Production et caractérisation de l'allergène recombinant Bet v 1 utilisé à des fins d'immunothérapie allergénique / PRODUCTION & CHARACTERIZATION OF Bet v 1 RECOMBINANT ALLERGEN INTENDED FOR IMMUNOTHERAPYNony, Emmanuel 09 January 2015 (has links)
L'allergie respiratoire au pollen de bouleau affecte un nombre important de personnes dans le monde. Il est estimé que 100 millions d'individus sont sensibilisés à l'allergène majeur du pollen de bouleau nommé Bet v 1. L’immunothérapie allergénique, basée sur l'administration répétée de l'allergène incriminé, permet la rééducation du système immunitaire du patient d’un profil TH2 vers un profil TH1/Treg et à terme la diminution des symptômes allergiques. Ces travaux de thèse avaient donc pour finalité de produire et de caractériser l'allergène recombinant Bet v 1, à des fins d’immunothérapie allergénique. Dans ce cadre, différentes méthodes analytiques ont été développées et appliquées afin d'optimiser le procédé de production via l'élimination de différentes impuretés liées au produit ou au procédé de production et de documenter la structure de l’allergène. En particulier, l'utilisation de la spectrométrie de masse a permis la détermination de la masse exacte de l'allergène ainsi que la vérification complète de sa séquence en acides aminés. Les travaux en spectrométrie de masse ont également contribué aux détections et identifications de diverses impuretés et produits de dégradations et ont ainsi conduit à plusieurs optimisations du procédé industriel de production de l'allergène recombinant. Les activités immunologiques de certains produits de dégradations ont également été investiguées. La structure tertiaire (spatiale) de l'allergène a été déterminée par diffraction aux rayons X. Enfin, ces travaux ont permis de documenter la qualité de l'allergène recombinant rBet v 1 afin i) de l'établir comme substance de référence pour la Pharmacopée Européenne et ii) de procéder à une étude clinique d’immunothérapie allergénique de phase II auprès de 483 patients allergiques au pollen de bouleau. / Respiratory allergy to birch pollen affects a large number of people in the world. It is estimated that 100 million people are sensitized to the major allergen from birch pollen, namely Bet v 1. Allergen immunotherapy, based on the repeated administration of the allergen of interest, allows the modification of the patient's immune response from a TH2 to a TH1/Treg pattern and thus the reduction of allergic symptoms. This study was therefore aimed to produce and characterize the recombinant Bet v 1 (rBet v 1) allergen, for immunotherapy purpose.In this context, various analytical methods have been developed and applied in order to optimize the production of rBet v 1 via the reduction of process or product-related impurities as well as to document the quality of the purified allergen. In particular, the use of mass spectrometry has allowed the determination of the exact mass of the intact allergen and the complete verification of its amino acid sequence. Mass spectrometry data have also contributed to the detection and identification of impurities and degradation products and have therefore led to several optimizations of the industrial process for the production of the recombinant allergen. Immunological activities of certain degradation products were also investigated and the allergen tertiary structure was determined by X-ray diffraction. Finally, this study was decisive in order i) to establish rBet v 1 as a chemical reference substance for the European Pharmacopoeia as well as ii) to perform a phase II clinical study conducted in 483 patients with birch pollen-induced rhinoconjunctivitis.

