Padronização de ténica para recuperação de ovos de Toxocara canis em solo / Padronization of technique for recovery of Toxocara canis in soil

The tests were done under standardized conditions on 1 g of previously sterilized soil samples contaminated experimentally with 200 eggs of T. canis. The following variables were evaluated in serial steps: sieving; type of wash; time of stirring; resuspention of sediment; solution flotation. Centrifuge-flotation in sodium nitrate (d= 1,20) was adopted as an initial technique, using Tween 80 (0.2%) and decinormal sodium hydroxide as solutions for washing the samples. Ten tests were made to compare the variables, using counting in triplicate. The sieving of the material reduced the recovery of eggs in a significant difference (p<0.001) and the number of eggs recovered were higher when the sediment was resuspended (p<0.05). After standardization, flotation solutions sodium chloride, zinc sulphate, sodium dichromate, magnesium sulphate, and sodium nitrate (d= 1,20) were compared. There was a significant difference between zinc sulphate and sodium chloride (p<0.05). In conclusion, the chance of recovering T. canis eggs from samples using flotation solutions can be increased by using resuspended sediment. On the other hand, the sieving procedure can drastically reduce the number of eggs. / O objetivo do estudo foi padronizar uma técnica para recuperação de ovos de ascarídeos (Toxocara canis) em solo, com a comparação de diferentes etapas. Amostras de solo foram previamente esterilizadas e divididas em alíquotas de um grama, sendo contaminadas com 200 ovos. Após a contaminação, foram comparadas, em etapas seriadas, as variáveis: filtragem, tipo de lavagem e re-suspensão do material. Como ponto de partida para comparação, utilizou-se uma técnica com lavagem de solo em Tween 80 (0,2%) e solução de hidróxido de sódio 0,1N; re-suspensão; e centrífugo-flutuação em solução de nitrato de sódio (d=1.20). Os ovos recuperados foram contados com 10 repetições, com três leituras para cada repetição. A filtragem reduziu significativamente a recuperação de ovos em relação ao material não filtrado (p<0,001), enquanto o número de ovos foi significativamente maior quando utilizada a re-suspensão do material (p<0,05). Após padronização, comparou-se as soluções de cloreto de sódio, dicromato de sódio, nitrato de sódio, sulfato de zinco, sulfato de magnésio, havendo diferença entre sulfato de zinco e cloreto de sódio (p<0,001). Dessa forma, as chances de recuperação de ovos de T. canis podem ser ampliadas pelo re-suspensão do sedimento, enquanto que a filtragem reduz consideravelmente o número de ovos.

A infecção em seres humanos por larvas de nematódeos do gênero Toxocara é uma antropozoonose endêmica em diversas localidades com prevalências superiores a 50% em diferentes grupos populacionais, tanto em países em desenvolvimento como nos desenvolvidos. O presente estudo avaliou a prevalência, fatores associados e distribuição espacial de infecção por Toxocara spp. em crianças residentes na área urbana de Acrelândia, Acre, Amazônia Ocidental Brasileira. Conduziu-se um estudo transversal de base populacional com 1112 crianças <11 anos de idade. Informações socioeconômicas e demográficas foram obtidas por meio de questionário estruturado. Medidas antropométricas e coleta de amostras biológicas (sangue e fezes) foram realizadas pela equipe de pesquisa. A avaliação de soroprevalência da infecção por Toxocara spp. utilizou o método imunoensaio enzimático (ELISA). Registraram-se as coordenadas geográficas pontuais de todos os domicílios participantes do inquérito para análise espacial de varredura. Razões de prevalência (RP; intervalo com 95% de confiança, IC95%) para fatores associados à antropozoonose foram estimadas por regressão de Poisson com seleção hierárquica das variáveis independentes. Verificou-se soroprevalência geral de 38% para anticorpos IgG anti-Toxocara spp.; crianças 5 anos de idade apresentaram maior prevalência para infecção por Toxocara spp., estatisticamente significante, em relação aos de menor faixa etária (RP, 1,70; IC95%, 1,442,00). Ausência de tratamento para água de beber (RP, 1,38; IC95%,1,111,71), deficiência de vitamina A (RP, 1,47; IC95%, 1,221,78) e presença de geo-helmintos (RP, 1,57; IC95%, 1,251,98) foram positivamente associados à infecção pelo nematódeo. Em contrapartida, maior quarto do índice de riqueza domiciliar (RP, 0,65; IC95%, 0,510,83) e maior escolaridade materna (RP, 0,71; IC95%, 0,580,88) foram inversamente associados à infecção por Toxocara spp. Presença de cão e/ou gato nas residências não foi associada à infecção pelo nematódeo. Análise da distribuição espacial detectou um aglomerado de baixa prevalência para infecção por Toxocara spp. na área investigada, compreendendo 36,0% das crianças com sorologia negativa para o nematódeo. Os resultados indicam que a prevalência da infecção por Toxocara spp. observada no presente estudo foi semelhante ao descrito em estudos anteriores na região da Amazônia Brasileira, mas superiores às observadas nas regiões sul e sudeste do país e em outros países. Prevalence ratios (PR; 95% confidence intervals, 95%CI) of factors associated to this anthropozoonosis were estimated using Poisson regression models, with hierarchical selection of independent variables. An overall prevalence of 38% for IgG anti-Toxocara spp. antibodies was found; children 5 years of age presented statistically significantly higher prevalence of Toxocara spp. infection when compared with younger children (PR, 1.70; 95%CI, 1.442.00). The absence of treatment of drinking water (PR: 1.38, 95%CI, 1.111.71), vitamin A deficiency (PR: 1.47, 95%CI, 1.221.78), and presence of geo-helminths (PR: 1.57, 95%CI, 1.251.98) were positively associated to this anthropozoonosis infection. In contrast, the highest wealth index quartile (PR: 0.65, 95%CI, 0.510.83), and the higher level of maternal education (PR: 0.71, 95%CI, 0.580.88) were inversely associated to Toxocara spp. infection. Presence of a dog or cat in the household was not associated with infection by the nematode. Spatial distribution analysis detected a significant low-prevalence cluster for Toxocara spp. infection in the area under investigation, comprising 36.0% of the seronegative subjects. The results indicate that the prevalence of Toxocara spp. infection observed in this study was similar to that found in the Brazilian Amazon region, but higher than rates in the south and southeast regions of Brazil, as well as those reported in others countries. It is noteworthy that the identification of factors associated with Toxocara spp. infection allowed the targeting of priority groups for appropriate intervention in this population.

