Analysis of Lighting Schemes in Public Assembly Rooms for Improved Energy Performance

The energy use of commercial building operations is a significant contributor to the total global energy demand, and lighting is one of the largest end uses of electricity in commercial buildings. Convention Centers as commercial buildings are examples of big energy consumers, and they present a unique opportunity to showcase benefits of green building techniques due to their large size and broad range of visitors; however, the individual nature of convention centers makes it challenging to find design improvements which will be helpful in general for many convention centers. One exception to this rule of individuality is the presence of public assembly rooms, which are ubiquitous in nature and are used heavily. Traditionally, public assembly rooms are fully enclosed to allow for full control of illumination levels with artificial lighting, particularly fluorescent light. This represents a missing opportunity to utilize daylight or energy efficient artificial light to reduce energy use while enhancing human visual comfort. The objective of this research is to analyze the potential impact of the use of daylight in public assembly rooms. We present the results of a set of computer simulations performed using IES VE software. The simulations are performed as an iterative process, using as base case a north-facing public assembly room in the Tucson Convention Center, and progressively updating the lighting scheme using the software to analyze the illuminance levels for each case. The results are used to propose a lighting scheme for the north-facing public assembly room which uses 100% daylight to achieve a uniform level of illumination and meets the compliance of illumination requirements for public assembly rooms.

Le Ve siècle de Plutarque / Plutarch's fifth century

Plutarque est un auteur grec des Ier-IIe siècles ap. J.-C. dont l'œuvre immense et variée constitue, en particulier, une source essentielle de notre connaissance de la période que nous appelons le Ve siècle, "âge d'or" de la Grèce. Plutarque possède-t-il une vision clairement définie de ce siècle en tant que période homogène et, par conséquent, est-il un des promoteurs de notre conception actuelle ? L'analyse de ses sources révèle qu'il est d'abord largement tributaire de la tradition historiographique du IVe siècle. Plutarque s'attache plus aux hommes qu'aux évènements et plus à leurs comportements qu'à leurs actes. L'étude des "grands hommes" du Ve siècle doit donc permettre de répondre à la problématique posée. Or cette étude révèle justement que Plutarque ne voit pas ce siècle d'une manière uniforme mais perçoit une rupture dans les décennies 440-430. Une des causes de cette rupture serait le renouvellement du personnel politique en particulier à Athènes. / Plutarch is a Greek author of Ist-IInd centuries A.D. whose numerous and various works are in particular an essential source of our knowledge on the period called Vth century, "golden age" of Greece. Does Plutarch have a clear and definite vision of this century as a homogeneous period and, consequently, is he responsible for our current view ? The analysis of his sources reveals that he is widely dependent on the historiographical tradition of the IVth century. Plutarch is more interested in men than in events and more in their behaviours than in their acts. The study of the "great figures" of the Vth century shall thus permit the answer to the previous questions. In fact, this study reveals that Plutarch does not apprehend this century in a uniform way but perceives a change in the decades 440-430. This change is among others due to the replacement of the political leaders especially in Athens.

Efektivní výukové strategie / Effective Instructional Strategies

The main aim of this work is to clarify the concept of educational effectiveness. In the theoretical part we first of all describe different conceptualizations of effectiveness. Then we focus on the "processual effectiveness" rather than the "result effectiveness" of the learning in lien with the Czech science literature. Searching for the optimal school instruction which engage student learning is the next step. We focus on three key factors and we try to explain their principles. In our concept an effective instruction arises when students understand and adopt the instructional objectives (setting objectives), learn actively (activity) and finally they reveal the meaning in the topic (meaningfulness). The qualitative factors mentioned above were put through the time factor and thus we tried to design a model o f the effective instruction. We identify five crucial stages of a lesson: setting objectives, activating prior knowledge, understanding the topic, retention of knowledge and concluding reflection. These stages create an effective learning cycle as a part of a learning spiral. For every stage we give some examples of particular instructional strategies and methods. The Effects of Sickle Erythrocytes on Endothelial Permeability

