Vliv pomocného přípravku Pentakeep na vybranou modelovou kulturu zeleniny

The aim of the Diploma thesis was to evaluate the effect of auxiliary preparation Pentakeep super on quality and yield of lettuce. The theoretical part of the thesis is trying to approach the issue of plant stress and effect of the Pentakeep super for plants. The practical part of the thesis describes the experiment, which was based at the Faculty of Horticulture in Lednice in Moravia in 2013. Tested crop was lettuce (Lactuca sativa var. Capitata L.) variety 'Lento' and 'Jupiter'. Effect of preparation Pentakeep super was studied on morphological parameters (weight, leaf area, yield) and ingredients (vitamin C, nitrate, percentage of dry matter). After statistical processing of results the effect of treatment was a demonstrable only on yield, weight and vitamin C content.

This thesis is a summary of the experience gained during my studies. It compares the functioning of student, independent and professional theater. The term diagnose is used as a possible metaphor for the approach in thinking about the acting profession and the acting experience itself. The thesis discusses the various types of work-related stress. It seeks to analyze and describe their origin. We ask: Can the acting profession be considered a job for the mentally ill? To what extent is this profession pathogenic? And we also attempt to find answers to questions considering phenomena of putting oneself into one's position, experiencing a role, the embodiment and trans-bodiement. It is based on a comparison of the experience of rehearsing completely different genres in completely different areas and types of functioning.

Ovlivnění odpovědi rostlin na teplotní stres modulovanými hladinami cytokininů - fenomická a protemická analýza

Cytokinins are important group of phytohormones regulating many physiological processes ranging from cell division to programmed cell death. This thesis is focused on effects of cytokinin levels in response to heat stress in Arabidopsis thaliana. Analysis of transgenic plants with regulated expression of ipt and HvCKX showed that cytokinins and their optimal levels play important role in the morphological alterations induced by heat stress. Seedlings with increased and decreased levels of cytokinins exhibit inhibition of petioles growth, decreased length of blades of true leaves and reduced leaf area. To obtain insights into molecular events underlying early response to heat stress LC-MS analysis of whole proteom was performed. Analysis revealed 57 differentialy regulated proteins in response to heat stress in Columbia ecotype. On the cellular level, most of the proteins were located in cytosol (47 %) nebo plastids (32 %). Coparative analysis between wild-type seedlings and seedlings with decreased level of cytokinins confirmed 31 proteinInfluencing plant responses to temperature stress modulating cytokinin levels - fenomic and proteomic analysiss regulated by cytokinins in response to heat stress. Among these proteins, desarurase 7 and Tudor SN1 protein were previously found as important factors in response to heat stress.

The aim of the diploma thesis is to assess and analyse the value of the character test in general, both in the Czech Republic and Sweden. Their quality to reveal the real personality, emotional stability, fear reactivity and aggressive reactions. Czech character test is required as a condition for breeding for both Belgian and Australian shepherds by the Belgian Shepherd Club of Czech Republic. The aim of the analysis is to measure the differences between the tasks and the results of the Czech character test and highly-developed Swedish Mental test that reveals the character in a more complex way and is generally more demanding also from time management side. Furthermore the analysis explores the possible relation between the results of the character tests and trial performance in dogs undergoing this research. The testing ran in three terms, while both tests were conducted in a single day for each participant. The target group were owners of Belgian and Australian shepherds with dogs between 1 to 4 years old. The total number of tested dogs is 17. The Swedish test proved higher connectivity of the tasks (high correlations) as wells as higher results variability between tested animals as opposite of the Czech test. The results of this pilot project study did not show any connection between the high score in the test and higher probability of passing the working exam, although this result can be influenced by low number of animals tested and their lower age. This study works as pilot applied research to increase the effectivity and simplify the testing of working dogs, however, it is essential for next stages of research to focus on concrete parts of the test and their connectivity to the dog behaviour in a given situation.

