Heterosídeos alcaloídicos de Solanum lycocarpum A. St.-Hil.: avaliação das atividades contra fungos dermatófitos e câncer de pele / Alcaloidic heterosides from Solanum lycocarpum A. St.-Hil.: evaluation of the activities against dermatophytic fungus and skin cancer

de pele é o tipo mais frequente, correspondendo a cerca de 25% de todos os tumores malignos registrados no Brasil. Há que se destacar que uma formulação de uso tópico foi desenvolvida pelo grupo para uso contra câncer de pele e o estudo da estabilidade acelerada desta formulação foi avaliado nesse trabalho. Os frutos da espécie S. lycocarpum foram coletados, secos, triturados, submetidos à extração ácido-base e o precipitado foi suspenso em etanol e filtrado. Os heterosídeos alcaloídicos, SS e SM, foram isolados por cromatografia em coluna a vácuo e purificados em CLAE semipreparativa. A formulação contendo extrato alcaloídico armazenada em temperatura ambiente (27 ± 2 oC) apresentou a melhor estabilidade física. Ensaios para avaliação da atividade antifúngica in vitro do extrato alcaloídico, SS, SM e aglicona (SD) foram realizados com cepas de fungos dermatófitos e Candida spp., sendo que a SM apresentou o menor valor de concentração inibitória. No ensaio de citotoxidade em células humanas de carcinoma espinocelular (A431) e células de fibroblastos de camundongos (L929) foi possível observar a toxidade do extrato alcaloídico e das substâncias, SS, SM e SD frente às células cancerígenas. No ensaio in vivo, os animais foram induzidos ao câncer de pele não-melanoma do tipo espinocelular com células A431 e verificou-se que apenas o grupo de animais tratados com a formulação Curaderm BEC 5 apresentou redução tumoral. Em contrapartida, o grupo tratado com formulação contendo extrato alcaloídico se comportou da mesma maneira que os grupos controle da formulação e controle negativo. Com auxílio da histologia confirmou-se que o câncer de pele induzido nos animais foi do tipo espinocelular. No ensaio imunoistoquímico foi utilizado o anticorpo caspase-3 para marcar as células em apoptose nos tumores, sendo que o maior número de células apoptóticas foi verificada no grupo tratado com a formulação Curaderm BEC 5. / Solanum lycocarpum A. Saint-Hilaire (Solanaceae), commonly known as wolf apple or wolf´s fruit, is a plant native of Brazil very common in the Brazilian savanna. High concentrations of steroidal alkaloids are found in S. lycocarpum fruits, and solasonine (SS) and solamargine (SM) are the most important ones. These two alkaloids present potential antifungal and anticancer activities. Dermatophytic fungi are the most common agents responsible for superficial mycoses in humans and animals, by infecting exclusively the stratum corneum of skin, hair and nails. Skin cancer is the most frequent type corresponding to roughly 25% of all the malignant tumors registered in Brazil. A topical formulation for skin cancer treatment was developed by our research group and its stability tests are reported in this work. The S. lycocarpum fruits were collected, dried, milled and submitted to acid-base extraction furnishing a precipitate, which was solubilized in ethanol and then filtered. The heterosidic alkaloids SS and SM were isolated by using vacuum column chromatography and purified by semi-preparative HPLC. The formulation containing the alkaloidic extract stored at room temperature (27 ± 2 oC) was more stable than the ones in other conditions. Antifungal in vitro test of the alkaloidic extract, SS, SM and the aglycone (SD) were performed with Candida spp and dermatophytic fungi strains. The alkaloid SM displayed the lower inhibitory concentration. The toxicity of the alkaloidic extract, SS, SM and SD was verified in a cytotoxicicity test performed with both cultured human cells of spinocellular carcinoma (A431) and mice fibroblast cells (L929). An in vivo cytotoxicicity test was performed by inducing non-melanoma skin cancer in mice with spinocellular cells A431, in which only the animals treated with a commercial formulation Curaderm BEC 5, showed tumor reduction. There was no statistical difference between the group treated with the formulation containing the alkaloidic extract and control groups. Histological analysis confirmed that the skin cancer induced in the animals were spinocellular type. In an immunohistoquimic test, caspase-3 antibody was employed to stain the cells in apoptosis in the tumors, showing that apoptotic cells were more numerous in the group treated with the formulation Curaderm BEC 5.

