São salmonelas de composição antigênica similar e a distinção de ambas tem sido baseada em provas bioquímicas. Entretanto, este procedimento é laborioso e o surgimento de estirpes com comportamento bioquímico atípico tem estimulado a procura por outros métodos de diagnóstico, como os testes moleculares. No presente estudo, analisou-se o método descrito na literatura envolvendo o gene rfbS em conjunto com testes desenvolvidos com base nos genes speC e speF. O gene rfbS codifica a enzima que atua na parede celular de bactérias e tem sido utilizado para diferenciar SP e SG. Com o propósito de padronizar a PCR para ser utilizada na rotina laboratorial, empregou-se dois métodos de extração de DNA e diferentes concentrações de reagentes. Entretanto, tendo em vista a dificuldade de padronização encontrada desde o início, foi concomitantemente desenvolvida a PCR para os genes speC e speF. Estes dois últimos estão relacionados com a produção da enzima ornitina-descarboxilase, a qual é ativa em SP e inativa em SG. Após testes inicias, foi possível padronizar a PCR do gene speC com posterior utilização da técnica de tratamento enzimático com a enzima de restrição Eco RI. Tanto para o gene rfbS quanto para os genes speC e speF, os resultados encontrados permitiram a diferenciação de 13 amostras de SP e 20 de SG isoladas no Brasil e duas cepas ATCC. Sendo assim, as ações sanitárias e a tomada de decisão no campo podem ser conduzidas de maneira rápida e segura. / Salmonella Pullorum (SP) is very similar to Salmonella Gallinarum (SG). They are respectively agents of Pulorosis and Avian Typhoid, both diseases responsible for economic losses to poultry production. Due to the fact that both salmonellas have similar antigenic composition, their differentiation has been based in biochemical tests. However, this fact that this kind of procedure is very troublesome and the occurence of strains showing atypical behavior have stimulated the search for others diagnostic methods, such as molecular tests. In the present study, a method described in the literature using gene rfbS was analyzed together with tests developed based on genes speC and speF. Gene rfbS encodes an enzyme that acts on the cell wall of bacteria and has been used in the differentiation between SP and SG. In order to standardize the PCR procedure to be used in laboratory routine, two methods for DNA extraction were used, as well as different concentrations of the reagents. However, due to the difficulty in the standardization observed from the beginning of the study, a PCR procedure was developed for genes speC and speF. These genes are related to the production of the enzyme ornithine-descarboxilase, active in SP and inactive in SG. After the initial tests, PCR of gene speC was standardized, and later on it was coupled with the enzymatic treatment with restriction enzyme Eco RI. Results obtained for gene rfbS as well as for genes speC and speF enabled differentiation of 13 SP and 20 SG strains isolated in Brazil and two strains ATCC. So, the sanitary actions and the decisions in the livestock can to be conduced with safety and rapidity.

Die Arbeit untersucht Einflussfaktoren auf die SPEC-CPU-
Benchmarks CINT95 und CFP95. Es werden die Messbedingungen
bei Messungen von Rechnerherstellern fuer SPEC-Veroeffentlichungen
und bei Messungen im Universitaetsrechenzentrum verglichen.
Aus den unterschiedlichen Bedingungen werden moegliche
Einflussfaktoren abgeleitet, die fuer die Abweichung der
Benchmarkergebnisse unter Lastbedingungen ursaechlich sein
koennten. Fuer diese Einflusskomponenten eines Rechnersystems
werden charakteristische Lastmasse erarbeitet.
Im experimentellen Teil der Arbeit werden 4 Rechner des
Universitaetsrechenzentrums mit 4 gezielt ausgewaehlten SPEC-
CPU-Benchmarks untersucht. Ueber mehrere Wochen hinweg werden
dabei Messungen unter verschiedensten Lastverhaeltnissen
durchgefuehrt, wobei jeweils die Benchmarklaufzeit und die
mittleren Werte fuer die definierten Lastmasse erfasst werden.
Auf Basis dieser 16 Messreihen von jeweils 20 bis 60
Messungen werden schliesslich Aussagen zu Einflugroessen auf
einzelne Benchmarks, zum generellen Einfluss einzelner
Teilsysteme, sowie zum differenzierten Lastverhalten der
einzelnen betrachteten UNIX-Rechnersysteme gewonnen.

