Previous issue date: 2014-07-16 / O polietileno de ultra alto peso molecular (PEUAPM) é um polímero com
excelentes propriedades físicas e mecânicas tendo como mais notáveis a sua
inércia química, a lubricidade, resistência impacto e resistência a abrasão.
Exibe um papel relevante no sucesso na Artroplastia Total de Joelho (ATJ) e
Artroplastia Total de Quadril (ATQ). A Artroplastia se caracteriza pela
substituição da articulação natural por uma articulação artificial. As articulações
artificiais são formadas por um conjunto de componentes, sendo fabricados de
matéria prima metálica ou cerâmica e um componente, normalmente feito em
PEUAPM, que é interposto entre componentes metálicos. Alguns fatores
contribuem para um maior desgaste do componente de polietileno. Os
componentes fabricados em polietileno PEUAPM não possuem
fotoestabilizadores. E em inspeções realizadas em fabricantes e distribuidores
destes produtos foi observado que durante seu processamento fabril e
comercialização eles não são adequadamente protegidos da iluminação direta.
As características consideradas importantes para o desempenho in vitro destes
dispositivos estão definidas em normas técnicas, mas muitas destas normas
somente recomendam determinados requisitos considerados importantes do
ponto de vista técnico. Atualmente a área de materiais de uso em saúde
trabalha em regulamentos que visam instituir os requisitos mínimos para
registro de implantes ortopédicos. Desta forma, este trabalho propôs avaliar as
possíveis alterações de componentes fabricados em polietileno de ultra alto
peso molecular utilizados em procedimentos cirúrgicos de artroplastia e joelho
e quadril, exposto a radiação ultravioleta. O trabalho foi dividido em 4 grupos:
grupo 1 (material não exposto a UV); grupo 2 (material exposto a UV por 1h);
grupo 3 (material exposto a UV por 24h) e grupo 4 (material exposto a UV por
48h). Todas as amostras foram caracterizadas por Difração de Raio X (DRX),
Espectroscopia de Absorção de Infravermelho por Transformada de Fourier
(FTIR), Microscopia Ótica (MO), Microscopia Eletrônica de Varredura (MEV)
com mapeamento por EDS e Microscopia de Força Atômica (AFM) com o
intuito de avaliar as possíveis degradações do PEUAPM submetidos a radiação
UV. Os resultados demonstraram que a exposição do polímero a radiação UV
em todos os tempos testados neste trabalho promoveu alterações nas
amostras, pois foi observado, principalmente, nos ensaios de DRX e EDS que
ocorreu respectivamente, um discreto aumento da cristalinidade do material e
diminuição no percentual de oxigênio. Desta forma, pode-se concluir que o
aumento da cristalinidade pode, pela ação da radiação UV, afetar nas
propriedades do material contribuindo para o aumento do desgaste e
diminuição do tempo de vida útil do implante. / The polyethylene, ultra high molecular weight (UHMWPE) is a polymer with
excellent physical and mechanical properties with the most notable its chemical
inertness, lubricity, impact resistance and abrasion resistance. Displays a
relevant role in the successful Total Knee Arthroplasty (TKA) and Total Hip
Arthroplasty (THA). The arthroplasty is characterized by replacement of the
natural joint with an artificial joint. Artificial joints are formed by a set of
components, being manufactured from metal or ceramic raw material and a
component normally done in UHMWPE, which is interposed between the metal
components. Some factors contribute to increased wear of the polyethylene
component. Components manufactured in polyethylene UHMWPE not have
photostabilizers. And inspections on manufacturers and distributors of these
products was observed that during its industrial processing and marketing they
are not adequately protected from direct light. Characteristics considered
important for the in vitro performance of these devices are set in technical
standards, but many of these standards only recommend certain requirements
considered important from a technical standpoint. Currently the area of
materials for use in healthcare working on regulations that seek to establish the
minimum requirements for registration of orthopedic implants. Thus, this study
aimed to evaluate the possible changes of components manufactured in
polyethylene, ultra high molecular weight used in surgical procedures and knee
arthroplasty and hip, exposed to ultraviolet radiation. The work was divided into
4 groups: group 1 (material not exposed to UV); group 2 (material exposed to
UV for 1 h); group 3 (material exposed to UV for 24 h) and group 4 (material
exposed to UV for 48 h). All samples were characterized by X-Ray Diffraction
(XRD), Absorption Spectroscopy Fourier Transform Infrared (FTIR), optical
microscopy (OM), scanning electron microscopy (SEM) with EDS mapping and
Atomic Force Microscopy (AFM) in order to evaluate the possible degradation of
UHMWPE subjected to UV radiation. The results showed that exposure of the
polymer to UV radiation at all times tested in this study modified the samples, it
was observed mainly in the experimental XRD and EDS which occurred
respectively a slight increase in crystallinity and decrease in the percentage
oxygen. Thus, it can be concluded that the increase in crystallinity can by the
action of UV radiation, affect the material properties contributing to increased
wear and reduced useful life of the implant.