Solanum lycocarpum A. St.- Hil. (Solanaceae) é uma espécie nativa do Brasil distribuída nas regiões sudeste e centro-oeste do país. O fruto desta planta é popularmente conhecido por fruta do lobo\". O consumo destes frutos impede o desenvolvimento de uma parasitose renal (Dioctophyna renale) que afeta lobos (Chrysocyon brachyurus). Estes frutos contêm dois principais heterosídeos alcaloídicos: solamargina (SM) e solasonina (SS), sendo ambos derivados da aglicona solasodina (SD). Doenças parasitárias, como leishmaniose, esquistossomose e estrongiloidíase estão diretamente ligadas a questões socioeconômicas representando um grande problema de saúde pública principalmente em regiões tropicais e subtropicais. Portanto, é extremamente importante a busca de novos fármacos para estas e outras doenças negligenciadas. O presente trabalho descreve a obtenção do extrato alcaloídico (EA) a partir dos frutos da lobeira, o isolamento dos heterosídeos alcaloídicos majoritários e da aglicona SD, bem como a avaliação de suas atividades in vitro leishmanicida, estrongiloicida e esquistossomicida. Além disso, a atividade esquistossomicida do EA também foi avaliada em modelo animal. O EA foi obtido a partir de frutos secos e triturado por meio de extração ácido-base. SM e SS foram isolados por cromatografia em camada delgada preparativa ou por cromatografia flash. SD foi obtida a partir da hidrólise ácida do EA, e purificada por cromatografia em coluna clássica utilizando eluição por gradiente. As estruturas dos compostos isolados foram confirmadas por análises de RMN e espectrometria de massas. Nas avaliações biológicas observaram-se atividade leishmanicida, estrongiloicida e esquistossomicida do EA. Os compostos isolados, SM, SS e a mistura equimolar foram ativos contra L. amazonensis. Além disso, esses compostos foram capazes de reduzir a atividade motora de vermes adultos de S. mansoni, separar os pares de verme adulto em macho e fêmea, causar a morte dos vermes, alterar o tegumento e reduzir o desenvolvimento dos ovos in vitro. A SD foi inativa contra L. amazonensis e vermes adultos de S. mansoni. O EA avaliado in vivo mostrou atividade esquistossomicida, diminuindo o número de ovos e granulomas no fígado dos animais tratados. / The Brazilian Solanaceae plant, Solanum lycocarpum A. St.-Hil. is distributed in the Southeast and Middle West of the country. The fruit of this plant is popularly known as fruta do lobo (wolf-fruit). The intake of these fruits prevents renal parasitism (Dioctophyna renale) which affects wolves (Chrysocyon brachyurus). These fruits contain two major alkaloid heterosides (solamargine (SM) and solasonine (SS)) bearing both the same aglycone, solasodine (SD). Among the parasitic diseases, such as leishmaniasis, strongyloidiasis and schistosomiasis stand out in terms of socioeconomic and public health importance in tropical and subtropical areas. Its extremely important to search for new drugs to treat these and other neglected diseases. The present work reports the obtainment of the alkaloidic extract (AE) of the S. lycocarpum fruit, the isolation of the major alkaloid heterosides and to obtain the aglycone. The obtained samples were evaluated for the in vitro leishmanicidal, strongyloicidal and schistosomicidal activities. Also, the schistosomicidal activity of the AE was evaluated in an animal model. The analysis of the obtained results for the biological evaluation showed that the AE displays leishmanicidal, strongyloicidal and schistosomicidal activities. The isolated compounds, SM, SS and their equimolar mix were actived against L. amazonensis. These compounds also were able to reduce the mobility, to separate the worm into male and female, to modify the tegumental and to cause the death of the adults worms besides reducing the development of eggs. The SD was inactive against L. amazonensis and S. mansoni adult worms. When AE was evaluated in vivo showed activity as schistosomicidal, decreasing the number of eggs and granulomas in the liver of the treated mice.

