Introdução. A identificação dos flebotomíneos baseia-se principalmente na morfologia do adulto, o que pode ser problemático quando as espécies são morfologicamente muito semelhantes. Psychodopygus é um gênero de flebotomíneos de grande interesse em saúde pública devido ao papel de algumas espécies na veiculação de Leishmania spp. no Brasil. No entanto, este gênero inclui espécies com fêmeas morfologicamente indistinguíveis que pertencem à Série Chagasi, sendo elas: P. chagasi, P. complexus, P. squamiventris maripaensis, P. squamiventris squamiventris e P. wellcomei. Objetivos. Investigar a possibilidade de distinguir essas espécies por meio de análises morfométrica e molecular, além de produzir uma distribuição geográfica atualizada para o grupo analisando a probabilidade de ocorrência das espécies através da análise de modelagem de nicho ecológico. Material e Métodos. Foi realizada a análise discriminante na morfometria geométrica (cabeça e asa) e linear, morfologia (usando microscopia óptica e eletrônica de varredura) e a análise do citocromo c oxidase subunidade 1 (COI), avaliando-se um total de 752 espécimes (460 fêmeas e 292 machos) dos seguintes estados Amapá, Amazonas, Ceará, Mato Grosso, Pará, Rondônia, Roraima e Tocantins. Mapas de distribuição foram produzidos através de dados obtidos do material analisado e de revisão bibliográfica. Resultados. A análise discriminante usando caracteres morfométricos lineares mostrou-se capaz de diferenciar todas as espécies, exceto P. complexus, que apresentou 2,2% de erro de identificação. A morfometria geométrica das asas foi incapaz de separar completamente as espécies através da conformação, mas o tamanho do centróide dos espécimes fêmeas falhou apenas em distinguir P. complexus de P. s. maripaensis. Por outro lado, a morfometria geométrica das cabeças foi capaz de distinguir todas as espécies com grande eficiência ao usar tanto a forma como o tamanho do centróide. A análise morfológica revelou que a coloração torácica, principalmente do pronoto e do pós-noto, pode ser usada para separar as cinco espécies em três grupos: P. chagasi, P. wellcomei / P. complexus e P. s. mariapaensis / P. s. squamiventris. Os resultados da análise de DNA Barcoding, mostraram um agrupamento semelhante ao observado na morfologia; embora os espécimes de P. wellcomei do estado do Ceará mostrem uma grande distância genética da população do estado do Pará, evidenciando que essa espécie possa representar um complexo. Quanto à microscopia eletrônica de varredura, foram avaliadas detalhadamente as estruturas das antenas, tórax e genitália masculina. Salientamos que no anepímero (tórax) foi observada uma escama tipo \"raquete\" modificada apenas em Psychodopygus s. squamiventris. A revisão da distribuição geográfica mostrou que as espécies possuem uma distribuição cis-andina, ocorrendo principalmente no bioma Amazônico. A nítida separação de algumas espécies pelo rio Amazonas, sugere que o surgimento do grupo ocorreu no período que se estende da orogênese dos Andes até a formação deste rio. Conclusões. O estudo possibilitou diferenciar completamente as fêmeas das cinco espécies da Série Chagasi utilizando o conjunto de dados obtidos por morfometria linear e geométrica e análises morfológicas e também apresentar novos caracteres morfológicos e padrões distribucionais que facilitarão a identificação de machos e fêmeas dessas espécies. / Introduction. The identification of sand flies is mainly based on adult morphology, which can be problematic when species are morphologically very similar. Psychodopygus is one of the sand fly genera of great interest in public health, due to the role of some species in the transmission of Leishmania spp. in Brazil. However, this genus includes species with morphologically indistinguishable females that belong to the Chagasi series, which includes: P. chagasi, P. complexus, P. squamiventris maripaensis, P. squamiventris squamiventris and P. wellcomei. Objectives. To investigate the possibility of distinguishing among these species by means of morphometric and molecular analyses in addition to producing an updated geographical distribution for the group, analyzing the probability of the occurrence of the species by the analysis of ecological niche modeling. Material and methods. The analyses of the cytochrome c oxidase subunit 1 (COI), geometrical (head and wing) and of linear morphometry