Transient State Monitoring and Fluorescence Correlation Spectroscopy of Flavin Adenine Dinucleotide

Many human diseases including cancer have been associated with altered cellular metabolism and a changed oxygen consumption in cells. Fluorophores are sensitive to their local environment due to their long life times in transient dark states. A recent study successfully utilized this sensitivity to image differences in oxygen concentrations in cells using transient state (TRAST) microscopy together with fluorescent labels [1]. A natural continuation of this study is to investigate the possibilities of using this method with natural fluorophores already present in cells and thereby avoid articial labeling. Flavin adenine dinucleotide (FAD) is an auto fluorescent coenzyme that is naturally present in cells and involved in cellular metabolism. This project is an exploratory pilot study for cellular measurements with the aim to investigate if FAD can be used to probe oxygen concentrations in aqueous solution using transient state monitoring and fluorescence correlation spectroscopy (FCS). This thesis includes the results from FCS and TRAST experiments on FAD in aqueous solutions with different oxygen concentrations as well as different ascorbic acid concentrations. The performed experiments showed that FAD monitored with TRAST is sensitive to differences in oxygen concentrations for the aqueous solutions used in this study.

Currently, many governments are developing programs and environmental initiatives aimed at reducing the level of harmful emissions into the atmosphere through the implementation of environmental enterprises. In 2018, the Ecology National Project was adopted in Russia, with a total budget of 54 trillion rubles. Within the framework of the National Project, 12 federal projects are being implemented, one of which, the Clean Air project, is aimed at reducing the level of air pollution in large industrial centers and at improving the existing monitoring system, since the reliability of the results obtained during implementation depends on its effectiveness. federal project "Clean Air" results. / В настоящее время правительства многих стран разрабатывают программы и экологические инициативы, направленные на снижение уровня выбросов вредных веществ в атмосферу за счет реализации природоохранных предприятий. В рамках Национального проекта реализуется 12 федеральный проектов, один из которых, проект «Чистый воздух», направлен на снижение уровня загрязнения атмосферного воздуха в крупных промышленных центрах и на совершенствование существующей системы мониторинга, так как именно от эффективности её деятельности зависит достоверность полученных в ходе реализации федерального проекта «Чистый воздух» результатов. Актуальность исследования определяется необходимостью разработки предложений по совершенствованию системы экологического мониторинга в ГО Нижний Тагил одного из городов-участников эксперимента по комплексному квотированию выбросов рамках Федерального проекта «Чистый воздух» и национального проекта «Экология».

QoS routing for mobile ad hoc networks using genetic algorithm

Mobile Ad Hoc Networks (MANETs) are a class of infrastructure less network architecture which are formed by a collection of mobile nodes that communicate with each other using multi-hop wireless links. They eliminate the need for central management, hence each node must operate cooperatively to successfully maintain the network. Each node performs as a source, a sink and a router. Future applications of MANETs are expected to be based on all-IP architecture, carrying a multitude of real-time multimedia applications such as voice, video and data. It would be necessary for MANETs to have an efficient routing and quality of service (QoS) mechanism to support diverse applications. This thesis proposes a set of cooperative protocols that provide support for QoS routing. The first is the on-demand, Non-Disjoint Multiple Routes Discovery protocol (NDMRD). NDMRD allows the establishment of multiple paths with node non-disjoint between source and destination node. It returns to the source a collection of routes with the QoS parameters. The second part of the protocol is the Node State Monitoring protocol for the purpose of monitoring, acquisition, dissemination and accumulation of QoS route information. The third part of the protocol implements the QoS route selection based on a Genetic Algorithm. The GA is implemented online with predetermined initial population and weighted-sum fitness function which operates simultaneously on the node bandwidth, media access delay, end to end delay and the node connectivity index (NCI). The term node connectivity index is a numerical value designed to predict comparatively the longest time a node-pair might be connected wirelessly.

Peut-on contraindre un État à sauver l'environnement? Si les récents jugements rendus en droit interne semblent vouloir répondre à cette question par l'affirmative, cela n'est pas évident en droit international. Érigée autour de ce plafond de verre hérité du traité de Westhphalie, la sphère juridique internationaliste se voit confrontée aux limites qu'elle s'est elle-même imposée : l'impossibilité de contraindre un État à agir contre sa volonté. Dans le but de brider les actions de ces entités supranationalement toutes puissantes dans leur propre domaine de compétence, maintes conventions environnementales ont été conclues. Au sein d'un processus de fabrication normatif contrôlé par le sujet et l'auteur de la disposition à venir, les organisations internationales se rendent compte de leur impuissance devant la difficulté de convenir d'une surveillance intrusive et de sanctions rigides, en somme d'un mécanisme de contrôle des obligations efficace. La surpuissance de l'État n'est pas pour autant synonyme d'invincibilité sur la scène internationale, en ce que divers moyens ne cessent d'apparaître dans l'optique de mener à une gouvernance environnementale mondiale désirée depuis des décennies. / Can we compel a State to save the environment? Whereas recents judgements made in domestic law seems to want to answer to this question in an affirmative way, it is not as clear in international law. Erected around the Westhpalian-inherited glass ceilling, the international legal sphere is confronted to limits that were self-imposed : the impossibilty to constrain a State to act against its will. In an attempt to restrain the acts of those supranational all-mighty entities in their competencies, many environnemental agreements were made. Inside a process of manufacturing new norms that is controled by the subject and the author of the future provision, the international organizations are aware of the fact that they are powerless. Especially regarding the difficulty to strike a intrusive surveillance or rigid sanctions, in sum anything that is included in an effective compliance mechanism. The State's supreme power is not tied with an invincibility on the international scene, as diverses means to lead to global environnemental governance are starting to emerge.