Solanum lycocarpum A. St.-Hil. (Solanaceae Solanum), popular lobeira, é espécie arbustiva nativa e característica do Cerrado brasileiro. Seus frutos são empregados na medicina caseira como diurética, calmante, anti-espasmódica, antiofídica e antiepilética. As espécies do gênero Solanum são produtoras de heterosídeos alcaloídicos, os quais possuem atividade antitumoral, incluindo-se anticâncer de pele. O câncer de pele tem preocupado as autoridades no mundo com os crescentes índices atuais e, por esse motivo, a busca por novos ativos anticâncer é primordial. Dos frutos da lobeira, pode-se produzir extrato alcaloídico, o qual se constitui de uma mistura de glicoalcalóides composta majoritariamente de solasonina e solamargina. Tais compostos têm sido estudados quanto ao potencial anticancerígeno, são relativamente seletivos para células tumorais e, em consequência, apresentam baixa toxicidade às células sadias. Por estes motivos, o potencial antitumoral desta espécie deve ser investigado. Para isso, os frutos foram coletados, secos e triturados. A droga vegetal resultante foi submetida à extração tipo ácido-base para obtenção do extrato alcaloídico. O método analítico empregado para quantificação dos heterosídeos alcaloídicos foi desenvolvida e validada em CLAE-UV. A partir do extrato alcaloídico padronizado, foram desenvolvidas formulações para uso tópico. Estas formulações foram avaliadas por meio de ensaios de permeação e retenção dérmica in vitro, cujo método analítico para quantificação de solasonina e solamargina foi validado. Para escolha da formulação mais promissora para ensaio anticâncer in vivo, as formulações de melhor desempenho in vitro foram avaliadas quanto à retenção dérmica in vivo em camundongos hairless. A formulação B, contendo 1% de extrato alcaloídico, 5% de monoleína (monoleato de glicerol), 5% de propilenoglicol em base de gel de Natrosol (pH 6,5), apresentou retenção dérmica total adequada para futuros ensaios anticâncer de pele in vivo. / Solanum lycocarpum A. St.-Hil. (Solanaceae - Solanum), popular wolf-fruit is a native shrub species and characteristic of the Brazilian Cerrado vegetation. Its fruits are used in folk medicine as diuretic, sedative, anti-spasmodic, antiepileptic and antiophidic. This species belongs to the genus Solanum, known to produce alkaloid heterosides, which possess antitumor activity, including skin anticancer. Skin cancer has concerned the authorities in the world because of its growing rates. Therefore, the search for new active anticancer is paramount. The wolf-fruit furnish an alkaloidic extract, which is rich in a mixture of heteroside alkaloids and composed mostly of solasonine and solamargine. Such compounds have been studied as potential anticancer, because they are relatively selective for tumor cells and, consequently, have low toxicity to health cell. For these reasons, the antitumor potential of this species should be investigated. For that, the fruits were collected, dried and crushed. The dried plant biomass was submitted to acid-base extraction to obtain the alkaloidic extract. The analytical method for the quantitation of the alkaloids in both crude alkaloid extract and plant biomass was developed and validated by HPLC-UV. From the alkaloidic extract, formulations were developed for topical use. These formulations were evaluated by in vitro tests of skin permeation and retention in Franz diffusion cell, for which an analytical method for quantifying solasonine and solamargine was developed, as well. The selection of the most promising formulation for anticancer assay in vivo was based in both, the permeation performance of the formulation in vitro and its skin retention in vivo in hairless health mice. The formulation B, containing crude alkaloid extract 1%, Monoolein 5%, propyleneglycol 5% in Natrosol gel (pH 6.5), showed total dermal retention, which is suitable for future trials of skin anticancer in animal models.

