Implementation of marker assisted breeding in triticale

Ntladi, Solomon Magwadi 12 1900 (has links)
Thesis (MScAgric)--Stellenbosch University, 2011. / ENGLISH ABSTRACT: Research into markers for the detection of genetic diversity and cultivar identification has become an important component of the genetic improvement of crops. However, the incorporation of marker assisted selection (MAS) as a tool for the identification and characterization of breeding material has not been fully implemented in the breeding of spring triticale at Stellenbosch University’s Plant Breeding laboratory (SU-PBL). The present study served as a case study in order to achieve this. The first part of the study concerned the detection of genetic diversity in 101 newly sourced triticale cultivars, from a USDA germplasm bank, together with five local control cultivars, in order to identify possible crossing parents. Eight SSR markers, including five derived from rye and three from wheat, and five agronomic characteristics were used to assess diversity. In seedling screening the foreign cultivars showed resistance towards the stem rust isolate used, but were mostly susceptible to the leaf rust isolate. Out of the 8 SSR markers tested, 7 markers were polymorphic and revealed 140 alleles varying from 12 to 26 with an average of 17.5 alleles per locus. The observed polymorphic information content (PIC) value ranged from 0.39 to 0.88 with an average of 0.70, indicative of the good discriminatory ability of the SSR markers. The data revealed that the South African cultivars were genetically closely related to cultivars from the USA and Canada. The second part of the study focused on the introgression of a blue aleurone layer gene (Ba), carried by a wheat cultivar, ‘Cltr1202STR’, and purple pericarp genes (Pp1; Pp3) also carried by a wheat cultivar, ‘Amethyst’, into a triticale background. Unfortunately the introgression of the purple pericarp genes failed. Two lines containing the blue aleurone layer, 11T023 and 11T028, were however successfully created. Molecular typing of these lines with SSR markers were able to show that BC4F1 line 11T023 (Ba) B was genetically similar to the recurrent parent ‘Agbeacon’; and that the BC4F1 11T028 line (Ba) A was closest to the ‘US2007’ recurrent parent. The study illustrated that MAS was a reliable tool for detecting genetic diversity in newly sourced germplasm, and assisted in making a backcross breeding effort more effective. The data generated from MAS could therefore clearly assist in making the SU-PBL breeding program more effective by moving, better informed, decision making toward data based partly on the genotype, thereby minimizing the risks associated with purely phenotypic based decisions.

Investigation into the suitability of spring triticale (×Triticosecale Wittmack) for bio-ethanol production in the Western Cape

Tsupko, Yuriy Vadimovich 12 1900 (has links)
MScAgric / Thesis (MSc (Genetics))--University of Stellenbosch, 2009. / ENGLISH ABSTRACT: In the Western Cape small grain cereals, triticale (×Triticosecale Wittmack ex A. Camus) in particular, appear to be among the most promising starch-carrying raw materials for the production of bio-ethanol. A core group of cultivars and lines from the Stellenbosch University Plant Breeding Laboratory spring triticale breeding programme were subjected to initial testing for the purpose of ethanol production. They underwent multi-location field-testing across six (season 2006–2007) and nine (season 2007–2008) locations representing the Western Cape cereal production area. Climatic conditions during the study were characterised as generally favourable, especially in the 2007 season. During the season, trials were visited in order to make in situ observations. Disease susceptibility was given specific attention. After harvesting, grain yield (kg.ha-1), test weight (kg.HL-1), total starch content in whole grain (%), amylose/amylopectin ratio, protein content (%), ethanol output (L.tonne-1) and ethanol yield (L.ha-1) were analysed. Near infra-red reflectance spectroscopy calibration models were developed for moisture and starch contents. The best calibration based on whole grain spectra for moisture content had RPD = 1.691, R2 = 0.657 and SEP = 0.271%, and for starch content RPD = 1.646, R2 = 0.634 and SEP = 1.356%. Calibrations developed from milled grain showed better results for moisture content RPD = 2.526, R2 = 0.843, SEP = 0.182%, and for starch content RPD = 1.741, R2 = 0.673, SEP = 1.277%. These calibrations are suitable for rough screening of samples. In the 2006 season, starch yield was highly positively correlated with grain yield (R2 = 0.988, P <0.001). Both starch yield and grain yield were positively correlated with days to heading (R2 = 0.533 and R2 = 0.556, respectively; P <0.001). The 2007 season was characterised by a generally higher starch yield (2952– 3142kg.ha-1, 95%CI) compared to the 2006 season (2077–2315kg.ha-1, 95%CI). Starch yield was strongly positively correlated with grain yield (R2 = 0.975, P <0.001). Test weight demonstrated weak positive correlation with ethanol yield (R2 = 0.238, P <0.01) and grain yield (R2 = 0.279, P <0.001). Mean ethanol output ranged between 466–477L.tonne-1 at the 95%CI. Ethanol output was demonstrated to be more dependent on starch and other polysaccharides accessibility to enzymatic digestion than on the total starch content as such. The best lines for ethanol output in the 2007 season were G2, D3 and H2 for the Swartland region, and D3, G2 and D1 for the Overberg region. The best triticale lines under investigation showed their potential from a biological point of view to be a suitable crop for ethanol production in the Western Cape, with the achieved ethanol yield ranging between 2446–2625L.ha-1 at the 95%CI. For the Swartland region the best genotypes for ethanol yield were D1, H1 and D2, and for the Overberg H1 and G2. The 23 best lines were selected from the elite and senior blocks, and then used for the establishment of a recurrent massselection pre-breeding block.

