A ?ltima d?cada foi particularmente favor?vel ao reconhecimento da exist?ncia do processo que leva um contingente cada vez maior de pessoas a viver em situa??o de mobilidade, fruto de fen?menos universais de ordem econ?mica, ecol?gica e sociopol?tica. Neste contexto s?o elaborados diversos esfor?os para possibilitar compreens?o e/ou esclarecimentos sobre a popula??o em situa??o de mobilidade enquanto sujeitos de direitos organizados em torno de suas pr?prias demandas. Tradicionalmente entendidos como inaptos para a organiza??o pol?tica, passam a elaborar suas pr?prias reivindica??es, de modo a obter reconhecimento como um segmento que necessita ser atendido segundo as especificidades deste modo de vida. A partir da pesquisa etnogr?fica iniciada em 2015, na cidade de Santo Ant?nio e Serrinha, cidades localizadas na Regi?o denominada pelo IBGE de Agreste do RN, a proposta tem enfoque particular em eventos recentes e na mem?ria de agentes envolvidos com o processo de mobilidade da popula??o do Agreste do Rio Grande do Norte. Por fim, trata-se de indicar apontamentos sobre como a pr?tica de viver em mobilidade, entendida por muito tempo como ?processo migrat?rio?, se transforma em objeto de pol?ticas p?blicas estatal a partir da mobiliza??o coletiva para o reconhecimento de suas especificidades. / Beaucoup a ?t? dit ? propos de l'?volution historique du ph?nom?ne qui aujourd'hui nous nous r?f?rons ? la mobilit?, que ce soit dans le diff?rend domestique ou dans des proportions plus importantes, relier les gens avec des endroits nombreux dans le monde, ce qui rend la d?finition de la mobilit? un sujet de d?bat et de controverse. La derni?re d?cennie a ?t? particuli?rement favorable ? la reconnaissance de l'existence du processus qui conduit un nombre croissant de personnes vivant dans une situation de mobilit?, le r?sultat de ph?nom?nes universels des droits ?conomiques, ?cologiques et socio-politique. Dans ce contexte, il est fait divers efforts pour permettre la compr?hension et / ou une clarification de la population en situation de mobilit? comme sujets de droits organis?s autour de leurs propres exigences. Previous issue date: 2017-05-03 / Este estudo tem por tema o Programa de Educação em Tempo Integral do município de Vitória. Quanto ao objetivo geral, pretendeu-se investigar como tem ocorrido a implementação desse programa nesse município, entre 2004 e 2016, buscando analisar as possibilidades e desafios no desenvolvimento de práticas educativas voltadas para educação em tempo integral. Com esse propósito, atuou-se no contexto da escola acompanhando o desenvolvimento escolar de um grupo de alunos atendidos pelo programa. O campo empírico da pesquisa foi um centro municipal de educação infantil, que presta atendimento em tempo integral a um grupo de 20 crianças, com faixa etária de 3 e 4 anos. Metodologicamente recorreu-se à pesquisa-ação como orientadora do percurso e à utilização do questionário, entrevistas semiestruturadas e observação em contexto como procedimentos de coleta de dados. Com base nesses instrumentos, procedeu-se à sistematização dos dados por meio de registros fotográficos, gravações de voz, registros das observações no diário de campo e pesquisa documental. Para o embasamento teórico do estudo, optou-se por usar a teoria sociológica figuracional de Norbert Elias, que contribui para as análises, apoiadas em conceitos centrais, como civilização, figuração, interdependência. Para Elias, as sociedades se estruturam com diferentes relações de interdependência, as quais produzem as mais diversas figurações sociais, cujas alterações são resultado de outras relações de interdependência existentes em seu interior. Nessa perspectiva, procedeu-se à análise do Programa de Educação em Tempo Integral no município de Vitória-ES. Os dados da pesquisa revelam a necessidade de tal programa passar por mudanças significativas de diferentes naturezas, entre as quais questões de concepção, questões estruturais e econômicas. De modo geral, observa-se que a perspectiva assistencialista ainda é predominante nos discursos e existe a necessidade de compreensão, por parte dos envolvidos, do que vem a ser um projeto educativo de atendimento à educação integral, pois somente a ampliação do tempo de escola não transforma por si mesma a qualidade ou quantidade de oportunidades oferecidas aos estudantes.

