La génération du stress oxydant comme stratégie thérapeutique anticancéreuse : Investigation des mécanismes d’action de la vitamine C, de l’auranofin et de leur combinaison / Generation of oxidative stress as an anticancer therapeutic strategy : Investigating the mechanism of action of vitamin C, auranofin and their combination

L’équilibre rédox entre les niveaux des espèces réactives de l’oxygène et de l’azote (ROS, RNS) et les espèces antioxydantes cellulaires est déterminant pour le fonctionnement normal de la cellule et sa viabilité. Le déséquilibre redox ou « stress oxydant » peut altérer les voies de signalisation cellulaires et générer des dommages sur les protéines, les lipides et l’ADN des cellules. Il est ainsi associé à de nombreuses pathologies, notamment les cancers. Les cellules cancéreuses présentent une dérégulation redox importante et un stress oxydant basal intrinsèque plus élevé par rapport aux cellules normales. Elles sont donc très dépendantes des systèmes antioxydants pour leur viabilité. Ainsi, l’administration de drogues qui i) génèrent des ROS / RNS additionnelles ou ii) inhibent les systèmes antioxydant cellulaires, permet une cytotoxicité sélective contre les cellules cancéreuses. C’est la base biologique de la « thérapie anticancéreuse basée sur la modulation de l’équilibre redox». Dans ce contexte, nos travaux ont pour but de décrypter les mécanismes redox derrière l’activité anticancéreuse de la vitamine C (VitC) et de l’auranofin (AUF), seuls ou en combinaison, dans le modèle du cancer du sein. La VitC à des concentrations pharmacologiques élevées présente des propriétés pro-oxydantes. Dans cette étude, l’activité anticancéreuse de la VitC contre les lignées du cancer du sein est associée à une génération extracellulaire et intracellulaire de peroxyde d'hydrogène (H₂O₂) accompagnée d'une oxydation intracellulaire du glutathion (GSH). L’approche protéomique «redoxome» a révélé que la VitC induit une altération de l'état rédox d’enzymes antioxydantes clés et d'un certain nombre de protéines contenant des cystéines, impliquées dans les métabolismes de l’ARN et l’ADN et dans les processus énergétiques. La VitC est également responsable d’un retard dans la progression du cycle cellulaire et d’une inhibition de la traduction. Finalement, des analyses bioinformatiques ont montré que les niveaux d'expression de la peroxiredoxine 1 (PRDX1) sont corrélés à la cytotoxicité différentielle de la VitC dans les cellules cancéreuses du sein. L'AUF, un antirhumatismal, est un inhibiteur des thiorédoxines réductases qui a reçu une attention croissante pour son activité anticancéreuse. Nos travaux montrent que l’AUF inhibe également le système antioxydant du GSH et que cette inhibition est primordiale pour son activité anticancéreuse. L’AUF modifie l'état redox de nombreuses protéines impliquées dans la prolifération et le cycle cellulaire, et provoque une déplétion des dNTPs et un arrêt du cycle cellulaire. De façon remarquable, nous avons démontré que la combinaison de l’AUF et de la VitC présente une cytotoxicité accrue, synergique, médiée par H₂O₂ dans les cellules MDA-MB-231 et d'autres lignées cellulaires du cancer du sein sans trop affecter les cellules normales. In vivo, l’efficacité de la combinaison AUF/VitC a été validée sur des xénogreffes de MDA-MB-231 chez les souris sans présenter une toxicité notable, tandis que l'administration de l’AUF ou de la VitC en monothérapie n’inhibe pas la croissance tumorale. Enfin, les analyses protéomiques, bioinformatiques et fonctionnelles ont identifié la prostaglandine réductase 1 (PTGR1) comme biomarqueur prédictif de la réponse des cellules cancéreuses du sein à la combinaison AUF/VitC. En résumé, ces résultats contribuent à une meilleure compréhension des mécanismes anticancéreux de la VitC et de l'AUF, seuls et en combinaison. En particulier, la combinaison de ces deux médicaments disponibles et non toxiques