and morphology (using optical microscopy and scanning electron microscopy) were carried out using a total of 752 specimens (460 females and 292 males) from the following states: Amapá, Amazonas, Ceará, Mato Grosso, Pará, Rondônia, Roraima e Tocantins. Distribution maps were produced on the basis of data obtained from the material analyzed and a bibliographical review. Results. The discriminant analysis using linear morphometric characters was able to differentiate among all the species, except for P. complexus, which presented a 2.2% error of identification. The geometric morphometry of the wings was unable to completely separate the species by means of the shape analyses, but the centroid size of the female specimens only failed to distinguish P. complexus from P. s. maripaensis. Otherwise, the geometric morphometry of the heads was sufficient to distinguish all the species with great efficiency, when using both the head-shape and the centroid size. The morphological analysis revealed that the thoracic coloration, mainly of the pronotum and the post-notum, can be used to separate the five species into three groups: P. chagasi, P. wellcomei / P. complexus, P. s. mariapaensis / P. s. squamiventris. The results of the Barcoding DNA analyses showed a cluster similar to that observed in the morphology; however, P. wellcomei specimens from the Ceará population showed a great genetic distance from the population of Pará, evidencing that this species may represent a complex. As for the scanning electron microscopy, the structures of the antennae, thorax and male genitalia were evaluated in detail. In the anepimerum (thorax) a modified \"racket\"-type scale was observed only in Psychodopygus s. squamiventris. The review of the geographical distribution showed that the species have a cis-Andean distribution, occurring mainly in the Amazonian biome. The separation of some species from the others by the Amazon river suggests that the appearance of the Chagasi series occurred in the period from the orogenesis of the Andes to the formation of this river. Conclusions. The results clearly differentiate the females of the five species of the Chagasi series using the data set of linear and geometric morphometry and morphological analyses, providing new morphological and distributional data that will facilitate the identification of the males and females of this group.

Phenotypic and molecular characterization of mice deficient in protein kinase A regulatory subunit type 1A (prkar1a) and catalytic subunit A (prkaca). / CUHK electronic theses & dissertations collection

A population of stromal cells that retains osteogenic capacity in adult bone (adult bone stromal cells or aBSCs) exists and is under intense investigation in relation to osteogenesis and relevant pathology. aBSCs may be different from their embryonic or neonatal counterparts, and are influenced by species-/age-specific and other factors. Mice heterozygous for a null allele of prkar1a (Prkar1a+/-, a gene encoding for cyclic adenosine mono-phosphate (cAMP)-dependent regulatory subunit of protein kinase A (PKA), developed bone lesions that resembled fibrous dysplasia (FD) originated from cAMP-responsive osteogenic cells. Prkar1a +/- mice were crossed with mice heterozygous for catalytic subunit Calpha (Prkaca+/-), the main PKA activity-mediating molecule and generated mouse model with double heterozygosity for prkar1a and prkaca (Prkar1a +/-Prkaca+/-). Unexpectedly, Prkar1a+/-Prkaca+/- mice developed a large number of osseous lesions starting at 2--3 months of age that varied from the rare chondromas in the long bones and the ubiquitous osteochondrodysplasia of tail vertebral bodies to the occasional sarcoma in older animals. Cells from these lesions were fibroblast- and FD-like, and almost always originated from an area proximal to the growth plate and adjacent to endosteal surface of the periosteum; they expanded gradually in the bone marrow space. These cells expressed osteogenic cell markers, showed higher PKA activity that was mostly type II (PKA-II) and display an alternate pattern of catalytic subunit expression, and surprisingly possessed higher cAMP levels. In addition, markers of bone synthesis and lysis were increased. Gene expression profiling not only confirmed an early (progenitor) osteoblastic nature for these cells but also showed a signature that was indicative of mesenchymal-to-epithelial (MET) transition and increased Wnt signaling, particularly the brachyury expression. These studies show that a specific subpopulation of aBSCs can be stimulated in adult bone by PKA-II and altered Calpha activity, generating the only available germline mutant mouse model of a disorder that has similarities to human FD. Estudo da invasão de hepatócitos de rato por Shigella flexneri: análise da influência da hipóxia sobre a injúria celular / Study of rat hepatocytes invasion by Shigella flexneri: analysis of hypoxia influence on cellular injury

