Estudio de factibilidad de distribución de productos Tesa Tape en Lima

El objetivo del presente “Estudio de Factibilidad”, fue demostrar la viabilidad de una distribuidora de cintas adhesivas especializadas industriales para el mercado de Lima en Perú. Por muchos años Perú ha sido un foco de progreso industrial, teniendo un PBI positivo durante los últimos 5 periodos, por encima del 5% y siendo reconocido a nivel mundial como una de las mejores economías emergentes de Latinoamérica. El mercado de cintas adhesivas tiene un potencial de más de 18 millones de dólares. Se destacan en el Perú solo tres empresas que atienden el mercado de cintas adhesivas con productos commodities, y solo una marca, de nombre 3M tiene productos especializados, con alrededor de 20 distribuidoras. De otro lado, una de las compañías más importantes en cintas adhesivas industriales de procedencia Alemana, identificó a un solo competidor directo dentro del mercado peruano y decidió incursionar en el. En ese sentido, en la actualidad la compañía Tesa Tape se encuentra en la búsqueda de un distribuidor que realice la labor de penetración y posicionamiento de sus cintas adhesivas, a través de su marca tesa, signo distintivo de gran reconocimiento internacional, siendo además líder en el mercado europeo. Las cintas adhesivas industriales, al ser productos de venta y aplicación especializada disponen de unos márgenes de ganancia que alcanzan entre un 30% a un 40%, asimismo como su concentración se da en el sector industrial no requieren de gran inversión en publicidad. Sin embargo, sí se hace necesaria la asesoría y acompañamiento técnico que será brindado por los asesores de la distribuidora, quienes deben estar muy bien capacitados, razón por la que la compañía tesa tape está dispuesta a invertir en su entrenamiento, ya sea que este se lleve a cabo en alguna de sus sedes del exterior, o de manera local. La distribuidora contaría con una estructura organizacional compuesta por 3 personas: una contadora-administradora, encargada de la administración y el control contable; dos representantes de ventas, quienes tendrían como objetivo cubrir geográficamente a la ciudad de Lima y un outsourcing de transporte. Asimismo se contará con una oficina comercial ubicada en una zona industrial y estratégica para la atención rápida y oportuna de los clientes. Con el plan financiero se desprende que con una inversión de capital de $ 300 mil dólares es viable realizar el proyecto simulado a un costo de capital del 13.4% el cual entrega un VAN de $ 77,725 dólares, una TIR de 19.3% y un Pay Back de 3.9 años.

Structural and Functional Characterization of Proteins involved in PDIM biosynthesis in Mycobacterium tuberculosis

Tuberculosis is an infectious disease caused by the pathogen Mycobacterium tuberculosis which is still a public health threat. Due to the appearance of multidrug-resistant M. tuberculosis strains, it is urgent to find new drugs to fight against tuberculosis, especially against the extensively drug resistant tuberculosis. The unique impermeable mycobacteria cell wall, rich in lipids, is a key barrier to anti-tuberculosis drug uptake. One of these lipids, the phthiocerol dimycocerosate (PDIM), is related to mycobacteria virulence, drug resistance and mycobacteria cell wall integrity. Therefore, research focusing on PDIM biosynthesis potentially provides a new route for the development of new anti-tuberculosis drugs.In this context, we investigated two proteins, the thioesterase TesA and the chaperonin GroEL1, that are involved in the PDIM biosynthesis, and studied their structural and functional properties. Firstly, using various substrates, we demonstrated that TesA displays thioesterase and esterase activities and showed that TesA undergoes substrate-dimerization. A new TesA inhibitor, methyl arachidonyl fluorophosphonate (MAFP), was identified that irreversibly binds to the TesA catalytic serine residue. Furthermore, MAFP can increase the susceptibility to vancomycin in a TesA inhibition dependent manner while it can also affect the M. bovis BCG biofilm formation through broader action mechanisms. Secondly, we observed that the mycobacterial GroEL1, unlike the other chaperonins, E. coli GroEL and mycobacteria GroEL2, increases copper tolerance. We demonstrated that the copper can specifically bind to the GroEL1 with a strong affinity for its natural carboxyl terminal histidine-rich region. The classical feature of chaperonins is also increased in the presence of copper. These results suggest that GroEL1 could help mycobacteria to tolerate high bactericidal metal concentrations during macrophage infection, potentially through a metal storage mechanism. Last but not the least, we identified new mycobacterial GroEL1 and GroEL2 activities. In one word, the biochemical properties of two essential proteins, GroEL1 and TesA, involved in PDIM biosynthesis in M. tuberculosis have been further elucidated. Estudo clínico da infecção natural por T. cruzi em cães residentes em uma área rural de Mato Grosso do Sul, Brasil

