatidos até aos 20 kg, desaleitados aos 60 dias, para obtenção de melhor qualidade de carcaça e carne. / The objective of the present study was to evaluate the influence of growth systems and slaughter weight on carcass characteristics, as well as physico-chemical characteristics and sensory of the meat 99 crossbred kids (½ Anglo Nubiana x ½ Pardo Alpina), being 50 whole males and 49 females distributed in a completely randomized design with factorial scheme 2x2x3+1, being two sexes, submitted to two growth systems (GS): suckled up to 60 days (SL) and suckled to slaughter (CL), three weights slaughter (WS): 20, 30 and 40 kg and more additional treatment: weaned to slaughter at 60 days. The goats were artificially suckled buckets with 1,5 L.d-1 natural goat milk and from the 10th day of life, they began to receive the experimental diet (28.1% CP, 2.8% EE, 46.0% NDF, 25.5% ADF and 67.2% TDN) ad libitum containing 700 g kg-1 of concentrate ground and 300 g kg-1 of Coast cross hay, in natural matter. In the carcass, the following characteristics were evaluated: hot carcass yield (HCY) and cold (CCY), loss of cooling, loin eye area, degree of finish and proportion of meat cuts. In the meat the physical-chemical and sensorial characteristics: humidity (H), crude protein (CP), ethereal extract (EE), ashes (ASH), pH, color, water retention capacity (WRC), weight loss by cooking (WLC), shear force (SF), aroma, flavor, tenderness, juiciness, chewiness and profile of fatty acids (FA): ω3 (omega-3), ω6 (omega-6), saturated fatty acids (SFA), monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA), ω6/ω3, MUFA/SFA, UFA/SFA, hypocholesterolemic/hypercholesterolemic (HH) ratios and atherogenicity (AI) and thrombogenicity (TI) index. The data collected were analyzed using statistical software SAS version 9.4 by the application of PROC GLM, at the level of 5% of significance. The GS CL compared to SL, provided increases in HCY (48.7 vs 47.5%) and CCY (47.7 vs 46.5%). WS 40 kg showed HCY and CCY of 49.6 and 48.6% higher than WS 20 and 30 kg, which presented values of 46.8 and 45.5% and 47.9 and 47.1%, respectively. For the early developmental cuts (palette and leg), WS 20 kg presented superior results in relation to 40 kg. However, it presented a decrease in relation to the late developmental cuts (neck and rib) when compared to WS 40 kg. The loin was not influenced by GS and WS. Male goats had superior HCY of females (49.14 vs 48.21%), followed by larger proportions of neck and shoulder cuts. The GS CL in relation to SL increased EE (2.12 vs 1.74%) and decreased CP (17.74 vs 18.10%) of meat. GS SL presented lower SFA (35.24 vs 38.82%), IA (0.45 vs 0.69) and TI (1.11 vs 1.34) in relation to CL. WS 20 kg presented higher H and lower MM in relation to WS 40 kg and lower CRA in relation to WS 30 kg. The WS 20 kg presented increase in PUFA and ω3 when compared to WS 40 kg. Male goats presented superior results for CP, pH, PUFA and ω3 in relation to females. There was no influence of GS in sensory, however WS 20 kg showed higher tenderness, juiciness and chewiness relative to WS 40 kg. It is recommended that crossbred goats kids should be slaughtered up to 20 kg, weaned at 60 days, in order to obtain better quality of carcass and meat. / FAPESP: 16/14512-1

O nitrogênio pode ser perdido facilmente no solo, seja por volatilização ou por lixiviação, sendo indicado que seu fornecimento ocorra juntamente com a aplicação de materiais ricos em matéria orgânica, parte no plantio e parte em cobertura. Diante do exposto, objetivou-se com este estudo avaliar o efeito das doses e épocas de aplicação de torta de mamona em cobertura na produção, características físico-químicas e acúmulo de macronutrientes na cultura da couve-flor em manejos orgânicos. Foram realizados dois experimentos um no município de São Manuel, SP, e o outro em Botucatu, SP. Em São Manuel, SP foram estudados 14 tratamentos, em esquema fatorial 3 x 4 + 2, sendo três doses de torta de mamona (2150, 3225 e 4300 kg ha-1), combinadas com quatro parcelamentos em cobertura (¼ + 2/4 + ¼; 1/3 + 1/3 + 1/3 e 1/3 + 2/3 e 2/3 + 1/3), além de duas testemunhas (sem adubação em cobertura e com adubação inorgânica). Em Botucatu, SP, foram 10 tratamentos, em esquema fatorial 3 x 3 + 1, sendo três doses de torta de mamona (1290, 2580 e 3870 kg ha-1) combinadas com três épocas de aplicação em cobertura (única aos 30 ou aos 45 dias após o transplante (DAT) e parcelada aos 30 e 45 DAT), além de uma testemunha (sem adubação em cobertura). Os experimentos foram no delineamento experimental de blocos ao acaso, com quatro repetições. Foram analisadas as seguintes características: ciclo (DAT), número de folhas, diâmetro e altura da inflorescência, massa da matéria fresca da inflorescência e massa da matéria fresca da parte vegetativa (caule e folhas), pH, acidez titulável (AT), ácido ascórbico (AA), sólidos solúveis (SS), índice de maturação (IM)/“Ratio”, teores de açúcares redutores (AR), proteína, massa da matéria seca, teor e acúmulo de macronutrientes na inflorescência da couve-flor. Foram realizadas análises de variância, de regressão, teste de Tukey e de Dunnet. Em São Manuel, obteve-se aumento linear da massa fresca da inflorescência quanto maior a dose de torta de mamona, enquanto para a altura e o diâmetro da inflorescência o efeito foi quadrático, com máximos valores estimados para as doses de 4300 e 3856 kg ha-1, respectivamente. Os fatores não afetaram o teor de sólidos solúveis, entretanto, o aumento das doses elevou os valores de pH, índice de maturação, proteína bruta e teor de açúcares redutores, enquanto reduziu a acidez titulável na inflorescência. A ordem decrescente dos nutrientes acumulados na inflorescência foi K>N>P≈Ca>Mg>S com máximas de 1,99; 1,94; 0,28; 0,27; 0,12 e 0,09 g planta-1. Em Botucatu, a máxima massa fresca da inflorescência foi estimada com a dose de 3015 kg ha-1, enquanto a altura e o diâmetro máximos foram atingidos nas doses de 3785 e 3335 kg ha-1, respectivamente. Os valores de sólidos solúveis e proteína bruta não foram afetados pelos fatores avaliados, enquanto que o aumento das doses de torta de mamona em cobertura elevou os valores de acidez titulável e reduziu o de açúcares redutores, ácido ascórbico e o índice de maturação da inflorescência de couve-flor. A ordem decrescente dos nutrientes acumulados em Botucatu foi K>N>P≈Ca>Mg>S, com máximas de 1,48; 1,03; 0,15; 0,14; 0,08 e 0,06 g inflorescência-1. / Currently, conventional cauliflower cultivation is still predominant; however, cultivation in alternative systems, such as organic, has been growing. Compared to other brassicas, it is the most demanding in fertile soils with a high content of organic matter. Nitrogen and potassium are the most accumulated nutrients in cauliflower. Nitrogen can be easily lost in the soil, either by volatilization or by leaching, and it is indicated that its supply occurs along with the application of materials rich in organic matter, part in planting and part in cover. In view of the above, this study aimed to evaluate the effect of doses and times of application of castor bean cake on yield, physical-chemical characteristics and accumulation of macronutrients in cauliflower crop in organics managements. Two experiments were carried out, one in the municipality of São Manuel, SP, and the another in Botucatu, SP. In the experiment 1 (São Manuel, SP), 14 treatments were studied in a 3 x 4 + 2 factorial scheme, with three doses of castor bean (2150, 3225 and 4300 kg ha-1), combined with four installments (¼ + 2/4 + ¼; 1/3 + 1/3 + 1/3; 1/3 + 2/3 and 2/3 + 1/3), besides two controls (without cover fertilization and with inorganic fertilization). In experiment 2 (Botucatu, SP), 10 treatments were used, in a 3 x 3 + 1 factorial scheme, three castor bean doses (1290, 2580 and 3870 kg ha-1) combined with three installments (one application at 30 or at 45 days after transplanting (DAT) and parceled at 30 and 45 DAT), in addition to one control (no fertilization in coverage). The experiments were in a randomized complete block design, with four replications. The following characteristics were analyzed: cycle (DAT), number of leaves, inflorescence diameter and height, inflorescence fresh matter weight and fresh matter weight of the vegetative part (stem and leaves), pH, titratable acidity (TA), acid ascorbic acid (AA), soluble solids (SS), maturation index (MI) / "Ratio", reducing sugars (RS), protein, dry weight of inflorescence, macronutrient content and accumulation in cauliflower inflorescence . Analyzes of variance, regression, Tukey and Dunnet tests were performed. It was observed that, for most of the characteristics, the best dose ranged between 3.2 and 3.9 t ha-1 of castor bean cake in coverage for both sites, whereas the parceling only had an effect on the cultivation of São Manuel, where 2/3 + 1/3 of the total dose applied at 20 and 40 DAT increased the fresh weight of the inflorescence. In general, the effects were more pronounced in S. Manuel, where soil is poorer in organic matter. Regarding the physico-chemical characteristics, it was verified that only pH was influenced by the splitting in both places. For the experiment carried out in São Manuel, the increase in doses reduced only the titratable acidity, while the other characteristics (SS, pH, RS and MI) increased. On the other hand, in Botucatu, TA increased with increasing doses and AA, RS and MI reduced. The difference in values between the sites was minimal, indicating that for these physico-chemical characteristics, the difference in organic matter content in the soil had little influence. The decreasing order of nutrients accumulated in the inflorescence in São Manuel and Botucatu was K>N>P≈Ca>Mg>S and K≈N>P≈Ca>Mg>S, respectively, with maximums of 1.99; 1.94; 0.28; 0.27; 0.12 and 0.09 g plant-1 in São Manuel and 1.48; 1.03; 0.14; 0.15; 0.08 and 0.06 g inflorescence-1 in Botucatu.