O presente estudo avaliou a capacidade de invasão de hepatócitos de rato por Shigella flexneri (S. flexneri) nas condições de normóxia e hipóxia. O estudo do microambiente de hipóxia tem grande importância, por estar presente em muitas doenças hepáticas, além de aumentar a translocação quando presente no lúmen intestinal. Bactérias invasivas como S. flexneri podem romper a barreira intestinal e chegar ao fígado através da circulação portal. O efeito da invasão bacteriana das células hepáticas é pouco conhecido. Neste trabalho buscamos pesquisar as alterações morfológicas e funcionais de hepatócitos de rato após infecção por S. flexneri na presença e na ausência de hipóxia. Para esta finalidade foram utilizados hepatócitos de rato cultivados pela técnica de cultura primária. Vários parâmetros foram analisados, tais como: taxa de invasão celular pela bactéria, quantificação da produção e liberação de DHL, produção de TNF-, taxa de morte celular por apoptose e a expressão do fator de transcrição HIF-1a. Os resultados mostraram que a metodologia empregada para a obtenção do microambiente hipóxico foi satisfatória, com redução de 70% da pO2 inicial (atingindo 43.2 mmHg in vitro ou 6.5% O2). A invasão de hepatócitos de rato por S. flexneri foi menor nas células previamente expostas à hipóxia quando comparada com a invasão das células cultivadas em normóxia. A viabilidade dos hepatócitos não apresentou diferenças significativas entre os grupos experimentais, variando entre 74 e 86%. A liberação de TNF- nas situações de normóxia e hipóxia foi similar, embora as células infectadas em normóxia tenham aumentado a liberação desta citocina. Na condição de hipóxia + infecção a liberação de TNF- foi menor do que na condição de normóxia + infecção, porém ambos os grupos produziram aumento significativo da citocina em relação aos controles normóxicos e hipóxicos. Este resultado sugere que a presença da bactéria no interior das células aumenta significativamente a liberação de TNF-pelos hepatócitos. A produção de DHL também foi maior de forma significativa no grupo hipóxico em relação ao grupo normóxico, porém não apresentou alteração nos grupos infectados por S. flexneri após uma hora. As taxas de apoptose aumentaram nos grupos hipóxia e nos grupos infectados com S. flexneri de maneira similar, variando entre 24 e 31%, quando comparados aos grupos controle em normóxia. A expressão do fator de transcrição HIF ocorreu nos grupos: hipóxia, normóxia + infecção e hipóxia + infecção, evidenciando que a infecção por S. flexneri induz a expressão deste fator. Em seu conjunto, nossos resultados buscam contribuir para o maior conhecimento da interação entre S. flexneri e hepatócitos em condição de hipóxia e normóxia. Tal conhecimento poderá ser útil na construção de futuras estratégias para auxiliar no combate a esta importante bactéria invasiva, principalmente nos casos de septicemia / This study evaluated the invasiveness of rat hepatocytes by Shigella flexneri (S. flexneri) in normoxia and hypoxia conditions. The study of hypoxia microenvironment is of great importance, since hypoxia is present in many liver diseases and increases bacterial translocation when present in intestinal lumen. Invasive bacteria such as S. flexneri can disrupt the intestinal barrier and reach the liver through portal circulation. The effect of bacterial invasion in liver cells is poorly understood. In this study we investigated the morphological and functional changes of rat hepatocytes after infection with S. flexneri in the presence and absence of hypoxia. For this purpose we used primary cultures of rat hepatocytes. Several parameters were analyzed, such as: bacterial invasion cell rate, quantification of LDH production and release, TNF-a production, cell death rate by apoptosis and expression of the transcription factor HIF-1a. The