Catch the dream Wave : Propagation of Cortical Slow Oscillation to the Striatum in anaesthetised miceFerreira, Tiago January 2014 (has links)
Under anaesthesia or in deep sleep, different parts of the brain have a distinctive slow oscillatory activity, characterised by states of high membrane potential and intensive spiking activity, the Up-states; followed by hyperpolarisation and quiescence, the Down-states. This activity has been previously described in vitro and in vivo in the cortex and the striatum, across several species. Here, we look into it, during anaesthesia, in the mouse brain. Using whole-cell patch-clamp recordings of cortical cells, it was possible to compare different signal processing methods used to extract the Up-and- Down states in extracellular recordings of the cortex. Our results show that the method based on the Multi-Unit Activity (> 200Hz) have better ac- curacy than High-Gamma Range (20 100Hz) or wavelet decomposition (< 2Hz band). After establishing the most robust method, this was used to compare the intracellular recordings of striatal cells to different parts of the cortex. The results obtained here support a functional connection between the dorsolateral striatal neurons and the ipsilateral barrel field. They also support a functional connection between dorsomedial striatal cells and the primary visual cortex. The analysis of delay between recordings allowed to establish temporal relationships between the contralateral barrel field, the ipsilateral barrel field, and the dorsolateral striatum; and between the ipsilateral barrel field, the ipsilateral primary visual field and the dorsomedial striatum. / <p>External Advisor: Dr. Ramon Reig, from Karolinska Institutet</p>

Les travaux entrepris au cours de cette thèse ont eu pour but d'étudier la modulation de la transmission synaptique inhibitrice au niveau des réseaux moteurs spinaux, à la fois au cours du développement et après lésion de la moelle épinière. Le nouveau-né présente des activités motrices spontanées qui jouent un rôle important dans la maturation des muscles et des réseaux de neurones de la moelle épinière. Dans une première étude, nous avons identifié l'un des mécanismes impliqués dans la genèse de ces activités chez le rat nouveau-né in vitro. Nous avons démontré que l'activation des récepteurs GABAᴀ au niveau des terminales d'afférences primaires joue un rôle majeur dans le déclenchement et la propagation de ces activités spontanées. Dans une deuxième étude, nous avons testé la robustesse des dépolarisations de nature GABAergique enregistrées in vitro, c'est-à-dire leur dépendance vis-à-vis des paramètres du milieu de perfusion. Nous avons démontré que l'action dépolarisante des neurotransmetteurs GABA/glycine au niveau des motoneurones et celle du GABA au niveau des terminales d'afférences primaires ne sont pas dues à une fourniture énergétique insuffisante. La dernière étude a été consacrée à la modulation de la transmission synaptique inhibitrice après lésion de la moelle épinière. Nous avons montré que l'activation des récepteurs 5-HT2 (R5-HT2), particulièrement celle de l'isoforme 5-HT2ᴀ, renforce le poids synaptique inhibiteur via une hyperpolarisation du potentiel d'équilibre des ions chlorure (ECl) et une augmentation d'expression de KCC2 au niveau de la membrane des motoneurones. / The aim of this thesis was to explore the modulation of the inhibitory synaptic transmission within the spinal motor networks, both during development and after SCI. Spontaneous movements are an ubiquitous feature of fetal and infant behavior. They provide signals that are important for the development of muscles and the assembly of neuronal networks in the spinal cord. In a first study, we characterized one of the mechanisms underlying spontaneous motor behaviors in the in vitro spinal cord preparation isolated from neonatal rats. We demonstrated that the GABA is playing a key role in promoting spontaneous activity through primary afferent depolarizations which reach firing threshold. In the second part of my thesis, we tested the robustness of the in vitro GABAergic depolarizations and their dependence on the aCSF parameters. We demonstrated that during development the depolarizing actions of GABA/glycine on motoneurons and GABA on primary afferent terminals are not due to inadequate energy supply. In the last part of my thesis, we focused on the modulation of the inhibitory synaptic transmission following SCI. We demonstrated that activation of the 5-HT2 receptors, particularly the 5-HT2ᴀ subtype, strengthens inhibitory synaptic transmission to spinal motoneurons by hyperpolarizing the reversal potential of Cl- ions (ECl) and by increasing the cell-membrane expression of KCC2. This phenomenon reduces spasticity after SCI in rats. Upregulation of KCC2 function by targeting 5-HT2ᴀ receptors therefore opens new therapeutic strategies for the treatment of spasticity following SCI.

