I deal with the subject of stress in my thesis. The objective was to ascertain how the management employees of the Charity of the Archdiocese in Prague (hereinafter only ``CAP{\crq}q) experience and handle the stress associated with their work. I deal with ascertaining how CAP management employees handle stress in the practical section. I set two research objectives. The first objective was ascertaining the presence and level of stress that management employees experience and the second was identifying the stressors that most affect management employees. In order to achieve research objectives and verify the stipulated hypotheses, I utilized the anonymous questionnaire technique. I utilized two standardized questionnaires that ascertain how individual employees subjectively experience stress. One was Meister{\crq}s questionnaire evaluating neuropsychic stress and the second was the Stress Scale. Both objectives were met. The results showed that employees experienced commensurate stress in many areas. Most respondents experienced stress in only four areas of their work. The results also show the different ways in which man and woman experience stress. Based on the results of the research I proposed recommendations regarding handling stress and its prevention.

Metabolom rostlin a jeho proměny v reakci na stres

Adverse environmental effects have a significant impact on plants' life and influ-ence the plants' metabolome. Specific stress-related responses are manifested both on plant development and its metabolites. This thesis entitled "Plant metabolome and its response to stress" outlines the composition of plant metabolome and reviews stress-related metabolites and metabolomics processes. The experimental part summarizes the results of two independent experiments. First, Hordeum vulgare response to stress was followed via targeted LC-MS metabolomics and proteome analysis. Next, a drought response was analyzed in Solanum lycopersicum via GC-MS metabolome profiling. Results highlighted a tissue-specific response to stress and pinpointed several putative abiotic stress markers

Iron is an essential element for living organisms. However, as it is a transition metal, it can participate in Fenton reaction resulting in generation of free radicals and oxidative damage to tissues. Antioxidants may prevent possible iron toxicity by chelating free iron or scavenging free radicals. Falvonoids are naturally occurring substances that are capable of formation of complexes with metals, including iron. T h e y have been show to possess antioxidant activity, which depends on molecular complexity of numerous types of flavonoids, e.g. quercetin and silibinin. Bisphosphonates are synthetic drugs used to treat various metabolic diseases of bones. Their principál effect is an inhibition of osteclast activity leading to a decreased bone resorption. Bisphosphonates have been however shown to exert some antioxidant activity in in vitro experiments, too. The aim of this PhD thesis was to investigate the role of iron in toxicity of other metals (cadmium) and the effect of flavonoids (quercetin and silibinin) and bisphosphonates (clodronate, etidronate and risedronate) on iron-induced oxidative damage in vivo. This thesis focuses on the stress loads and stressors of the Police of the Czech Republic. In the theoretical part, stress, stress history, types of stress and where stress comes from is closely described. The next part explains the influence of the load and stress on health, and the consequences of stress overload. The end of the theoretical part is focused on the unitary armed security corps. Work stress, stressors and factors at Police of the Czech Republic are defined there. The empirical part discusses a research of the stress level in the workplace of the Police of the Czech Republic that was carried out in Chomutov, Jirkov, Ústí nad Labem and Prague via questionnaire survey. This part of the work provides an inspection of the general characteristics of the sample, its statistic evaluation and graphs with an interpretation of the results. At the end, the stress level is evaluated and stressors which affect the workplace of selected group of Police of the Czech Republic are identified. Finally, the measures for their limitation are proposed.

Subsequent goal is to carry out the empirical research on target group of university students and managers and to find out their approach against stress, stress management techniques and the level of psychohygiene in their lives. Results of the empirical research will be the base for the final recommendations.

