O TNF atua diretamente sobre a pineal inibindo a via biossintética da melatonina. De forma semelhante o LPS, potente ativador da resposta imune inata, tem efeito inibitório sobre a produção de melatonina e seu precursor. A supressão da síntese noturna de melatonina é admitida de forma a permitir a montagem da resposta inflamatória, tanto durante o dia como à noite. Dados do grupo tem demonstrado que o NFKB é um fator constitutivamente expresso na glândula pineal e que o ritmo diário de translocação pode ser determinante para o início da síntese de melatonina. A inibição de sua ativação potencia a síntese de melatonina enquanto que fatores que classicamente promovem a sua ativação inibem esta produção. Nesta dissertação, verificamos as moléculas envolvidas no reconhecimento e na de sinalização desta citocina na pineal. Demonstramos que a pineal está apta a responder ao TNF, células gliais da pineal e as células secretoras, os pinealócitos, expressam constitutivamente o receptor TNF-R1. O TNF promove a translocação nuclear dos dímeros p50/p50 e p50/RelA do NFKB em glândulas pineais em cultura. Confirmamos a ativação desta via em pinealócitos pela análise da proteína inibitória IKB. Vimos que a ativação desse fator é essencial para expressão da iNOS e a produção de NO induzida por TNF em pinealócitos isolados. Além disso, mostramos que o TNF promove a redução da expressão do receptor TNF-R1 na membrana de pinealócitos. Em resumo, mostramos neste trabalho que a pineal está instrumentalizada a responder ao TNF, e que os pinealócitos são alvos diretos para o seu reconhecimento. Confirmamos a relevância do fator NFKB na resposta da pineal em situações de injúria, ampliando o conceito de que a pineal está apta a detectar moléculas do processo inflamatório. / TNF acts directly on the pineal gland by inhibiting the biosynthesis of melatonin. Similarly, LPS, a potent activator of the innate immune response has an inhibitory effect on the production of melatonin and its precursor. The suppression of nocturnal melatonin synthesis is admitted to allow the full mounting of the inflammatory response both during the day and night. Data from our laboratory have shown that the NFKB is constitutively expressed in the pineal gland and that the daily rate of translocation can be determinant for the onset of melatonin synthesis. Inhibition of their activation potentiates melatonin synthesis whereas factors that promote its activation classically inhibit this production. In this dissertation we find the molecules involved in recognition and signaling of this cytokine in the pineal. We demonstrated that the pineal is responsive to TNF, the pineal glial cells and secretory cells, the pinealocytes, constitutively express the TNF-R1. TNF promotes nuclear translocation of p50/RelA and p50/p50 NFKB dimers in the pineal glands in culture. We confirmed the activation of this pathway in pinealocytes by analyzing the inhibitory protein IKB. We have seen that the activation of this factor is essential for iNOS expression and NO production induced by TNF in isolated pinealocytes. Furthermore we show that TNF promotes the reduction of the expression of TNF-R1 receptor on the membrane of pinealocytes. In summary, we show here that the pineal is instrumented to respond to TNF and that the pinealocytes are direct targets for its recognition. We confirm the relevance of NFKB in the pineal response in situations of injury extending the concept that the pineal is able to detect molecules in the inflammatory process and respond accordingly.