Previous issue date: 2012-08-18 / Multi-Electrode Array, MEA, was developed more than thirty years ago. This planar device of multiple microelectrodes has been used to detect local electric potential variations created by the ion movement through the protein channels that traverses the cell membranes in the near neighborhood. MEA offers the possibility of non invasive registering of the cell and their network s activities, allowing to know how the neurons start to connect through the synapses forming a network and generating spontaneous electrophysiological activities. In order to understand the neural network dynamics in face of the spontaneous activities and its evolution in dissociated hippocampal cell cultures, important properties in the synaptic plasticity, in this work it is proposed the analysis of the evolution and modeling of the hippocampal cell cultures in MEA using the theory of Partial Directed Coherence and Complex Networks. There were used the electrophysiological records obtained using MEA60 System, of the dissociated neurons of 18 days old Wistar rat embyo, in an experiment denoted as 371, realized at the University of Genoa, Italy. As the results obtained using the Partial Directed Coherence approach, it was verified that the method is capable to detect neuronal connectivity in the neuron cultures using MEA, even with the noisy signals. It was also verified that different time delays between signals during application of the PDC method do not affect directly on the results of the causality. PDC allowed to show that in MEA the amount of direct connections resulted is less than the amount of indirect connections, through the microelectrodes. This can indicate that the neurons prefer to communicate through existing connections than creating new connections. It was also observed that it is easier to lose direct connections than indirect connections between microelectrodes through the time. Through the experiments it can be observed that the culture in 25 DIV (Days In Vitro) developed more amount of connections between neighboring electrodes, with less overall connections than the culture in 46 DIV, that had more overall connections with less neighborhood connections. Since one of the PDC features is the directionality detection between connections, it was observed direction changes through the connections through the time, even though we do not know the physiological meaning of these changes in the cognitive process. It was also observed that the established connections do not follow random patterns, showing an indicative of a free scale network, although we used small statistical measures to characterize the networks. / Há mais de 30 anos a Matriz de microeletrodos (Multi-Electrode Array,MEA) foi desenvolvida. Este dispositivo planar de múltiplos microeletrodos tem permitido detectar as variações do potencial elétrico local que são criadas pelo movimento de íons através dos canais de proteínas que atravessam as membranas das células neuronais em sua vizinhança imediata. A MEA oferece a possibilidade de gravação não-invasiva da atividade das células e de redes de células, permitindo-nos conhecer como os neurônios se comunicam através de sinapses formando uma rede e disparando atividades eletrofisiológicas espontâneas evocadas. No sentido de poder entender melhor a dinâmica das redes neurais em termos de atividades espontâneas e sua evolução em culturas de células hipocampais dissociadas, que são as propriedades mais importantes na plasticidade sináptica, neste trabalho é proposta a análise da evolução e o modelamento de culturas de células hipocampais na MEA usando a teoria de Coerência Parcial Direcionada (PDC) e de Redes Complexas. Foram usados os registros eletrofisiológicos obtidos usando o sistema MEA60, de neurônios dissociados de embrião de rato Wistar, de 18 dias de vida em um experimento denotado como 371, realizado na Universidade de Gênova, Itália. Como resultados obtidos ao utilizar o método de Coerência Parcial Direcionada, verificou-se que o método é capaz de detectar conectividade neuronal nas culturas de neurônios da MEA, mesmo com sinais com presença de ruído. Também verificou-se que tempos de atraso diferentes na aplicação do PDC não tem uma influência direta nos resultados de causalidade. O PDC permitiu mostrar que na MEA a quantidade de conexões diretas estabelecidas foram sempre em menor quantidade do que as conexões indiretas, através de caminhos. Isso pode ser um indicativo de que os neurônios preferem estabelecer comunicações usando caminhos ja existentes do que criando novas conexões. Também observou-se que com o passar do tempo é mais fácil se perderem conexões diretas do que conexões intermediárias entre neurônios. Através dos experimentos realizados pode se obervar que a cultura no 25 DIV (dias in vitro) desenvolveu maior quantidade de conexões com nós vizinhos, possuindo menos conexões em total, do que a rede no 46 DIV que teve maior quantidade de conexões total mas menor quantidade de conexões com nós vizinhos. Como uma das características do PDC é a deteção da direcionalidade nas conexões, observouse mudanças no sentido das conexões ao longo do registro, embora não saibamos ainda o significado fisiológico dessas mudanças em processos cognoscitivos. Também observou-se que as conexões estabelecidas não seguiam um padrão aleatório, encontrando-se um indicativo de um comportamento de redes livres de escala, embora não possamos afirmar isso por ter utilizado poucas medidas estatísticas para caracterizar as redes.