Changes in the Rpb3 Interactome Caused by the Deletion of RPB9 in Saccharomyces cerevisiaeTalbert, Eric A. 02 August 2016 (has links)
Indiana University-Purdue University Indianapolis (IUPUI) / RNA Polymerase II (Pol II) is the primary actor in the transcription of mRNA from genes. Pol II is a complex composed of twelve protein subunits. This study focused on the changes in the interactome of Rbp3 in S. cerevisiae when the Pol II subunit Rpb9 is removed. Rpb3 is one of the core subunits of Pol II, and any significant changes to the Rpb3 incteractome due to the loss of Rpb9 can be used to infer new information about Rpb9’s role in the Pol II complex.
Rpb3 was pulled down using FLAG purification from both wild type and rpb9Δ S. cerevisiae cultures. Rpb3 and the proteins complexed with it were then analyzed using multi-dimensional protein identification technology (MudPIT), a form of liquid chromatography-mass spectrometry (LC-MS). This data was searched using the SEQUEST database search algorithm, and the results were further analyzed for likelihood of interaction using Significance Analysis of INTeractome (SAINT), as well as for post-translational phosphorylation. Deletion of rpb9 did not present any changes in Pol II phosphorylation however it did cause several changes in the interaction network. The rpb9Δ strain showed new interactions with Rtr1, Sen1, Vtc4, Pyc1, Tgl4, Sec61, Tfb2, Hfd1, Erv25, Rib4, Sla1, Ubp15, Bbc1, and Hxk1. The most prominent of these hits are Rtr1, an Rpb1 C-terminal domain phosphatase linked to transcription termination, and Sen1, an RNA/DNA nuclease that terminates transcription. In addition, this mutant showed no interaction with Mtd1, an interaction that is present in the wild type. In all cases, these hits should be considered fuel for future research, rather than conclusive evidence of novel interactions.

Molecular basis of viroid RNA-templated transcriptionDissanayaka Mudiyanselage, Shachinthaka D 09 December 2022 (has links) (PDF)
Transcription is a fundamental process catalyzed by DNA-dependent RNA polymerases (DdRPs). Interestingly, some DdRPs can use both DNA and RNA as templates for transcription. This RNA-dependent RNA polymerase (RdRP) activity of DdRPs is used by RNA-based pathogens such as viroids and hepatitis delta virus for replication. In addition, RdRP activity of DdRPs widely occurs in various organisms to regulate gene transcription. Despite the importance of this intrinsic RdRP activity of DdRPs, associated factors and mechanisms are in their infancy stage. We employed potato spindle tuber viroid (PSTVd) as a model to study RNA-templated transcription. Here, we present evidence showing that circular PSTVd templates are critical for the synthesis of longer-than-unit-length (-) strand products. Further, we show transcription factor IIS is dispensable for PSTVd replication supporting de novo transcription on PSTVd RNA templates. The absence of canonical general transcription factor, TFIIS from PSTVd-templated transcription complex led to the hypothesis that RNA-templated transcription has a distinct organization on the RNA template. To test this hypothesis, we used our well-established in vitro transcription (IVT) system and demonstrated that RNA polymerase II (Pol II) accepts minus-strand for transcription. In addition, transcription factor TFIIIA-7ZF is needed to aid Pol II transcription activity. Further analyses of the critical zinc finger domains in TFIIIA-7ZF revealed that the first three zinc finger domains are pivotal for template binding. Notably, we identified a remodeled Pol II complex for viroid transcription that is missing Rpb4, Rpb5, Rpb6, Rpb7, and Rpb9. General transcription factors for DNA-templated transcription are also absent in the transcription complex on the RNA template. This remodeled Pol II complex still possesses the transcription activity on PSTVd RNA template. Collectively, our data illustrate a distinct organization of Pol II complex on viroid RNA templates, providing new insights into viroid replication, the evolution of transcription machinery, as well as the mechanism of RNA-templated transcription.

