Performance of Hereford and Holstein heifers on kikuyu pasture (Pennisetum clandestinum), using n-alkanes for determination of digestibility and dry matter intake.

Kikuyu pasture (Pennisetum clandestinum) is potentially the most important source of roughage used to feed dairy heifers in summer in KwaZulu-Natal. It is commonly believed that on kikuyu pasture beef breed females grow at a faster rate than those from dairy breeds when no supplementation is given. Little conclusive evidence is, however, available to support this. Explanations as to why such differences may exist are also limited. Eight Hereford and eight Holstein heifers of similar age and maturity stage were used in a trial. The trial was run over a twenty week period. For the first ten weeks all the animals in the trial grazed ad libitum kikuyu pasture with no supplementation except for a mineral lick. Over this (grass only) period the two breed groups formed the two treatments. During the second ten week period of the trial all of the Holsteins and four of the Herefords were fed a restricted but equivalent amount (1 .7 kg) of a maize meal based concentrate. The use of a computerized, mobile feeding system allowed concentrate intake of individual animals to be measured. Animal height, weight and condition score readings were taken weekly over the grass only and the concentrate (final seven weeks) periods of the trial. Herbage intake and digestibility were estimated using n-alkanes as indigestible markers in two experiments conducted during the grass only and concentrate periods. The Herefords had a significantly higher ADG than the Holsteins (0.82 vs. 0.04 kg/day; P < 0.01) over the grass only period. During the concentrate period the rate of mass gain of the Holstein treatment did not differ significantly (P >0.05) from the Hereford treatment receiving concentrate. The Herefords receiving concentrate were also not significantly different (P > 0.05) in rate of mass gain from the Herefords not receiving concentrate. Rate of height gain was not significantly different (P> 0.05) between treatments over either the concentrate or the grass only periods. During the grass only period the Holsteins lost condition (0.07 condition score units per week) whilst the Herefords gained condition at an equivalent rate. The voluntary intake of concentrates was not significantly different (P > 0.05) between the Herefords and Holsteins (19.19 vs. 16.40 g/kg/L.W(liveweight) (0.75)). Regression coefficients relating level of concentrate intake to rate of mass gain were also not significant (P > 0.05) for either of the treatments receiving concentrate. The use of n-alkanes as indigestible markers showed the intake of the Holstein treatment to have an intake 55% (P < 0.0 1) higher than the Herefords (185.4 vs. 120.5 g/kg L.W(0.75)) over the first experiment where both treatments were grazing ad lib. kikuyu alone (grass only period). During the concentrate period intake of the Herefords receiving concentrate exceeded that of the Holsteins (P < 0.01) by 23% (139.1 vs. 113.1 g/kg L.W(0.75)). Review of the literature, suggests that the double alkanes technique greatly over-estimated intake. Errors in herbage sampling (accentuated by pasture rotation in the first experiment), a low daily dose of the synthetic alkane (C(32)) and incorrect estimation of the C(32) content in the daily doses are identified as possible causes of the over-estimation of intake. Faecal recoveries of the herbage n-alkanes were demonstrated to increase with increasing chain length and hence C(35) was proposed as the most reliable herbage alkane for dry matter digestibility determination. Digestibility differences between treatments estimated using the C(35) alkane were not significantly different (P > 0.05) in either the first or second experiments. The mean digestibility estimates (using the C(35) alkane) for the first and second experiments were 64.9 and 56.61 %, respectively. In conclusion, higher growth rates of Herefords on kikuyu pasture would seem to be primarily due to differences in the dry matter intake of the grazed herbage. Further work using other breeds of dairy and beef animals is required. Biological control of the common house fly Musca domestica L. in horse stables, using Bacillus thuringiensis serovar israelensis and Beauveria bassiana.

