One of the key measures of the IP.TMTx/ Rx is reliability is link availability (LA), and a key element of LA is time to resynchronize after RF link loss.

Integrating XML and RDF concepts to achieve automation within a tactical knowledge management environment

Approved for public release, distribution is unlimited / Since the advent of Naval Warfare, Tactical Knowledge Management (KM) has been critical to the success of the On Scene Commander. Today's Tactical Knowledge Manager typically operates in a high stressed environment with a multitude of knowledge sources including detailed sensor deployment plans, rules of engagement contingencies, and weapon delivery assignments. However the WarFighter has placed a heavy reliance on delivering this data with traditional messaging processes while focusing on information organization vice knowledge management. This information oriented paradigm results in a continuation of data overload due to the manual intervention of human resources. Focusing on the data archiving aspect of information management overlooks the advantages of computational processing while delaying the empowerment of the processor as an automated decision making tool. Resource Description Framework (RDF) and XML provide the potential of increased machine reasoning within a KM design allowing the WarFighter to migrate from the dependency on manual information systems to a more computational intensive Knowledge Management environment. However the unique environment of a tactical platform requires innovative solutions to automate the existing naval message architecture while improving the knowledge management process. This thesis captures the key aspects for building a prototype Knowledge Management Model and provides an implementation example for evaluation. The model developed for this analysis was instantiated to evaluate the use of RDF and XML technologies in the Knowledge Management domain. The goal for the prototype included: 1. Processing required technical links in RDF/XML for feeding the KM model from multiple information sources. 2. Experiment with the visualization of Knowledge Management processing vice traditional Information Resource Display techniques. The results from working with the prototype KM Model demonstrated the flexibility of processing all information data under an XML context. Furthermore the RDF attribute format provided a convenient structure for automated decision making based on multiple information sources. UDP-sugar metabolizing pyrophosphorylases in plants : formation of precursors for essential glycosylation-reactions

UDP-sugar metabolizing pyrophosphorylases provide the primary mechanism for de novo synthesis of UDP-sugars, which can then be used for myriads of glycosyltranferase reactions, producing cell wall carbohydrates, sucrose, glycoproteins and glycolipids, as well as many other glycosylated compounds. The pyrophosphorylases can be divided into three families: UDP-Glc pyrophosphorylase (UGPase), UDP-sugar pyrophosphorylase (USPase) and UDP-N-acety lglucosamine pyrophosphorylase (UAGPase), which can be discriminated both by differences in accepted substrate range and amino acid sequences. This thesis focuses both on experimental examination (and re-examination) of some enzymatic/ biochemical properties of selected members of the UGPases and USPases and UAGPase families and on the design and implementation of a strategy to study in vivo roles of these pyrophosphorylases using specific inhibitors. In the first part, substrate specificities of members of the Arabidopsis UGPase, USPase and UAGPase families were comprehensively surveyed and kinetically analyzed, with barley UGPase also further studied with regard to itspH dependency, regulation by oligomerization, etc. Whereas all the enzymes preferentially used UTP as nucleotide donor, they differed in their specificity for sugar-1-P. UGPases had high activity with D-Glc-1-P, but could also react with Frc-1-P, whereas USPase reacted with arange of sugar-1-phosphates, including D-Glc-1-P, D-Gal-1-P, D-GalA-1-P, β-L-Ara-1-P and α-D-Fuc-1-P. In contrast, UAGPase2 reacted only with D-GlcNAc-1-P, D-GalNAc-1-P and, to some extent, with D-Glc-1-P. A structure activity relationship was established to connect enzyme activity, the examined sugar-1-phosphates and the three pyrophosphorylases. The UGPase/USPase/UAGPase active sites were subsequently compared in an attempt to identify amino acids which may contribute to the experimentally determined differences in substrate specificities. The second part of the thesis deals with identification and characterization of inhibitors of the pyrophosphorylases and with studies on in vivo effects of those inhibitors in Arabidopsis-based systems. A novel luminescence-based high-throughput assay system was designed, which allowed for quantitative measurement of UGPase and USPase activities, down to a pmol per min level. The assay was then used to screen a chemical library (which contained 17,500 potential inhibitors) to identify several compounds affecting UGPase and USPase. Hit-optimization on one of the compounds revealed even stronger inhibitors of UGPase and USPase which also strongly inhibited Arabidopsis pollen germination, by disturbing UDP-sugar metabolism. The inhibitors may represent useful tools to study in vivo roles of the pyrophosphorylases, as a complement to previous genetics-based studies. The thesis also includes two review papers on mechanisms of synthesis of NDP-sugars. The first review covered the characterization of USPase from both prokaryotic and eukaryotic organisms, whereas the second review was a comprehensive survey of NDP-sugar producing enzymes (not only UDP-sugar producing and not only pyrophosphorylases). All these enzymes were discussed with respect to their substrate specificities and structural features (if known) and their proposed in vivo functions.

Effects of Amino Acid Insertion on the Substrate and Regiospecificity of a Citrus paradisi Glucosyltransferase

Glucosyltransferases, or GTs, are enzymes which perform glucosylation reactions. These glucosylation reactions involve attaching a UDP-activated glucose molecule to acceptor molecules specific to the enzyme. The products of these reactions are observed to have a myriad of effects on metabolic processes, including stabilization of structures, solubility modification, and regulation of compound bioavailability. The enzyme which our lab focuses its research on is a flavonol-specific 3-O-GT found in Citrus paradisi, or grapefruit. This enzyme is part of the class of enzymes known as flavonoid GTs, which are responsible for, among other things, the formation of compounds which can affect the taste of citrus. Our lab focuses its research on performing site-directed mutagenesis on Citrus paradisi 3-O-GT in an attempt to modify its substrate specificity and regiospecificity. In this poster, we report our findings thus far concerning the addition of specific residues to the 3-O-GT's amino acid sequence based on an alignment with the sequence of a putative flavonoid GT found in Citrus sinensis.