The presence of ultraviolet patterns on body, as well as perception of ultraviolet spectrum by special photoreceptors, is part of sensory ecology of many animal species, including reptiles. Most current research discusses the importance of ultraviolet signs in coloration of diurnal species. The aim of our study was to find out what character have the reflective signs in ultraviolet spectrum in overall coloration of common leopard gecko (Eubplepharis macularius) through a digital photography. The reflective pattern is present in both adults and juveniles and passes as well as the rest of the coloration by significant ontogenetic changes. Another aim was to evaluate the role of ultraviolet reflecting signs in the biology of this crepuscular-nocturnal species. We expect that the pattern contributes to their antipredatory strategies during their first few months of life, and also a white reflecting surface is preserved in adulthood, especially on their tail, which is differently coloured than the rest of the body.

Evaluation of the effects of solar ultraviolet radiation on the growth of vibrio cholerae and on the secretion of the cholera toxinSsemakalu, Cornelius Cano 09 1900 (has links)
Cholera is a water-borne disease that continues to ravage resource poor communities around the world especially those in developing countries. The disease is caused by Vibrio cholerae microorganisms whose natural habitat is the aquatic ecosystem. It is believed that this microorganism prior to becoming the primary cause of cholera acquired virulence factors expressed by two separate genetic elements. These genetic elements are known as VPIФ and CTXФ were acquired in that order for known physiological reasons. However only V. cholerae in possession of the CTX genetic element are capable of causing cholera disease. At present only two serotypes are known to have the ability to cause cholera and these are V. cholerae serotypes O1 and O139. SODIS (Solar disinfection) is an extremely low cost refined technology that can be used for the disinfection of water especially in areas where there is a considerable amount of sunshine. Although this technology is a composite of various factors the underlying principle is the use of solar ultraviolet radiation (SUVR). The preliminary target of SUVR is the cytoplasmic membrane and this was confirmed by flow cytometric analysis. The consequences of leaky cytoplasmic membrane include cellular death to the microorganism as well as an increase in cholera toxin secretion. The main objective of this study was to investigate the effect of solar ultraviolet radiation on the growth of V. cholerae and on the secretion of cholera toxin and to provide supporting information for the use of SODIS in South Africa while observing the possible role that climate may play in the onset of cholera disease. The initial part of the study evaluated the culturability, biomass increase and cholera toxin secretion in both a nutrient poor and a nutrient rich media by two toxigenic and one non toxigenic strain of V. cholerae. A series of pH and temperature combinations were used to achieve this objective. The result revealed that the microorganisms survived in both media. An increase in biomass was observed for all the bacteria grown in the nutrient rich media whereas in the poor nutrient media the bacteria remained culturable but no increase in biomass was observed. Interestingly lower temperatures seemed to provide more optimal growth conditions while high temperature on most occasions favoured cholera toxin secretion, in both media.The second part of the study required the exposure of the microorganisms to SUVR. A SODIS approach was used with a few modifications. The V. cholerae strains were exposed to solar radiation during all the seasons of the year. Evaluation of the viability, the increase in biomass and the detection of cholera toxin secretion was determined after each exposure to solar radiation. The results seem to suggest that the effect of SUVR depended on the season of the year, the nature of the media, strain, solar conditions and in the duration of solar exposure, in no particular order. The secretion of cholera toxin was mainly dependent on the media used, the season of the year and on the serotype of the strain. This study represents the first report on the evaluation of SUVR for the disinfection of water under South African conditions (Pretoria area) during all seasons of the year with variations in solar radiation levels and temperature. Furthermore what actually happened to V. cholerae during solar exposure in terms of cell morphology, cell viability and secretion of cholera toxin is also reported and this can give an insight of the possible role that SUVR may play in the onset of cholera. The main recommendation emanating from this study is the sensitisation of communities worldwide about the capacity that, SUVR carries to lighten