House flies (Musca domestica L.) are common pests affecting horses and their owners. Control of house flies in stable yards is currently based on the use of pesticides. However, the development of resistance by these flies to most pesticide groups has motivated horse owners to seek alternative methods of fly control. An entomopathogenic fungus, Beauveria bassiana (Bb) and an entomopathogenic bacterium, Bacillus thuringiensis var. israelensis (Bti) are two biological agents known to have activity against house flies. The broad objective of this study was to evaluate the effect of these two biological control agents on house flies in an equine environment. Using a structured questionnaire, presented in Chapter 2, thirty horse owners in KwaZulu-Natal were asked about the nuisance value of house flies, their current control measures, the potential market for biocontrol agents against house flies, and each owner’s perception of biocontrol methods. The horse owners were using three methods of house fly control namely, physical, chemical and biological. Most horse owners (97%) wanted access to effective biocontrol agents for control of house flies. Most horse owners (80%) stabled their horses at night, some or all of the time. The resultant manure piles in the stable yard were considered to be the primary cause of house fly problems. About 64% of the horse owners were dissatisfied with the currently available methods of controlling house flies in this situation. Chapter 3 covers two observational trials in which varying doses of Bacillus thuringiensis var. israelensis (Bti) were fed to horses, in order to identify a baseline dosage to give to horses in order to adequately control house fly populations growing in horse manure. The bacterium Bti, grown on wheat bran, was fed to six miniature horses at doses of 0, 0.125, 0.25, 0.5, 0.75, and 1.0 g per meal in Trial 3a, and at 0, 0.5, 1, 2, 4 and 8 g per meal in Trial 3b. Faeces were collected three times a week for 11 weeks and placed in incubation trays to allow the number of emerging adult house flies and closed pupae to be counted. In Trial 3a, there was a significant reduction in the number of closed pupae with an increase in Bti in the feed. The regression equation suggests that there will be 3.1 times as many closed pupae in the faeces when horses are fed 1 g of Bti in their feed, than when horses are fed no Bti. This dosage is the minimum baseline dosage for future trials. v Using manure from horses dosed in Trials 3a and 3b, the survival of the bacterium through the gut of horses was evaluated using a standard isolation technique. The growth of Bt colonies on the manure after the Bt isolation technique showed that some of the bacterial cells survived transition through the digestive tract of the horse. This study was qualitative in nature and did not attempt to quantify the level of Bti spore survival. These two observations suggest that Bacillus thuringiensis var. israelensis has the potential to be used as a biocontrol agent, applied via horse feed, for the control of house flies in stable yards. Future clinical trials, with appropriate replication, should be conducted using 1 g Bti/meal as the lowest test dosage. The objective of Chapter 4 was to determine whether spraying Bti or Bb on to horse manure is effective in the control of house flies. Over a six week period, two spraying trials were conducted in which increasing doses of Bb and Bti were sprayed on to 500 g samples of horse manure. Counts of house fly pupae and adults were taken. The doses of Bb and Bti tested were 0, 1, 2, 4 g in Trial 4a, and 0, 4, 8 and 12 g in Trial 4b. The research reported in Chapter 4 was characterized by the unexpectedly high levels of biological variation in egg, larvae and pupae numbers that were found in samples of horse manure, taken from the same skip two days apart. The statistical design of the two trials conducted was inadequate to cope with the high level of variation about treatment means for fly and larval counts. However, despite the lack of significant differences between treatment means, there is observational evidence that suggests that both Bb and Bti do have an effect on house fly survival. A simplified statistical model, which compared the number of hatched house flies on untreated manure, with the number on manure treated with any level of Bb (1 to 4 g /250 ml water), found a significant reduction in the number of hatched flies on treated manure. There was no significant corresponding reduction in the number of closed pupae, which suggests that Bb acts primarily before the larva pupates. The optimal dose of Bb and Bti to be sprayed on to manure could not be determined because of the high variation about treatment means. It is suggested that, in future trials similar dosages for Bb could be tested, but that higher dosages of Bti (starting at 2 g/250 ml water) should be used. Trial periods should be extended and replication increased dramatically to reduce variation about treatment means. The effects of dietary lysine, crude protein, energy and feed allocation on broiler breeder hen performance.