Hospodaření na ekologické farmě v podhůří Šumavy a biodiverzita vybraných agroekosystémů / Biodiversity of model agroecosystems on the ecological farm in the foothills of Šumava Mts.

OUŘEDNÍK, David January 2014 (has links)
The content of the thesis was research and study of the epigeal beetle communities at three different sites in the cadastral municipality Mačice near village Soběšice in Western Region. Habitats were managed under organic farming. Specifically there were three different sites, a field sown triticale, meadow and pasture. Method of pitfall traps was used for the collection of biological material. In total, there were intercepted and identified 640 individuals from 57 species and 14 families in the reporting period. In all of the habitats, species of beetles family (Carabidae) (23 species) were mostly represented, followed by family rove beetles (Staphylinidae) (11 species) and family carrion (Silphidae) (6 species). Most individuals were found at the field habitat with the numbers in total of 255 (30 species from 8 families), followed by habitat meadow with 198 individuals (36 species from 9 families) and the pasture habitat with 187 individuals (34 species from 10 families). The species spectrum of epigeal beetles was divided into groups according to the sensitivity to anthropogenic influences. Species that do not have special requirements on the quality and nature of the environment (group E) dominated in all habitats with 48 species. Total of nine species inhabiting habitats moderately affected by human activities (group R2) were found in all habitats, the least of them occurred in the field (1 species) , pasture (5 species) and at the meadow were found most (6 species). Index of anthropogenic impacts shows that these are very heavily modified habitats. When comparing individual sites, field habitat (ISD 1,67 %) was found to be the most affected. The least affected was the meadow habitat (ISD 8,34 %). As a result we can conclude that the hypothesis that organic farming has a positive impact on biodiversity was not confirmed.