Características fisico-químicas e celulares na secreção láctea de caprinos da raça Saanen durante o processo de secagem da glândula mamária / Physico-chemical and cellular characteristics on milk secretion of Saanen goats during the mammary gland drying off

Para avaliar a influência do processo de secagem da glândula mamária sobre as características físico-químicas e celulares da secreção láctea foram avaliadas 216 amostras provenientes de 27 glândulas mamárias de 15 cabras da raça Saanen consideradas sadias, sendo que essas mamas foram separadas de acordo com o resultado do exame microbiológico da secreção láctea, caracterizando dois grupos: com e sem isolamento bacteriano, composto por 17 e 10 mamas, respectivamente. A partir do dia da interrupção da ordenha e em mais sete momentos (1, 3, 5, 7, 10, 15 e 30 dias após o início da secagem) foi feita a inspeção e a palpação das glândulas mamárias e posterior avaliação do aspecto da secreção láctea. Imediatamente após o exame clínico foram colhidas amostras de leite para a realização do exame microbiológico e das determinações das características físico-químicas e celulares (pH, eletrocondutividade, cloreto, lactose, proteínas, gordura, sólidos totais, índice cloreto/lactose, California Mastitis Test (CMT) e contagem de células somáticas). As glândulas mamárias apresentaram sua maior distensão no terceiro dia após a interrupção da ordenha, mesmo período em que foi possível observar o aparecimento de edema na base dos tetos e em toda a glândula, desaparecendo após o quinto dia, quando teve início a involução do úbere e a freqüência de mamas distendidas começou a declinar, sendo que ao final do processo 37,04% das mamas observadas eram classificadas como não distendidas. A secreção láctea inicial com aspecto de leite foi substituída a princípio por uma de coloração creme clara e mais tardiamente por uma de aspecto aquoso, sendo que ao final do processo as duas últimas eram mais observadas que a com aspecto de leite. Apenas o pH, a eletrocondutividade, os teores de cloretos, lactose, proteínas e índice cloreto/lactose sofreram uma significativa influência do processo de secagem, sendo que os valores do pH, eletrocondutividade, cloretos, proteínas e índice cloreto/lactose aumentaram e os valores de lactose declinaram. O CMT também sofreu uma influência do processo de secagem, mais especificamente as respostas \"traços\" e \"duas cruzes\". Estas, de maneira geral, aumentaram no decorrer da secagem, sendo que no último momento de colheita ocorreu uma queda na freqüência da resposta \"traços\" e a freqüência da resposta \"duas cruzes\" teve uma pequena diminuição no meio do processo. Ainda foi observado que, apesar de existirem diferenças estatísticas significativas entre valores das características físico-químicas e celulares das amostras de secreção láctea com e sem isolamento bacteriano, as diferenças são isoladas e limitadas a algumas características estudadas, impossibilitando afirmar que o processo de secagem dos dois grupos difere de maneira significativa. / To evaluate the influence of the mammary gland drying off on the physiochemical and cellular characteristics in the milk secretion they were appraised 216 coming samples of 27 mammary glands of 15 Saanen goats considered healthy, those mammas were separate in agreement with the result of the microbiological test of the milky secretion characterizing two groups: one with 17 mammas without bacterial isolation and other with 10 mammas that presented positive bacterial isolation. Starting from the day of the interruption of the milking and in more seven moments (1, 3, 5, 7, 10, 15 and 30 days after the beginning of the drying off) it was made the inspection and palpation of the mammary glands with subsequent evaluation of the aspect of the milky secretion. Immediately after the exam they were picked samples of milk for the accomplishment of the microbiological test and of the determinations of the physiochemical and cellular characteristics (pH, electrical conductance, chloride, lactose, proteins, fat, total solids, chloride/lactose ratio, California Mastitis Test (CMT) and somatic cell count). The mammary glands presented its largest distention in the third day after the milking interruption, same period in that was possible to observe the edema emergence in the teats base and in the whole gland, that disappeared after the fifth day, when involution of the udder begins and the frequency of distended mammas began to refuse, and at the end of the process 37,04% of the observed mammas they were classified as no distended. The initial milky secretion with aspect of milk was substituted at first for one of coloration cream clear and, so late, for one of aqueous aspect, and in the end of the process those two last they were more observed that the secretion with aspect of milk. Just the pH, electrical conductance, chloride, lactose, proteins and chloride/lactose ratio suffered a significant influence of the drying off, and the values of the pH, electrical conductance, chloride, proteins and chloride/lactose ratio increased and the lactose values refused. CMT also suffered an influence of the drying off, more specifically the answers \"lines\" and \"two crosses.\" These, in a general way, increased in elapsing of the drying off, and in the last moment studied it happened a fall in the frequency of the answer \"lines\" and the frequency of the answer \"two crosses\" had a small decrease in the middle of the process. It was still observed that in spite of significant statistical differences exist among values of the physiochemical and cellular characteristics of the samples of milky secretion with isolation and without bacterial isolation, the differences are punctual and limited to some studied characteristics, disabling to affirm that the process of drying of the two groups differs in a significant way.