Desenvolvimento de nanopartículas lipídicas sólidas no carreamento de extrato alcaloídico de Solanum lycocarpum e avaliação biológica in vitro em células de câncer de bexiga / Development of solid lipid nanoparticles in loading alcaloídico extract of Solanum Lycocarpum and biological evaluation in vitro in bladder cancer cells

Solanum lycocarpum é uma espécie brasileira conhecida por sua diversidade em glicoalcalóides (GA). Os GA solasonina (SS) e solamargina (SM), extraídos dos frutos desta planta, apresentaram propriedades antioxidantes e anticancerígenas atuando em diferentes mecanismos moleculares para a terapia do câncer. No entanto, a baixa solubilidade em água destas moléculas e a sua toxicidade limita a aplicação destes glicoalcalóides. Desta forma, o desenvolvimento de uma formulação nanoestruturada contendo GA pode possibilitar a sua aplicação clínica na terapia do câncer de bexiga (CB). Assim, este projeto visa o desenvolvimento e caracterização de carreadores lipídicos nanoestruturados (CLN) como sistemas de carreamento do extrato alcaloídico de S. Lycocarpum (EA), rico em GA, e avaliação da citotoxidade in vitro em células de câncer de bexiga. Os CLN foram preparadas pelo método de emulsão à quente e sonicação e otimizados utilizando um planejamento fatorial 23. No planejamento avaliou-se a influência de três variáveis independentes: tipo e quantidade de lipídio e tipo de surfactante nos fatores de resposta diâmetro e índice de polidispersão (PdI). CLNs foram caracterizadas de acordo com diâmetro médio, distribuição de tamanho e potencial zeta por espalhamento dinâmico de luz (DLS), índice de recristalização (IR) por calorimetria exploratória diferencial (DSC), porcentagem da eficiência de encapsulamento (EE%) por LC-MS/MS, avaliação morfológica por microscopia de força atômica (AFM), distribuição e concentração de nanopartículas por análise de rastreamento de nanopartículas (NTA), perfil de liberação do EA encapsulado em CLN pelo ensaio de perfil de liberação in vitro em células de Franz, e estabilidade ao longo do tempo. A citotoxidade dos glicoalcalóides foi avaliada pelo ensaio de captura do corante de vermelho neutro utilizando células de câncer de bexiga (RT4). As células foram tratadas com EA livre e encapsulado no CLN nas concentrações de 5 ?g/mL à 20 ?g/mL, assim como CLN vazia no período de 24h e 48h e foram calculadas suas respectivas concentrações inibitórias médias (IC50). O CLN desenvolvido apresentou tamanho inferior a 80 nm e potencial zeta negativo. O PdI foi inferior a 0,2 indicando uma baixa polidispersão. O CLN foi estável por 60 dias em relação ao tamanho, PdI e potencial zeta. O índice de recristalização (IR) do CLN com EA foi 47,23%, indicando uma estrutura menos cristalina do lipídio no CLN que propicia uma alta eficiência de encapsulamento de SS (86%) e SM (89,1%) e evita a expulsão dos ativos durante a estocagem. Os CLNs apresentaram-se semiesféricas como observado por AFM. A concentração de partículas, medida por NTA, foi de 9,74x 1011 partículas/mL e o diâmetro médio foi de 125 nm. Os glicoalcalóides presentes no EA encapsulado em CLN apresentam perfil de liberação in vitro sustentado após 36 h de liberação em condição \"sink\". O mecanismo de liberação dos GA foram diferentes, sendo que para o SS foi não-fickano enquanto que o SM foi fickano. O EA livre e encapsulado (CLN-EA) foi citotóxico frente células RT4, sendo que o CLN-EA foi mais citotóxico que o EA livre após 48h de tratamento. O IC50 das CLN-EA, após 24h de tratamento, foi de 15,94 ?g.mL-1 e 12,35 ?g.mL-1 após 48 h de tratamento. Esta redução no valor do IC50 está relacionada ao perfil de liberação dos glicoalcalóides SS e SM das nanopartículas. Os resultados demonstraram que a CLN desenvolvida é um sistema interessante para encapsular EA com potencial para futura terapia do câncer de bexiga / Solanum lycocarpum is a native species of the Brazilian savannah known for its diversity in glycoalkaloids (GA). The GA solasonine (SS) and solamargine (SM), extracted from the fruits of their plant, have antioxidant and antitumoral activity; acting on different molecular mechanisms interesting for cancer therapy. However, the low water solubility of these molecules and their toxicity limits their application. Thus, the