Detecção e expressão dos genes supressores p53 E c-Myc em tumores palpebrais de cães

Foram utilizadas 10 amostras de tumores que foram fixados em formol e incluídos em parafina. O material foi obtido junto aos arquivos do Serviço de Patologia Veterinária, sendo nove amostras de tumores localizados nas pálpebras e terceira pálpebra e uma de tumor mamário para controle. Todos os tumores tiveram o seu diagnóstico firmado empregando-se a coloração de H.E e imunoistoquímica para citoqueratina AE1/AE3 e vimentina (V9), marcadores de tecido epitelial e mesenquimal, respectivamente. Os resultados indicaram que os tumores palpebrais e da terceira pálpebra aqui estudados verificou-se a presença do gene supressor p53 em 8 amostras (88,8%, n=8), e entre as amostras positivas (n=8), ele esteve expresso em 75 % delas. O gene supressor c-Myc esteve presente em 5 amostras (55,5%) e com expressão em 100% delas (n=5). Foi possível concluir que os tumores palpebrais e da terceira pálpebra de cães expressam o p-53 e c-Myc identificados pelas técnicas de PCR e RT-PCR, no entanto, as técnicas de PCR ELISA e RT-PCR ELISA foram mais importantes para avaliação da presença e expressão do oncogenes estudados, pois permitiram identificar produtos amplificados que não foram visualizados em gel de agarose. / The aims of this study were to detect the presence and the expression of p53 and c-Myc suppressor genes in eyelid tumors of dogs, by the PCR, RT-PCR, PCR-ELISA and RT-PCR-ELISA techniques, which until then they were not described in these tumors and in this specie. Ten samples of tumors were fixed in formalin and included in paraffin. The material was obtained from the archives of the Department of Veterinary Pathology, being nine samples of epithelial tumors located in the eyelids and the third eyelid, and a breast tumor which was used as a positive control of the reactions. All the samples had reached their diagnosis employing up the HE technique, and the immunohistochemistry for cytokeratin AE1/AE3 and vimentin (V9). The results showed that the eyelid and the third eyelid tumors, here studied, (88.8%, n=8) of them demonstrated the presence of the p53 gene and between the positive samples (n=8), the expression was around 75%. The c-Myc gene was present in 55.5% (n=5) of the samples, with 100% of expression (n=5). Thus, it was possible to conclude that the eyelid and the third eyelid tumors of dogs express the p53 and c-Myc genes, identified by the techniques of PCR and RT-PCR, however, the PCR ELISA and RT-PCR ELISA techniques were of extreme importance for assessing the presence and expression of these studied genes, and they allowed to identify amplified products that were not visible on the electrophoresis on the agarose gel.