results showed that the methodology used to obtain the hypoxic microenvironment was satisfactory, with 70% reduction of initial pO2 (to 43.2 mmHg in vitro or 6.5% O2). The invasion of rat hepatocytes by S. flexneri was lower in cells previously exposed to hypoxia compared with the invasion of cells grown in normoxia. The viability of hepatocytes showed no significant differences between experimental groups, ranging between 74% and 86%. The release of TNF-a in situations of normoxia and hypoxia was similar, although the infected cells in normoxia have increased the release levels of this cytokine. In hypoxia + infection condition the release of TNF-a was lower than normoxia + infection condition, but both groups produced a significant increase in cytokine release when compared to normoxic and hypoxic controls. This result suggests that the presence of bacteria inside the cells significantly increases the release of TNF-a by hepatocytes. DHL production was also significantly greater in the hypoxic group compared to the normoxic group, but had no change in the groups infected with S. flexneri after an hour. The apoptosis rates increased in hypoxia and infected groups in a similar way, varying between 24% and 31% when compared with control group in normoxia. The expression of HIF- 1a transcription factor occurred in hypoxia, normoxia + infection and hypoxia + infection groups, indicating that infection with S. flexneri induces the expression of this factor. Overall, our results sought to contribute to a greater understanding of the interaction between S. flexneri and hepatocytes under hypoxia and normoxia conditions. Such knowledge may be useful in building future strategies to assist in combating these major invasive bacteria

Νόσος του Parkinson και γνωστική δυσλειτουργία : συσχέτιση με τον κινητικό φαινότυπο και το γονίδιο της α4 υπομονάδας του νευρωνικού νικοτινικού υποδοχέα της ακετυλοχολίνης

ΜΕΛΕΤΗ Α΄ Στόχος: Να διερευνηθεί αν ο κινητικός υπότυπος της αστάθειας κορμού και δυσχέρειας της βάδισης (ΑΚΔΒ) σχετίζεται με τη γνωστική δυσλειτουργία που εμφανίζουν οι ασθενείς με νόσο του Parkinson (NP) χωρίς άνοια. Μέθοδοι: Χορηγήσαμε μια συστοιχία επιλεγμένων νευροψυχολογικών δοκιμασιών σε δύο ομάδες μη ανοϊκών ασθενών με ήπια έως μέτριας βαρύτητας νόσο κατηγοριοποιημένους είτε στον υπότυπο της ΑΚΔΒ είτε σε υπότυπο μη ΑΚΔΒ, καθώς και σε μια ομάδα υγιών μαρτύρων. Οι ομάδες εξισώθηκαν κατά το δυνατόν όσον αφορά δυνητικούς συγχυτικούς παράγοντες που επηρεάζουν τις νευροψυχολογικές επιδόσεις. Αποτελέσματα: Δε διαπιστώθηκαν σημαντικές διαφορές μεταξύ των δύο ομάδων ασθενών στην επίδοση σε οποιαδήποτε από τις χορηγηθείσες νευροψυχολογικές δοκιμασίες. Παρόλα αυτά, σε σχέση με τους μάρτυρες υπήρξε μια τάση διαφοροποίησης ως προς το κυρίαρχο πρότυπο της γνωστικής δυσλειτουργίας. Η ομάδα με τον υπότυπο της ΑΚΔΒ είχε βραδύτερες επιδόσεις σε μια δοκιμασία ψυχοκινητικής ταχύτητας και γνωστικής ευελιξίας, ενώ η ομάδα με υπότυπο της νόσου μη ΑΚΔΒ είχε χειρότερες επιδόσεις στις μετρήσεις της λεκτικής μάθησης και της οπτικοχωρικής αντίληψης. Συμπεράσματα: Ο υπότυπος της ΑΚΔΒ δε συσχετίσθηκε με σοβαρότερα γνωστικά ελλείμματα και έτσι είναι πιθανό οι μηχανισμοί της γνωστικής δυσλειτουργίας να είναι, έως ένα ορισμένο βαθμό, κοινοί ανεξάρτητα από τον κινητικό υπότυπο της νόσου. ΜΕΛΕΤΗ Β Στόχος: Να διερευνηθεί αν υπάρχει συσχέτιση μεταξύ της ΝΡ και του γονιδίου CHRNA4, το οποίο κωδικοποιεί την α4 υπομονάδα του α4β2 νικοτινικού υποδοχέα της ακετυλοχολίνης (nAChR). Mέθοδοι: Στη μελέτη συμμετείχαν 100 ασθενείς με ΝΡ και 105 μάρτυρες, εξισωμένοι ως προς την ηλικία και το φύλο και ανήκοντες στην ίδια πληθυσμιακή ομάδα με τους ασθενείς. Ο γενετικός δείκτης που εξετάσθηκε είναι ένας μονονουκλεοτιδικός πολυμορφισμός στο 5ο εξόνιο του γονιδίου CHRNA4 (dbSNP rs1044396). Έγινε απομόνωση DNA γονιδιώματος από περιφερικό αίμα και ακολούθησε ανάλυση μεγέθους περιοριστικών τμημάτων μετά από αλυσωτή αντίδραση πολυμεράσης και κατάτμηση των προϊόντων αυτής με το ένζυμο Hha I. Μια υποομάδα 42 ασθενών υποβλήθηκαν επίσης σε λεπτομερή κλινική και νευροψυχολογική εκτίμηση. Η στατιστική ανάλυση για τη σύγκριση της συχνότητας των αλληλομόρφων και των γονοτύπων μεταξύ των ομάδων έγινε με τη δοκιμασία χ2, και τον ακριβή έλεγχο Fisher εάν έστω ένα κελί είχε n<5. Υπολογίστηκαν οι σχετικοί κίνδυνοι και τα κατά 95% διαστήματα αξιοπιστίας τους που αντιστοιχούσαν στα αλληλόμορφα και τους