Contaminação ambiental por geo-helmintos em praças do município de Botucatu, São Paulo

Orientador: Elizabeth Moreira dos Santos Schmidt / Resumo: Os geo-helmintos são um grupo de parasitas em que parte do ciclo evolutivo ocorre necessariamente no solo, que desempenha o papel de fonte de infecção. O solo pode ser responsável pela transmissão das geo-helmintíases, principalmente quando o local é compartilhado por animais e seres humanos, como em parques e/ou praças públicas. Em relação aos geo-hemintos de potencial zoonótico, destacam-se os gêneros Toxocara e Ancylostoma. Entretanto, outros parasitos intestinais, como Trichuris spp. e Strongyloides spp., que possuem parte de seu ciclo biológico no solo, também desempenham importante papel na contaminação de ambientes públicos. Desta forma o objetivo deste trabalho foi investigar a contaminação do solo por geo-helmintos em praças do município de Botucatu-SP, identificando a prevalência dos principais gêneros e sua ocorrência em praças de diferentes áreas do município. As praças foram catalogadas e distribuídas entre as regiões administrativas (norte, sul, leste, oeste e central). As coletas foram realizadas em dois momentos: momento I (de outubro a dezembro de 2017) e momento II (de fevereiro a março de 2018). No presente trabalho foram coletadas amostras de solo e avaliadas por meio das técnicas de centrífugo-sedimentação, centrífugo-flutuação com sulfato de zinco, e o método de Baermann-Moraes na busca por ovos e larvas de geo-helmintos. A partir das análises das amostras do solo obtidas nas praças estudadas, foram observados ovos do gênero Toxocara e ovos de ancilostom... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Geohelminths are a group of parasites in which part of the life cycle necessarily occurs in the soil, playing a role as a source of infection. Soil may be responsible for the transmission of geohelminthiasis, especially when the site is shared by animals and humans, such as in parks and/or public squares. Regarding geohelminths of zoonotic potential, we highlight the genera Toxocara and Ancylostoma. However, other intestinal parasites, such as Trichuris spp. and Strongyloides spp., which also have part of their biological cycle in the soil, also play an important role in the contamination of public environments. This study aimed to investigate the soil contamination by geohelminths in squares of the municipality of Botucatu-SP, identifying the prevalence of the main genera and their occurrence in squares of different areas of the municipality. The squares were cataloged and distributed among administrative regions (north, south, east, west, and central). The samples were carried out in two time point: I (from October to December 2017) and II (from February to March 2018). In this study soil samples were collected and evaluated by means of centrifugalsedimentation techniques, zinc sulfate centrifugal flotation , and the BaermannMoraes method searching for geohelminths eggs and larvaes.