Entwicklung und Charakterisierung von TNFR2-spezifischen Agonisten / Development and characterisation of TNFR2-specific agonists

Liganden und Rezeptoren des Körpers spielen eine multifaktorielle Rolle in der Regulierung zellulärer Prozesse des Körpers. Der Tumornekrosefaktor (TNF), ein proinflammatorisches Zytokin, bindet natürlicherweise an zwei Rezeptoren, den TNF-Rezeptor 1 (TNFR1) oder den TNFR-Rezeptor 2 (TNFR2) und kann durch Aktivierung vielfältiger Signalwege unterschiedliche Zelleffekte im Körper auslösen. Während TNF in membrangebundener Form vorkommend TNFR1 sowie TNFR2 optimal stimulieren kann, ist lösliches TNF in der Lage zwar an beide Rezeptoren zu binden, natürlicherweise jedoch nur den TNFR1 zu stimulieren. Da eine unkontrollierte Bindung bzw. Aktivierung von beiden Rezeptoren schwere unerwünschte Nebenwirkungen wie Inflammationen haben kann, wurden zur konkreten Aktivierung der einzelnen Rezeptoren TNFR1 und TNFR2 spezifische TNF-Mutanten, wie TNF80 zur Bindung an TNFR2 und TNF60 zur Bindung an TNFR1 konstruiert. Durch die TNF-Mutante TNF80 gelingt es die TNFR2 Wirkungskette zu aktivieren, während die TNFR1-Stimulation verhindert wird. Die Aktivierung des TNFR2-Rezeptors hat eine Stimulierung von regulatorischen T-Zellen (Tregs) zur Folge. Im Rahmen dieser Dissertation wurden einerseits die TNF-TNC-Formen weiterentwickelt, indem die konstante Domäne der schweren Antikörperkette des humanen IgG1 (Fc) hinzukloniert wurde. Hier wurde primär der Effekt der Oligomerisierung mit der aktivierenden Wirkung auf TNFR2 erforscht. Weiterhin wird jedoch durch die Bindungsspezifität des Fc-Fusionsproteins von TNF80 an Tregs eine antitumorale Wirkung ausgelöst, indem durch das ausgelöste ADCC die Tregs zerstört werden. Andererseits wurden Kombinationskonstrukte von TNF80 und IL2 kloniert um die Bindungsspezifität des Fusionsproteins auf TNFR2, ebenso wie den IL2-Rezeptor welcher auf regulatorischen T-Zellen hoch exprimiert wird, herzustellen. Die spezifische Stimulation von Tregs würde der Therapie von Autoimmunerkrankungen dienen. In der Abteilung für Molekulare Innere Medizin in Würzburg wurde eine kovalent verknüpfte, nonamere Form von TNF, nämlich eine single-chain-TNF-TNC-Form hergestellt, sodass auch die Aktivierung von TNFR2 durch lösliches TNF möglich ist, was zur klinischen Anwendung (durch Injektionen) notwendig ist. Nach Klonierung und Produktion der Konstrukte in HEK293-Zellen erfolgte deren Aufreinigung und Quantifizierung. Letztendlich wurde mittels Bindungsstudien die Funktionalität der aufgereinigten Fusionsproteine überprüft. Zukünftige Studien müssen nun aufklären, ob die IL8-Produktion durch TNF80(h)-Flag-IL2(h) bzw. TNF80(mu)-Flag-IL2(mu) stimuliert wird, nachdem der IL2-Teil der Konstrukte den IL2-Rezeptor gebunden hat. / The tumornekrosisfactor (TNF) is a cytocine which has a multifunctional role in the immune system. TNF can bind two receptors to activate different signaling pathways: TNFR1 (TNF receptor type 1) or TNFR2 (TNF receptor type 2). TNF occurs as a membrane-integrated form or as a soluble homotrimeric cytokine. TNFR1 can be activated by the membrane-bound and the soluble trimeric form, whereas TNFR2 can be acivated by the membran-bound form of TNF. For specific regulation and a better control of the TNF-binding TNF-mutants were developed. While TNF60 specifically binds TNFR1, TNF80 specifically binds TNFR2 only. Binding TNF80 to TNFR2 results in an expansion of regulatory T cells. Within the frame of this dissertation a TNFR2-specific agonist with the functional domain IL2 was developed and characterised, on the one hand, to achieve an expansion of regulatory T-cells. The specific stimulation of regulatory T-cells could be used as therapeutic medium for autoimmune disease. On the other hand, fusion proteins with TNF80 and the antibody fragment of IgG-Fc were created to achieve a depletion of regulatory T cells by provoking an antibody dependent cellular cytotoxicity (ADCC). This process could be used in therapy against cancer.