γονότυπους. Χρησιμοποιήθηκε η λογιστική ανάλυση παλινδρόμησης εάν ήταν απαραίτητη η προσαρμογή για την ηλικία ή το φύλο. Αποτελέσματα: Οι συχνότητες των γονοτύπων στην ομάδα των ασθενών (TT 34%; CT 58%; CC 8%) σε σύγκριση με τις συχνότητες των γονοτύπων στην ομάδα των μαρτύρων (TT 28.6 %; CT 47.6%; CC 23.8 %) παρουσίασαν στατιστικά σημαντική διαφορά (χ2 = 9.48, df = 2, p = 0.009). Η ομοζυγωτία CC συσχετίσθηκε με χαμηλότερο κίνδυνο παρουσίας της ΝΡ (CC vs φορείς T: OR = 0.28; 95% CI = 0.12–0.65; p = 0.002; στατιστική ισχύς 93.1%). Παρατηρήθηκε επίσης απόκλιση στην κατανομή των αλληλομόρφων μεταξύ των ασθενών και των μαρτύρων. Υπήρχε σημαντικά χαμηλότερη συχνότητα του αλληλόμορφου C μεταξύ των ασθενών (37%) σε σχέση με τους μάρτυρες (47.6%) (χ2 = 4.73; df = 1; OR=0.65; 95% CI = 0.44–0.96; p = 0.03). Η ανάλυση διαστρωμάτωσης έδειξε ότι η διαφορά στην κατανομή των γονοτύπων μεταξύ ασθενών και μαρτύρων ήταν στατιστικά σημαντική και συγκριτικά μεγαλύτερη στο θήλυ φύλο σε σχέση με το άρρεν φύλο και στους ασθενείς με εκδήλωση ΝΡ σε όψιμη ηλικία ( > 50 ετών) σε σχέση με αυτούς που εμφάνισαν πρώιμης έναρξης νόσο (< 50 ετών). Οι ασθενείς με ΝP που ανιχνεύθηκαν να φέρουν το γονότυπο CC και υποβλήθηκαν σε νευροψυχολογική αξιολόγηση έτειναν να έχουν καλύτερα διατηρημένες τις γνωστικές λειτουργίες που σχετίζονται με την προσοχή και την ταχύτητα επεξεργασίας των πληροφοριών. Συμπεράσματα: Η παρουσία του αλληλομόρφου C (dbSNP rs1044396) του γονιδίου CHRNA4 συνδέεται με μειωμένο κίνδυνο ΝΡ κατά 35%. Επίσης, τα άτομα με το γονότυπο CC εμφανίζουν σχεδόν τρισήμισυ (3,5) φορές χαμηλότερο κίνδυνο νόσου του Parkinson. Η ποικιλομορφία του γονιδίου CHRNA4 φαίνεται ότι σχετίζεται ιδιαίτερα με την επιρρέπεια εκδήλωσης της ΝΡ με ηλικιακά όψιμη έναρξη της νόσου, και επίσης με τις γνωστικές λειτουργίες των ασθενών χωρίς άνοια, ειδικά αυτές που εξαρτώνται από την προσοχή και την οπτικοκινητική αντίληψη. / Study A Aim: To investigate whether there is an association of the postural instability and gait difficulty (PIGD) motor subtype with cognitive dysfunction in non-demented Parkinson’s disease (PD) patients. Methods: We administered a battery of selected neuropsychological tests to assess attention, psychomotor speed, executive functions (set shifting ability and inhibitory control), visuospatial perception and visual constructive ability to two groups of non-demented patients with mild to moderate disease classified either as PIGD or as non-PIGD subtype and to a group of healthy controls. Groups were matched on potential confounders of neuropsychological performance. Results: No significant differences were revealed between the two groups of patients in the performance of any of the administered neuropsychological tests. However, relative to controls there was a tendency towards a differential pattern of cognitive dysfunction. The PIGD group had slower performance in a test of psychomotor speed and cognitive flexibility, whilst the non-PIGD group performed worse in measures of verbal learning and visuo-spatial perception. Conclusions: The PIGD subtype was not associated with more severe cognitive deficits and may to a certain extent share common mechanisms of cognitive dysfunction with non-PIGD subtypes. Study B Aim: to investigate whether there is an association between PD and a variation in the CHRNA4 gene coding for the α4 subunit, the primary subunit of the α4β2 brain nicotinic acetylcholine receptors. Methods: Patients (N=100) and controls (N=105), matched on the basis of sex, age and ethnicity, were genotyped for a single nucleotide polymorphism at cDNA position 1860 lying within the 5th exon of the CHRNA4 gene. DNA was extracted from peripheral blood samples and genotyping was done by PCR-based restriction fragment length polymorphism analysis. A subset of 42 patients also received detailed clinical and cognitive assessments. Comparisons of allele and genotype frequencies between groups were performed using the χ2 test, and the Fisher exact test if one cell had n<5. The relative risk for genotypes and alleles was estimated through calculation of odds ratios (ORs) with 95% confidence intervals (CIs). Logistic regression analysis was used if adjustment for age or sex was necessary. Results: The genotype frequencies in the patients group (TT 34%; CT 58%; CC 8%) vs. the genotype frequencies in the control group (TT 28.6 %; CT 47.6%; CC 23.8 %) demonstrated a statistically