Dissertation deals with three areas of modernization of reading and writing which I chose, considering them the most actual. The work is divided into theoretical and practical part. Theoretical part of the dissertation is concerned with basic terms of the given topic and is generally devoted to the theory of initiatory iteracy of reading and writing. Great emphasis is put on the field of research, especially on handwriting Comenia Script, evaluation of reading and writing on the 1st level of Elementary School and on the projects which are tematically connected to the subject matters focused particularly at reading and writing. Practical part of the dissertation compounds of quantitative research and its objective is finding out how the given areas are applied in the process of learning. This part of the research is concerning the three areas of modernization which I consider the most prevalent. The research project was accomplished by question-forms, observation and test of the readers' literacy. The work is in a way a probe, testing how Comenia Script is enforced at the 1st level of Elementary School with help of transcript and reading of the unfamiliar text. The main goal in reversing the allergy epidemic is the development of effective prophylactic strategies. Early life events, such as exposures to microbes, have a major influence on the development of balanced immune responses. Due to their ability to interact with host immune system and to modulate host immune responses probiotics, mainly bifidobacteria and lactobacilli have been used with some success in prevention of allergic disease. In order to be referred to as probiotic, bacterial strain has to undergo rigorous testing. We have selected three new Lactobacillus (L.) strains out of 24 human isolates according to their antagonistic activity against pathogenic bacteria, resistance to low pH and milieu of bile salts. Safety of these strains was proven upon intragastric administration to mice; moreover, we have shown their ability to shift cytokine Th1 - Th2 balance towards non-allergic Th1 response in isolated splenic cells. Allergen specific prophylaxis using probiotics as vehicles for mucosal delivery of recombinant allergen is an attractive concept for development of well-tolerated and effective allergy vaccines. Promise and Freedom, Flesh and Slavery: Paul's Hermeneutical Key in Galatians 4:21-5:1 in Light of the Themes and the Structure of GalatiansWallaker, Douglas R. 02 January 2018 (has links)
The story of Sarah and Hagar has been treated as one of the foundational stories for the nation of Israel. Yet, Paul, in one deft allegory, will attempt to overturn centuries of theological and historical interpretation. Against other interpretations, I argue that Paul’s untraditional uniting of Hagar, Sinai, and the present Jerusalem together was accomplished through the interpretive key provided in 4:23: the one who comes from a slave has been born because of the flesh, and the one who comes from a free woman has been born through the promise.
Paul no longer reads the story of Sarah and Hagar through a lens of separation based on physical realities, but rather through the lens of promise and flesh, as helpfully stated in 4:23. Ishmael was not “cast out” because of his sinfulness or supposed ethnic deficiencies, but rather because he was born “κατὰ σάρκα” and not “δι᾽ ἐπαγγελίας.” These comparative realities are not introduced here in the allegory for the first time; rather, Paul has been building these comparisons as his primary argument since the foundational 2:15-21. This dissertation, therefore, not only interprets the allegory in consideration of the lens of 4:23, but seeks to set the argument within both the thematic and schematic content of Galatians.
While the first chapter summarizes and categorizes the typical interpretations of the difficult Galatians 4:21-5:1, the second chapter seeks to critique these approaches to the allegory. The value of applying Paul’s interpretive lens (Gal 4:23) to the allegory is argued for in the third chapter. By using the hermeneutical lens of 4:23, many of the allegory’s difficulties are eased, and the meaning and import of the allegory is clarified. The fourth chapter seeks to set the allegory within the epistle, both thematically and schematically, centering the main propositio around Paul’s remarks to Peter in Galatians 2:15-21. Finally, the last chapter looks at some limited applications of the thesis to OT hermeneutics, specifically in literal and allegorical readings of the Sarah/Hagar incident.

La découverte d'une structure vivante de l'être qui invite à avancer toujours plus dans sa vérité a été rendue possible grâce à l'inscription du processus dans le cadre de la recherche heuristique. Par la suite, la pratique réglée de lecture appelée sémiotique a permis de construire, à partir de la péricope de Luc 5, 1–11, des significations éclairantes quant aux possibilités, pour la Bible, d'être signifiante parce que révélatrice de ('expérience humaine et de la présence de Dieu au cœur même de cette dernière.

The work examines formation of the Catholic Church's social teaching from the end of the nineteenth century until the end of the World War II. It pays attention to official documents denoting its realm as well as to the response to the programme within the Belgian milieu. On the example of La jeunesse ouvrière chrétienne the work tries to indentify foundations of the Church's social teaching along with changes, shifts and adaptations that appeared in their unique form in the JOC movement. The work also notices origins, formation and development of JOC itself. This historic and religious topic is set in wider social and cultural context. Latter part of the work is devoted to particularities in the acceptance of Church's social teaching in Czechoslovakia in between the world wars.