This study was conducted to investigate the effects of varying levels of dietary lysine and feed allocation, on the performance of female broiler breeders. In trial 1, 900 Cobb breeders from 26 to 45 weeks of age were used. The first four treatments had a fixed level of lysine throughout the experimental period, with 1200, 1070, 930 and 800 mg Lysine/bird/day and the last two treatments had their levels of lysine changed every two weeks, with treatment five started off with 1200 mg lysine/bird/day followed by a gradual decrease up to 975 mg lysine/bird/day at 45 week old, while treatment six started with the lowest level of lysine, 800 mg/bird/day and at 45 week old the intake was 1025 mg/bird/day. The rations provided 1900KJ ME/ day, the birds received 160 g of feed/day. Dietary lysine did not affect body weight, egg production, or egg composition. There were no significant differences in age at 50%, or peak production. Birds receiving 1070 and 1025 mg lysine/bird.day had a slightly, but not significantly, higher production in the current study. Birds receiving 1200 mg lysine/bird.day had the highest body weight and the lowest egg production. In trial 2, 900 broiler breeder hens were used. Protein intake and feed allocation were changed for each treatment at 26, 38 and 50 weeks and ending at 60 weeks. Birds on T1 were fed a constant CP content (145 g protein/kg) but a decreasing feed allocation (175 to 145 g/d). Those on T2 had a constant feed allocation (160 g/d) and a decreasing CP (166 to 124 g/kg) while those on T3 had varying levels of protein (166, 124 and 166 g/kg) and feed allocation (160, 160, 145 g/d). Mean cumulative protein intakes were 5.2, 5.3 and 4.8 kg/bird and mean energy intakes were 417, 412 and 402 MJ/bird for T1, T2 and T3 respectively. Body weights differed significantly at the end of the trial (P<0.05) with T2 showing the highest weight gain. However, egg production, egg weight and egg output were not affected by treatment. These breeders did not benefit from the additional protein and energy provided in the first and second periods by T1 and T2 (vs. T3), nor did they benefit from the additional energy provided in the final ten weeks of production by T2. / M.Sc.Agric. Morphometric characteristics and consumer acceptability of meat from Potchefstroom Koekoek, Black Australorp, Venda and Ovambo chickens.

Indigenous chicken production receives little institutional support and resources because of lack of information on the socioeconomic importance, morphometric characteristic and meat quality of indigenous chickens. A cross sectional survey was conducted to highlight the major constraints to production. A structured questionnaire was administered to 126 households selected from communities of Mnambiti-Ladysmith and Impendle local municipalities of KwaZulu-Natal Province of South Africa. The mean flock size per household was 20 and 17 for Mnambiti-Ladysmith and Impendle, respectively. The cock to hen ratio was 1:2:5. The chickens were mostly kept for meat and income ranked 1 and 2 respectively. Generally, adult females made the majority of decisions on chicken management and marketing (61%), with youths playing a minor role. Scavenging was the major feeding system, seasonally supplemented with cereal grain. The majority of the farmers (87%) provided birds with drinking water. Mortality of chickens was prevalent (46%) in both seasons. In experiment 2, the objective of the study was to compare morphometric characteristics of Black Australorp, Potchefstroom Koekoek, Venda and Ovambo chickens. A flock of 200 indigenous chickens, 50 each of Black Australorp, Potchefstroom Koekoek, Venda and Ovambo breeds were reared under semi-intensive system for 22 weeks. The chickens were slaughtered at 22 weeks of age by manual neck cut, bled for 2 minutes and de-feathered. Body weights, organ weights and linear body measure were estimated using flexible tape prior to slaughter. The body weight for the Black Australorp were higher (P<0.05) than the other breeds. There was no significant difference between the lung, heart kidney and spleen weights among breeds. Linear regression revealed that measurements of linear body parts can be used to predict weight of the birds. The objective of Experiment 3 was to compare consumer acceptability of meat from chickens that are indigenous to South Africa compared to Black Australorp and the broiler. A flock of 200 unsexed freely ranging indigenous chickens of Potchefstroom Koekoek, Venda, Black Australorp and Ovambo breeds were reared under an improved semi-intensive system for 22 weeks. The acceptability of cooked meat samples from each breed was rated on a 9 point Hedonic scale by 69 consumer pannellists drawn from the University of KwaZulu-Natal, Cedara College of Agriculture and the Depart of Agriculture. Age of consumer had no effect on all the sensory attributes of the meat evaluated (P> 0.05). Chicken breed had a significant effect on taste and overall acceptability (P< 0.05) with the Venda, Broiler and Black Australorp, but it had no significant difference on colour, texture and aroma acceptability. Gender of the consumer had a significant effect (p<0.01) on taste, colour and texture acceptability of the meat, but no significant effect on aroma and overall acceptability. Female respondents gave lower scores for taste than did the males (P< 0.01). There was no interaction between gender of consumer and chicken breed on meat texture. Prediction of gut capacity of weaner and finishing pigs using physicochemical measurements of bulkiness of fibrous feeds.