pourrait être efficace contre le cancer du sein triple négatif et potentiellement d'autres cancers présentant des propriétés redox similaires. Ainsi, une évaluation préclinique et clinique de ces traitements ouvrira la voie à des nouvelles thérapies anticancéreuses basées sur la modulation de l’équilibre redox cellulaire. / Reactive oxygen and nitrogen species (ROS, RNS) homeostasis and intracellular reductive/oxidative (redox) dynamics play a key role in regulating cell fate and are critical for normal cellular functions. Oxidative stress via the disruption of redox homeostasis can lead to aberrant cell signaling and toxic oxidative damage of DNA, lipids and proteins, and is therefore associated with human pathologies such as cancers. Cancer cells experience extensive redox deregulation and generally exhibit higher intrinsic basal oxidative stress than normal cells, as a consequence, they are more dependent on their antioxidant systems for survival. Thus, the administration of a drug generating additional ROS / RNS or inhibiting cellular antioxidant systems will exert a selective cytotoxicity towards cancer cells while sparing their normal counterparts. This is the biological basis for « redox-based anticancer therapy ». The work described here aims to investigate the redox-based anticancer activity of vitamin C (VitC) and auranofin (AUF), as single drugs or in combination, in breast cancer model. VitC at high pharmacological concentrations shows pro-oxidant properties. In this study, we showed that VitC anticancer activity against breast cancer cell lines was associated to extracellular and intracellular generation of hydrogen peroxide (H₂O₂), accompanied by the oxidation of intracellular glutathione (GSH). A “redoxome” proteomics approach revealed that VitC induces alterations of the redox state of key antioxidant enzymes and a number of cysteines-containing proteins including many proteins involved in RNA and DNA metabolisms and energetic processes. Cell cycle arrest and translation inhibition are associated with VitC-induced cytotoxicity. Finally, bioinformatics analysis and biological experiments identified that peroxiredoxin 1 (PRDX1) expression levels correlate with VitC differential cytotoxicity in breast cancer cells. AUF, an antirheumatic drug and known inhibitor of thioredoxin reductases, has been repurposed recently as a potent anticancer drug. We showed that AUF acts on both the thioredoxin and GSH systems and its impact on GSH system is essential for its anticancer activity. AUF alters the redox state of a number of nucleic acid-binding proteins involved in cell proliferation, cell division and cell cycle, triggering dNTP depletion and cell cycle arrest. Importantly, we observed that the combination of AUF and VitC reveals a synergetic and H₂O₂-mediated cytotoxicity towards MDA-MB-231 cells and other breast cancer cell lines without much impact on normal cells, thus decreasing the cytotoxic concentrations of AUF or VitC single drug. The anticancer potential of AUF/VitC combinations was validated in vivo on MDA-MB-231 xenografts in mice without notable side effects, while administration of AUF or VitC as a single agent failed to suppress tumor growth. Finally, SILAC proteomics, bioinformatics analysis, and functional experiments linked prostaglandin reductase 1 (PTGR1) expression levels with breast cancer cell response to AUF/VitC combination, thus identifying a potential predictive biomarker. Overall, these results provide new insights into the anticancer mechanisms of VitC and AUF, as single drugs and in combination. In particular, this combination of two non-toxic and commonly available drugs could be efficient against triple-negative breast cancer and potentially other cancers with similar redox properties. Further assessment in preclinical and clinical studies of these drugs and combinations could open new avenues for redox-based anticancer therapy.