La pollution odorante est une problématique majeure, deuxième source de plaintes après le bruit. La réduction des émissions d'odeurs est incontournable, contraignant le plus souvent les industriels à la mise en œuvre d'un procédé de traitement performant et économiquement acceptable. Parmi les techniques disponibles, les procédés biologiques en général et la biofiltration en particulier sont les plus utilisés à l'échelle industrielle. Les performances de ce procédé sont étroitement liées aux paramètres opératoires. Le matériau support apparaît comme un élément clé. Du fait des critères qu'il doit satisfaire, son choix est difficile et reste encore empirique. Ces travaux de thèse visent par conséquent à rationaliser le choix d'un matériau en s'intéressant plus particulièrement à la relation entre les caractéristiques physico-chimiques et hydrodynamiques du matériau support et les composantes macroscopique et microscopique du procédé. Trois matériaux modèles ont été sélectionnés : deux granulaires et un fibreux différant par leur propriétés physico-chimiques et hydrodynamiques. Les travaux ont été menés sur des unités pilote alimentées par un effluent gazeux contenant un mélange de trois composants volatils différant par leur solubilité et leur biodégradabilité. Si une influence des caractéristiques physico-chimiques du matériau a bien été observée, il n'a pas été possible de mettre en évidence un effet des propriétés hydrodynamiques. Concernant la composante microbiologique, le niveau de diversité global est similaire, quel que soit le matériau considéré. L'analyse de la structure de la communauté bactérienne totale met en évidence trois communautés distinctes, chacune étant associée à un matériau support. Il est fort probable que la communauté endogène participe largement à l‘établissement de ces communautés, rendant ainsi plus difficile de répondre aux questions posées. La communauté bactérienne totale ne semble donc pas être un indicateur adapté. Par le suivi de la communauté fonctionnelle, des réponses plus précises pourraient être apportées. Par conséquent, la recherche de niches écologiques spécifiques à la dégradation biologique d'un composé soufré (DMS) a fait l'objet d'une attention particulière dans le cadre de cette thèse. / Odour nuisance is a major issue, second cause of complaints after noise. Reduction of odor emissions is essential, binding the industry to implement a process for an economically acceptable treatment in terms of reduction of odours. Among the available techniques, biological ones and particularly biofiltration are the most used on industrial scale. The performances of this process are closely related to operating conditions. Packing material appears as a key factor, its choice is difficult and remains empirical because of the multiple criteriait must satisfy. This thesis work aimed to streamline the selection of a material with a focus on the relationship between physico-chemical and hydrodynamic caracteristics of the packing material and macroscopic and microscopic components of this process. Three materials were selected as models: two granulars and one fibrous differing in their physico-chemical and hydrodynamic caracteristics. The work was carried out on pilot units fed by an effluent gas containing a mixture of three volatile components differing in their solubility and biodegradability. The influence of physico-chemical characteristics of the packing material on the performances seems to be confirmed. However, the impact of the hydrodynamic characteristics of the material on these criteria has not been demonstrated. Regarding the microbiological component, the overall level of diversity is similar, regardless of the material considered. Analysis of the structure of the total bacterial community reveals the existence of three distinct communities, each associated with a carrier material. It is likely that the endogenous community participates largely to the establishment of these communities, making it difficult to give appropriate answers to the questions adressed. The total bacterial community does not seem to be a suitable indicator. By monitoring the functional community, more precise answers could be given. Therefore, the research for specific ecological niches in the biological degradation of a sulfur compound (DMS) has been the subject of special attention in the context of this thesis.