Physicochemical properties of different fibrous ingredients were used to determine the influence of feed bulk on voluntary feed intake and gut capacity in weaner and finishing pigs. Physicochemical measures of bulkiness determined on feed ingredients were DM, crude protein (CP), ether extract, ash, water holding capacity (WHC), bulk density, crude fibre (CF), neutral detergent fibre (NDF) and acid detergent fibre (ADF). Among the fibre sources, maize stover had the highest WHC, followed by veld grass, then lucerne hay, maize cob, sunflower husks, groundnut haulms, rice bran and saw dust. The greatest bulk densities (1.4 g DM/ml) were observed in lucerne hay and saw dust, whilst sunflower husk had the lowest (0.7 g DM/ml) (P < 0.001). Rice bran, maize cob and groundnut haulms were the most fermentable fibrous ingredients (P < 0.05). Based on differences in physicochemical properties, six fibres namely lucerne hay, maize cob, maize stover, veld grass, saw dust and sunflower husk were selected and used in formulating fibrous diets fed to growing pigs based on wideness in ranges of their bulk properties. Thirty-one complete diets were formulated by dilution of a conventional weaner feed with increment levels of each fibre source at 0, 80, 160, 240, 320 and 400 g/kg. Each of the diets was offered ad libitum to four of 124 pigs weighing 18.1 (s.d. 1.37) kg body weight, in individual pens, for four weeks. There was a linear decrease in scaled feed intake (SFI) (P < 0.001) as bulk density of the feeds increased. There was a quadratic relationship between SFI and WHC, NDF and ADF, respectively, whereby SFI increased up to a point when it reached its maximum and then started to decrease as bulkiness increased (P < 0.001). By use of the broken stick model, the maximum SFI marking the gut capacity of pigs was attained when WHC = 4.5 ± 1.25 g water/g DM (P< 0.001), NDF = 367 ± 29 g/kg DM (P < 0.001) and ADF = 138 ± 77 g/kg DM (P < 0.01), respectively. The SFI decreased linearly with an increase in bulk density of the feeds (P < 0.001). Four of 84 finishing pigs in individual pens, at 65 (s.d. 1.37) kg body weight were given, ad libitum to each of 21 diets containing graded levels of lucerne, maize cobs, saw dust and sunflower husk. There was a linear decrease in SFI (P < 0.001) as WHC increased. There was a quadratic decrease in SFI as CF (P < 0.001) and NDF (P < 0.01) increased. As CP increased, there was a quadratic increase in SFI (P < 0.01). In weaner pigs, an increase in WHC, NDF, ADF and bulk density constrains feed intake, thereby providing relationships that can be used to predict gut capacity. Influence of feed bulk on physicochemical properties of digesta in pigs.