Presented diploma thesis is dedicated to the determination of selected chemical parameters of fruits of new currant varieties. The theoretical part is focused on the characteristics of currant, the biologically active substances of its fruits and also the utilization of current in the food industry. Further part of the theory deals with the main chemical parameters which were examined: polyphenols, anthocyanins, ascorbic acid and antioxidant activity. In the practical part there were determined selected chemical parameters in six white, twelve red and thirteen black varieties of currant. Anthocyanins and polyphenols have been determined spectrophotometrically. The content of ascorbic acid in selected varieties of currant was determined by HPLC. As a technique for the determinationof the antioxidant activity was selected the method of free radical DPPH.

Les fruits et légumes constituent la principale source de vitamine C dans l’alimentation humaine, mais leur concentration en vitamine C varie fortement en fonction de la saison et des conditions de culture. Au cours de cette thèse, nous avons testé successivement différentes hypothèses afin de mettre en évidence le rôle de la lumière dans ces variations, en prenant comme modèle le fruit de tomate. Nous avons supposé i) un effet direct du rayonnement intercepté par les fruits sur le métabolisme de la vitamine C (synthèse, recyclage, dégradation), ou ii) un effet du rayonnement sur les feuilles augmentant le transport de molécules (sucres, vitamine C, …) favorisant l’accumulation de vitamine C dans le fruit. Nos résultats ont souligné la complexité de la régulation de la teneur en vitamine C dans les fruits par la lumière, qui dépend principalement du microclimat radiatif du fruit et dans une moindre mesure du rayonnement intercepté par les feuilles, en interaction avec le stade de développement du fruit. L’étude de la relation sucres/vitamine C dans les fruits a montré que les sucres n’étaient pas limitants pour la synthèse de vitamine C. L’impact du rayonnement sur le métabolisme de la vitamine C a été étudié, en interaction avec la température, sur fruits détachés. Le rayonnement augmente les teneurs en vitamine C des fruits pour les températures inférieures ou égales à 23°C en liaison avec l’augmentation des expressions des gènes de la voie de biosynthèse et des activités des enzymes de recyclage, particulièrement à 12°C. À forte température (31°C), la lumière ne modifie pas la teneur en vitamine C du fruit malgré l’augmentation de l’expression de certains gènes de la voie de synthèse, mais on observe une diminution du recyclage de la vitamine C (DHAR) et une augmentation d’un produit de dégradation de la vitamine C (thréonate). Les données recueillies ont permis d’initier un modèle d’accumulation de la vitamine C au cours du développement du fruit qui dans le futur prendra en compte les facteurs de l’environnement / Fruits and vegetables are the major source of vitamin C in human diet; however, their vitamin C content varies with environmental conditions and agricultural practices. In this work, we successively tested different hypotheses concerning light impact on these variations, using tomato fruit as model. We hypothesized that i) light reaching fruit could have a direct impact on vitamin C metabolism (synthesis, recycling and degradation) or that ii) light reaching leaves could increase the transport of molecules triggering vitamin C accumulation in fruit (sugars, vitamin C…). Our results showed that vitamin C variations with light are complex and depend mostly on light reaching the fruit and to a lesser extent on light reaching leaves,according to fruit developmental stage. The study of vitamin C/sugars relationship in fruit showed that sugars were not determinant in explaining variations in vitamin C. Light impact on vitamin C metabolism were studied, in interaction with temperature, on off-vine fruit ripening. Light increased fruit vitamin C content for temperature lower or equal to 23°C byincreasing transcripts of vitamin C biosynthetic pathway and activity of vitamin C recycling enzyme, particularly at low temperature (12°C). At high temperature (31°C), light did not increase fruit vitamin C content but it decreased DHAR activity and increased threonate content likely produced from vitamin C degradation. The data obtained were used to initiate the building of a model describing vitamin C content during fruit development that will integrate environmental factors in the future

Consommation alimentaire d’antioxydants et risque de cancer du poumon : une étude cas-témoins montréalaise