development of a nanostructured formulation containing these glycoalkaloids may allow their clinical application in bladder cancer therapy (BC). Thus, this project aims at the development and characterization of nanostructured solid lipid (NLC) as carrier systems of alkaloidic extract of S. lycocarpum (AE), rich in GA, and their cytotoxic evaluation in order to obtain a nanostructured system for future intravesical therapy of bladder cancer. The NLC were prepared by the emulsion and sonication method and optimized using a factorial design 23. The influence of three independent variables was evaluated as type and amount of lipid and type of surfactant in response factors diameter and polydispersity index (PdI). NLCs were characterized according to average diameter, size distribution and zeta potential by dynamic light scattering (DLS), recrystallization index (RI) by differential scanning calorimetry (DSC), percentage of encapsulation efficiency (EE%) by LC MS/MS, morphological analysis by atomic force microscopy (AFM), distribution and concentration of nanoparticles by nanoparticle tracking analysis (NTA), the in vitro release profile of GA from NLC was done by Franz\'s cells diffusion, and stability over time by DLS. Cytotoxicity assay was evaluated by neutral red uptake assay in bladder cancer cells (RT4). Cells were treated with different concentrations of GA free and encapsulated in NLC (5-40 ?g/mL) and empty NLC for 24h, 48h and 72h. The NLC developed showed diameter smaller than 80 nm and zeta potential of -9.26 mV. The PdI was less than 0.2 indicating dispersion with low polydispersity. The NLC was stable for 60 days regarding to size, PdI and zeta potential. Recrystallization index (RI) of NLC-AE was 47.23%, indicating a less crystalline structure of the lipid in the particle, which provides high encapsulation efficiency of SS (86%) and SM (89.1%). This low RI prevents the expulsion of GA from NLC during the storage. NLCs exhibited semi-spherical form as observed by AFM. The particles concentration, measured by NTA, was 9.74 x 1011 particles/mL and the diameter was about 125 nm. The GA present in NLC-AE showed an in vitro sustained release profile after 36 h of release in a sink condition. The release mechanisms of SS and SM were different. The release mechanism of SS was non-Fickan while SM was Fickan. The AE free and NLC-AE was cytotoxic to RT4 cells, and the NLC-AE was more cytotoxic than the AE free after 48 hours of treatment. The IC50 of NLC-AE, after 24 hours of treatment, was 15.94 ?g.mL-1 and 12.35 ?g.mL-1 after 48 h of treatment. This reduction in IC50 value can be related to the release profile of the GA (SS and SM) of the nanoparticles. The results demonstrate that NLC developed is an interesting system to encapsulate AE with potential for future therapy of bladder cancer

A Solanum lycocarpum é uma planta encontrada em quase todos os estados do Brasil, podendo ser ingerida por animais em idade fértil e em condições naturais. Os frutos desta planta possuem glicoalcalóides esteroidais com propriedades semelhantes aos esteróides. Neste estudo investigou-se os efeitos na esfera reprodutiva causados pela ingestão de S. lycocarpum durante a puberdade de ratos. Os frutos secos foram administrados na ração (10%) desde o desmame até a idade adulta (70dias). Foram avaliados o peso corpóreo e de órgãos, o comportamento e a orientação sexual, os níveis de hormônios sexuais nas fezes, a bioquímica sérica, os níveis de neurotransmissores centrais e os achados anatomopatológico, tanto de ratos machos como de fêmeas. Observou-se os seguintes resultados: 1) Redução no peso corpóreo dos machos e das fêmeas; 2) Em machos, o peso relativo aumentou nos epidídimos, vesícula seminal e testículos; 3) Não houve diferenças no hemograma entre os grupos controle e experimental de ambos os sexos; 4) Alterações na bioquímica sangüínea sugerem leve toxicidade em células musculares e hepática; 5) Em fêmeas, não constatou-se nenhuma alteração no ciclo estral e no comportamento sexual; enquanto nos machos houve melhora do comportamento sexual e 6) Os níveis de neurotransmissores do estriado foi modificado mas não os do hipotálamo. Esses achados indicam que a exposição a S. lycocarpum durante a puberdade