the burden of communicable water borne diseases especially, in resource limited areas through the implementation of SODIS. / Life and Consumer Sciences / M. Fundação de Amparo a Pesquisa no Estado do Rio Grande do Sul / On the last decades, amphibian populations have been suffering a global decline, and different factors can be related to this process. Parallel observations between the decrease of stratospheric ozone concentration in the late 1970s and amphibian decline in early 1980s aroused interest to study the possible association among increased incidence of ultraviolet radiation (UV) and amphibian decline. The UV radiation has important biological impact on living organisms due to their ability to induce lesions in the DNA molecule, which may result in the induction of mutations or cell death. Photoreactivation is an important DNA repair mechanism that promotes the reversal of UV-induced DNA damage by photolyase enzymes after the absorption of UVA and visible light photons. In this study, we evaluated the biolocial effects of UVB and UVA through morphological, histological, locomotor and survival of Hypsiboas pulchellus (Anura: Hylidae) tadpoles. Tadpoles kept in the laboratory were submitted to acute and chronic treatments, composed of different doses of UVB and UVA exposures. The results of survival curves indicate that the H. pulchellus is very sensitive to UVB radiation comparative to UVA. However, despite the high tadpole survival after UVA, they showed serious malformations after metamorphosis. Tadpoles body weight also suffered influence of different UV radiation doses, however the body length was bit affected. The locomotor capacity (horizontal and vertical speed) of UVB-exposed tadpoles has been significantly changed. In addition, UVB radiation had severe impact on the skin (stratum corneum) and mouth (rows of teeth and jaw sheaths) keratin structures of tadpoles, indicating that these should be important impacts of solar UV radiation in reducing the performance of these organisms. In parallel, it was also possible to observe the extreme importance of activation of photolyases for the repair of DNA damage after treatment with UVB but not UVA. Therefore, in this study, we present a wide characterization of the biological effects of solar UVB and UVA on the performance of H. pulchellus tadpoles. / Durante as últimas décadas, as populações de anfíbios vêm sofrendo um declínio generalizado e diferentes fatores são sugeridos como responsáveis por esse efeito. Observações paralelas entre o rápido decréscimo da concentração do ozônio estratosférico no final da década de 1970, com o declínio de anfíbios no início da década de 1980, despertaram grande interesse para se estudar as possíveis associações entre o aumento da incidência de radiação ultravioleta (UV) na superfície e a redução do número de espécies de anfíbios. Sabe-se que a radiação UV solar tem importante impacto biológico na vida dos organismos devido a sua grande capacidade de induzir lesões na molécula de DNA, que podem levar a indução de mutações ou morte celular. Um dos principais mecanismos de reparo de DNA é a fotorreativação que ocorre por meio das enzimas fotoliases, as quais promovem reversão dessas lesões ao absorver fótons de luz UVA e luz visível. Neste projeto, avaliou-se a ação biológica da radiação UVB e UVA a partir de dados morfológicos, histológicos, locomotores e de sobrevivência de girinos da espécie Hypsiboas pulchellus (Anura: Hylidae). Girinos coletados e mantidos no laboratório foram submetidos a tratamentos agudos e crônicos, compostos por exposições a diferentes doses de luz UVB e UVA. Os resultados de sobrevivência indicam que os girinos de H. pulchellus são muito sensíveis à radiação UVB em relação à UVA. Entretanto, apesar da alta sobrevivência dos indivíduos submetidos à radiação UVA, estes apresentaram sérias malformações em longo prazo. A massa corporal dos girinos também sofreu influência das diferentes doses de radiação UV aplicadas, entretanto o comprimento total foi pouco influenciado. A capacidade locomotora (vertical e horizontal) dos organismos expostos à luz UV foi bastante alterada, onde se destaca a grave redução da atividade locomotora dos indivíduos tratados com radiação UVB. Adicionalmente, a radiação UVB teve impacto severo nas estruturas de queratina da pele (estrato córneo) e da boca (queratodontes e mandíbula) dos girinos, indicando que esses devem ser impactos principais da radiação UV solar na redução da performance desses organismos. Em paralelo, também foi possível observar a extrema importância da ativação das enzimas fotoliases para a reparação dos danos de DNA, assim como os seus efeitos após os tratamentos com radiação UVB, mas não com UVA. Portanto, no presente trabalho, foi possível apresentar uma ampla caracterização dos efeitos biológicos da radiação UVB e UVA solar na performance de girinos de H. pulchellus.

