Biometria da rádula e padrão dos fragmentos de restrição do DNA de espécies do gênero Phyllocaulis (mollusca : gatropoda : veronicellidae)

No. of bitstreams: 1 000401361-Texto+Completo-0.pdf: 1361469 bytes, checksum: 329fa0580621e6b8b6156c8d6e01f764 (MD5) Previous issue date: 2001 / The genus Phyllocaulis Colosi, 1922 has being differenciated into five species based on anatomical and morphological analysis: P. variegatus (Semper, 1885), P. tuberculosus (Martens, 1868), P. boraceiensis Thomé 1976, P. soleiformis (Orbigny, 1835) and P. gayi (Fischer, 1872). Using classification emphasis is given to the form of the penis, spathe and the penal and pedal glands. However such criteria have been proved to be insufficient for identification of more complex groups, where the differentiating characteristics are not evident. There are five forms of Phyllocaulis soleiformis, A, B, C, D and E, representing populations from distinct geographical regions. In this investigation the entire genus was studied using scanning electron microscope (SEM) to examine the mandible, lateral and rachidian teeth and the radula. This examination together with the application of statistical analysis permitted the reclassification of form C of P. soleiformis as a new species. A complementary study based on the compilation of enzyme standards obtained from fragments of fourteen restriction enzymes which were applied in the regions ITS1 of 18S rDNA. The resulting gene distance tree showed form C of P. soleiformis to be a link between the two principal groups. Therefore, the application of these different methodologies led to the discrimination of the species P. variegatus, P. tuberculosus, P. boraceiensis, and P. gayi, forms A, B, D, and E and the differentiation of form C. As a result of these findings we propose that, P. soleiformis form C be renamed Phyllocaulis renschi Thomé, 1965. / O gênero Phyllocaulis Colosi, 1922 vem sendo diferenciado em cinco espécies, segundo análises morfológicas: P. variegatus, P. tuberculosus, P. boraceiensis, P. soleiformis e P. gayi. A ênfase é dada à forma do pênis, espata, glândula peniana e glândula pediosa. Esse critério vem se mostrando insuficiente para a resolução de grupos mais complexos, onde os caracteres diferenciais não são evidentes. Em P. soleiformis adotou-se formas A, B, C, D e E para populações que apresentam variabilidade morfológica e são oriundas de regiões geográficas distintas. Nesta investigação, abrangeu-se todas as espécies do gênero Phyllocaulis analisandose as ripas da mandíbula e os dentes laterais e raquidianos da rádula, a partir de microscopia eletrônica de varredura (MEV). Esta análise juntamente com tratamentos estatísticos permitiu reclassificar a forma C de P. soleiformis como uma nova espécie. Em outra investigação determinou-se o mapa físico empregando 14 enzimas de restrição de P. variegatus, P. tuberculosus, P. boraceiensis, P. soleiformis e, para fins comparativos, Belocaulus angustipes e Sarasinula linguaeformis. Esse mapa foi obtido através dos fragmentos da região ITS1 do 18S rDNA. A árvore de distâncias gênicas apresenta a forma C de P. soleiformis como um elo de ligação entre os dois principais agrupamentos por possuir um grande número de caracteres comuns com todo os outros táxons. Isto demonstra que a forma C sofreu poucas alterações ao longo de seu processo evolutivo. As diferentes técnicas levaram à caracterização das espécies P. variegatus, P. tuberculosus, P. boraceiensis, P. soleiformis com a diferenciação da forma C. Os resultados permitem propor a revalidação do nome Phyllocaulis renschi para a forma C de P. soleiformis.
