Seleção genômica em bovinos de corte / Genomic selection in beef cattle

Objetivou-se com este trabalho: (1) avaliar parâmetros de diversidade genética e de estrutura populacional com base nas raças Angus, Devon, Hereford e Shorthorn. Taxa de endogamia (ΔF), tamanho efetivo (Ne), grau de parentesco, entre outros parâmetros, foram estimados para fornecer subsídios aos programas de melhoramento. Os parâmetros indicaram adequada diversidade genética, com Ne variando entre 128 no Devon e 303 no Shorthorn e ΔF variando entre 1,50 no Angus e 3,92 no Devon; (2) avaliar estratégias de imputação de genótipos utilizando dados de Braford e Hereford através de painéis de baixa densidade (3K, 6K, 8K, 15K e 20K) para os painéis de 50K e 777K. Para o painel de 777K, também foram utilizados na imputação os painéis de 50K, 90iK e 90tK. Os resultados indicaram que, com exceção do painel de 3K, todos os demais painéis de baixa densidade poderiam ser utilizados como base visando à imputação para o painel de 50K e também que os painéis de média densidade (50K, 90iK e 90tK), poderiam ser utilizados como base na imputação para o painel de 777K. Esses painéis mostraram-se eficientes e possuem, em geral, custos compatíveis com a atividade pecuária; (3) avaliar a acurácia de predição dos valores genômicos utilizando alguns painéis de baixa densidade (8K e 15K) imputados para o painel de 50K, relacionando os resultados com o uso do painel original de 50K. A acurácia do valor genômico direto (DGV) e do valor genético genômico (GEBV) com o valor genético (EBV) utilizando painéis imputados ou não, indicaram que não houveram diferenças em acurácia e as perdas em acurácia por utilizar os painéis imputados ficaram entre -0,0002 e -0,0021 dependendo do painel, do cenário e da característica analisada; (4) usar marcadores moleculares na seleção genômica testando dois métodos BLUP (procedimento de passo único ou de multi passo) com dados simulados de bovinos de corte. Os resultados demonstraram, com base nos parâmetros estudados, igualdade de resultados entre os dois procedimentos; (5) avaliar a viabilidade do uso da seleção genômica usando dados de campo de animais Braford e Hereford, testando os dois métodos BLUP (passo único e multi passo). Os resultados com base nos parâmetros estudados, mostraram que as acurácias de predição do DGV e do GEBV foram iguais nos dois procedimentos, porém no método multi passo as predições genômicas foram menos viesadas. / The aim of this work were: (1) to evaluate parameters of genetic diversity and population structure based on Angus, Devon, Hereford and Shorthorn breeds. Inbreeding rate (ΔF), effective size (Ne), relatedness, among other parameters, were estimated to provide subsidies for breeding programs. The parameters indicated a good genetic diversity, with Ne ranging from 128 in Devon to 303 in Shorthorn and ΔF ranging from 1.50 in Angus to 3.92 in Devon; (2) to evaluate strategies of genotype imputation with Braford and Hereford beef data using low density panels (3K, 6K, 8K, 15K and 20K) for 50K and 777K panels. These panels have been efficient and have, in general, compatible costs of the beef cattle operation; (3) to evaluate the accuracy of genomic prediction using some low density panels (8K and 15K) imputed to the 50K panel, relating the results with 50K original panel. The accuracy of direct genomic value (DGV) and genomic estimated breeding value (GEBV) with estimated breeding value (EBV) using imputed or not panels, indicated that there were no differences in accuracy and the losses in accuracy by using the imputed panels ranged from -0.0002 to -0.0021 depending on the panel, the scenario and the trait; (4) to use molecular markers in genomic selection testing two BLUP methods (single and two steps) with simulated beef cattle data. The results showed, based on the parameters studied, equality of results between the two methods; (5) to evaluate the viability of using the genomic selection using Braford and Hereford beef cattle and testing the two BLUP methods (single and two steps). The results, based on the parameters studied, showed that DGV and GEBV accuracies were similar in both methods, but the genomic predictions were less biased with then two-step method.

Dvojstupňové slinování pokročilých keramických materiálů s různými krystalovými strukturami / Two-step sintering of advanced ceramic materials with various crystal structures

The influence of Two-Step Sintering (TSS) process on the final microstructure of oxide ceramics materials with two different crystal structures was studied. Tetragonal zirconia (stabilized with 3mol%Y2O3, particle size 90nm and 140nm ) and cubic zirconia (8mol%Y2O3, 70nm) powders were cold isostatically pressed and pressure less sintered with different heating schedules. The microstructures achieved with TSS method were compared with microstructures achieved with conventional Single-Step Sintering schedule (SSS). The results showed that the efficiency of the TSS of these oxide ceramics was more dependent on their crystal structure than on their particle size and green body microstructure. The method of TSS brought only negligible improvement of the microstructure of tetragonal zirconia ceramics. On the other hand, TSS was successful in the sintering of cubic zirconia ceramics; it led to a decrease in grain size by a factor of 2.