significant difference (χ2 = 9.48, df = 2, p = 0.009). CC homozygosity was associated with a lower risk of PD (CC vs T carriers: OR = 0.28; 95% CI = 0.12–0.65; p = 0.002). Also, the allelic distribution was significantly different between patients and controls. There was a significantly lower frequency of the C allele among the patients with PD (37%) as compared with the controls (47.6%) (χ2 = 4.73; df = 1; OR=0.65; 95% CI = 0.44–0.96; p = 0.03). Stratified analysis showed that the difference in the genotypic distribution between cases and controls was significant among females but did not reach significance among males. The frequency of CC homozygotes was also significantly lower in the group of patients with late onset PD than in the controls, but it was not significantly different between the early onset group of patients and the controls. CC homozygotes also tended to have better performance than T carriers on measures of attention and psychomotor speed (Trail Making Test part A and Symbol Digit Modalities Test). Conclusions: The presence of the C allele at SNP rs1044396 of the CHRNA4 gene is associated with a decreased risk for PD by 35%. Moreover, the CC genotype lowers the risk for PD by ~ 3.5 fold. Variation in the CHRNA4 gene may particularly influence susceptibility for late onset PD and further be associated with measurable effects on overt cognitive performance of yet not-demented PD patients, specifically the part loading on attentional capacities.

Structural aspects of the ribosome evolution and function

L'année suivante, la structure aux rayons X de l'ensemble du ribosome bactérien a été publiée. Ces grandes réalisations ont ouvert une nouvelle ère dans l'étude des mécanismes de la synthèse des protéines. Dès lors, il est devenu possible de relier différents aspects de la fonction du ribosome à des éléments particuliers de sa structure tertiaire. L'établissement de la relation structure-fonction peut toutefois être problématique en raison de l'immense complexité de la structure du ribosome. En d'autres termes, pour que les données cristallographiques sur la structure tertiaire du ribosome soient vraiment utiles à la compréhension du fonctionnement du ribosome, ces données devraient elles-mêmes faire l'objet d'une analyse approfondie. Le travail, présenté ici, peut être vu comme une tentative de ce genre. En appliquant l’analyse systématique des structure cristallographiques du ribosome disponibles, nous avons essayé de résoudre deux problèmes fondamentaux de la biologie ribosomale concernant (1) la nature des réarrangements du ribosome qui ont lieu à différentes étapes de son cycle de fonctionnement et (2) la possibilité de reconstitution de l'évolution du ribosome du monde-à-ARN jusqu’à nos jours. Dans le premier projet, nous avons systématiquement comparé les structures du ribosome disponibles et de sa sous-unité afin d'identifier les domaines rigides, qui ont toujours la même conformation, et les régions flexibles dont la conformation peut varier d'une structure de ribosome à une autre. Il y a deux types de réarrangements structuraux connus dont nous voulions comprendre les mécanismes: le « ratchet-like movement » et la «fermeture de domaines ». Le premier a lieu au cours de la translocation du ribosome et est plus ou moins perçu comme une rotation d'une sous-unité par rapport à l'autre. Le deuxième se produit dans la petite sous-unité et est associé à la reconnaissance codon-anticodon au site A. La comparaison des conformations ribosomales disponibles a révélé les mécanismes spécifiques des deux réarrangements. Bien que la sélection de l'aminoacyl-ARNt appropriée au site A et la translocation du ribosome n'ont jamais été considérés comme ayant quelque chose en commun, nous démontrons ici que les réarrangements de la structure des ribosomes associés au premier processus répète les réarrangements associés au deuxième mais dans l’ordre inverse. En d'autres termes, pendant le cycle d'élongation, la fermeture de domaine et le « ratchet » peuvent ii être considérés comme un mouvement de va-et-vient, qui renvoie finalement le ribosome à sa conformation initiale. Dans le second projet, nous avons fait une tentative de reconstitution de l'évolution de l'ARNr 23S, du monde-à-ARN jusqu`à nos jours. Ici nous nous sommes basés sur la supposition que l'évolution de cette molécule a procédé par des insertions aléatoires des régions