Analysis of PMCH and LEP genotypes and study of the ITM2A gene as a basis for selection of beef replacement heifers2015 December 1900 (has links)
The need for a more reliable method to select beef heifers to retain as replacement dams has become a concern in the beef industry. Two polymorphisms described in leptin (LEP), p.Arg25Cys, and pro-melanin concentrating hormone (PMCH), g.-134A>T, have already been shown to improve carcass quality in beef cattle. This study was designed to evaluate any additional advantages of these polymorphisms in terms of heifer conception and calving success and lactational milk yield measured indirectly by their calves’ early ADG while they were primarily on lactation.
A dominant effect of the dam’s PMCH T allele was observed on improved calf early ADGs in Simmental heifer dams, although not in Angus heifer dams. This effect could be useful in cow-calf operations where calves were suckling their dams for a longer period before sending the cow-calf pairs out to summer pasture. The dam LEP genotype did not show an effect on their calves’ ADG. This was assumed to be due to low body fat reserves of the heifer dams at the age of two years, allowing for only low levels of leptin. Even though heifer conception was not affected by their LEP and PMCH genotypes, it would be worth evaluating their rebreeding success in the presence of these SNPs in the future.
The Integral Membrane Protein 2A (ITM2A) was hypothesized as a candidate gene for frame size in cattle. DNA fragments from 20 cattle, matching the predicted exons of the cattle ITM2A gene, were sequenced to determine whether genetic variation existed. However, the sequence obtained based on the predicted cattle ITM2A sequences appeared to be a
pseudogene, rather than the actual cattle ITM2A gene, because exons 1, 2, 3 and 5 contained stop codons. Since frame size has been reported to be associated with the reproductive performance of beef dams and their calves’ growth characteristics, it would be useful to characterize this gene once an improved cattle genome assembly is available.

Tendo esse contexto como pano de fundo, esta dissertação discute todo o processo linguístico e linguístico-computacional de construção de ontoléxicos - constructos formais cuja natureza léxico-conceitual possibilita o desenvolvimento de investigações teóricas (estudos lexicogramaticais) e aplicadas (construção de recursos lexicográficos e para o processamento computacional de informação textual disponível na Web). Em particular, constrói um ontoléxico exploratório que entrelaça conceitos e itens lexicais do domínio léxico-conceitual da Indústria do Bordado de Ibitinga. O embasamento teórico-metodológico assenta-se nos estudos de semântica lexical (wordnets) e de frames (framenets), pura e computacional, de processamento automático de língua natural e de ontologias. / Abstract: Ibitinga, which is a Brazilian town nationally known for its outstanding position in the embroidery business, has its economy almost totally based on the production of a broad selection of embroideries. With this embroidery industry in the backdrop, this master thesis describes the whole linguistic and computational-linguistic process of construction of ontolexicons - formal constructs whose lexical-conceptual nature allows for both theoretical (lexical-grammar construction) and applied (lexicographical and ontolexical resource construction) research. In particular, the study focuses on the design and implementation of a toy ontolexicon for the Ibitinga Embroydery Industry lexical-conceptual domain. Ce travail étudie les discours sur les songes dans la philosophie spéculative du siècle des Lumières jusqu’à Diderot, en revenant préalablement aux sources du cartésianisme. Puis il s’intéresse aux songes brefs comme récits-cadres des années 1730 à 1790.Dans un premier temps, il montre que le rêve est de plus en plus étudié comme un épisode de la vie de l’esprit ; il gagne la veille et en vient à éclairer le fonctionnement de l’esprit, dans les opérations de la mémoire ou de l’imagination. C’est un fonctionnement pétri par la matière et par le monde, qui devient comme chez Denis Diderot un modèle pour aborder l’esprit et un modèle de découverte scientifique. Dans un second temps, nous nous tournons vers les songes littéraires comme récits-cadre, petits textes désinvoltes, rassemblés en série comme chez d’Argens ou Mercier. Cette forme favorise l’exploration des confins obscurs du sujet, dans le pèlerinage amoureux, la satire ou le songe philosophique : hédonisme triomphant, sentiment océanique, pulsion scopique, pulsion de mort dirigé contre les autres ou contre soi. / The Dream in the Eighteenth Century or the Testing of the Self This thesis investigates the discourse about dreams in philosophy of mind of the Enlightenment to Diderot, after an initial examination of the sources of Cartesianism. Next it proceeds to study the dream frame narratives produced in the 1730-1790 period.First, it shows that the dream is increasingly studied as an episode in the life of the mind; dreams pervade the wake, soon unfolding the operations of our mind, memory and imagination. They involve world and matter, and become a model reflecting all aspects of the mind, finally turning into a model of scientific discovery in Denis Diderot’s writings. Secondly, we turn to literary dreams as frame narratives, short flippant texts, significantly grouped together in series by Argens or by Mercier. This form favours the exploration of the dark confines of the self, in the love pilgrimage, in satirical or philosophical dreams : triumph of hedonism, Oceanic Feeling, scopic drive, drive towards death and self destruction.