prejudicou parâmetros da esfera reprodutiva, em particular, dos machos. / Solanum lycocarpum St. Hil, a toxic plant common in almost all states of Brazil, may be consumed by animals in fertile age and in natural condition. The dried fruits were administered in the diet (10%) from the weaning up to adult age (70 days). The body and organs weights, sexual behavior as well as sexual orientation, the feces hormonal levels, hemogram and biochemical blood evaluations, hypothalamic and striatal neurotransmitter levels, and anatomopathological studies of male and female rats were performed. The results showed: 1) a reduced body weight in male and female; 2) in male, an increased epydidimis, seminal vesicle and testis relative weights; 3) no differences of controls and experimental groups of both sex in the hemogram; 4) blood biochemical alterations meaning slight toxicity in muscular and hepatic cells; 5) in female, no interference was detected in estrous cycle and sexual behavior; an improvement of male sexual behavior was observed; 6) that striatal neurotransmitter levels were modified but not in the hypothalamus. These results indicate that exposure to S. lycocarpum during puberty prejudiced the reproductive system, particularly in male rats.

The objective of this work is the use of starch extracted from the fruit of S. lycocarpum as a new source for the production of ethanol by fermentation. The starch in the fruit was extracted by grinding lobeira mesocarp citric acid 0.8% (w / v) subsequent decantation. Thereafter the starch was hydrolyzed using the α-amilise and immobilized glucoamylase-glutaraldehyde polyaniline, with an average rate of glucose production of approximately 80% after 60 h. Glucose solutions at concentrations of 1%, 5% and 9% (w / v) were fermented by the yeast Saccharomyces bayanus INCQS 40235, provided by the National Institute of Quality Control in Health Oswaldo Cruz Foundation. Ethanol production was quantified by the following methods: by reducing the weight due to release of CO2 and the maximum yield obtained after 48 h of fermentation, reaching a production of 6.1704 g in 48 hours; reduction of 75.63% of the total sugar content in the fermentation broth reaching 48.4 mg mL-1 after 48 hours. The fermentation process was also evaluated by the growth of yeast cells that reached 1,407 x 108 cells in 48 hours of fermentation. Results of this work have shown that the starch extracted from the fruit of lobeira is a promising source for the production of ethanol, yielding 10.5% of ethanol after 48h fermentation using a 9% solution of hydrolyzed starch under industrial fermentation simulation. / A planta Solanum lycocarpum, conhecida popularmente como lobeira, é um arbusto ou árvore de até 5 m frequente em todo o Cerrado brasileiro, e responsável pela produção do fruto do lobo. O objetivo deste trabalho é a utilização do amido extraído do fruto da S. lycocarpum como uma nova fonte para a produção de etanol por processo fermentativo. O amido do fruto da lobeira foi extraído por trituração do mesocarpo em ácido cítrico 0.8% (p/v) e posterior decantação. Posteriormente o amido foi hidrolisado utilizando-se a α-amilase e a glicoamilase imobilizadas em polianilina-glutaraldeído, com uma produção média de glicose de aproximadamente 91%, após 60 h de reação. Soluções de glicose nas concentrações 1%, 5% e 9% (p/v) foram fermentadas pela levedura Saccharomyces bayanus INCQS 40235, fornecida pelo Instituto Nacional de Controle de Qualidade em Saúde da Fundação Oswaldo Cruz. A produção de etanol foi quantificada pela redução do peso devido ao desprendimento de CO2, sendo o máximo rendimento obtido após 48 h de fermentação, de 6,17g/ 100 mL; o que apresenta uma redução de 75,63% na concentração de açúcares totais no caldo fermentativo, chegando a 48,4 mg mL-1 após 48 horas. O processo fermentativo também foi avaliado pelo crescimento do número de células da levedura que atingiu 1,407 x 108 células em 48 horas de fermentação. Os resultados deste trabalho permitem concluir que o amido extraído do fruto da lobeira é uma fonte promissora para a produção de etanol, com rendimento de 8,64% de etanol produzido após 48h de fermentação partindo de uma solução a 9% de glicose numa simulação de escala industrial.