The thesis deals with the formation of the initial graphomotor skills in the last year of kindergarten education with connection to the system of teaching writing in the first grade of primary school. The work consists of two parts. The theoretical part is generally aimed at preschool age and a system of preschoolers preparation, especially in the formation of hygiene and work habits. It also deals with the issue of left-handedness. The second part of my thesis is based on research of practical use. The research was conducted in kindergarten schools with a focus on the health and work habits (proper grip and pen direction for right-handers and left-handers, proper seating, etc.). The attention was also paid to uppercase block letters (mirror writing) and graphomotor development (graphomotor exercises). The results are compared with theoretical part and supported by extensive appendix.

Thesis (PhD)--Stellenbosch University, 2013. / ENGLISH ABSTRACT: This dissertation is an attempt to address the problem of the relationship between Yahweh and Hope in Isaiah 5:1-7 in the light of parallelism in the book Isaiah. It is an attempt to prove that our hypothesis, that hope in the book Isaiah can be explained theologically in a special manner by means of parallelism (synonymous, antithetical and synthetical); that hope can also not only theologically be understood in concordance (synonymous and synthetical), but also in contradiction (antithetical); that Isaiah 5:1-7 can be used as a good example of antithetical parallelism and that the social, cultural and ideological texture give cause to the different accents of the theological understanding of hope in Isaiah. For this study we used the socio-rhetorical approach of Vernon Robbins.
In Chapter 1 we look more closely at the socio-rhetorical approach, we look for possible flaws and make some suggestions. For the clarification of the concepts "hope" and "parallelism" we focus on the meaning of these concepts as included in the hypothesis of this dissertation. An overview of the research of Isaiah 5:1-7 is given and how the history of interpretation has been characterised by three approaches, namely the historical-critical, literary and theological approaches. By the use of the socio-rhetorical approach we attempt to study more fully "God and Hope in Isaiah 5:1-7 in the light of parallelism in the book Isaiah".
In Chapter 2 we are focusing on the inner texture of Isaiah 5:1-7. We are looking to the following: - The repetitive texture and pattern; we are looking for the occurrence of words and phrases more than once in a unit.
- The progressive texture and pattern; there is a progressive building up from verse 1 to verse 7 in words and phrases.
- The narrational texture and pattern; Isaiah 5:1-7 has been characterised by two structural forms, namely a love song and a juridical case.
- The opening-middle-closing texture and pattern; the exposition looks as follows: - opening: vv. 1-2d;
- middle: vv. 2e-4;
- closing: vv. 5-7. - The argumentative texture and pattern; Yahweh is building up an argument. He is using the example of a vineyard and court terms to explain and build up His case.
- The sensory-aesthetic texture and pattern; the relationship between Yahweh and his people become sour and we are talking of contradicting emotions and acts, namely excitement and disappointment, pressing (of the grapes) and judgement. In Chapter 3 we are focussing on the intertexture of Isaiah 5:1-7 and its connection with hope. We are looking to the dating of Isaiah 5:1-7. The possible intertextual connections between Isaiah 5:1-7 and the other texts in Isaiah are "recitation", "recontextualization" and "thematic elaboration".
Considering that "parallelism" is part of the theme of this dissertation, we are taking antithetical parallelism as point of departure to concentrate on four focus texts, namely Isaiah 5:1-7 (main focus texts), Isaiah 27:2-6, Isaiah 40:27-31 and Isaiah 59:1-21. In terms of parallelism and dating we typify the four texts as follows: - Isaiah 5:1-7: antithetical parallelism; late post-exilic.
- Isaiah 27:2-6: late pre-exilic.
- Isaiah 40:27-31: antithetical parallelism; exilic.