Manufacturing and characterization of single cell intermediate-temperature solid oxide fuel cells for APU in transportation application / Synthèse et caractérisation de cellules pour pile à combustible de type IT-SOFC utilisée en tant que système APU dans les transports

La fabrication de cellules de piles à combustible IT-SOFC de large dimension par un nouveau procédé simple et peu coûteux est présentée dans ce manuscrit. L’optimisation de ce nouveau procédé en regard de l’utilisation d’agents de porosité, d’épaisseur de couches et de température de frittage a été réalisée. Les résultats des tests électrochimiques sur des cellules de surface active 10 cm2 réalisés dans le dispositif Fiaxell semi-ouvert ont été détaillés pour différentes cellules. Des tests de performance de longue durée ont également été menés sur le dispositif Fiaxell, présentés et discutés. La préparation et la réalisation d’un nouveau banc de test de stack a également été mené et présenté dans ces travaux. / The fabrications of large area IT-SOFC planar cell by new simple and cost effective process were explained. The optimization of the new process with respect to pore formers, thickness of layers, sintering temperature were performed. The electrochemical results of 10cm2 performed in Fiaxell open flange set up were detailed with respect to different configuration. Long term ageing performance tests of single cells were conducted in Fiaxell device and results are discussed. Preparation of new test bench and stacking process performed till now were briefed.

Desnitrificação autotrófica com o uso de sulfeto e integração com o processo de nitrificação em um único reator / Autotrophic denitrification with sulphide and the use of integration with the process of nitrification in a single reactor