relativement courtes dans différentes parties de la chaîne poly-nucléotidique. Pour cela, nous avons élaboré des critères de l'intégrité de la structure ribosomale et présumé que lors de l'évolution, la structure du ribosome s’est toujours adaptée à ces standards. Nous avons examiné l'interaction de type A-mineur, un arrangement fréquent dans la structure de l’ARN ribosomique, constitué d'un empilement d’adénosines non-appariées, attachées à une double hélice. Nous avons supposé que dans toutes les interactions A-mineurs existantes dans le ribosome, la double hélice est apparue avant ou au moins simultanément avec la pile d’adénosines correspondantes. L'application systématique de ce principe à la structure tertiaire de l’ARN 23S a permis d'élucider de manière progressive l'ordre dans lequel les parties différentes de l’ARN 23S ont rejoint la structure. Pris ensemble, les deux projets démontrent l'efficacité de l'analyse systématique in-silico de la structure tertiaire du ribosome et ouvrent la voie à de futures découvertes. / In the year 2000, the first high-resolution structures of the individual ribosomal subunits became available to the public. The following year, the X-ray structure of the complete bacterial ribosome was published. These major achievements opened a new era in studying the mechanisms of protein synthesis. From then on, it became possible to attribute different aspects of the ribosome function to particular elements of its tertiary structure. However, establishing the structure-function relationships is problematic due to the immense complexity of the ribosome structure. In other words, in order to make the crystallographic data on the ribosome tertiary structure really useful for understanding of how the ribosome functions, it must be thoroughly analyzed. Here, based on systematic analysis of the available X-ray conformations of the ribosome we have tried to resolve two fundamental problems of the ribosome biology: concerning (1) the nature of rearrangements in the ribosome that take place at different steps of its functional cycle, and (2) the reconstruction of the ribosome evolution from the RNA world to present time. In the first project, we systematically compared the available structures of the ribosome and its subunits to identify rigid domains, which always have the same conformation, and flexible regions, where the conformation can vary from one ribosome structure to another. There were two known types of structural rearrangements whose mechanisms we wanted to understand: the ratchet-like motion and the so-called domain closure. The ratchet-like motion takes place during the ribosomal translocation and is roughly seen as a rotation of one subunit with respect to the other. The domain closure occurs in the small subunit and is associated with the cognate codon-anticodon recognition in the A-site. Comparison of the available ribosome conformations revealed the detailed mechanisms of both rearrangements. Although the selection of the cognate amino-acyl-tRNA in the A-site and of the ribosomal translocation have never been thought to have anything in common, we demonstrate that the rearrangements in the ribosome structure associated with the first process repeat in reverse order the rearrangements associated with the second process. In other words, during the ribosome elongation cycle, the domain closure and the ratchet-like motion can be seen as a back-and-forth movement, which eventually returns the ribosome to the initial conformation. iv In the second project, we attempted to reconstruct the evolution of the 23S rRNA from the RNA world to present time based on the presumption that the evolutionary expansion of this molecule proceeded though random insertions of relatively short regions into different regions of the polynucleotide chain. We developed criteria for integrity of the ribosome structure and presumed that during the evolutionary expansion, the ribosome structure always matched to these standards. For this, we specifically considered the A-minor interaction, a frequent arrangement in the rRNA structure consisting of a stack of unpaired adenosines tightly attached to a double helix. We presumed that in all A-minor interactions present in the ribosome, the double helix emerged before or at least simultaneously with the corresponding adenosine stack. The systematic application of this principle to the known tertiary structure of the 23S rRNA allowed us to elucidate in a step-vise manner the order in which different part of the modern 23S rRNA joined the structure. Taken together, the two projects demonstrate the effectiveness of the systematic in-silico analysis of the ribosome tertiary structure and pave the way for future discoveries. / Les résultats ont été obtenus avec le logiciel "Insight-2" de Accelris (San Diego, CA)

