Apesar de Culex quinquefasciatus ser um eficiente vetor de doenças tais como a filariose linfática, febre do Nilo Ocidental e outras várias neuroviroses, poucas pesquisas sobre sua fisiologia têm sido conduzidas. Como em todos os animais ovíparos, o desenvolvimento embrionário dos mosquitos depende da degradação dos nutrientes armazenados no ovo, sendo a catepsina B uma protease que tem sido identificada e caracterizada em vários insetos como envolvida nesta função. Neste trabalho identificamos, por espectrometria de massas, duas catepsinas B de Culex quinquefasciatus, parcialmente purificadas por autoproteólise de extrato total de ovos. Segundo a anotação de suas sequências no banco de dados específico para vetores, o VectorBase, elas são enzimas parálogas e suas sequências apresentam 77% de homologia. Denominadas neste trabalho como CatB1 e CatB2, ambas são expressas simultaneamente no corpo gorduroso de todas as fêmeas vitelogênicas de nossa colônia e sua atividade pode ser detectada nos ovários vitelogênicos, sugerindo sua origem materna. A transcrição de ambas as enzimas se inicia após o repasto sanguíneo (ARS), alcançando o pico de expressão às 36 h ARS, de forma bastante semelhante à da vitelogenina. / Despite Culex quinquefasciatus be an efficient vector of diseases such as lymphatic filariasis, West Nile fever and other various neurotrophic viruses, little research on its physiology have been conducted. As in all oviparous animals, embryonic development of mosquitoes depends on the degradation of the nutrients stored in the egg. Cathepsin B is a protease that has been identified and characterized in a number of insects as involved in this function. In this work we have identified, by mass spectrometry, two cathepsins B of Culex quinquefasciatus, partially purified by self-proteolysis of total egg extract. According to the annotation of their sequences in the specific vector database, the VectorBase, they are paralogue enzymes and their sequences have 77% homology. Named in this work as CatB1 and CatB2, both are expressed simultaneously in the fat body of all vitellogenic females of our colony and its activity can be detected in vitellogenic ovaries, suggesting a maternal origin. The transcription of both enzymes starts post blood meal (PBM), reaching their peak of expression at 36 h PBM, quite similar to vitellogenin.

Insetos vetores apresentam diminuição na produção de ovos quando infectadas por plasmódios. Esse fenômeno foi demonstrado em diferentes modelos de laboratório utilizando mosquitos Aedes aegypti, Anopheles gambiae e Anopheles stephensi. Durante o desenvolvimento de Plasmodium gallinaceum ocorrer alterações em diferentes processos fisiológicos de Aedes aegypti, tais como redução nos níveis de ecdisona (precursora do hormônio 20-hidroxiecdisona) na hemolinfa e alterações nos níveis transcricionais em genes envolvidos com vitelogênese, transporte de lipídeos e resposta imune. Os dados obtidos também mostram alterações nas quantidades de proteínas hemolinfáticas minoritárias e majoritárias em fêmeas infectadas. Embora maior parte do desenvolvimento do parasita acontece no trato digestivo, outros tecidos como corpo gorduroso, ovários e hemolinfa são afetados. This thesis comprises three published papers and one manuscript, all focused on the role of juvenile hormone (JH), the JH receptor Methoprene-tolerant (Met) and its target gene Krüppel-homolog 1 (Kr-h1) in insect development and reproduction. The JH-Met-Kr-h1 pathway is critical for metamorphic transition in hemimetabolan Pyrrhocoris apterus (Hemiptera) and holometabolan Bombyx mori (Lepidoptera) but seems to be dispensable during early larval postembryonic development. The results also show that Met and its heterodimeric partner Taiman (Tai) but not Kr-h1 are critical for ovarian development and vitellogenesis in Pyrrhocoris females. In vitro, in vivo and cell-based techniques in Drosophila melanogaster have demonstrated that Met and its paralog Gce are a bona fide receptor for JH. Only Gce capable of binding JH rescued Drosophila deficient for Met and Gce proteins, and the capacity of Gce to bind JH was necessary for JH-dependent transcriptional activation by Gce and Tai.