Forecasting The Onset Of Cloud-ground Lightning Using S-pol And Nldn DataRamakrishnan, Kartik 01 January 2004 (has links)
The maximum number of thunderstorms in the United States occur in Central Florida. The cloud-ground lightning from these storms is responsible for extensive damage to life and property. The lightning from these storms is also responsible for delays and cancellations of space shuttle launch attempts at the Kennedy Space Center (KSC) and the 45th Space Wing unmanned launches at Cape Canaveral launch facilities. For these and other reasons accurate forecasting of cloud-ground lightning is of crucial importance. The second phase of NASA's Tropical Rainfall Measuring Mission Texas and Florida Underflights project (TEFLUN-B) was conducted between 1st August and 30th September, 1998. The S-band dual-polarization radar (S-Pol) belonging to the National Center for Atmospheric Research (NCAR) was part of the surface based facilities during this project, and was located at Melbourne, Florida. This provided an excellent opportunity to observe Florida thunderstorms with the help of a dual-polarization radar. This project aims at developing cloud-ground lightning forecasting signatures by analyzing S-Pol data for 10 thunderstorms that occurred over the Kennedy Space Center. Time-height trends of reflectivity, ice and graupel-hail as well as electric potential trends for these storms are taken into consideration while developing the forecasting signatures. This thesis proposes that a 35dBZ echo at the -5[degree]C temperature level is the best indicator of imminent CG lightning with a POD of 90%, an FAR of 10% and a CSI of 81.8%. An electric potential level of approximately 1000 V/m also indicates the onset of cloud-ground lightning. An analysis of the microphysical structure of the thunderstorms reveals that the presence of graupel-hail at the -10[degree]C temperature level is necessary in order for cloud-ground lightning to occur.

The nonlinearity is of the zero-memory symmetric-cut-off type following a cube law.</p> <p> An experimental oscillator was built, and curves of the frequency shift of the oscillator fundamental against the magnitude of the input forcing signal are shown, both for a sinusoidal input and for a narrow band noise input. An empirical result is derived.</p> <p> The case of the sinusoidal input is examined theoretically. Thermal and Electrical Considerations for the Design of Highly-Integrated Point-of-Load ConvertersBall, Arthur 11 May 2009 (has links)
DC/DC Power converter design has been following a trend of reducing size while also increasing performance for the last several years. This push for higher power output and smaller footprint and profile requires integration and higher switching frequencies in order to continue. Higher frequencies require physical integration to eliminate problems induced by parasitics, which increase losses.
GE's Power Overlay and Philip's PCB integration schemes have been clear steps in the quest to reduce size with new system design techniques. However, both have downsides. GE Power Overlay embeds the devices inside a milled AlN ceramic cavity and then layers interconnections on top using polyimide dielectric interlayers. The milling of AlN ceramic is a very costly and time consuming task due to the brittleness of the material, and the interlayers add additional complexity to the fabrication process.
Philip's PCB integration was primarily aimed at integrating passives along with the PCB process for reduction of size. Inductor windings and capacitive layers were built up along with FR4 epoxy layers using typical PCB fabrication methods. However, unlike GE's Power Overlay, the substrate material was several times lower in thermal conductivity which invariably has corresponding thermal penalties.
The work presented here reconciles the good of both integration techniques. Initially called Embedded Power, alumina ceramic was used as the substrate and rather than milling holes for the devices, holes were laser cut all the way through and interconnections were made by using interlayers and sputtered copper deposition, similar to GE's method. Integration of passives was done using LTCC ferrite to make an inductor of thin profile, rather than embedding cores and windings inside PCB. However, fabrication remained time consuming due to numerous solder masking and sputtering steps and thermal performance was not optimized due to the use of alumina ceramic.
A revised design method called Stacked Power is presented in this dissertation that follows on the work of Embedded Power, but improves on it by simplifying fabrication through the elimination of thermally-restrictive interlayers, as well as time consuming sputtering and electroplating of copper interconnections. Instead, AlN Direct Bonded Copper is used as a multifunctional material thanks to its many-times-greater thermal conductivity than PCB or alumina, solderable device dies are implemented in a vertical fashion, and interconnections are simply made using copper straps soldered into place. For applications where moisture contamination and breakdown isolation are potential problems, dip conformal coating can easily be applied, replacing laborious solder masking.
The work in this dissertation describes the fabrication methodology for Stacked Power, demonstrates the thermal advantages, and shows examples of high-frequency buck converters that achieve super-high levels of power density in the smallest of volumes and require no more thermal management than modest airflow. The added cost incurred with aluminum nitride is traded for distinct advantages in terms of low-profile, low airflow requirements for the power output, capability of natural convection for use in locations where fans are prohibitive and compact size for ease of implementation. / Ph.