The Molecular Mechanisms for Maintenance of Cancer Stem Cells in Chronic Myeloid Leukemia: A Dissertation

Chronic myeloid leukemia (CML) is a clonal hematopoietic stem cell disorder associated with the Philadelphia chromosome (Ph) that arises from a reciprocal translocation between chromosomes 9 and 22, thereby resulting in the formation of the chimeric BCR-ABL oncogene encoding a constitutively activated tyrosine kinase. BCR-ABL tyrosine kinase inhibitors (TKIs) induce a complete hematologic and cytogenetic response in the majority of chronic phrase CML patients. However, TKIs cannot efficiently eradicate leukemia stem cells (LSCs) because of the insensitivity of LSCs to TKIs. Therefore, developing new strategies to target LSCs is necessary and critical for curing CML, and success of this approach depends on further understanding the molecular mechanisms by which LSCs survive and are maintained. In Chapter I, I briefly introduce CML disease, BCR-ABL oncoprotein, and TKIs. I also describe the identification and features of LSCs. Several key pathways in LSCs including Wnt/ß-catenin, hedgehog, FoxO, Bcl6 and HIF1, are discussed. I also propose our strategy to identify unique molecular pathways that are important for LSCs but not their normal stem cell counterparts. In Chapter II, I describe our finding about the function of the positive regulator, HIF1α, in CML development and LSC survival. I show that loss of HIF1α impairs the maintenance of CML through impairing cell cycle progression and inducing apoptosis of LSCs, and I also report that p16Ink4a and p19Arf mediate the effect of HIF1α on LSCs, as knockdown of p16Ink4a and p19Arf rescues the defective colony-forming ability of HIF1α-/- LSCs. As detailed in Chapter III and IV, through comparing the global gene expression profiles of LSCs and HSCs, I find two novel regulators, Blk and Scd1, which act as tumor suppressors in CML development. In Chapter III, I show that Blk is markedly down-regulated by BCR-ABL in LSCs, and that c-Myc and Pax5 mediate this down-regulation. Deletion of Blk accelerates CML development; conversely, Blk overexpression significantly delays the development of CML and impairs the function of LSCs. I also demonstrate that p27, as a downstream effector, is involved in the function of Blk in LSCs. Blk also functions as a tumor suppressor in human CML stem cells, and inhibits the colony-forming ability of human CML cells. In Chapter IV, I investigate the function of another negative regulator, Scd1, in CML LSCs, and find that expression of Scd1 is down-regulated in mouse LSCs and human CML cells. We report that Scd1 acts as a tumor suppressor in CML, as loss of Scd1 causes acceleration of CML development and overexpression of Scd1 delays CML development. Using a colony-forming assay, I demonstrate that Scd1 impairs the maintenance of LSCs due to the change of expression of Pten, p53 and Bcl2. Importantly, I find that both Blk and Scd1 do not affect normal hematopoietic stem cells (HSCs) or hematopoiesis. Taken together, our findings demonstrate that HIF1α is required for the maintenance of CML LSCs, and conversely that Blk and Scd1 suppress the function of LSCs, suggesting that combining TKI treatment with specific targeting of LSCs will be necessary for curing CML.

The rock cavy SCN is innervated in its ventral portion by terminals from the predominantly contralateral retina. It also contains neurophisin and vasoactive intestinal polypeptide immunoreactive cell bodies and neuropeptide Y and enkephalin immunopositive fibres and terminals and is marked by intense GFAP immunoreactivity. The IGL receives a predominantly contralateral retinal projection, contains neuropeptide Y and nitric oxide synthase producing neurons and enkephalin immunopositive terminals and is characterized by dense GFAP immunoreactivity. This is the first report examining the neural circadian system in a crepuscular rodent species for which circadian properties have been described.
