Immunotherapy has revolutionized the treatment of certain cancers by facilitating the antitumor immune response and represents today one of the mainstays of cancer therapy. However, only a subset of patients responds to immunotherapy, which can also lead to serious complications. The tumor microenvironment is composed of multiple and complex cellular and molecular interactions providing to cancer cells not only a supportive framework but promoting also many steps of immunosuppression and tumor progression. To date, the mechanisms that drive the acquisition of these immunosuppressive features are still poorly defined. Tumor-associated macrophages can be highly represented in the tumor microenvironment where they are shaped and become key players in the innate and adaptive immune escape of the tumor cells.Heme oxygenase-1 is the rate-limiting enzyme that catabolizes heme into three major biologically active byproducts which display cytoprotective, antioxidant and immunomodulatory effects. We hypothesized that tumor-associated macrophages might suppress anti-tumor T-cell response through heme oxygenase-1 induction in the tumor microenvironment and macrophage polarization. We showed that heme oxygenase-1 is highly expressed in tumor-associated macrophages. By using a subcutaneous EG7-OVA lymphoma model on genetically engineered mice with a conditional deletion of heme oxygenase-1 in macrophages, our data show that myeloid-restricted heme oxygenase-1 deficiency improves the effect of a therapeutic antitumor immunization by enhancing tumor-infiltrating antitumor CD8+ T-cell proliferation and cytotoxicity and represses tumor growth. Our data suggest a major role of myeloid heme oxygenase-1 in the differentiation and the phenotypic, functional, transcriptional and epigenetic reprograming of tumor-associated macrophages. Myeloid HO-1 inhibition might be considered as a new myeloid HO-1-mediated immune checkpoint blockade. Tumor Associated Macrophages in a MaFIA Mouse Model

Recent evidence has shown the important role of macrophages in both tumor development and progression. To investigate the role of macrophages we used a mouse model known as MaFIA (Macrophage Fas Induced Apoptosis) mice that allows for the selective deletion of macrophages. Mice were given melanoma cells at various stages of depletion. Tumor mass was measured and organs were processed for flow cytometry to measure melanoma cell migration. The results show that mice receiving depletion treatment have larger tumor sizes and weights than those mice retaining their macrophage population. We detect metastasis in both the lung and kidney in both macrophage depleted and non depleted mice. The more macrophages in an organ the larger the amount of melanoma positive cells are detected.

Avaliação de macrófagos e eosinófilos associados ao tumor como fatores prognósticos em mastocitomas cutâneos caninos / Tumor-associated macrophages and eosinophils evaluation as prognostic factors in canine mast cell tumors

O mastocitoma é a neoplasia cutânea mais comum em cães. Acredita-se que as células inflamatórias associadas ao tumor desempenham papéis importantes no comportamento biológico do mastocitoma. Este estudo teve como objetivo verificar a presença de macrófagos associados ao tumor (TAMs) e eosinófilos na região intratumoral e comparar à graduação histopatológica e à evolução clínica dos casos para avaliar se são marcadores prognósticos para MCTs cutâneos caninos. Foram avaliados 38 MCTs provenientes de 30 animais submetidos à cirurgia, sem nenhuma outra terapia complementar. Os tumores foram submetidos à análise imuno-histoquímica para detecção de CD68 e à contagem de eosinófilos em lâminas coradas por hematoxilina e eosina. Os resultados foram comparados à classificação histopatológica segundo os critérios de Patnaik et a. (1984) e Kiupel et al. (2011) e à sobrevida e mortalidade em função do tumor. O número de TAM detectado pela imunomarcação de CD68 não apresentou diferenças significativas entre os graus histopatológicos, assim como não foi um indicador de mortalidade em função do tumor, nem para sobrevida, o mesmo sendo observado para a contagem de eosinófilos. Os números de TAMs e eosinófilos não apresentaram correlação. Nossos resultados sugerem que a presença de TAM e eosinófilos em MCTs cutâneos caninos não são bons indicadores prognósticos para esta doença. / Mast cell tumors are the most common skin cancer in dogs. It is believed that tumor-associated inflammatory cells play important roles in biological behavior of this neoplasm. This study aimed to verify the presence of the tumor associated macrophages (TAMs) and eosinophils in the intratumoral region in comparison with the histopathologic grading and clinical evolution of cases in order to assess their prognostic value for canine cutaneous MCTs. Thirty-eight MCTs from 30 dogs treated by surgery with no complementary therapy were investigated. The tumors were submitted to immunohistochemical analysis for CD68 detection and eosinophil count on HE-stained slides. The results were compared with histopathological classification, according to the criteria of Patnaik et al. (1984) and Kiupel et al. (2011), and with survival and mortality due to the disease. The number of TAMs and eosinophils showed no significant differences between the histopathologic grades, as well as it was not a predictor of disease-related mortality or survival. The number of TAMs and eosinophil count showed no correlation. Our results suggest that the presence of TAMs and eosinophils in cutaneous canine MCTs are not good prognostic indicators for this disease.