Analyse spatiotemporelle de données MEA pour l'étude de la dynamique de l'activité de la moelle épinière et du tronc cérébral immatures chez la souris / Spatiotemporal analysis of MEA data for the characterisation of the dynamics of developing hindbrain and spinal cord activity in the mouseAbdoun, Oussama 20 July 2012 (has links)
Tous les réseaux de neurones immatures génèrent une activité dite « spontanée »qui persiste même en l’absence de toute afférence et est impliquée dans de nombreux processus développementaux. Cette activité apparaît in vitro sous formes de vagues calciques ou électriques pouvant se propager sur de grandes distances et embraser toute la préparation. Toutefois, sa dynamique a été assez peu étudiée jusqu’à présent. En vue de combler quelque peu cette lacune, nous avons utilisé des matrices de microélectrodes (MEA) pour caractériser l’activité rythmique spontanée dans la moelle épinière embryonnaire de souris, sur des préparations aigues et en culture incluant également le tronc cérébral.Les enregistrements MEA produisent des volumes de données très importants qui nécessitent des outils d’analyse performants et adaptés à l’information que l’on souhaite extraire. Nous avons donc développé des méthodes pour la détection, la classification et la cartographie des patrons spatiotemporels d’activité dans les données multicanaux. Notre approche cartographique utilise l’interpolation par splines et est orientée vers la production de cartes multimodales combinant l’activité électrique et des données anatomiques ou biochimiques (marquages). Ces méthodes d’analyse nous ont permis de décrire très précisément l’évolution de l’activité spontanée aux stades précoces (E12.5–E15.5). Nous avons également montré que, à E14.5, l’activité est initiée dans le bulbe, plus précisément dans une région riche en neurones 5-HT, suggérant un nouveau rôle des voies sérotoninergiques descendantes dans la maturation des réseaux spinaux.Enfin, nous avons analysé les mouvements embryonnaires à E14.5 et avons découvert des caractéristiques analogues à la dynamiques spatiotemporelle des activités intraspinales. / Immature neural networks generate a peculiar type of activity that persists even in the absence of electrical inputs and was termed for this reason “endogenous”or “spontaneous”. This activity is ubiquitous and was found involved in a wide range of developmental events. In vitro, it can be observed as calcium or electrical waves propagating over great distances, often invading the whole preparation,but its dynamics remain poorly described. In order to somewhat fill this gap,we used multielectrode arrays (MEAs) to characterise the spontaneous rhythmic activity in the mouse developing spinal cord, in both acute and cultured isolated hindbrain-spinal cord preparations.To extract relevant information from the massive amounts of data yielded by MEA recordings, adapted analysis tools are needed. Thus, we have developedmethods for the detection, classification and mapping of spatiotemporal patternsof activity in multichannel data. Our mapping approach is based on the thin plates pline interpolation and includes the possibility to combine maps of activity with anatomical or stained data for multimodal imaging.These methods allowed us to analyse in great detail the evolution of spontaneousactivity at early stages (E12.5–E15.5). In addition, we have localised theinitiation site of E14.5 activity in the medulla and shown that it matches a densemidline population of serotoninergic neurons, suggesting a new role for 5-HTpathways in the maturation of spinal networks. Finally, we have recorded andtracked spontaneous limb movements of E14.5 embryos and found that features of motility were consistent with patterns of spinal activity.