Tumor necrosis factor (TNF) is a pleiotropic cytokine that has several functions in the defense mechanisms of the host, among which promote inflammation and, consequently, inhibit the growth of microorganisms. In protozoa, TNF can affect the growth of these through their lectin domains that are spatially distinct from TNF receptors. To our knowledge, a direct TNF effect on yeasts, which lack TNF receptors, has never been reported. Thus, the present study aims to demonstrate that TNF affects the in vitro growth of Candida albicans through a mechanism independent of the TNF receptor. For this, four strains biofilm producerCandida albicans(two strong producer and two weak producer) were cultured in the presence or absence of TNF solutions. Susceptibility to TNF was evaluated using the broth microdilution method. Biofilms were grown on 96-well microtitre plates and evaluated using XTT (2,3-Bis-(2-Methoxy-4-Nitro-5-Sulfophenyl)- 2H-Tetrazolium-5-Carboxanilide) assay, optical, confocal and scanning and transmission electron microscopy.Biochemical biofilm characteristics such as protein quantification and hydrophobicity index (HI) were also evaluated.Thus,it was observed thatTNF did not alter Candida albicans planktonic growth. However, TNF dose-dependently inhibited growing but not established biofilm development, virtually halting yeast filamentation. Preincubation with the TNF antibody adalimumab but not with the immunoglobulin G coupled TNF soluble receptor etanercept blocked TNF activity. Preincubation with the carbohydrate N,N’-diacetylchitobiose, a major component of the yeast cell wall, that binds to TNF lectin domain, abrogated TNF effect against Candida albicans growing biofilm. In conclusion, this is the first demonstration of a receptor independent TNF and no mammalian cells activity, on other words, direct effect against C. albicans biofilm growth. Blockade of TNF effect by N,N’-diacetylchitobiose suggests that it is mediated via the lectin domain of the cytokine. / A Candida albicans é um fungo polimórfico que exprime como principal fator de virulência a formação de biofilmes, especialmente, em implantes médicos na superfícies de pacientes imunossuprimidos, causando infecções potencialmente fatais. O fator de necrose tumoral (TNF) é uma citocina pleitrópicaque denota diversas funções nos mecanismos de defesa do hospedeiro, dentre os quais promover a inflamação e, consequentemente, inibir o crescimento de microrganismos. Em protozoários, o TNF pode afetar o crescimento destes por meio dos seus domínios semelhante à lectina, espacialmente distintos dos receptores de TNF. No conhecimento que se tem, não há relatos de um efeito TNF direto em leveduras, que não possuem receptores de TNF. Assim, este trabalho objetivou demonstrar que o TNF afeta o crescimento in vitro de Candida albicanspor meio de um mecanismo independente do receptor de TNF. Para tal, quatro cepas de Candida albicans produtoras de biofilme (duas fortes e duas fracas formadoras) foram cultivadas na presença ou ausência de soluções de TNF. A susceptibilidade ao TNF foi avaliada utilizando o método de microdiluição em caldo. Os biofilmes foram cultivados em placas de 96 poços e avaliados empregando ensaio de redução de sal de XTT (2,3-Bis- (2-Metoxi-4-Nitro-5-Sulfofenil) -2H-Tetrazolium-5-Carboxanilida), microscopia óptica de campo claro, microscopia confocal e microscopia eletrônica de varredura e de transmissão. Também foram avaliada características bioquímicas do biofilme, como quantificação de proteína e o índice de hidrofobicidade (IH). Assim, foi observado que o TNF não alterou o crescimento planctônico de Candida albicans. O TNF inibiu, contudo, de maneira dose-dependente, a formação de biofilme,por intermédio da modulação da transição de hifa-filamento. A pré-incubação com o anticorpo contra TNF, adalimumab, mas não com o receptor solúvel de TNF acoplado a imunoglobulina G, etanercept,causou um bloqueioda atividade do TNF. A pré-incubação com o carboidrato N, N'-Diacetilquitobiose, um dos principais componentes da parede celular da Candida albicans, que se liga ao domínio semelhante à lectina do TNF, bloqueou o efeito do TNF contra o crescimento de biofilme de Candida albicans.Em conclusão, esse é o primeiro trabalho que demosntra um efeito direto do TNFcontra a formação do biofilme de Candida albicans, ou seja, sem a participação de seus receptores clássicos ou células mamíferas. O bloqueio do efeito do TNF pela N, N'-diacetilquitobiose sugere que este efeito é mediado por meio de um domínio lectina da citocina.