Physicochemical properties of fibre-based diets were used to determine the influence of feed bulk on physicochemical properties of digesta within each segment of the gastrointestinal tract and digesta in pigs. In the first experiment, three pigs (14 ± 1.2 kg body weight (BW) were allocated to each of six diets containing maize cob levels at 0, 80, 160, 240, 320 and 400 g/kg DM inclusion levels for four weeks. All pigs were fed ad libitum. They were slaughtered, eviscerated and weights of the gut compartments were recorded, then contents of digesta from each segment were sampled for the determination of water concentration, water holding capacity (WHC) and swelling capacity (SWC). The WHC of digesta in the stomach, ileum and caecum decreased (P < 0.05) with maize cob inclusion level. The SWC in the stomach decreased with the inclusion level of maize cob meal. The SWC of caecal digesta increased with maize cob inclusion (P < 0.05). Physicochemical properties of digesta increased (P < 0.05) from the stomach to ileum then decreased as the digesta moved through the hindgut. In the second experiment, four fibres namely maize cob, lucerne hay, sunflower husk and citrus pulp were used. These fibres were used in formulating diets for finishing pigs. Twenty-one complete diets were formulated by dilution of a conventional feed with increment levels of each fibre source at 0, 80, 160, 240, 320 and 400 g/kg. Each of the diets was offered ad libitum to four of 84 pigs weighing 80.8 ± 8.15 kg body weight, in individual pens, for 30 days. Stomach weights increased linearly with an increase in neutral detergent fibre (NDF) but increased with quadratic functions with an increase in SWC of the diet (P <0.05). An increase in WHC of the fibrous diets increased linearly the WHC of the proximal colon (P <0.01) at a faster rate compared to the WHC of the distal colon (P <0.001). As the SWC of the diets increased, linear increases in SWC of the digesta in the stomach (P <0.01) and caecum (P <0.001) were observed. The WHC of the digesta was negatively correlated to SWC (P < 0.001) in the stomach. Scaled feed intake (SFI) decreased linearly with an increase in SWC of the diet (P < 0.001). There was no relationship between WHC of the diet and SFI (P > 0.05). There was a linear decrease (P < 0.01) in SFI of finishing pigs as the SWC of the digesta. It can be concluded that the swelling capacity of the diets and stomach digesta in stomach are accurate predictors of scaled feed intake. Swelling capacity had great influence in the stomach weights whilst other bulking properties, such as WHC and neutral detergent fibre, affected the weight and digesta properties in the caecum, proximal and distal colon. / M.Sc.Agric. Modelling broiler populations for purposes of optimisation.

With the narrow margin of profit in the broiler enterprise, how can producers increase profit potential? It is not an easy task to answer this question since the net financial return depends on many factors; some are related to the animal, some to the feed, some to the environment and others are outside the production system, like availability and cost of labour and capital. Many researchers have attempted to improve the efficiency of the system using alternative management strategies and to develop a unified theory that could simultaneously evaluate all the relevant factors and the interactions between them. Simulation models are seen as the most promising means of moving this subject forward. Geneticists are continually improving the potential growth rate of broilers, yet there has been little change in feed specifications for these birds over the past few decades. Only recently has it been possible to make use of simulation models to optimise the feeds and feeding programs of modern broiler strains at a commercial level, but little testing of these programs has been carried out. What is needed is a thorough investigation of these models, which at present are based on an individual, as opposed to a population response. Modelling plays an increasingly important part in animal science and research as a way of organizing and evaluating the large body of existing knowledge. With the use of an accurate description of the potential growth rates of broiler genotypes, it is possible to make more efficient use of growth models which are becoming more abundant in the industry and which, in turn, enable the nutritionist or producer to predict the performance of animals when subjected to a given feed or feeding programme. The predictions made by most of the growth models now available are based on individual animals, and the results obtained may be inadequate in optimising the nutrient requirements of a broiler population because of the variation that exists in these populations. Variation in performance traits in broilers may be the result of variation in the genotype, in the environmental conditions within the house, and in the composition of the feed offered to the birds, and these sources of variation cannot all be accommodated in a model that simulates the food intake and growth of just one bird. But if variation is to be incorporated into growth models, it is necessary to ascertain the effects of variation in the various genetic parameters on the mean response of the population. A sensitivity analysis is useful in accomplishing this objective. Similarly, it is important to know what the optimum size of a simulated population should be, that takes account both of the accuracy of the simulation and the time taken to complete the exercise. This is especially important when optimisation routines are followed, as such calculations are time consuming. As a means of addressing these issues, simulation exercises were conducted using EFG Broiler Growth Model version 6 and EFG Broiler Optimiser Model version 1 (EFG Software, 2006) to determine: (a) whether it is worth generating a population when optimising feeds and feeding programs for broilers, rather than using the average individual, (b) the size of the population required to obtain an accurate estimate of the population response when optimising the feeding program for different objective functions, (c) the effect of changing