Adicionalmente, objetivou-se a realização do diagnóstico molecular de C. felis e estabelecimento do diagnóstico diferencial da clamidiose felina em relação à infecção pelo herpesvírus felino tipo 1 (HVF-1), vírus da Imunodeficiência felina (FIV) e vírus da leucemia felina (FeLV). Anticorpos anti-Chlamydiaceae foram detectados pela RIFI COMERCIAL em 81% dos animais (162/201), pela RIFI OVO em 55% (111/201) e pelo ELISA em 31% (69/201) das amostras. Verificou-se que o antígeno de Chlamydophila abortus inoculado em ovos embrionados não é indicado para utilização em técnicas imunológicas, devido à baixa sensibilidade, especificidade e correlação entre os testes estudados. Das amostras de suabes de conjuntiva submetidas à PCR, em 5,1% (12/235) foi amplificado o gene MOMP de C. felis e três amostras (1,3%) foi detectado DNA de herpesvírus. Houve coinfecção para os dois agentes em 0,4% (1/235) das amostras analisadas. Ademais, 150 amostras de soro foram avaliadas, sendo anticorpos anti-FIV detectados em 8 amostras (5,3%) e o antígeno de FeLV detectado em 11 amostras (7,3%). Não houve coinfecção por C. felis e HVF-1 com os vírus imunossupressores, demonstrando não haver correlação entre a infecção de FIV e/ou FeLV e o complexo respiratório felino / This study aimed at perfoming the serological diagnosis of Chlamydophila felis by three serological approaches, using a commercial kit for Indirect Fluorescent Antibody Test (IFAT COMMERCIAL), and Indirect Fluorescent Antibody Test (IFAT EGG) and ELISA prepared with Chlamydophila abortus with antigen produced in SPF embryonated chicken eggs. Additionally, our study aimed at performing the molecular diagnosis of C. felis and establishing the differential diagnosis of feline chlamydiosis in relation to Feline Herpesvirus type-1 (HVF-1) in cat conjunctival swab samples, as well as investigate the presence of antibodies to Feline Immunodeficiency Virus (FIV) and presence of Feline Leukemia Virus (FeLV) antigen in cat serum samples. IgG antibodies to Chlamydophila sp. were detected by in 81% (162/201), 55% (111/201), and 31% (69/201) of serum samples, by IFA Chlamydiaceae COMMERCIAL, IFA EGG and ELISA, respectively. Os objetivos deste ensaio foram os de identificar a espécie Leishmania (Leishmania) chagasi, utilizando o método PCR-RFLP em amostras de tecido de cães com leishmaniose visceral; seqüenciar o fragmento amplificado de DNA das amostras de linfonodos de diferentes animais, em busca de homologia e polimorfismos; comparar os métodos de imunoistoquímica e de PCR dos rins, na identificação da Leishmania e comparar as técnicas sorológicas em relação à PCR como auxílio diagnóstico da leishmaniose. Para tanto, foram utilizadas amostras de 48 cães com leishmaniose, diagnosticados por meio de testes IFI, ELISA e por PCR. Realizou-se PCR com amostras de linfonodo poplíteo, baço e rins, utilizando ‘primers’ específicos para o gênero Leishmania. A partir de amostras amplificadas, o DNA foi submetido à digestão enzimática (técnica PCR-RFLP) com as enzimas Hae III, Bsr I e Rsa I. Para o seqüenciamento de DNA, produtos de PCR foram amplificados com ‘primer sense’, precipitados e submetidos ao seqüenciamento automático. Com fragmentos histológicos renais, realizou-se a técnica imunoistoquímica utilizando anticorpo policlonal anti-Leishmania produzido em coelho. O método PCR-RFLP permitiu identificar a espécie Leishmania (Leishmania) chagasi em todas as amostras de DNA dos diferentes tecidos. No seqüenciamento de DNA, os fragmentos amplificados mostraram-se pertencentes às espécies causadoras de leishmaniose visceral, porém não foi possível diferenciá-las. A técnica de imunoistoquímica mostrou presença de formas amastigotas íntegras em meio ao infiltrado inflamatório intersticial em dois (4%) dos 48 animais avaliados, enquanto a PCR confirmou Leishmania sp em 77% dos cães. Conclui-se que a técnica... / Leishmaniasis is a disease caused by organisms belonging to the genus Leishmania. Clinical forms of leishmaniasis are found as being cutaneous, mucocutaneous or visceral. The objectives of the present study were to identify the Leishmania (Leishmania) chagasi species in tissue specimen from dogs presenting visceral disease diagnosed by PCR-RFLP assay; to determine the fragment sequence (120bp) from lymph node samples from different animals searching for homologies and polymorphism; and to compare immunohistochemistry method to PCR assay with renal tissue and Leishmania local identification. Forty eight samples from dogs clinically diagnosed positive to leishmaniasis by IFAT, ELISA and PCR assays were used in this study. The PCR were performed with samples of popliteo lymph node, spleen and kidneys, and specific oligonucleotides to genus Leishmania. PCR products were digested with enzymes Hae III, Bsr I and Rsa I. The nucleotide sequences were determined automatically. For immunohistochemical purposes the sections of kidneys and anti-Leishmania polyclonal antibody were used. PCR-RFLP assay allowed us to identify the Leishmania (Leishmania) chagasi specie in all DNA samples from different tissues samples. DNA sequencing revealed products amplified belonging to specie responsible to cause visceral leishmaniasis, but it was not possible to distinguish the species. The immunohistochemical study revealed the presence of amastigotes organisms on intersticial inflammatory infiltrate from two dogs (4%), while the PCR assay detected the Leishmania sp in 37 dogs (77%). Based on the PCR-RFLP results, we concluded that it characterized as being Leishmania (Leishmania) chagasi species, etiologic agents of visceral leishmaniasis, in this group of animals; DNA sequence presented homology of 55 bp among all Leishmania spp studied. Ocorrência e caracterização molecular de Cryptosporidium spp. em bezerros leiteiros da região noroeste do Estado de São Paulo

Realizou-se a análise microscópica pela técnica de coloração negativa com verde malaquita em todas as amostras de fezes. Para identificação molecular de Cryptosporidium, utilizou-se a reação de nested PCR, utilizando primers específicos para amplificação de fragmentos do gene codificador da subunidade 18S do RNA ribossômico e do gene da glicoproteína GP 60, submetendo o produto da PCR a sequenciamento e análise filogenética. Por meio de exame de fezes, foi observada positividade de 2% (4/196), por meio de microscopia, e de 10,7% (21/196) pela nested PCR. Como resultado do sequenciamento, foram identificadas quatro espécies de Cryptosporidium: C. parvum (subtipo IIa15G2R1) C. ryanae, C. bovis e C. andersoni. Esta descoberta de C. parvum subtipo IIaA15G2R1 na região estudada ilustra a utilidade da subtipagem. Embora estes resultados iniciais baseados em subtipos com GP60 sejam úteis para compreender a dinâmica de transmissão das espécies de Cryptosporidium, o número de amostras analisadas é relativamente pequeno e muito mais ainda pode ser pesquisado. É interessante notar que as informações de tipagem molecular com o gene 18S rRNA e a subtipagem com o gene GP60, permitem que os epidemiologistas possam rastrear as fontes de surtos das diferentes espécies de Cryptosporidium. / A total of 196 fecal samples from crossbred calves, 7-30 days old of both sexes were collected in 31 properties with the objective of determining the prevalence of Cryptosporidium in the northwestern region of São Paulo, as well as characterizing the molecular species involved in this parasite. We performed microscopic analysis by the technique of negative staining with malachite green in all stool samples. For molecular identification of Cryptosporidium, we used the reaction of nested PCR using specific primers for amplification of gene fragments coding for the 18S subunit ribosomal RNA gene and the glycoprotein GP 60 by subjecting the PCR product sequencing and phylogenetic analysis. By stool examination, positivity was observed 2% (4 / 196) by means of microscopy, and 10.7% (21/196) by nested PCR. As a result of sequencing, we identified four species of Cryptosporidium: C. parvum (subtype IIa15G2R1) C. Ryanair, C. bovis and C. andersoni. This discovery of C. IIaA15G2R1 parvum subtype in the studied region illustrates the usefulness of subtyping. Although based on these initial results with GP60 subtypes are useful for understanding the transmission dynamics of the species of Cryptosporidium, the number of samples is relatively small and much more can still be searched. It is interesting to note that the information of molecular typing with 18S rRNA gene and subtyping with the GP60 gene, allow epidemiologists can track sources of outbreaks of different species of Cryptosporidium.