O carcinoma espinocelular de boca (CEB) é a neoplasia maligna mais comum da cavidade oral, correspondendo à aproximadamente 94% dos casos dessa região. Apesar dos diversos estudos moleculares e celulares do CEB, a taxa de sobrevida dos pacientes é de aproximadamente 50%, devido principalmente ao tamanho do tumor, metástase em linfonodos regionais, grau de diferenciação das células e sítio anatômico. O microambiente tumoral do CEB, é extremamente complexo e diversificado, tendo como característica principal um estado inflamatório crônico imunossupressivo. Este microambiente é sustentado pela liberação de diferentes citocinas inflamatórias, como IL-6, TNF- - atividades exercidas tanto pelas células tumorais quanto pelas estromais. Dentre essas atividades, tem sido relatado na literatura que as citocinas inflamatórias são capazes de aumentar a migração e a capacidade de invasão das células tumorais. Entre as células estromais, os macrófagos são as mais abundantes e participam da manutenção do microambiente tumoral. De acordo com o estímulo, podem ser polarizados M1, com papel pró-inflamatório e antitumoral, e M2, com papel anti-inflamatório e pró-tumoral. O objetivo desta tese foi compreender o papel dos macrófagos no prognóstico de CEB e das citocinas inflamatórias IL-6, TNF- - linhagens celulares de CEB. Para verificar o papel dos macrófagos no prognóstico, foi realizada uma revisão sistemática na qual foram incluídos apenas os estudos que utilizavam amostra de pacientes com CEB e avaliavam o prognóstico com marcadores para macrófagos. Foi observado que maiores concentrações de macrófagos CD68+ e CD163+ estavam relacionados com pior prognóstico de pacientes com CEB, embora não tenha sido possível concluir qual região tumoral a presença destas células seja mais importante 7 para o desfecho. Para analisar o papel das citocinas inflamatórias IL-6, TNFILensaios in vitro utilizando duas linhagens celulares, SCC25 e Cal27, em condições promotoras de migração sob a influência dessas citocinas. Foi observado que a citocina IL-6 foi capaz de aumentar a velocidade de migração e a direcionalidade tanto da SCC25 quanto da Cal 27 e que esta melhora na capacidade migratória ocorreu através de um crosstalk entre a via de sinalização relacionada a IL6 (STAT3) e a via reguladora de migração celular, Rho GTPase Rac1. The OSCC tumor microenvironment is extremely complex and diverse, with the main characteristic being an immunosuppressive chronic inflammatory state. This microenvironment is supported by the release of different inflammatory cytokines, such as IL-6, TNF- - and enhance the activities of both tumor and stromal cells. Among these activities, it has been reported in the literature that inflammatory cytokines are capable of increasing migration and invasiveness of tumor cells. It was observed that IL-6 was able to increase the speed migration and directionality of both SCC25 and Cal 27 and that this improvement in migratory capacity occurred through a crosstalk between the IL6-related signaling pathway (STAT3) and cell migration-related pathway, RhoGTPase Rac1. These data reinforce the role of the tumor microenvironment in the tumor progression process and suggest potential therapeutic targets such as the modulation of the profile of the macrophages population and the role of interleukins in the control of tissue invasion and metastasis.

Regulatory T Cells Promote Breast Cancer Progression Through Inhibiting Classical Activation of Macrophages

Transient ablation of regulatory T cells has been shown to be effective at hindering tumor growth and metastasis in murine breast cancer model. Based on our lab’s previous work, we have demonstrated that NK cells and CD8+ cytotoxic T cells were not required for the protective effect of Treg cell ablation. However, we also reported that CD4+ helper T cells and IFN-γ were required for the protective effect of Treg cell ablation. Furthermore, we observed that CD11B+ cells responded to Treg ablation therapy by up-regulating target genes of IFN-γ. Therefore, this study aimed to investigate the connection between the myeloid cell compartment and IFN-γ signaling after regulatory T cell ablation therapy. Through a combination of conditional knockout mouse models, cellular fate mapping experiments, adoptive transfers, and co-injection experiments, we demonstrated that tumor-associated macrophages (TAMs), derived from the bone marrow via CCR2/CCL2 axis, were responsible for the therapeutic effect of regulatory T cell ablation. In addition, we determined that IFN-γ signaling was required for the TAMs to mediate the protective phenotype seen after regulatory T cell ablation. Furthermore, based on our findings, we developed a genetic signature based on TAMs from treated or untreated tumors that had a predictive value for patient survival. Thus, our findings indicated a strong connection between IFN-γ release, classical activation of TAMs, and depletion of regulatory T cells, and taken together, our data could offer potential clinical strategies to mimic regulatory T cell ablation.
