It aims to define the range of companies using the methods of intercompany comparison, especially the simple method of ranking, scoring, standard variable and distance from a fictive object. Furthermore the companies are sorted into the groups by the use of the classification analysis and selected outputs graphically depicted using the spider analysis.

Characterization of the large diameter fibers in egg case silk : identification of a core fibroin, TuSp1, and localization of fibroin-like molecules, ECPs, from the black widow spider, latrodectus hesperus

Araneoid spiders use specialized sets of abdominal silk glands to produce up to seven different types of silks, each with diverse functional properties. At the time of these studies, fibroin eDNA sequences that encode egg case silk had not been reported in the literature. This study used conventional nucleic acid-nucleic acid screening of a eDNA library to isolate a novel gene, named tubuliform spidroin 1 , from the black widow spider Latrodectus hesperus. TuSp 1 was demonstrated to be selectively expressed in the tubuliform gland (the gland suspected for egg case silk production), and examination of the amino acid sequence revealed highly homogeneous repeats (184 amino acid ensemble repeats), a characteristic feature of fibroin sequences. Analyses of the ensemble repeats within the amino acid sequence of TuSp 1 revealed the lack of long stretches of polyalanine and glycine-alanine sub-repeats, which are commonly found in minor ampullate and major ampullate silks. Polyserine blocks and short polyalanine stretches were highly represented in the TuSp 1 amino acid sequence. Our data support the assertion that TuSp 1 represents the main constituent within egg case silk. This supposition is supported by the observation that the amino acid composition of raw egg case silk was strikingly similar to the amino acid composition predicted from the translated TuSp1 eDNA. Two additional constituents identified in black widow egg case, egg case protein 1 (ECP-1) and egg case protein 2 (ECP-2), were also partially characterized in this study. Using immunohistochemical approaches, we demonstrate that ECPs predominantly localize to the exterior of the large diameter fibers of egg cases. Additionally, these studies revealed smaller amounts of ECPs localized to the interior portion of the fibers. Collectively, these results support TuSp1 as the predominant fibroin within egg sacs as well as reveal a structural role for the ECPs, providing clues regarding the supramolecular structure of egg case fibers.