Brassinosteroids (BRs) are a class of polyhydroxysteroids that have been recognized as a sixth class of plant hormones. In specific conditions BRs can positively influence the plant growth and development. The first isolated brassinosteroid was natural Brassinolide from rapeseed (Brassica napus L.). Recently BRs are extracted primarily synthetically; they are effective even in very low concentrations. Several studies of different plant species show positive influence of BRs on growth, flowering, germination and resistance to stress factors. Existing scientific studies that deal with BRs impact on plants were predominantly applied on agricultural crops. Until presence, the similar research in forest seed treatment and forest nursery was almost missing. In the Czech Republic the Department of Silviculture of the Faculty of Forestry and Wood Sciences of the Czech University of Life Sciences Prague newly started to deal with the use of BRs in forestry. The aim of this master thesis is to elaborate literature review on BRs and to summarize existing research of their impact on plants. Second part of the thesis solves influence of BRs on Scots pine (Pinus silvestris) seeds germination. Tests are performed in normal and in stress conditions. Results of the practical part show that BRs affect positively seed germination in normal conditions; seeds treated with BRs show higher germination rate than control seeds. Positive impact was not registered when the seeds were exposed to stress factor -- heat. Any research paper showing significant positive influence on forest seedlings in Central European conditions was not published yet.

Možnosti využití rostlinných stimulátorů jako prostředku pro omezení důsledků vláhového deficitu při pěstování zeleniny / The possibilities of using growth stimulants as agents for limiting the consequences of water deficit with growing vegetable

Field and laboratory experiments were conducted to investigate the influence of a synthetic brassinolide and Atonik on the growth and yield of onion (Allium cepa L.; cultivars ´Alice´ and ´Lusy´) and head lettuce (Lactuca sativa L. var. capitata; cultivars ´Maršálus´, ´Mars´ and ´Santoro´) grown at two irrigation levels. The treatment with brassinolide (1 nM) and Atonik (0.05 %) promoted germination at the beginning of the experiment in the onion (cultivar ´Alice´). The results also showed varietal differences in response to the treatments. In field trials under conditions with optimal irrigation the influence of the treatments with synthetic brassinolide in the onion (cultivar ´Alice´) was proven on the mass of the individual bulbs and yield in 2013. In the case of reduced irrigation, the treatment with a concentration of 1 nM had a statistically significant positive influence on the mass of the individual bulbs (2013) and yield (2012) in the cultivar ´Alice´. With the cultivar ´Lusy´, the treatment with a concentration of 1 nM and 0.01 nM significantly increased the marketable yield relative to the control variant (2012). The results of laboratory experiments showed that treatment of lettuce plants in the initial stage of the development with 1nM solution of synthetic brassinolide can be used to limit the consequences of reduced moisture conditions. Significant increase of marketable heads was noted after treatment with synthetic brassinolide in filed experiments (head lettuce - cultivar ´Santoro´). There were no significant differences among the ascorbic acid /nitrates contents of the treated vegetables. The results confirmed that it was possible to reduce the negative impact of the water deficit in the cultivation of onion and lettuce with growth stimulants. It is, however, necessary to bear in mind which parameter of the plants should be affected and last but not least also the varying sensitivity of the species and cultivars to the treatment.

The diploma thesis deals with the issue of a stress load in the working environment, specifically of employees of the criminal police. The theoretical part provides a general definition of basic concepts such as stress, personality or resistance. It introduces the various types of stress, its symptoms, causes and possible responses to psychological stress. The larger section is devoted to stress in the working conditions. It describes the most common stressors, risk of a burnout and possibility of stress reduction in general and directly for the police officers. The knowledge gained from the theoretical foundations is applied in the empirical part. The aim of the empirical part is to find the level of stress load among respondents from the ranks of the criminal police using qualitative methods of investigation, interview, in combination with the use of a questionnaire Meister. This section focuses on the positive and negative factors in the workplace, preventive measures and assesses differences based on gender, age, length of practice or marital status.

This paper is targeted on influence of various factors on biochemical blood parameters of horses, especially influence of housing management, physical aktivit, or stress. Its goal is to evaluace knowledge about blood content and use of biochemical indicators in horse dinase diagnostics. Also, brief chapter is dedicated to alternative pasture management, so called paddlock paradise. The second part of this work is experimental. The experiment took place on Tlumačov stud farm. There we took samples from 18 horses in period of 4 months. Results were established in laboratory and later evaluated by one-way ANOVA statistical method.
