(in English): The aim of the work was to perform in-depth analysis of M2KO mice both at baseline and upon a challenge with a cold stress and to explore the role of opposing receptors (i.e. adrenoceptors) in adaptation to lacking M2-receptors in the heart. We have performed receptor binding studies, study of receptor gene expression, echocardiography, telemetric monitoring of heart rate, body temperature and activity, heart rate variability and biorhythm analysis, analysis of heart rate response to the application of drugs (carbachol, atropine, isoprenaline, propranolol), assessment of adenylyl cyclase and NO synthase activity, measurement of catecholamine blood concentration and gene expression of catecholamine-synthesizing enzymes. We have found that the disruption of M2-receptor gene caused a compensatory decrease of cardiostimulatory β1-adrenoceptors and β2-adrenoceptors with corresponding down-regulation of the gene expression, M3-receptors were down-regulated as well. Missing M2-receptors were functionally replaced by the main cardioinhibitory β3-adrenoceptors that were up-regulated, not by cardioinhibitory M4-receptors. β3-adrenoceptors were found to signal through adenylyl cyclase instead of NO synthase. Cardiovascular diseases (CVDs) are the most widely spread diseases of modern civilization. Mechanisms involved in the protection of myocardial tissue are for that very reason in the focus of cardiovascular research. The adaptation to chronic hypoxia has been studied for many years in the context of its positive effects on heart function and its increased tolerance to ischemia-reperfusion (I/R) injury. This Master thesis describes the role of Akt kinase in the mechanisms leading to myocardial protection against I/R injury using the model of adaptation to chronic normobaric hypoxia (CNH). The hearts from male Wistar rats, that were kept in normoxic or hypoxic conditions (O2 0.1) for the period of 3 weeks, were retrogradely perfused by oxygenated Krebs-Henseleit solution and then subjected to 10 min of ischemia and 10 min of reperfusion. Samples prepared from left ventricles (LV) of experimental hearts were later used for protein analyses. The adaptation to CNH leads to increased phosphorylation of Akt kinase on Ser473, but it did not affect the phosphorylation on Thr308 nor the total protein level of Akt. and keywords The aim of this work was to evaluate the effect on rat myocardial β-adrenergic system of short-term (10 hours and 3 days) and long-term (5 weeks) cold exposure of rats with possible subsequent two-week recovery at normal temperature. The subtypes of β-adrenergic receptors (β-AR), their cognate G-proteins and adenylate cyclase have been characterized. β-AR are important components of cardiac regulatory mechanisms. They are involved in stimulating G-protein (Gs) and adenylate cyclase to increase cardiac contractility and frequency during stressful situations, including cold exposure. Heart tissue contains all three β-AR subtypes (β1-AR, β2-AR, β3-AR). While β1-AR couples with only Gs, β2-AR and β3-AR interact with the inhibitory G-protein (Gi). Electrophoresis and Western blotting showed a significant increase in β1-AR after a three-day exposure to cold. There was also a significant increase in β3-AR concentrations after a five-week cold exposure and this increase lasted for two weeks. There were no significant changes in the amnounts of β-AR cognate G-proteins (Gαs, Gβ, Gαi1/2 a Gαi3). This thesis focuses on cardiac imaging and quantification of T1 relaxation time in rat hearts. Its main focus is to investigate available methods for such quantification and their application in the development of quantification tools. The large impact is given to methods of acquisition synchronization, mainly with respect to cardiac motion and breathing using retrospective gating, where the navigator signal is obtained solely from the acquired data, so without any external equipment such as the ECG or respiratory sensors. This paper takes into account situations where steady-state has been reached and also those where it has not, by means of contrast agent injection or by inversion pulses.

Author: Bc. Michaela Tejnecká Title: The effect of regularly applied Nordic walking on selected cardiorespiratory parameters in adults after heart valve surgery. Aim: The aim of this work was to analyze the effect of six months of Nordic walking intervention on selected casrdiorespiratory parameters in adults after heart valve surgery. Methods: The study included ten individuals (mean age 62 years) who were at least five years apart after heart valve surgery. Blood pressure (BP) was measured using a pulsed wrist tonometer, blood oxygen saturation (BOS) and heart rate (HR) were measured using a pulse oximeter, and the ejection fraction of the heart (EF) was evaluated echocardiographically. Nordic walking training units took plaxe once a week for 6 months. During this intervention, the level of training gradually increased both in distance and elevation of the terrain. Results: In all probands there was an average reduction in systolic blood pressure during exercise by an average of 10,3 mmHg and a reductionin heart rate during exercise by an average of 24 beats per minute. The resting ejection fraction increased by an average of 5,5% after six month intervention. These changes were statistically significant (p<0,01). Master’s thesis deals with theme of heartbeats. Monitoring and evaluation of heartbeat is important for diagnostician cardiac dysfunction. First part of thesis is focused on anatomical description of heart and his operation. Another part is description of electrical activity of the heart and methods of capture. There is also mention of the cardiac rhythm and its disorders. One of the principal objective of this thesis is to study methods for sensing heart rate. These methods are analyzed and then selected a particular method for further solutions. The core thesis is to design functional cardiotachometer. Cardiotachometer derives the heart rate signal from phonocardiographical signal, which is acoustic manifestation of heart. Implementation takes place in the Labview, which is graphical programming environment. Suggested the device measures the current and average heart rate and the addition of HRV analysis in time domain.

The first two chapters are devoted to theoretical analysis of issues relating to portable EKG monitor. This chapters are aimed to further acquaint the reader with the issue for subsequent easier of understanding on further steps in the implementation of the device which will be contain in the following chapters. The first chapter is mainly a medical nature. We describe the anatomy and functional properties of the heart. In the next chapter we are deal with detail of electrocardiograph and the conditions for its proper function. Other chapters are devoted to the actual implementation of the facility. We are dealing with the issue of build functional portable EKG monitor. This practical part begins by fourth chapter, which consists block diagram of the device and discusses the required parameters. Next most extensive chapter is devoted to a detailed breakdown of the electrical circuit, including the calculation of the individual components. The last chapter describes the implementation and outcome measurements on test persons.