Sickle cell anemia is a hematological disorder that is caused by a single point mutation in the beta-globin chain of hemoglobin. It results in several complications related to the small and large vessels in patients with the disease. Large vessel complications include cerebral infarcts, which are observed in children under ten years old. The mechanism behind this complication is not completely understood. It is the goal of this project to begin to understand the role sickle erythrocytes may play in causing endothelial dysfunction as a precursor to sickle related complications. The hypothesis of this work is that exposure of large vessel endothelium to sickle erythrocytes causes an increase in endothelial permeability through loosening of adherens junctions. In the first goal of this work, bovine aortic endothelial cells (BAECs) are grown on coverslips and exposed to sickle erythrocytes for 5 minutes and either immediately fixed or incubated in 30 minutes and then fixed. Immunofluorescent studies labeling VE cadherin show changes in VE cadherin dynamics, suggesting sickle erythrocytes may be involved in this observation. Next, BAECs were grown on transwell inserts and exposed to sickle erythrocytes for 5 minutes. The erythrocytes are washed off and the BAEC are incubated with 10,000 MW dextran conjugated to lucifer yellow or FITC-BSA or to determine BAEC permeability. When dextran is used as the test molecule, endothelial permeability did not show a significant change from baseline. However, when BSA is used as the test molecule, increases in endothelial permeability are observed. Explanations into the differences between the transport mechanisms of the two molecules are discussed. These experiments show changes in VE cadherin localization due to sickle erythrocyte exposure. This may cause increases in endothelial permeability and an experimental model and preliminary studies are performed. This study provides potential mechanisms to explain the changes in VE cadherin localization and provide suggestions for further studies to test the effect of sickle erythrocytes on endothelial permeability. This work provides a strong foundation for continuing studies on the effects of sickle erythrocytes on endothelial dysfunction within the confines of sickle related complications.

PROTEIN KINASE A, EXCHANGE PROTEIN ACTIVATED BY cAMP 1, AND PHOSPHODIESTERASE 4D ALL ASSOCIATE WITH VE-CADHERIN TO REGULATE ENDOTHELIAL BARRIER FUNCTION

Vascular endothelial cells (VECs) play an essential role in regulating the passage of macromolecules and cells between the blood stream and underlying tissues. The second messenger 3’, 5’ cyclic adenosine monophosphate (cAMP) regulates numerous events in VECs, including permeability. Since human VECs express several distinct cAMP-hydrolyzing phosphodiesterases (PDEs), and these are the only enzymes that catalyze the inactivation of cAMP, we investigated if selective pharmacological inhibition of PDEs could impact VEC permeability. Interestingly, we found that PDE4 inhibitors decreased human aortic VEC (HAEC) permeability and PDE4 and PDE3 inhibitors decreased human microvascular VEC (HMVEC) permeability. Consistent with a role for both protein kinase A (PKA) and exchange protein activated by cAMP (EPAC) in regulating VEC permeability, selective activators of these enzymes significantly decreased permeability. Since neither PDE4 nor PDE3 inhibitors significantly increased cAMP in these cells, our data are consistent with the idea that PDE inhibition causes small localized increases in “pools” of cAMP that regulate permeability. In order to test if PDE4 enzymes could act locally on pools of cAMP that regulated permeability, we selectively isolated the adherens junctional protein VE-cadherin from confluent monolayers of HAECs or HMVECs, and immunoblotted these isolates for cAMPeffectors and PDEs. Briefly, we found that each PKA-II, EPAC1, and a PDE4D variant, but not PDE3 enzymes, each could be isolated in VE-cadherin-based complexes from these cells. These novel findings identify PKA-II, EPAC1, and PDE4D as members of VE-cadherin-based signaling complexes in human VECs and are consistent with the idea that localized cAMP-signaling regulates permeability in these cells. / Thesis (Master, Pathology & Molecular Medicine) -- Queen's University, 2007-09-14 15:52:20.216