Studien undersöker hur det interaktiva elementet i TV-spelet Spec Ops: The Line påverkar och formar spelets narrativ, och försöker utifrån det dra mer allmänna slutsatser rörande vilka effekter interaktiva element i TV-spel kan ha på såväl spelet som dess spelare. Resultaten visar att interaktiva aspekter tillåter spelet att i högre grad förankra sitt budskap i spelaren som individ och dennes beslut, i kontrast till att göra en poäng på mänskligheten i allmänhets bekostnad. I diskussionen uppmanas lärarvärlden att ta till vara på de unika möjligheter som spel erbjuder för att fördjupa, utvidga och nyansera inte minst en diskussion om narrativ, samt för att förmedla en del av de berättelser och lärdomar som i skolan traditionellt har förmedlats genom litteraturen, om inte annat som en inkörsport till litteratur.

Modelling, Simulation and control of a Shortest Path Enthalpy Change Air Handling Unit with Liquid DesiccantWilkens, Marcus January 2021 (has links)
In an abundance of industries, production lines and facilities, a correct indoor climate is an integral part to ensure the quality of the products and also very important for its occupants. To ensure a high quality air climate it is not enough to control the air temperature, air humidity is of at least equal importance. Most heating, ventilation and air conditioning systems (HVAC), are based on multiple treatment steps that each change both temperature and humidity. Due to the correlation between the temperature and relative humidity this results in system control difficulties, and unnecessary realignments to achieve the desired outputs. The novel Shortest path enthalpy change system (SPEC), developed by AirWaterGreen (AWG) with Controlled vapor pressure technology (CVP), aims to eliminate these problems. This is done by utilizing a method that may achieve the desired air quality with only one conditioning step, transferring heat and humidity simultaneously to or from the air. To validate this technology the work of this thesis is focused on producing a viable simulator model with verification, performed by comparing to a prototype system with the same technology. By verifying the simulator, it is also possible to continue development of the system by implementing system controllers within the Simulink environment. This provides the opportunity to ultimately obtain a highly efficient HVAC system with accurate setpoint control. In addition, the possibility of optimizing system dimensions is also investigated to allow for specific case configurations. The results show that high simulator accuracy is achievable, leading to a high confidence in the results, and showing that a PID or even proportional feedback controller is able to realize satisfactory system control. The optimization of dimensions shows that parallel computing is a fast way to reduce the amount of possible configurations that satisfy specific criteria. To support further development of the control and optimization of the system and ultimately hardware implementation, this thesis suggests points of interest for future work. These points include the improvement of ancillary simulator systems and further development of the pads physical equations, to increase the accuracy of process simulations, and the continued development of more complex controllers as well as their hardware implementation. Lastly, while focus of this thesis has been on the development of a functioning and controllable system, an important addition would be a study on the energy usage of the system and identifying points of interest for decreasing the total energy used from the system.

Utvärdering av analys av pankreas-specifikt lipas hos hund och katt med Vcheck V200 : en prospektiv komparativ studie / Evaluation of analysis of pancreas-specific Lipase in dogs and cats with Vcheck V200 : a prospective comparative studyCarlsson, Felicia January 2021 (has links)
Pankreatit anses vara en vanligt förekommande sjukdom hos hundar och katter och kan diagnostiseras genom mätningar av koncentrationen Canine Pancreas-specific Lipase (cPL) respektive Feline Pancreas-specific Lipase (fPL) i serum. Utifrån dessa koncentrationer graderas patienten enligt normalvärde, gråzon eller indikation på pankreatit. Golden standardmetoden för att analysera cPL/fPL är Spec cPL respektive Spec fPL. Nya metoder har utvecklats för kvantitativ mätning av pankreas-specifik lipas såsom Vcheck V200, ett instrument, som analyserar cPL/fPL med en fluorescerande immunoassay. Syftet med studien var att utvärdera analys av cPL/fPL på Vcheck V200 samt jämföra resultatet från detta instrument med värdena från ett referenslaboratorium i Tyskland för att se om det fanns en signifikant skillnad mellan metoderna. Koncentrationen cPL i hundserum (n=37) och koncentrationen fPL i kattserum (n=29) analyserades på Vcheck V200. Dessa prover skickades även till referenslaboratoriet där analysen Spec cPL respektive Spec fPL utfördes. Spridningen var stor kring bias i Bland-Altman diagram för både cPL och fPL och jämförelsen mellan metoderna för de specifika koncentrationerna av cPL/fPL bedömdes vara statistiskt signifikant (p<0,05). 27% av hundproverna graderas olika enligt de båda metoderna och skillnaden var signifikant (p<0,05). 24% av kattproverna graderades olika men skillnaden var inte signifikant (p=0,257). Studien tyder på att jämförelsen mellan de båda metoderna var signifikant förutom vid graderingen av kattproverna. Beaktande detta och det faktum att kvalitetssäkringen brister vid analys av fPL på grund av avsaknad av kontroller kan i dagsläget inte cPL/fPL på Vcheck V200 ersätta den nuvarande golden standardmetoden, trots att ingen signifikant skillnad sågs vid gradering av kattprover. / Pancreatitis is a common disease in canine and felines and can be diagnosed by measuring the concentration of Canine Pancreas-specific Lipase (cPL) or Feline Pancreas-specific Lipase (fPL) in serum. Based on the concentration of cPL/fPL, the patient is then classified in different diagnostic categories (normal value, gray zone or indication of pancreatitis). Spec cPL and Spec fPL is currently the golden standard method for analysis of cPL and fPL. New methods have been developed for the quantitative measurement of pancreatic lipases. Vcheck V200, being one example, utilizing a fluorescent immunoassay for quantification of the lipase. The aim of this study was to evaluate the cPL and fPL analysis on the Vcheck V200 and to examine if there was a significant difference (p≤0,05) when comparing the result from Vcheck V200 with the results from a reference laboratory. The concentration of cPL (n=37) and fPL (n=29) in serum from canine or felines were analyzed using Vcheck V200. The samples were also sent to the reference laboratory where Spec cPL and Spec cPL were performed. A Bland-Altman plot comparison between the two methods showed a large spread for both analysis of cPL and fPL. Comparison of the specific values for analysis of cPL and fPL between the two methods revealed a significant difference (p<0,05). 27% of the dog samples were categorized differently according to the two methods and this difference was significant (p<0,05). 24% of the cat samples were categorized differently and no significant difference were observed (p=0,257). This study indicates that the difference between the two methods was significant, besides the classification of cat samples. Considering this and the lack of quality assurance regarding analysis of fPL due to lack of controls, the cPL/fPL analysis on Vcheck V200 cannot replace the Spec cPL or Spec fPL at present.