the value of genetic parameters such as mature protein weight, rate of maturing, feathering rate and the maximum lipid:protein ratio in the gain on the optimum amino acid contents and nutrient densities of broiler feeds, and (d) the effect of variation in nutrient composition of different batches of feed, which have the same nutrient profile but different qualities of the main protein source, on broiler performance. A review of sources of variation in the nutrient content of poultry feed was conducted, and simulation exercises were carried out to determine to what extent broiler performance is affected by the segregation or breakage of pellets into small pieces at the time of delivery and along the feed conveyor within the broiler house, by the change in nutrient quality that might occur along the conveyor, and by the microclimates that develop in a longitudinally ventilated broiler house. The tendency in broiler marketing in most parts of the world is to sell broilers cut up, as portions or deboned after evisceration, rather than selling whole birds. Estimation of the growth rates of carcass parts is therefore of considerable importance if simulation models are to be useful in optimising the feeds and feeding programmes of broilers under different conditions. Allometric equations are used in the EFG broiler growth model to predict the weights of these carcass parts from the weight of body protein at the time. These equations are based on data collected many years ago, and it would be useful to determine whether they are still relevant in the face of announcements by the major broiler breeding companies that tremendous strides have been made in improving breast meat yield, for example, by judicious selection. For the purpose of this investigation it was important to determine to what extent the weights of the physical parts varied at the same body protein weight, thereby enabling a more accurate estimation of the variation that could be expected in these weights when developing a population response model. Towards this end, experiments were conducted to determine the effect of dietary protein content on the performance of Cobb and Ross broilers, including mortality and uniformity, and on the allometric relationships between the physical and chemical components of the body and body protein. The photoperiodic response of male broiler breeders.

The expression of photorefractoriness, a feature of seasonal breeding in birds, is important in production species, where egg production and fertility are affected by the photoperiod. Years of selection in meat-type birds have reduced the potential reproductive function, and the environmental manipulation of such genotypes is essential to maximise productivity. While it is known that egg-type pullets no longer exhibit photorefractoriness, and that female broiler breeders do, there is not much information about the response of male broiler breeders to photoperiod. Such information is important when designing lighting programmes for breeding stock, and this study aimed to gain more insight into the male response to photoperiod. A series of experiments was designed in order to achieve this. In the first the response of male broiler breeders to rearing on constant photoperiods was measured, and in the second the effects of age at photostimulation on age at sexual maturity were assessed. The response to an increase in photoperiod during the production cycle was investigated in the third trial. Evidence of photorefractoriness in males was observed, but this was not the case in all birds, possibly due to high variation in some of the fertility traits measured. It seems possible that males do not respond to photoperiod to the same extent as females. Unexpected observations of a response in some birds to early photostimulation, and the high variation observed prompted another experiment in which the heritability of the response to early photostimulation was measured as a potentially revolutionary method of selection, whilst checking that this response is not negatively linked to broiler growth rates. Left and right testis weight data, collected in the above experiments, were analysed for asymmetry. There was no consistent response in testes asymmetry to photoperiod, or evidence that asymmetry in testis weights is as a result of the imposition of genetic or environmental stress. A dynamic mechanistic anaylsis of the thermal interaction between a broiler chicken and its surrounding environment.

Chickens, being open thermodynamic systems, maintain a constant exchange of energy and matter with their surrounding environment. In order to avoid reaching thermodynamic equilibrium with the environment the bird makes use of homeostatic mechanisms. These ensure the reduction of the entropy of the system to values that guarantee its integrality. The thermoregulatory response is a major component of the homeostatic machinery of living systems. This induces modifications of physiological parameters of the bird, taking the system “bird” to a new steady state. The achievement of this new state is possible only if the thermoregulatory mechanisms of the birds are able to counteract the environmental demand/burden. A successful thermoregulatory response depends not only on the achievement of that steady state, but also on the compatibility of the value of those parameters with life (especially regarding the value achieved by body temperature) as well as on the time of exposure to the environmental perturbation. Based on those premises, this thesis presents a mechanistic analysis of the thermal interaction between a broiler and its surroundings. The first section of the document introduces the reader to the general concepts of thermodynamics of living systems and physics of heat exchange. The second use mechanistic simulation techniques to represent the environment, the thermal and thermoregulatory properties of a broiler chicken and the interaction between bird and environment. Finally, the third section describes a conceptual simulation model able to predict, over a given period of time, the response of a bird to environmental conditions above those associated with least thermoregulatory effort.