Após a colheita, os frutos foram selecionados para tornar o lote homogêneo. Foi realizada a caracterização do fruto verde e amadurecido quanto a acidez titulável, pH, umidade, teor de lipídios, cinzas (minerais), proteína (nitrogênio bruto), açúcar total e coloração. Foram realizados 3 experimentos com frutos recém colhidos. Experimento I: imersão dos frutos em água a 45ºC por 5, 10, 15 e 20 minutos; Experimento II: radiação gama nas doses 0,2; 0,4; 0,6 e 1,0kGy e Experimento III: radiação ultravioleta (UVC) por 5, 10, 15 e 20 minutos. Após os tratamentos, os frutos foram armazenados em temperatura ambiente (21ºC±1 e 70±5% de UR) e refrigerado (10ºC±1 e 90±5% de UR). Os frutos foram analisados quanto à capacidade antioxidante pelo método DPPH e compostos fenólicos totais aos 0, 3, 9 e 12 dias. A coloração foi avaliada aos 0, 3, 6, 9, 12 e 15 dias. Os resultados foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey ao nível de 1% de probabilidade. Nas condições em que os experimentos foram realizados, pode-se concluir que os frutos do tratamento testemunha e mantidos sob refrigeração apresentaram maior capacidade antioxidante e teor de compostos fenólicos totais ao longo do período de armazenamento; o tratamento térmico por 5 e 10 minutos foi efetivo na manutenção da capacidade antioxidante e dos compostos fenólicos; a dose de 0,6kGy não foi efetiva na manutenção da capacidade antioxidante; a radiação ultravioleta (UV-C) não manteve a capacidade antioxidante dos frutos, no entanto ocasionou aumento na quantidade de compostos fenólicos; o tratamento térmico e a radiação ultravioleta não afetaram a coloração da polpa / This study aimed to determine the antioxidant activity, total phenolic compounds and color in avocado 'Hass' submitted to heat treatment, gamma and ultraviolet radiation. After harvest, fruits were selected to make the homogeneous portion. The characterization was performed of the green fruit and mature as acidity, pH, moisture, lipids, ash (minerals), protein (nitrogen gross), total sugar and coloring. 3 experiments were performed with freshly harvested fruits. Experiment I: immersion of fruits in water at 45 ° C for 5, 10, 15 and 20 minutes; Experiment II: gamma radiation doses 0.2, 0.4, 0.6 and 1.0 kGy and Experiment III: ultraviolet radiation (UV-C) for 5, 10, 15 and 20 minutes. After treatments, fruits were stored at room temperature (21±1ºC and 70±5% HR) and refrigeration (10ºC±1 and 90±5% HR). The fruits were analyzed for their antioxidant capacity by DPPH method and phenolic compounds at 0, 3, 9 and 12 days. The color was measured at 0, 3, 6, 9, 12 and 15 days. Data were subjected to analysis of variance and mean comparison using Tukey’s test at 1% probability. In conditions where the experiments were performed, one can conclude that the fruits of control treatment and kept under refrigeration had higher antioxidant capacity and content of phenolic compounds during the storage period, the heat treatment for 5 and 10 minutes was effective the maintenance of antioxidant capacity and phenolic compounds, the dose of 0.6 kGy was not effective in maintenance the antioxidant capacity, ultraviolet radiation (UV-C) did not maintain the antioxidant capacity of fruits, however caused an increase in the content of phenolic compounds, heat treatment and ultraviolet radiation did not affect the pulp color