Estimating Charging on a Sounding Rocket Experiment Using Plasma Simulation

The aim of this project is to model current volt-age characteristic curves for different plasma conditions (i.eelectron density, electron temperature, ion temperature andplasma potential) that can be found in active auroras. This isdone by simulating the charging of a FFU with a connectedLangmuir probe in the software SPIS. These I-V curves wereused to determine the plasma properties of the auroras in whichthe sounding rockets SPIDER-1 and SPIDER-2 were launched.Through the simulations it was also studied how the differentparameters effects the I-V curves.The results showed that the plasma SPIDER-1 was launchedin most likely had properties close to nominal conditions and forSPIDER-2 there was colder electrons in the plasma. A conclusionthat only the electron temperature affects the shape of the I-Vcurves for the values simulated in this project can be drawnas well as the conclusion that the geometry of the probe doesnot affect the shape of the I-V curves. Another result showsthat electron temperature also affect how the hull of the FFUcharges. A higher electron temperature gives the hull a morenegative charge. / Syftet med detta projekt är att modellera ström-pänningskaraktäristisk kurvor för olika plasmatillstånd som finns i aktiva auroror. Detta görs genom att simulera laddning på en FFU med en ansluten Langmuir-prob i SPIS. Dessa I-V-kurvor används för att bestämma plasma egenskaperna för aurororna sondraketerna SPIDER-1 och SPIDER- 2 skjöts upp i. I-V kurvorna används också för att bestämma hur plasma parametrarna elektron temperatur, jon temperatur och elektrondensitet samt hur probens geometri påverkar I-V- kurvornas utseende. Resultaten visade att den plasma SPIDER-1 blev uppskjuten i troligen hade nominella förhållanden och att den SPIDER-2 blev uppskjuten i troligtvis hade kallare elektroner. En slutsats om att endast elektron temperaturen påverkar formen på IV- kurvorna kan dras, såväl som slutsatsen att probens geometri inte påverkar formen på IV-kurvorna. Ett annat resultat visar att elektron temperaturen också påverkar ytpotentialen på FFUn. Recently, the biotechnological potential of the toxins was explored by its use in clinical test, in the study of inflammatory response, as a research tool in cell biology, as a biopesticide and in immunotherapy, through the production of antiserum. In this context, immunotherapy is broadly spread through the production of vaccines, available for the treatment of deleterious reactions developed in accidents. In the present work, we investigated if immunological tolerance induction to dermonecrotic recombinant toxin (LiRecDT1) and its mutated form (LiRecDT1 H12A), through its oral administration, could modulate inflammatory and deleterious responses triggered by dermonecrotic toxin. For this purpose, an oral tolerance protocol was designed, consisting in the administration of 10 μg of LiRecDT1 and LiRecDT1 H12A three times in a week, for three weeks. Adult Swiss mice were further immunized, and oral tolerance induction was observed by reduction in serum levels of IgG antibody anti-toxin when compared with control group. It was observed that mice tolerant to LiRecDT1 H12A present a reduction in paw edema, caused by the injection of 6 μg of dermonecrotic toxin in plantar surface hind paw. Mice tolerized with LiRecDT1 and challenged with 50 μg of dermonecrotic toxin, exhibited higher survival, when compared to control group. This effect was not observed in mice tolerized to LiRecDT1 H12A. The present findings suggested that oral tolerance induction to LiRecDT1 H12A was able to alleviate inflammatory responses triggered by dermonecrotic toxin in paw edema and oral tolerance to LiRecDT1 increase survivability in challenge. The results shown that LiRecDT1 and LiRecDT1 H12A can be explored as a tool in the induction and study of oral tolerance phenomena. The generation of T regulatory cells (Tregs) and following involvement of immunosuppressive cytokines might take part in the modulation of immune response. / O veneno de aranha-marrom (Loxosceles sp.) apresenta-se como uma mistura complexa de toxina, capazes de causar necrose com espalhamento local, intensa resposta inflamatória, indução de edema e aumento da permeabilidade vascular in vivo. Recentemente, o potencial biotecnológico das toxinas foi explorado através de seu uso em análises clínicas, no estudo da resposta inflamatória, como ferramenta de pesquisa na biologia celular, como biopesticidas e na imunoterapia, através da produção de anti-soro. No presente trabalho, foi investigado se a indução de tolerância imunológica à toxina dermonecrótica recombinante (LiRecDT1) e sua forma mutada (LiRecDT1 H12A), através de sua administração oral, poderia modular as respostas inflamatórias e deletérias causadas pela toxina dermonecrótica. Para tal, foi desenvolvido um protocolo para indução de tolerância oral, consistindo na administração de 10 μg de LiRecDT1 e LiRecDT1 H12A três vezes por semana, durante três semanas. Camundongos Swiss fêmeas adultas foram posteriormente imunizadas, e a indução de tolerância foi confirmada pela diminuição nos níveis de anticorpos IgG anti-toxina em relação ao grupo controle, resultado que aponta a obtenção de sucesso na indução de tolerância imunológica. Observou-se que animais tolerantes a LiRecDT1 H12A apresentaram uma diminuição no edema desenvolvida na pata, causado pela aplicação de 6 μg de LiRecDT1 na superfície plantar traseira. Animais tolerizados com LiRecDT1 e desafiados com 50 μg intraperitoneal de toxina dermonecrótica apresentaram maior índice de sobrevivência, quando comparados ao grupo controle. Esse efeito não foi observado em animais tolerizados com LiRecDT1 H12A. Os dados obtidos no presente trabalho sugerem que a indução de tolerância oral à LiRecDT1 H12A é capaz de atenuar o desenvolvimento da resposta inflamatória no edema de pata, enquanto a tolerância oral a LiRecDT1 foi capaz de aumentar a sobrevivência em animais desafiados com LiRecDT1. Os resultados demonstram que as toxinas LiRecDT1 e LiRecDT1 H12A podem ser exploradas como ferramenta na indução e estudo da tolerância oral. A geração de células T regulatórias (Tregs) e subsequente participação de citocinas imunossupressoras devem estar envolvidas na modulação da resposta imune.