Foram utilizados métodos sorológicos convencionais e não convencionais em 75 cães residentes na área e avaliação cardiovascular e da bioquímica sérica em quatro animais confirmadamente chagásicos. Foram usados os testes de reação de imunofluorescência indireta (RIFI), ELISA com antígenos da forma epimastigota do T. cruzi (EAE ELISA) e ELISA com antígeno excretado e secretado da forma tripomastigota do T. cruzi (TESA-ELISA), que detectaram anticorpos em 45,33% (n=34), 24,0% (n=18) e 12,0% (n=09) dos cães, respectivamente. A real prevalência da infecção foi confirmada como 10,7% (n=08) pela técnica de immunoblotting com antígeno secretado e excretado da forma tripomastigota do T. cruzi (TESA-blot). O teste com melhor índice de concordância (Kappa; 0,93), sensibilidade (100%) e especificidade (98,5%) foi o TESA-ELISA, que quando associado à RIFI igualou-se aos dados obtidos com o TESA-blot (10,7%). Três dos quatro animais chagásicos apresentaram aumento da silhueta cardíaca direta, aumento da duração de onda P e do complexo QRS. Um cão apresentou bloqueio de ramo direto e outro, bloqueio atrioventricular de primeiro grau e parada sinusal. Notou-se, também, espessamento de parede livre de ventrículo esquerdo em diástole, redução da fração de encurtamento e inversão dos picos de velocidade das onda E e A, indicando disfunção sistólica e diastólica... / This study was conduted to describe the clinical characteristics of the natural infection caused by the T. cruzi in dogs residing in a rural area of Mato Grosso do Sul, Brazil. Conventional and nonconventional diagnosis methods were used in 75 dogs living in this area for screening T. cruzi infection. Cardiovascular and biochemical serum were also performed in four confirmed positive dogs. The following tests were used: indirect immunofluorescence test (IFAT), enzyme-linked immunosorbent assay with T .cruzi. epimastigote antigens (EAE ELISA) and enzymelinked immunosorbent assay with T.cruzi excreted-secreted trypomastigote antigens (TESA-ELISA) with detected antibodies in 45,33% (n=34), 24,0% (n=18) and 12,0% (n=09) of the dogs, respectively. The real prevalence of the infection was confirmed as 10,7% (n=08) by immunoblotting test with T. cruzi excreted-secreted antigens (TESAblot). The test with the best concordance index (Kappa; 0,93), sensibility (100%) and specifity (98,5%) was the TESA-ELISA, which, when associated with IFAT had the same as results those obtained with the TESA-blot (10,7%). Three out of the four chagasic animals showed enlarged cardiac silhouette on x-ray and an increase of the P-wave duration and QRS complex in electrocardiogram. K-12 Teachers' Perceptions of the TESA Program and its Impact on Teacher-Student Relationships

Administrators in a school district in north central Kansas implemented the Teacher Expectation and Student Achievement (TESA) professional development program (PD) to address ineffective instructional practices of K-12 teachers. TESA PD was designed to build and promote teacher-student interactions, enhance students' academic performance, teach students self-discipline, and improve the class environment so that students can work and study in diverse settings. The purpose of this qualitative case study was to explore the classroom experiences and perceptions of 10 teachers who integrated teaching interactions from the TESA program into their daily lessons. Brophy and Good's expectation theory holds that teacher interactions with students are impacted by exchanges between teacher and student and served as the conceptual framework. Qualitative data were gleaned from in-depth interviews, observations, and questionnaires and were analyzed using open coding and category construction for patterns, relationships, and themes. Findings indicated that TESA PD assisted these 10 teachers in how to build relationships with their students; how relationship building impacted teacher-student relationships; and how teacher expectations of students, regardless of students' achievement level and diverse backgrounds, impacted student academic performance. To improve relationship building of teachers and students, it is recommended that the TESA PD program be ongoing. Implementing the TESA interactions may contribute to positive social change by allowing students to connect to and communicate with the teacher; accept direction and praise from the teacher;, and trust the teacher, which, ultimately may lead to higher levels of academic success.

Produção de embriões humanos através da injeção intracitoplasmática de espermatozóides obtidos do ejaculado, epidídimo ou testículo / Production of human embryos with intracytoplasmic injection of sperm obtained from ejaculate, epididymis and testis

This retrospective study compared rates of fertilization, cleavage and quality of human embryos obtained by intracytoplasmic sperm injection (ICSI), with sperm from ejaculate, epididymis and testis. A total of 398 cycles of patients with some kind of male infertility were included and 3991 oocytes were inseminated. The spermatozoa from epididymis and testis were surgically obtained. Fertilization and cleavage rates with sperm from ejaculate (75% and 73%, respectively) were higher (P<0,0001) than those with sperm from the testis (59,7% e 56,8%, respectively) and from epididymis (61,9% e 59,3%). The production of viable embryos (grade I and II) was higher (P=0,0135) when the oocytes where inseminated with ejaculate sperm (89,4%) in relation to sperm from epididymis (82,9%) and testis (85,7%).The percentages of pregnancy were not different among groups (37,8% EJAC group, 27,6% EPID and 36,8% TEST group). In conclusion, sperm from ejaculate have a higher production of viable embryos. However, when sperm from the testis and epididymis are used for ICSI, the production of viable embryos is also considerable and can be seen as success in the assisted reproduction techniques. / Este estudo retrospectivo avaliou as taxas de fertilização, clivagem e qualidade de embriões humanos obtidos através da injeção intracitoplasmática (ICSI) de espermatozóides provenientes do ejaculado, epidídimo ou testículo. Foram incluídos 398 ciclos de pacientes que apresentaram algum tipo de infertilidade masculina, sendo inseminados 3991 oócitos. Os espermatozóides do epidídimo e do testículo foram recuperados cirurgicamente. As taxas de fertilização e de clivagem com espermatozóides do ejaculado (74,5% e 73%, respectivamente) foram maiores (P<0,0001) do que as do testículo (59,7% e 56,8%, respectivamente) e do epidídimo (61,9% e 59,3%). A produção de embriões viáveis (grau I e II) foi maior (P = 0,0135) quando os oócitos foram inseminados com espermatozóides do ejaculado (89,4%), em relação ao epidídimo (82,9%) e testículo (85,7%). As taxas de gestação não foram diferentes entre os grupos (37,8% no grupo EJAC, 27,6% EPID e 36,8% no grupo TEST). Em conclusão, espermatozóides do ejaculado produzem um maior número de embriões viáveis, com maiores taxas de fertilização e de clivagem. Entretanto, quando utilizados espermatozóides do epidídimo e do testículo na ICSI, a produção de embriões viáveis é considerável, obtendo êxito nas técnicas de reprodução assistida.