Polimorfismo da região do fator de necrose tumoral (TNF) na síndrome da lipodistrofia associada à terapia anti-retroviral em portadores do HIV-1 / Polymorfism of the Tumoral Necrosis Factor (TNF) in antiretroviralassociated lipodystrophy syndrome in HIV-1-infected patients

Apesar de causar um enorme impacto na história da evolução e prognóstico a partir da infecção pelo HIV, a terapia anti-retroviral altamente potente prolongada apresenta vários efeitos colaterais. Dentre esses, a síndrome da lipodistrofia (SL) caracterizada por alterações metabólicas e morfológicas. Embora tenha sido descrito que a adesão à terapia esteja associada, sua patogenia ainda permanece desconhecida. Um enfoque têm sido dado aos mediadores pró-inflamatórios, como o Fator de Necrose Tumoral (TNF), sugerindo que o aumento nos níveis dessa citocina esteja associado com o desenvolvimento da SL. Como os sítios polimórficos têm sido associados com a magnitude da produção de citocinas, no presente estudo avaliamos a freqüência de alguns sítios polimórficos na região do gene que codifica o TNF em portadores do HIV/aids apresentando ou não a SL. Para avaliar o polimorfismo genético dos microssatélites TNFa-e e da região promotora do TNF (TNF-308 e TNF-238) foram estudados 117 portadores do HIV-1 usando terapia anti-retroviral (67 com SL e 50 sem SL) e 131 controles saudáveis. Os microssatélites e região promotora do TNF foram tipificados usando DNA genômico hibridizado com iniciadores específicos. Os pacientes foram arrolados no Hospital das Clínicas da Universidade de São Paulo (HCFMRP-USP). A analise estatística foi realizada utilizando-se o teste exato de Fisher. Quando consideramos as comparações das freqüências dos alelos dos microssatélites da região do TNF podemos inferir que a presença do alelo TNFa5 pode conferir proteção aos indivíduos portadores do HIV/aids no desenvolvimento da SL. Já as comparações dos alelos da região promotora do TNF nos sugerem que a presença do alelo TNF-308G, assim como seu homozigoto TNF-308GG, podem conferir susceptibilidade para o desenvolvimento da SL. A presença do haplótipo TNFe3 d3 -238G -308A c1 a5 b7 sugere proteção para o desenvolvimento dessa síndrome. Esse é o primeiro estudo associando o polimorfismo dos microssatelites do TNF com a SL e aponta diversas associações entre alelos da região do gene que codifica o TNF com a SL. Embora os mecanismos relacionados com a participação do TNF no desenvolvimento da SL não estejam bem esclarecidos, este estudo sugere que fatores imunogenéticos associados com a magnitude de expressão do TNF e, provavelmente da expressão de outras citocinas pró-inflamatórias, estejam envolvidas no desenvolvimento da SL em portadores do HIV/aids. / Despite causing a significant impact in the history of evolution and prognosis after HIV infection, the highly potent antiretroviral therapy causes various side effects, which include lipodystrophy syndrome (LS), characterized by metabolic and morphologic changes. Although it has been reported that treatment compliance is associated, LS pathogenesis remains unknown. Special attention has been given to proinflammatory mediators, such as the Tumoral Necrosis Factor (TNF), suggesting that the increase in levels of this cytokine is associated with the development of LS. Since polymorphic sites have been associated with the magnitude of cytokine production, in the present study we evaluated the frequency of some polymorphic sites in the gene region that codes TNF in HIV/aids-infected patients presenting LS or not. In order to evaluate the genetic polymorphism of TNFa-e microsatellites and of the TNF promoter region (TNF-308 and TNF-238), 117 HIV-1 infected patients using antiretroviral therapy (67 with LS and 50 without LS) and 131 healthy controls were studied. Microsatellites and the TNF promoter region were typified using hybridized genomic DNA with specific initiators. Patients were selected at the Clinics Hospital of the University of São Paulo (HCFMRP-USP). Statistical analysis was performed using the Fisher\'s exact test. Comparisons of the frequencies of microsatellite alleles of the TNF region suggest that the presence of the TNFa5 allele could provide HIV/aids patients with protection against developing LS. In addition, comparisons of alleles of the TNF promoter region suggest that the presence of TNF-308G allele, as well as of its homozygote TNF-308GG could pose susceptibility to developing LS. The presence of the haplotype, TNFe3 d3 -238G -308A c1 a5 b7, suggests protection against developing that syndrome. The present study is the first to associate TNF microsatellite polymorphism with LS, indicating several associations among alleles of the gene region that codes TNF with LS. Although the mechanisms regarding the participation of TNF in the development of LS are not clear, this study suggests that immunogenetic factors associated with the TNF expression magnitude and probably the expression of other proinflammatory cytokines are involved in the development of LS in HIV/aids infected patients.