Objectif : Examiner l’association entre la consommation alimentaire de caroténoïdes (β-carotène, α-carotène, β-cryptoxanthine, lutéine/zéaxanthine, lycopène) et de vitamine C et le risque de cancer du poumon, selon le sexe, l’intensité de tabagisme et le sous-type histologique de la tumeur. Méthodes : Les données proviennent d’une étude cas-témoins menée à Montréal, Canada. Des entrevues ont été effectuées auprès de 1 105 cas incidents de cancer du poumon et 1 449 témoins issus de la population générale. Leur fréquence de consommation moyenne de 49 fruits et légumes deux ans auparavant a été convertie en apports en antioxydants. Les rapports de cotes (RC) et les intervalles de confiance (IC) à 95% caractérisant l’association entre les antioxydants et le risque de cancer du poumon ont été estimés à l’aide de modèles de régression logistique et polytomée, en tenant compte de facteurs de confusion potentiels. Résultats : Une consommation élevée en antioxydants était généralement associée à une diminution du risque de cancer du poumon de l’ordre de 30%. Un effet protecteur a été observé chez les hommes et les femmes, pour les non fumeurs, les fumeurs quelque soit l’intensité de tabagisme, ainsi que pour les carcinomes à petites cellules, épidermoïde et l’adénocarcinome. Conclusions : Plusieurs antioxydants alimentaires protégeraient du cancer du poumon. Les efforts de prévention bénéficieraient de cibler la promotion de la consommation de fruits et de légumes riches en caroténoïdes et en vitamine C. / Objective: To investigate the association between dietary intake of carotenoids (β-carotene, α-carotene, β-cryptoxanthin, lutein/zeaxanthin and lycopene) and vitamin C, and risk of lung cancer according to sex, smoking intensity and tumor histological subtype. Methods: In the course of a case-control study conducted in Montreal, Canada, in-person interviews elicited dietary data from 1,105 incident lung cancer cases and 1,449 population controls. Usual frequency of intake of 49 fruit and vegetables two years prior to diagnosis or interview was estimated and converted to antioxidant intakes. Odds ratios (OR) and 95% confidence intervals (CI) between intake variables and lung cancer were estimated using logistic and polytomous regression models, adjusting for potential confounding factors. Results: High intakes of antioxidants were generally associated with some 30% reduction in lung cancer risk. A protective effect was observed among men and women, among never smokers, smokers regardless of intensity, and for small cell carcinoma, squamous cell carcinoma and adenocarcinoma. Conclusions: Results from this study suggest several dietary antioxidants may protect against lung cancer. Prevention programs should promote increased intakes of fruit and vegetables rich in carotenoids and vitamin C.

Antioxidants as Risk Factors for Gingival Bleeding

Background: Studies of gingival bleeding and the effects of antioxidants on extracellular matrix and immunologic and inflammatory responses provide a rationale for hypothesizing that antioxidants reduce the risk for gingival bleeding.Methods: This study evaluated the role of antioxidants as contributing risk factors for gingival bleeding utilizing the Third National Health and Nutrition Examination Survey (NAHNES III). A sample of 18,825 adults (20 to ≥ 90 years of age), with dental measurement and assessment of serum levels of antioxidants were included in the study. Gingival bleeding was defined as those who had more than 30 percent of gingival bleeding in 28 sites examined. SPSS version 11.0 software and Epi-info 2000 were used to perform the statistical analysis.Results: Using multiple logistic regression in five separate antioxidants, the study showed an association between increased plasma levels of vitamin C (ascorbic acid) and decreased risk for gingival bleeding (OR= 0.33; 95% CI 0.15 to 0.72). An inverse relationship was also found between gingival bleeding and serum levels of beta carotene (OR=1.93; 95% CI 1.05 to 3.54). However, negative association was found between gingival bleeding and vitamin A (OR=2.60; 95% CI 1.04 to 6.50). No statistically significant association was observed between gingival bleeding and serum levels in vitamin E (alpha tocopherol) and selenium.Conclusion: Antioxidants, vitamin C, vitamin A, and beta carotene, were significant risk factors for gingival bleeding. This should be emphasized for improving the oral health of the U.S. adult population.