Régulation originale de la balance énergétique du rat Lou/C : un modèle d’hyperactivité et de résistance à l’obésité / Original regulation of energy balance in lean Lou/C rats : a model of hyperactivity and resistance to obesityBelouze, Maud 17 December 2009 (has links)
Le rat Lou/C, issu de la souche Wistar, reste maigre tout au long de sa vie. Le but de ce travail était de caractériser la balance énergétique du rat Lou/C et d’établir quel(s) étai(en)t le(s) tissu(s) thermogène(s) impliqué(s) dans la dissipation de l’énergie alimentaire ingérée en excès. Si la quantité d’énergie ingérée, rapportée par unité de masse corporelle, n’était pas différente entre les deux souches de rats, la dépense énergétique du Lou/C était supérieure au repos, suite à un repas et lors de l’exercice physique. Le rat Lou/C montrait également une hyperactivité locomotrice spontanée volontaire bien supérieure à celle du Wistar. De façon inattendue, le tissu adipeux brun (BAT) des Lou/C était peu actif, comme l'ont montré des approches fonctionnelles in vivo, biochimiques in vitro ou moléculaires. L'absence d'activation du BAT du rat Lou/C n’était pas liée à une déficience du tissu puisqu’il était aisément activable par une exposition prolongée au froid. La forte activité physique spontanée du Lou/C ne s’accompagnait pas de l’activation de processus thermogènes particuliers des mitochondries isolées de BAT ou de muscle squelettique. En revanche, nous avons mis en évidence un mécanisme potentiel de découplage des oxydations phosphorylantes mitochondriales dans le foie des Lou/C. Contre toute attente, le rat Lou/C disposait de capacités de synthèse des acides gras équivalentes à celles du Wistar dans le foie et supérieures dans le tissu adipeux blanc avec de fortes capacités d’oxydation de ces substrats dans ces tissus, suggérant un possible cycle futile entre la synthèse des acides gras et leur oxydation dans le foie et le tissu adipeux blanc. Le rat Lou/C représente donc un modèle original de régulation de la balance énergétique qui n’est pas basé sur l’activité thermogène du BAT. Le muscle squelettique, le foie et le tissu adipeux blanc du Lou/C pourraient participer à un métabolisme actif des lipides et contribuer à la dissipation accrue de l’énergie ingérée en excès. / Lou/C rat, an inbred strain of Wistar origin, remains lean throughout life. Our study aimed to characterize the energy balance of Lou/C rats and determine the tissue(s) that could be implicated in the dissipation of excess energy intake. Food intake, expressed per unit body mass, was not different between the two strains of rats but resting metabolic rate, diet-induced thermogenesis and exercise-associated energy expenditure were higher in the Lou/C strain. Moreover, the spontaneous activity of Lou/C rats was amazingly higher than that of Wistar rats. Unexpectedly, the thermogenic brown adipose tissue (BAT) of Lou/C rats was not over-stimulated as demonstrated by functional in vivo, biochemical in vitro or molecular approaches. Nevertheless, Lou/C BAT was not deficient as it could easily be stimulated by prolonged cold exposure. The high spontaneous activity of Lou/C rats was not correlated with an activation of specific thermogenic processes in isolated mitochondria of BAT or skeletal muscle. However, oxidation and phosphorylation were partly uncoupled in liver mitochondria of Lou/C rats. Unexpectedly, Lou/C rats displayed similar capacities for fatty acid synthesis in liver but higher capacities in white adipose tissue than Wistar rats, in association with high capacities for oxidation of these substrates in these tissues. These results suggested a possible mechanism of futile cycling between fatty acid synthesis and oxidation in liver and white adipose tissue. Lou/C rats therefore represent an original model of regulation of energy balance that is not based on the thermogenic activity of BAT. Skeletal muscle, liver and white adipose tissue of Lou/C rats could contribute to a higher lipid metabolism and the dissipation of excess energy intake.

Neural basis and behavioral effects of dynamic resting state functional magnetic resonance imaging as defined by sliding window correlation and quasi-periodic patternsThompson, Garth John 20 September 2013 (has links)
While task-based functional magnetic resonance imaging (fMRI) has helped us understand the functional role of many regions in the human brain, many diseases and complex behaviors defy explanation. Alternatively, if no task is performed, the fMRI signal between distant, anatomically connected, brain regions is similar over time. These correlations in “resting state” fMRI have been strongly linked to behavior and disease. Previous work primarily calculated correlation in entire fMRI runs of six minutes or more, making understanding the neural underpinnings of these fluctuations difficult. Recently, coordinated dynamic activity on shorter time scales has been observed in resting state fMRI: correlation calculated in comparatively short sliding windows and quasi-periodic (periodic but not constantly active) spatiotemporal patterns. However, little relevance to behavior or underlying neural activity has been demonstrated. This dissertation addresses this problem, first by using 12.3 second windows to demonstrate a behavior-fMRI relationship previously only observed in entire fMRI runs. Second, simultaneous recording of fMRI and electrical signals from the brains of anesthetized rats is used to demonstrate that both types of dynamic activity have strong correlates in electrophysiology. Very slow neural signals correspond to the quasi-periodic patterns, supporting the idea that low-frequency activity organizes large scale information transfer in the brain. This work both validates the use of dynamic analysis of resting state fMRI, and provides a starting point for the investigation of the systemic basis of many neuropsychiatric diseases.