A remoção de nitrogênio acoplada à oxidação de sulfeto pode ser uma opção adequada para o pós-tratamento de efluentes de reatores anaeróbios, os quais contêm nitrogênio amoniacal, que deve ser nitrificado, e sulfeto, que poderia ser utilizado como doador de elétrons endógeno para a desnitrificação autotrófica. Com base nessa constatação, esta pesquisa propôs a aplicação da nitrificação e desnitrificação autotrófica acoplada à oxidação de sulfeto, em um único reator, para a remoção de nitrogênio de efluentes de reatores anaeróbios tratando esgoto sanitário. Visto que existem lacunas na literatura referente ao processo desnitrificante autotrófico citado, as bases teóricas para a determinação das condições operacionais partiram da caracterização cinética e de aspectos fundamentais da desnitrificação autotrófica com uso de sulfeto como doador de elétrons. Numa primeira etapa, avaliou-se o efeito da concentração de sulfeto na desnitrificação, com uso de nitrato e nitrito como receptores de elétrons, em reatores verticais de leito fixo. Os resultados revelaram que compostos intermediários de enxofre foram principalmente formados quando se aplicou excesso de sulfeto, fato que foi mais evidente com o uso de nitrato. Evidências visuais sugeriram que enxofre elementar foi o principal intermediário formado, o qual também estava sendo utilizado quando aplicadas concentrações estequiométricas de sulfeto relativas a nitrato/nitrito. De modo geral, a desnitrificação autotrófica não foi afetada pela desnitrificação heterotrófica residual via atividade endogênica. Numa segunda etapa, determinou-se a cinética intrínseca da desnitrificação autotrófica via nitrato e nitrito com uso de diferentes concentrações de sulfeto em reatores diferenciais de leito fixo. Este bioprocesso pôde ser descrito por modelo cinético de ordem ½ para biofilmes. As constantes cinéticas variaram entre 0,425-0,658 mg N1/2 / L1/2 h para desnitrificação via nitrito e entre 0,190-0,609 mg N1/2 / L1/2 h para desnitrificação via nitrato. Neste último, o menor valor foi devido ao uso de elétrons doados a partir de compostos intermediários de enxofre formados. Numa terceira etapa, utilizou-se um reator de leito fixo operado em batelada alimentada seqüencial, com ciclos de 8 horas, submetido à aeração intermitente e empregando a desnitrificação autotrófica com uso de sulfeto presente no efluente sanitário, pré-tratado anaerobiamente, como doador de elétrons. O prévio estabelecimento da nitrificação com posterior aplicação de baixas concentrações de sulfeto foi a melhor estratégia de partida do reator. A alimentação em batelada alimentada com aplicação de sulfeto em excesso apenas nos períodos anóxicos foi a melhor estratégia de alimentação, proporcionando eficiência média de 85,7% e 53,0% para nitrificação e desnitrificação, respectivamente. O acúmulo de nitrito foi observado após aplicação de carga de choque de sulfeto, que inibiu as bactérias oxidadoras de nitrito. No entanto, houve dificuldade em se estabelecer a desnitrificação via nitrito em função da toxicidade deste composto aos organismos desnitrificantes instalados no reator. A baixa eficiência global de remoção de nitrogênio e algumas restrições operacionais indicaram que a desnitrificação autotrófica usando sulfeto em um único reator operado em bateladas seqüenciais não foi adequada para a proposta desta pesquisa. / Nitrogen removal coupled with sulfide oxidation may be suitable for the post treatment of effluents from anaerobic reactors. These effluents contain ammonium, which must be nitrified, and sulfide, which could be used as an endogenous electron donor for autotrophic denitrification. Since there are gaps in literature regarding the mentioned autotrophic denitrifying process, the theoretical basis for determination of operating conditions came from the characterization of kinetics and fundamentals aspects of autotrophic denitrification using sulfide as electron donor. In a first step, the effect of sulfide concentration on this bioprocess using nitrate and nitrite as electron acceptors in vertical fixed-bed reactors was evaluated. The results showed that intermediary sulfur compounds were mainly produced when excess of electron donor was applied, which was more evident when nitrate was used. Visual evidences suggested that elemental sulfur was the intermediary compound produced. There was also evidence that the elemental sulfur previously formed was being used when sulfide was applied in stoichiometric concentration relative to nitrate/nitrite. For all conditions assayed, autotrophic denitrification was not affected by residual heterotrophic denitrification via endogenic activity, occurring as a minor additional nitrogen removal process. In a second step, the intrinsic kinetics of sulfide-oxidizing autotrophic denitrification via nitrate and nitrite in systems containing attached cells was determined. Differential reactors were fed with nitrified synthetic domestic sewage and different sulfide concentrations. This bioprocess could be described by a half-order kinetic model for biofilms. The half-order kinetic coefficients ranged from 0.425 to 0.658 mg N1/2 / L1/2 h for denitrification via nitrite and from 0.190 to 0.609 mg N1/2 / L1/2 h for denitrification via nitrate. In this latter, the lower value was due to the use of electrons donated from intermediary sulfur compounds formed. In a third step, a sequencing fed-batch biofilm reactor of 8-h cycles was operated under intermittent aeration, applying autotrophic denitrification using sulfide present in the sanitary effluent, anaerobically pre-treated, as electron donor. The effect of the start-up period and the feeding strategy were evaluated. The previous establishment of nitrification process with subsequent application of sulfide in low concentrations was the best start-up strategy. The fed-batch mode with sulfide application in excess only in the anoxic periods was the best feeding strategy, providing average efficiencies of 85.7% and 53.0% for nitrification and denitrification, respectively. Nitrite accumulation was observed after application of shock loading of sulfide, which inhibited nitrite-oxidizing bacteria. However, it was difficult to establish denitrification via nitrite due to the toxicity of this compound to denitrifying organisms developed inside the reactor. The low overall efficiency of nitrogen removal and some operational constraints indicated that autotrophic denitrification using sulfide in a single sequencing fed-batch reactor was not suitable for the purpose of this research.

Comparison And Evaluation Of Three Dimensional Passive Source Localization Techniques

Passive source localization is the estimation of the positions of the sources or emitters given the sensor data. In this thesis, some of the well known methods for passive source localization are investigated and compared in a stationary emitter sensor framework. These algorithms are discussed in detail in two and three dimensions for both single and multiple target cases. Passive source localization methods can be divided into two groups as two-step algorithms and single-step algorithms. Angle-of-Arrival (AOA) based Maximum Likelihood (ML) and Least Squares (LS) source localization algorithms, Time- Difference-of-Arrival (TDOA) based ML and LS methods, AOA-TDOA based hybrid ML methods are presented as conventional two step techniques. Direct Position Determination (DPD) method is a well known technique within the single step approaches. In thesis, a number of variants of DPD technique with better computational complexity (the proposed methods do not need eigen-decomposition in the grid search) are presented. These are the Direct Localization (DL) with Multiple Signal Classification (MUSIC), DL with Deterministic ML (DML) and DL with Stochastic ML (SML) methods. The evaluation of these algorithms is done by considering the Cramer Rao Lower Bound (CRLB). Some of the CRLB expressions given in two dimensions in the literature are presented for threedimensions. Extensive simulations are done and the effects of different parameters on the performances of the methods are investigated. It is shown that the performance of the single step algorithms is good even at low SNR. DL with MUSIC algorithm performs as good as the DPD while it has significant savings in computational complexity. AOA, TDOA and hybrid algorithms are compared in different scenarios. It is shown that the improvement achieved by single-step techniques may be acceptable when the system cost and complexity are ignored. The localization algorithms are compared for the multiple target case as well. The effect of sensor deployments on the location performance is investigated.