In this study, we are testing the basis of substrate septicity of Cp3GT. We hypothesize that incorporation of fi ve amino acids specifi c to Citrus sinensis GT (CsGT) into Cp3GT at 308th position may facilitate mCp3GT to use anthocyanidins as one of the substrates. We report our fi ndings thus far concerning the addition of specifi c residues to the Cp3GT’s amino acid sequence based on an alignment with the sequence of a putative fl avonoid GT found in Citrus sinensis.

PACKET FILTER APPROACH TO DETECT DENIAL OF SERVICE ATTACKS

Denial of service attacks (DoS) are a common threat to many online services. These attacks aim to overcome the availability of an online service with massive traffic from multiple sources. By spoofing legitimate users, an attacker floods a target system with a high quantity of packets or connections to crash its network resources, bandwidth, equipment, or servers. Packet filtering methods are the most known way to prevent these attacks via identifying and blocking the spoofed attack from reaching its target. In this project, the extent of the DoS attacks problem and attempts to prevent it are explored. The attacks categories and existing countermeasures based on preventing, detecting, and responding are reviewed. Henceforward, a neural network learning algorithms and statistical analysis are utilized into the designing of our proposed packet filtering system.

Vi har valt att titta på hur företag använder tekniken samtidigt som vi tittar på vilka nya tjänster och möjligheter som finns. Hur ser tjänsteutbudet ut idag och hur påverkar detta sättet att kommunicera inom företagen? Vilken funktionalitet används och hur drar man nytta av de fördelar och möjligheter som den nya tekniken erbjuder? Med förändrade kommunikationssätt ser vi hur verksamheter både kan effektiviseras och utvecklas.</p><p>När vi tittar på hur leverantörer bidrar till att utveckla sina kunders verksamheter så ser vi att leverantörerna inte är delaktiga inom detta område i någon större utsträckning. Vi ser dock att man inte hunnit så långt i utvecklingen av VoIP, samt att leverantörer har vissa problem med att nå ut och marknadsföra de tjänster och möjligheter den nya tekniken faktiskt erbjuder. Fokus ligger istället mer på de kostnadsbesparingar man gör genom att kommunicera över internet (IP) istället för med traditionell telefoni (PSTN).</p><p>Tekniken är beroende av kvaliteten på bredbandsuppkoppling samt den interna infrastrukturen hos företagen. Mycket av de problem så som eko och dålig samtalskvalitet beror på undermålig utrustning internt på företagen. Dessa problem ser man i branschen som något övergående då ny hårdvara utvecklas och förses med bättre stöd för VoIP. Många leverantörer erbjuder även sina kunder en helhetslösning för att få kontroll över både telefoni och internetförbindelse.</p><p>Slutligen ser vi att VoIP öppnar upp för en omstöpning av hur företag kommunicerar, möjligheterna har bara börjat utforskas. Avaliação da experiência do usuário com base na arbitragem de parâmetros de QoS em fluxo multimídia, em cenários com roteadores Cisco e placa NetFPGA

A streaming video has features that differs from other streams: the occupation of large amount of bandwidth, and the possible variation of scenes with consequent increase (or decrease) in amount of bytes transmitted. Applications for video streaming transmission (YouTube, Vimeo, Netflix, Telecine Play, etc.) have aroused the interest of the scientific community regarding the behavior of networks. Our proposal is to measure, from a video stream, the user experience (QoE) based on arbitration QoS parameters in a controlled environment which use NetFPGA. The results of the experiments has shown the efficacy of TCP over UDP, which at the stage of choosing the most appropriate protocol for testing, showed to be ineffective to the Gigabit Ethernet rate. These experiments occurred in distinct physical topologies; only Cisco routers, with and without QoS, with and without injection of traffic generated by software iperf in the network, and using NetFPGA board set to router only with and without traffic via iperf, once the design developed to make the board work as a router does not allow QoS settings. in these scenarios, there was the need to start the client running at different times, setting their respective caches different networks so that did not coincide scenes, avoiding high bandwidth peaks in stretches with a lot of movement. / Um streaming de vídeo tem características que o difere dos demais fluxos: a ocupação de grande quantidade de banda, e a possíveis variações de cenas com consequente aumento (ou diminuição) na quantidade dos bytes transmitidos. Aplicações para transmissão de streaming de vídeo (YouTube, Vimeo, NetFlix, Telecine Play, e etc.) tem despertado o interesse da comunidade científica quanto ao comportamento das redes. Nossa proposta é correlacionar, a partir de um fluxo de vídeo, a experiência do usuário com base na arbitragem de parâmetros de QoS em ambiente controlado. O resultado dos experimentos demonstrou a eficácia do protocolo TCP sobre o UDP, que na fase da escolha do protocolo mais adequado para os testes, mostrou-se ineficiente até a taxa Gigabit Ethernet. Esses experimentos ocorreram em topologias físicas distintas; apenas com roteadores Cisco, com e sem QoS, tendo ou não injeção de tráfego gerado pelo software iPerf na rede, e usando a placa NetFPGA no modo router, apenas com e sem tráfego via iPerf, uma vez que o projeto desenvolvido para fazer a placa atuar como roteador não permite configurações de QoS. Nesses cenários, constatou-se a necessidade de iniciar a execução dos clientes em momentos diferentes, configurando seus respectivos caches de redes diferentes, para que não coincidissem as cenas, evitando altos picos de banda em trechos com muita movimentação.