I dagsläget har uppdragsgivaren ingen automatiserad metod för att samla in brutna länkar på deras hemsida, utan detta sker manuellt eller inte alls. Detta projekt har resulterat i en praktisk produkt som idag kan appliceras på uppdragsgivarens hemsida. Produktens mål är att automatisera arbetet med att hitta och samla in brutna länkar på hemsidan. Genom att på ett effektivt sätt samla in alla eventuellt brutna länkar, och placera dem i en separat lista så kan en administratör enkelt exportera listan och sedan åtgärda de brutna länkar som hittats. Uppdragsgivaren kommer att ha nytta av denna produkt då en hemsida utan brutna länkar höjer hemsidans kvalité, samtidigt som den ger besökare en bättre upplevelse. / Today, the customer has no automated method for finding and collecting broken links on their website. This is done manually or not at all. This project has resulted in a practical product, that can be applied to the customer’s website. The aim of the product is to ease the work when collecting and maintaining broken links on the website. This will be achieved by gathering all broken links effectively, and place them in a separate list that at will can be exported by an administrator who will then fix these broken links. The quality of the customer’s website will be higher, as all broken links will be easier to find and remove. This will ultimately give visitors a better experience.

The dissertation work concentrates on an intercompany comparison of Czech Post Office with other companies whose subject of activity are similar and to find out its standing ratio within the evaluation standard of the companies and the best industry values. The theoretical part concentrates on the ways and methods of comparison including the evaluation within its industry, logistics and storage. In the practical part, the analysis of 15 companies is presented based on ratio indicators from 2010 to 2014. Using a cluster analysis tool, the whole file was reduced and then one with the best values was chosen by using a math-statistical method. The last but not least important step was using the Spider analysis to compare the best company, with good average within the industry branch, with the specified company, and on those basics, certain measures were suggested towards the improvement of the company's situation.

Föreliggande kandidatarbete har till syfte att undersöka fenomenet Webskrapning. Webbskrapnings-program (också kända som Web Wanderers, Crawlers, Spiders eller skrapare) är program som söker igenom webben automatiskt för att extrahera information från webbsidor. Ett exempel på web skrapning är när ett företag samlar in data om prissättningar på en vara eller en tjänst och sen använder informationen för att producera billigare erbjudanden. Detta ger företaget en fördel så att de kan fokusera mera på att marknadsföra sin sida/tjänster. Utöver detta så blir de utsatta företagens servrar också hårt belastade med trafik (skrapning) från ”icke kunder”. Efter att ha genomsökt både akademiska och allmänna källor via informationsinsamling, av denna information så dras slutsatsen att man inte fullt ut kan hindra skrapning av hemsidor. Detta på samma sätt som man inte fullt ut kan hindra någon IT-attack, det finns inga 100 % vattentäta system. Av utfallet ifrån informationssökningen var det bara ett akademiskt arbete, av de hundra, som genomsöktes som hade inriktat sig på att förhindra skrapningsbotar.

The first part introduces the topic of this master's thesis and the aim of the thesis. The methodological part describes the economic analysis, its users and sources of information. It also deals with basic methods, such as analysis of absolute, differential and relative indicators, balance sheet rules, summary indexes of ratings, creditworthy and bankruptcy models and comparing the company with the industry through Spider analysis. At the beginning the practical part characterizes analyzed company, then the individual methods of economic analysis applied to a particular corporation. In conclusion the results will be summarized of the economic situation of the corporation.