In the internet age, privacy and security have become major concerns since an increasing number of transactions are made over an unsecured network. Thus there is a greater chance for private data to be misused. Further, insider attacks can result in loss of valuable data. Hence there arises a strong need for continual, non-intrusive, quick user re-authentication. Previously, a number of studies have been conducted on authentication using behavioral attributes. Currently, few successful re-authentication mechanisms are available since they use either the mouse or the keyboard for re-authentication and target particular applications. However, successful re-authentication is still dependent on a large number of factors such as user excitation level, fatigue and using just the keyboard or the mouse does not mitigate these factors successfully.
Both keyboard and mouse contain valuable, hard-to-duplicate information about the user's behavior. This can be used for analysis and identification of the current user. We propose an application independent system that uses this information for user re-authentication. This system will authenticate the user continually based on his/her behavioral attributes obtained from both the keyboard and mouse operations. This re-authentication system is simple, continual, non-intrusive and easily deployable. To utilize the mouse and keyboard information for re-authentication, we propose a novel heuristic that uses the percentage of mouse-to-keyboard interaction ratio. This heuristic allows us to extract suitable user-behavioral attributes. The extracted data is compared with an already trained database for user re-authentication.
The accuracy of the system is calculated by the number of correct identifications to total number of identifications. At present, the accuracy of the system is around 96% for application based user re-authentication and around 82% for application independent user re-authentication. We perform black box, white box testing and Spec# verification procedures that prove the robustness of the proposed system. On testing POCKET, a privacy protection software for children, it was found that the security of POCKET was inadequate at the user level. Our system enhances POCKET security at the user level and ensures that the child's privacy is protected. / Master of Science

The paper analyses the problems that occur in the relationship between families, which raise blind children, and medical (or other) specialists and indicates the possible directions of improving and deepening these relations. The purpose of the paper: to analyse the experience of the relationship between the families, which raise the children with visual disabilities, and medical specialists. The subject of the paper: what is the experience of the relationship between the families, raising blind children, and medical specialists in the process of helping the child. The paper tried to purify the experience of each family, to sense the meanings, which parents endow their empathies with, to ascertain what suspenses and hopes they foster.
The qualitative analysis was taken for analysing the experience of the families and the specialists and the instrument for compilation the data was a qualitative interview. 7 women, raising the children with visual disabilities, participated in the research. The problems are researched in three aspects: firstly, the factors are identified, which help or disturb the families, raising the blind children, to overcome the psychological and spiritual discomfort, which is connected with the birth of the disabled child and his/her development, and which can have the influence to the health of the family members; secondly, the efforts are made to reveal the real requirements of these families, which are related to the services of medical and...