Solanum lycocarpum A. St.-Hil. (Solanaceae Solanum), popular lobeira, é espécie arbustiva nativa e característica do Cerrado brasileiro. Seus frutos são empregados na medicina caseira como diurética, calmante, anti-espasmódica, antiofídica e antiepilética. As espécies do gênero Solanum são produtoras de heterosídeos alcaloídicos, os quais possuem atividade antitumoral, incluindo-se anticâncer de pele. O câncer de pele tem preocupado as autoridades no mundo com os crescentes índices atuais e, por esse motivo, a busca por novos ativos anticâncer é primordial. Dos frutos da lobeira, pode-se produzir extrato alcaloídico, o qual se constitui de uma mistura de glicoalcalóides composta majoritariamente de solasonina e solamargina. Tais compostos têm sido estudados quanto ao potencial anticancerígeno, são relativamente seletivos para células tumorais e, em consequência, apresentam baixa toxicidade às células sadias. Por estes motivos, o potencial antitumoral desta espécie deve ser investigado. Para isso, os frutos foram coletados, secos e triturados. A droga vegetal resultante foi submetida à extração tipo ácido-base para obtenção do extrato alcaloídico. O método analítico empregado para quantificação dos heterosídeos alcaloídicos foi desenvolvida e validada em CLAE-UV. A partir do extrato alcaloídico padronizado, foram desenvolvidas formulações para uso tópico. Estas formulações foram avaliadas por meio de ensaios de permeação e retenção dérmica in vitro, cujo método analítico para quantificação de solasonina e solamargina foi validado. Para escolha da formulação mais promissora para ensaio anticâncer in vivo, as formulações de melhor desempenho in vitro foram avaliadas quanto à retenção dérmica in vivo em camundongos hairless. A formulação B, contendo 1% de extrato alcaloídico, 5% de monoleína (monoleato de glicerol), 5% de propilenoglicol em base de gel de Natrosol (pH 6,5), apresentou retenção dérmica total adequada para futuros ensaios anticâncer de pele in vivo. / Solanum lycocarpum A. Its fruits are used in folk medicine as diuretic, sedative, anti-spasmodic, antiepileptic and antiophidic. This species belongs to the genus Solanum, known to produce alkaloid heterosides, which possess antitumor activity, including skin anticancer. Skin cancer has concerned the authorities in the world because of its growing rates. Therefore, the search for new active anticancer is paramount. The wolf-fruit furnish an alkaloidic extract, which is rich in a mixture of heteroside alkaloids and composed mostly of solasonine and solamargine. Such compounds have been studied as potential anticancer, because they are relatively selective for tumor cells and, consequently, have low toxicity to health cell. For these reasons, the antitumor potential of this species should be investigated. For that, the fruits were collected, dried and crushed. The dried plant biomass was submitted to acid-base extraction to obtain the alkaloidic extract. The analytical method for the quantitation of the alkaloids in both crude alkaloid extract and plant biomass was developed and validated by HPLC-UV. From the alkaloidic extract, formulations were developed for topical use. These formulations were evaluated by in vitro tests of skin permeation and retention in Franz diffusion cell, for which an analytical method for quantifying solasonine and solamargine was developed, as well. The selection of the most promising formulation for anticancer assay in vivo was based in both, the permeation performance of the formulation in vitro and its skin retention in vivo in hairless health mice. The formulation B, containing crude alkaloid extract 1%, Monoolein 5%, propyleneglycol 5% in Natrosol gel (pH 6.5), showed total dermal retention, which is suitable for future trials of skin anticancer in animal models.