Vliv kadmia na oxidační stres u rostlin / The effect of cadmium on oxidative stress in plants

The two ecotypes of Noccaea caerulescens originating from two differently contaminated sites (Redlschlag, Austria; Ganges, France), N. praecox (Mežica, Slovenia), Arabidopsis halleri and Spinacia oleracea were studied to compare Cd accumulation and tolerance. After 30, 60, 90 and 120 days of cultivation in Cd contaminated soil (30, 60 and 90 mg Cd/kg soil) for hyperaccumulators and 25, 40, 55 and 75 days of cultivation in Cd contaminated soil (30, 60 and 90 mg Cd/kg soil) for spinach gas-exchange parameters (net photosynthetic rate, transpiration rate, stomatal conductance, intercellular CO2 concentration), fatty acids, macro- and microelements were determined as well as N utilization by plants. Noccaea species and A. halleri showed similar changes in yield of biomass, Cd content and total content of amino acids. These species confirmed differences in the profile and contents of individual free amino acids. The comparison between Noccaea species showed that Cd stress resulted in similar changes of gas-exchange parameters. Contrasting responses of plants to Cd contamination were confirmed in macro- and microelements contents, fatty acids and amino acids metabolism. Spinach showed contrasting response to cadmium compared to hyperaccumulators, especially in profile of free amino acids and fatty acids.

Empirical part will deal with the effects of stress on the driver's performance and resistance to him. methodics The theoretical part will be based on a compilation of texts from literature. In the empirical part will be used method of standardized questionnaires. For research purposes is determined hypothesis, which will be checked by statistical methods. The results of empirical research will be commented and presented in the summary tables and graphs.

Role cytokininů v regulaci odpovědi na nedostatek síry u Arabidopsis thaliana

Sulfur deficiency stress induces a number of adaptive responses which must be coordinated. Phytohormone cytokinin has been previously shown to regulate sulfur aquisition and utilizati-on, however, the cytokinin-regulation of sulfur nutrition remains elusive. In this diploma the-sis, it was reported that cytokinin induces a sulfur-deficiency-like gene expression which is accompanied by a decrease in pool of the key sulfur-containing compound glutathione. Further, it was found that the sulfur deficiency responses involve altered cytokinin status to coordinate developmental and/or metabolic adaptations. Arabidopsis plants deficient in cytokinin signaling showed disrupted root elongation in response to sulfur starvation and, unexpectedly, the root elongation in response to sulfur starvation was further stimulated by a low cytokinin concentration. Further, the role of cytokinin in sulfur nutrition was indicated in cytokinin signaling-deficient plants which accumulated more glutathione under normal nu-tritive conditions in comparison to wild-type, and vice versa, the decrease of glutathione pool was exaggerated during sulfur starvation. Altogether, results of this diploma thesis suggest that cytokinins participate in nutritional sulfur homeostasis control.

Vliv tepelného stresu na kvalitu ejakulátu kanců

The aim of the study was to investigate the effect of high temperature on selected parameters of semen quality of duroc boars at the insemination station in Velké Mezirici (N 49°23.46667', E 15°52.70135') in season from May to September. For purpose of the experiment were chosen 20 boars of the Duroc breed, divided into two groups. Group A (the control group; n = 10) has average quality of semen and group B (the experimantal group; n = 10) showed below-average long-term quality of semen. Analysed parametres were volume of ejaculate (ml), concentration of sperm (thousands/ml), total account of sperm in ejaculate (mld. ks), motility (%) and rate of abnormal sperm (%). In the stable the temperature (°C) and relative humidity (%) were monitored at hourly intervals for whole period of this study. The results of the experiment shows that the volume of ejaculate from both monitored groups increased at the same rate (P > 0.05) and in group A from 198 ml to 252 ml; in group B from 203 ml to 241 ml. Concentration of sperm of group A decreased (from 499 000/ml to 436 000/ml), whereas concentration of sperm of group B was at the same level during the experiment. The motility of sperm of group A at the end of the experiment increased (from 71.4 % to 74.0 %) and motility of sperm of group B has intensively decreased (from 67.3 % to 62.2 %). The decreation of sperm motility was statisticaly significant (P < 0.05). The influence of ambient temperature in this case had no significant effect on boar semen with average values (group A), but has been reported statistical significance in deterioration of sperm motility (P <0.05) and number of morphologically abnormal sperm (P < 0.05) in group of boars, who showed below-average long-term semen quality (group B). In both groups there was lower sperm concentration, which could be due to the increased volume of ejaculate in the summer months.

The presented thesis about using of auxiliary products for selected culture of vegetable is mainly engaged in evaluating the effectiveness of various products or their combination on yield. From morphological parameters were evaluated: the weight of the aboveground parts, tuber weight, weight of root, leaf area length, width of leaf area, height and diameter of the tuber. In terms of content substances was evaluated content of: dry matter, ascorbic acid, carotenoids, fiber, minerals (potassium, sodium, calcium and magnesium), flavonoids and phenolic compounds, refractive solids content and the total antioxidant capacity. The thesis additionally evaluates the economic costs of auxiliary products for one treatment. The reader is thus presented a brief description of the efficiency of individual products and comparison of financial performance of individual measures.