In Chapter 5 we are focussing on the ideological and sacred texture of Isaiah 5:1-7 and its connection with hope. The following issues cast light on how hope must be understood in terms of the ideological texture, namely "the conflict between communities", "the ideological conflict concerning Zion theology", "the people circumstances in the exile" and "class conflict". In terms of the theme, "God and Hope in Isaiah 5:1-7 in the light of parallelism in the book Isaiah", the role of Yahweh is crucial in in the way hope must be understood. In the sacred texture of Isaiah 5:1-7 Yahweh plays the following roles: "Gardener-Viticulturist", "Creator", "King", "Judge", "Warrior" and "Husband".
In Chapter 6 we summarise our study and also give a conclusion to the study, the contribution of the dissertation to the field of study in comparison with existing research and suggestions with reference to further research.
Therefore, the answer on the question, "How can hope been theologically understood in the book Isaiah in consideration of the literary and historical textures of Isaiah 5:1-7?" is as follows:

Aangesien daar op parallelisme gefokus word met antitetiese parallelisme as die uitgangspunt, word daar 4 fokustekste behandel, naamlik: Jesaja 5:1-7 (hooffokusteks), Jesaja 27:2-6, Jesaja 40:27-31 en Jesaja 59:1-21. In terme van parallelismes en datering word dit soos volg getipeer: - Jesaja 5:1-7: antitetiese parallelisme, laat post-eksilies.

Die volgende kwessies werp lig op hoe hoop in terme van die ideologiese verband verstaan word, naamlik: "die konflik tussen gemeenskappe", "ideologiese konflik betreffende die Sionsteologie", "die volk se omstandighede in ballingskap" en "klassekonflik". In terme van die tema, "God en Hoop in Jesaja 5:1-7 aan die hand van parallelismes in die boek Jesaja", is die rol van Jahwe van deurslaggewende belang in hoe hoop verstaan word. In die teologiese verband van Jesaja 5:1-7 vertolk Jahwe die volgende rolle: "Tuinier-Wingerdboer", "Skepper", "Koning", "Regter", "Kryger" en "Man".
In Hoofstuk 6 word 'n opsomming gegee, asook 'n gevolgtrekking van die studie, die bydrae van die proefskif tot navorsing in vergelyking met bestaande navorsing en voorstelle met betrekking tot verdere navorsing.

Esta tese analisa as normas processuais aplicáveis à arbitragem. O reconhecimento da prevalência da autonomia da vontade das partes e, subsidiariamente, dos árbitros para estabelecer regras para condução do procedimento arbitral, afastou a aplicação das regras processuais do local da realização da arbitragem. A ausência de uma lei processual de regência ou da aplicação mandatória das regras processuais da lex fori possibilitou, por um lado, a harmonização da prática processual arbitral e, por outro lado, embates recorrentes sobre a forma adequada para a condução do procedimento. A ausência de uma lei de regência não significa que o processo arbitral possa ser discricionário. Existe uma pluralidade de fontes normativas que influenciam a tomada de decisão processual nas arbitragens. Propõe-se a análise destas fontes normativas à luz do direito transnacional. O método transnacional adotado inclui normas nacionais e internacionais, bem como outras fontes normativas que não se enquadram tipicamente em uma categoria tradicional, admitindo que o quadro regulatório do processo arbitral seja formado por uma confluência de fontes que operam em ordens jurídicas e planos normativos diversos. Diante dessa pluralidade de fontes normativas, defende-se a necessidade de maior normatização in concreto, incentivando que as partes e os árbitros realizem uma conferência preliminar para definir a melhor forma de conduzir o procedimento em cada arbitragem. / This thesis analyzes the procedural rules that apply to arbitration. The recognition that the parties and, subsidiarily, the arbitrators are free to establish the rules for conducting the arbitral proceedings has prevented the procedural rules of the place where the arbitration is held from being applied. The absence of a governing procedural law or the mandatory application of the procedural rules of the lex fori has, on the one hand, made the harmonization of arbitration procedure possible and, on the other, led to recurrent conflicts regarding the appropriate way to conduct an arbitration. The absence of a governing law does not mean that the arbitration procedure could be discretionary. The existence of a plurality of normative sources that influence procedural decision-making in arbitration is verified. The analysis of these normative sources in light of transnational law is proposed. The transnational method adopted includes domestic and international rules, as well as other normative sources that do not typically fit in a traditional category, allowing the regulatory framework of an arbitration proceeding to be formed by a confluence of sources that operate in different legal systems and on various normative planes. In light of this plurality of normative sources, the need for more active case management and rule-making is defended, encouraging the parties and the arbitrators to hold a preliminary conference to determine the best method for conducting the arbitration in each arbitration proceeding.