Estrogenic Activity of Chlordecone, O,P'-DDT and O,P'-DDE in Juvenile Rainbow Trout: Induction of Vitellogenesis and Interaction With Hepatic Estrogen Binding Sites

Persistent organochlorines such as chlordecone (CD), DDT, and DDT degradation products bioaccumulate in fish and potentially impair reproduction or development via estrogenic actions. We evaluated the estrogenicity of CD, o,p'-DDT, o,p'-DDE, and p,p'-DDE injuvenile rainbow trout by assessing their potential to induce vitellogenesis; estrogen-regulated hepatic synthesis of the yolk-protein precursor, vitellogenin (Vg). In order to compare the sensitivities of various markers of estrogen stimulation, trout were injected with 17β-estradiol (0-10 mg kg-1) on days 0 and 3 and were sampled on days 3-12. Estradiol (5 mg kg-1) increased plasma Vg (2400%; 640 μg l-1), liver somatic index (200%) and hepatic cytosolic estrogen binding site levels (EBS, 300%) on day 6. These results suggested plasma Vg was the most sensitive marker of estrogen exposure. Chronic dietary exposure to CD (0.4 mg kg-1 day-1, 33 weeks) elevated plasma Vg (0.9 μg l-1), but not hepatic EBS concentrations, and resulted in relatively high hepatic CD concentrations (16 μg g-1). The in vivo estrogenicity of DDT was examined by injecting trout at 14 day intervals with single or triplicate doses of o,p'-DDT, o,p'-DDE or p,p'-DDE (0, 5, 15 or 30 mg kg-1) and monitoring vitellogenesis 14 days after the final injection. Plasma Vg and hepatic EBS concentrations were significantly elevated by o,p'-DDT and o,p'-DDE (total dose 45 and 90 mg kg-1; 23-24 μg Vg l-1) but not p,p'-DDE. Target organ doses were estimated by conducting a disposition study in which trout were injected with three doses of 14C]p,p'-DDE (30 mg kg-1), at 14 day intervals. Hepatic [14C]p,p'-DDE equivalent concentrations, 14 days after the final injection, averaged 14 μg g-1 Additionally, we evaluated the relative affinity of CD and DDT or DDE for trout hepatic EBS utilizing in vitro competitive binding assays. CD had relatively low affinity (1000-fold less than moxestrol, a synthetic estradiol) for trout hepatic EBS. o,p'-DDT and o,p'-DDE, but not p,p'-DEE also exhibited low EBS affinity (approximately 156000-fold less than moxestrol). Collectively, these results indicated that CD, o,p'-DDT and o,p'-DDE were weakly estrogenic in juvenile trout.

Caenorhabditis elegans possui quatro genes de kpcs (kex2/subtilisin-like proprotein convertases): kpc-1, kpc-2/egl-3, kpc-3/aex-5, kpc-4/bli-4. Em C. elegans, dois dos quatro polipeptídeos de vitelogenina encontrados dentro dos ovócitos, YP115 e YP88, se originam a partir de um precursor polipeptídico (VIT-6) clivado pós-traducionalmente após o motivo RGKR. Nematóides transgênicos foram produzidos com construções repórteres transcricionais de GFP. Foi verificada expressão de kpc-1 tanto em neurônios quanto em células musculares e intestinais. Esses dados, aliados aos dados da literatura para os outros genes kpc de C. elegans, sugerem o envolvimento de KPC-1 no processamento de VIT-6, que é secretada por células intestinais. Ensaios de Western-blot compararam o processamento de VIT-6 em nematóides selvagens, mutantes e knock-down por RNAi para os diferentes genes kpc. Vitellogenesis in two caryophyllidean cestodes Caryophyllaeus laticeps and Khawia sinensis, parasitizing cyprinid fishes, were examined using light(LM)and transmission electron microscopy(TEM)and cytochemical staining for glycogen. Mature vitelline folicles consist of vitelline cells at various stages of development and an interstitial tissue. Maturing and mature vitellocytes contain vitelline material in the form of single small shell globules, which fuse and give rise to the large shell globule clusters.Glycogen was present in the cytoplasm and in the nucleus of the mature vitellocytes. Small lipid droplets were found in the cytoplasm of C. laticeps. "Lamellar granules" were observed in the cytoplasm of the mature vitellocytes in K. sinensis.