Protein composition-functionality relationships using novel genetic lines

Jonnala, Ramakanth S January 1900 (has links)
Doctor of Philosophy / Department of Grain Science and Industry / Finlay I. MacRitchie / Novel genetic materials were used to deduce gluten protein composition-functionality relationships. The Pegaso bread wheat near-isogenic lines (NILs) included addition, variation and/or deletion of major loci coding for HMW-GS, LMW-GS and gliadins. The waxy wheat lines (Svevo and N11 set) included wild, partial and complete waxy lines. Triticale translocations include 1R.1D and 1A.1D lines (GDS7, Trim, Rhino and Rigel sets) with HMW-GS 5+10 and 2+12. The main goal of the study was to establish the usefulness of NILs as appropriate materials to investigate the structure-function relationships of wheat proteins and to evaluate the performance of unique triticale translocations and waxy wheat lines. Effect of genetic variation on phytochemical (phenolic acid and policosanol) contents was also studied. Innovative methods like MALLS, Lab-on-a-chip and micro (10 g) baking were utilized along with traditional analytical methods. Results confirmed the potential of using NILs in understanding the effects of certain proteins coded at specific loci that might often be targeted in breeding programs. Removal of expected chain terminators at Gli-1/Gli-2 loci causes a shift in MWD to higher values, reflected in higher UPP and dough strength. Lines with HMW-GS 5+10 were clearly separated from 2+12 lines in terms of dough strength and UPP. The present study obtained evidence that modified ω-gliadins acts as chain terminators and cause reduction of protein polymer size and thus shifts in MWD. Marked differences in terms of milling characteristics, protein composition and ultimately in end-use functionality were observed with various waxy wheat null lines. Loaf volumes with waxy wheat flour alone were higher than a 50% blend with commercial wheat; however, breads were unacceptable to consumers in all aspects. Poor milling quality, very low mixing times with low bread loaf volumes were typical of all the triticales studied. However, translocation of the HMW-GS from wheat chromosome 1D increased dough strength, particularly the HMW-GS 5+10. Among the phytochemicals studied, double nulls at Gli-1 loci of Pegaso NILs had the highest total policosanols and total phenolic acid contents.Slight variation to wheat phenolic acid composition and contents were observed with waxy wheat and triticale lines.

No. of bitstreams: 2 Sarah Maria Hoppen 2017.pdf: 773988 bytes, checksum: 54b7784e5987fc7f5a70f27152d52cb7 (MD5) license_rdf: 0 bytes, checksum: d41d8cd98f00b204e9800998ecf8427e (MD5) Previous issue date: 2017-02-22 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / Temperate forages are widely used in cold regions as a form of nutritious and inexpensive food to the producer, often in a consortium manner in order to increase their productivity. However, this activity must be studied to prove its effectiveness. So this is the objective of this study, in order to evaluate the structural, productive and nutritional characteristics of these forages, in single or intercropping cultivation. The study was conducted between April and August 2015, in a randomized complete block design, with five treatments, four replicates and three plots subdivided in time. The treatments were: AE - white oat IPR Esmeralda, AP - black oat IAPAR 61, T - forage triticale Tpolo 981, AE (T) - white oat + forage triticale, and AP (T) - black oat + forage triticale. The evaluations occurred at 46, 82 and 119 days post emergence (DPE), representing the three evaluation periods. There was a productive difference (P <0.05) between treatments AP and AE (T) in the last period, at which time all treatments presented decreases in number of live leaves (NFV) and tillers per plant, and rate of appearance and Tiller survival. The consortium favored the increase of the apical meristem and survival of tillers of white oats in the last period, compared to their single use, as well as increasing plant height in the final third, and favored higher NFV of black oat IAPAR 61 in the second period, while reducing population density. The longevity of the forage triticale can be observed in NFV and population density higher than the others at the end of the experiment. All species showed excellent quality, increasing the dry matter (DM) concentration as a function of time, reaching 22.98%. Protein was higher in the treatments AP, AP (T) and T, with averages of 18.46%, 18.80% and 19.93%, respectively. The organic matter did not differ between the means in the treatments, increasing gradually in the time, unlike the total digestible nutrients, that maintained average of 79.69%. The fibrous components (FDNcp and FDAp) presented higher concentrations in AE and AE (T), compared to the others. FDNcp, as well as hemicellulose, presented a gradual increase throughout the study. The cellulose was constant in all aspects, being related to lignin, which differed only between the periods, with superiority of the third (2.03%) in relation to the second (1.77%). The in vitro digestibility of DM presented a general decrease in concentration as a function of time, with values between 68.29% and 88.10%. Among the minerals K was higher in the first period, with a gradual decrease over time. The treatments containing AE presented the lowest concentrations of P, Zn and Mn. All minerals have met the plant requirements, however, mineral supplementation is indicated for grass-fed animals only. Due to the increase of unwanted structures, such as stalk and dead material, the emerald white IPR oats had their nutritional quality reduced, nevertheless, presenting values within the indicated for animal feed. Oats, when unmarried, can be supplied to animals from June to early September, and can be combined with triticale to increase longevity.