As perdas pós-colheita de alimentos hortícolas justificam a adoção de técnicas de conservação. Uma vez beneficiados, esses produtos permitem agregar valor à produção primária e se tornam de conveniência ao consumidor. Essa pesquisa teve como objetivo definir as metodologias do processamento mínimo de cenoura, pimentão e salada mista (batata e vagem), quanto ao tipo e intensidade de adoção das técnicas de barreiras e monitorar o processamento, via controle microbiológico, sensorial, físico-químico e nutricional. As hortaliças foram lavadas em água corrente, descascadas e cortadas com facas afiadas de aço inoxidável. Para a sanificação, foram imersas por 15 min em água refrigerada (± 7ºC) com 100 mg L-1 de cloro livre a pH ajustado para 7,0; a seguir foram centrifugadas a 550-900 G, por 5 min. As hortaliças foram acondicionadas, manualmente, em sacos plásticos de filme multicamada laminado, do tipo BOPP/PEBD (polipropileno biorientado/polietileno de baixa densidade). As embalagens foram fechadas em seladora modelo AP-500, marca Tec Maq, sob ar atmosférico, vácuo e atmosfera modificada (2% de O2, 10% CO2 e 88% de N2) e a seguir foram armazenados sob refrigeração a 1ºC ± 1ºC. A vitamina C não foi afetada em nenhum dos tratamentos nos três produtos. Os teores de b-caroteno mantiveram-se praticamente constantes em cenoura e salada mista; no pimentão, após a primeira semana de armazenamento, sofreram um decréscimo, permanecendo praticamente inalterados ao longo do armazenamento. Quanto à análise sensorial, os resultados para a cenoura e salada mista, foram satisfatórios até 21 dias de armazenamento para todos os tratamentos; exceto para o pimentão. Os três produtos obtiveram em todos os tratamentos, contagens para psicrotróficos variando de 10 2 a 10 5 UFC/g, 10 1 a 10 3 UFC/g e 10 3 a 10 6 UFC/g, respectivamente. Anaeróbios mesófilos e coliformes totais, foram constatados em pimentões minimamente processados. Porém, as baixas contagens obtidas, em todos os tratamentos, mostraram que a vida útil destes prolongou-se até o 14º dia de armazenamento. Para a cenoura e salada mista minimamente processadas, não foram constatados coliformes totais e fecais, anaeróbios mesófilos e Salmonella em nenhum dos tratamentos. / The postharvest losses of horticultural products justify the adoption of preservation techniques. Once these products are processed, they add value to the primary products and they become convenient to the consumer. This research had the purpose of defining the methodologies for minimal processing of carrot, green pepper and mixed salad of potato and string bean in relation to the type and intensity of adoption of the barrier techniques. In addition, it was our objective to monitor the processing, through by microbiological, sensory, physical-chemical and nutritional evaluation. The vegetables were washed in running water, peeled and cut with sharp stainless steel knives. For the sanitization they were submerged, for 15 min, in cold water (± 7ºC) with 100 mg L -1 of free chlorine at pH 7,0; next, they were centrifuged at 550-900 G, for 5 min. The product was hand packed in plastic bags BOPP/LDPE (biorientated polypropylene/low-density polyethylene). The packages were sealed under air, vacuum and modified atmosphere (2% O2, 10% CO2 and 88% N2) and stored under refrigeration at 1ºC±1ºC. The vitamin C content was not affected in any of the treatments. The values for b-carotene remained stable for carrot and mixed salad. For the green pepper, the values decreased after the first week of storage. In relation to the sensory analysis, carrot and mixed salad were considered satisfactory until 21 days of storage for all treatments; but for green pepper. In all treatments, counts for the three products ranged psychrotrophic ranging from 10 2 to 10 5 UFC g -1 , 10 1 to 10 3 UFC g -1 , 10 3 to 10 6 UFC g -1 , respectively. Anaerobic mesophiles and total coliforms were observed in minimally processed green peppers. However, the low count obtained in all treatments indicated a shelf life of 14 days. For the minimally processed carrot and mixed salad, no total or fecal coliforms, anaerobic mesophiles and Salmonella were observed in any of the treatments.

Impact of Vitamin C on Genistein-Induced Apoptosis in Prostate Cancer

This study determined the impact of vitamin C dose on genistein-induced apoptosis in LNCaP cancer cells at various treatment regimens in vitro. Although the linear regression of viability assay (MTT) indicated a p-value = 0.11; NBT assay reveal a declining SOD activity during cell death. Apoptosis induction was the main mode of treatment induced cell death. The overall data showed the trend of treatment efficacy as;(Gen 10uM + Vit C 40uM) > (Gen 30uM + Vit C 40uM) > (Gen 70uM + Vit C 40uM) > 10uM genistein > 70uM genistein. The chi-square test for comparing necrosis, apoptosis and life cells showed that Vitamin C could impact genistein-induced apoptosis in LNCaP cells (p = 0.0003).