Quando consideramos as comparações das freqüências dos alelos dos microssatélites da região do TNF podemos inferir que a presença do alelo TNFa5 pode conferir proteção aos indivíduos portadores do HIV/aids no desenvolvimento da SL. Já as comparações dos alelos da região promotora do TNF nos sugerem que a presença do alelo TNF-308G, assim como seu homozigoto TNF-308GG, podem conferir susceptibilidade para o desenvolvimento da SL. A presença do haplótipo TNFe3 d3 -238G -308A c1 a5 b7 sugere proteção para o desenvolvimento dessa síndrome. Esse é o primeiro estudo associando o polimorfismo dos microssatelites do TNF com a SL e aponta diversas associações entre alelos da região do gene que codifica o TNF com a SL. Embora os mecanismos relacionados com a participação do TNF no desenvolvimento da SL não estejam bem esclarecidos, este estudo sugere que fatores imunogenéticos associados com a magnitude de expressão do TNF e, provavelmente da expressão de outras citocinas pró-inflamatórias, estejam envolvidas no desenvolvimento da SL em portadores do HIV/aids. / Despite causing a significant impact in the history of evolution and prognosis after HIV infection, the highly potent antiretroviral therapy causes various side effects, which include lipodystrophy syndrome (LS), characterized by metabolic and morphologic changes. Although it has been reported that treatment compliance is associated, LS pathogenesis remains unknown. Special attention has been given to proinflammatory mediators, such as the Tumoral Necrosis Factor (TNF), suggesting that the increase in levels of this cytokine is associated with the development of LS. Since polymorphic sites have been associated with the magnitude of cytokine production, in the present study we evaluated the frequency of some polymorphic sites in the gene region that codes TNF in HIV/aids-infected patients presenting LS or not. In order to evaluate the genetic polymorphism of TNFa-e microsatellites and of the TNF promoter region (TNF-308 and TNF-238), 117 HIV-1 infected patients using antiretroviral therapy (67 with LS and 50 without LS) and 131 healthy controls were studied. Microsatellites and the TNF promoter region were typified using hybridized genomic DNA with specific initiators. Patients were selected at the Clinics Hospital of the University of São Paulo (HCFMRP-USP). Statistical analysis was performed using the Fisher\'s exact test. Comparisons of the frequencies of microsatellite alleles of the TNF region suggest that the presence of the TNFa5 allele could provide HIV/aids patients with protection against developing LS. In addition, comparisons of alleles of the TNF promoter region suggest that the presence of TNF-308G allele, as well as of its homozygote TNF-308GG could pose susceptibility to developing LS. The presence of the haplotype, TNFe3 d3 -238G -308A c1 a5 b7, suggests protection against developing that syndrome. The present study is the first to associate TNF microsatellite polymorphism with LS, indicating several associations among alleles of the gene region that codes TNF with LS. Although the mechanisms regarding the participation of TNF in the development of LS are not clear, this study suggests that immunogenetic factors associated with the TNF expression magnitude and probably the expression of other proinflammatory cytokines are involved in the development of LS in HIV/aids infected patients.
