1	Functional analysis of the acsD gene for understanding cellulose biosynthesis in Gluconacetobacter xylinus Mehta, Kalpa Pravin 23 October 2012 (has links) The acsD gene is a unique gene present in the cellulose biosynthesis operon in G. xylinus. With the use of homologous recombination, the acsD gene disruption mutation was created in the G. xylinus genome. Phenotypic characterization of the acsD gene mutant was investigated with the assistance of light and electron microscopy observations, carboxymethyl cellulose alterations, and lower temperature incubation. The microscopic analysis of the cellulose ribbons secreted from the acsD gene mutant shows that the polymerization and the crystallization components in mutant cells were functional. Observations of the mutant cells after incubation with carboxymethyl cellulose and temperature changes indicate that the arrangements of the pores on the cell surface have been altered. These arrangements led to decreased cellulose secretion capacity of the mutant cells. Successful complementation was achieved by using gene expression plasmids with green fluorescence protein tag in the acsD mutant background. Anti-GFP antibodies were used to determine the in vitro localization of the protein. Using subcellular fractionation and western blotting, the AcsD protein was found to be localized in the periplasm of the cells. De vill även finnas på Windows Phone 8 och därmed stödja de tre största smartphoneplattformarna. Den här rapporten syftar till att undersöka hur en applikation kan utvecklas med hjälp av ett användarcentrerat arbetssätt för att uppnå hög användbarhet. Prototyping, card sort, användbarhetskrav är några av de metoder som tillämpats. Rapporten beskriver metoderna genom dess teori, hur de tillämpats i projektet samt dess resultat. Det grundläggande genomförandet var att samla in användarinformation för att sedan utveckla en prototyp som testades på användare. Sedan gjordes en ny prototyp utifrån resultatet från användartestet och denna process itererade fram fyra prototyper. Användbarhetsproblem från testerna samt tid för genomförande av specificerad uppgift redogörs. Resultatet är en prototyp för Windows Phone 8 som nästintill är helt färdig för publicering. Användartest är den metod som haft mest påverkan på resultatet av applikationen. Varje test har frambringat problem med användbarheten som sedan har kunnat åtgärdas för att förbättra applikationen. 3	Coffeepot for Masochists : A Study in User-Centered System Design / Kaffepanna för Masochister : En studie i Användarcentrerad Systemdesign Abbasi, Azad Ismail January 2015 (has links) This master thesis is carried out in the field of “Human-Computer interaction”, more specifically the area “User-centered system design”. The focus has been on “usability” and useful graphical user interfaces. Current theories and definitions in the field have been considered. Literature studies contain well known authors and organisations in domains mentioned above; Jakob Nielsen, Donald A Norman and International Organization for Standardization ISO to mention some. Another source for this work from which the theories and way of working have been used is the book “User-Centered System Design” written by Jan Gulliksen and Bengt Göransson. The work started with a literature study followed by looking at methods to use. The next step was to do task and user analysis which followed by the development phase. The user has been given a central role in this project and, just as recommended, also been involved through the whole cycle. A useful method to get feedback from users, in addition to interviews and workshops, has been the “Heuristic Evaluation”. The final result and conclusion shows that the user-centered system design is a powerful tool to adapt when designing and developing interactive user interface. / Detta examensarbete har utförts inom området för ”Människa-Datorinteraktion” mer specifikt ”Användarcentrerad Systemdesign”. Fokus har varit på ”användbarhet” och användbart grafiskt användargränssnitt. Aktuella teorier och definitioner har iakttagits. Litteraturstudien har omfattat välkända författare och organisationer i ovannämnda domäner; Jakob Nielsen, Donald A Norman och Internationella standardiseringsorganisationen ISO för att nämna några. En annan källa vars teorier och arbetssätt har tillämpats i detta arbete är boken ”Användarcentrerad Systemdesign” av författarna Jan Gulliksen och Bengt Göransson. Arbetet började med en litteraturstudie följd av val av lämplig metod. Nästa steg innebar att utföra uppgifts- och användaranalyser och därefter var det dags för utvecklingsfasen. Användaren har haft en central roll i detta projekt och har, precis som rekommenderat, involverats i samtliga moment. En lämplig och nyttig metod för att få återkoppling från användarna, förutom intervjuer och workshop, har varit ”Heuristisk Utvärdering”. Det slutliga resultatet och slutsatsen visar att användarcentrerad systemdesign är ett kraftfullt verktyg att nyttja när det kommer till design och utveckling av interaktivt användargränssnitt. 4	Usability guided development of a participant database system / Användbarhetsledd utveckling av ett deltagardatabassystem Falk, Joakim January 2018 (has links) This project consisted of the development of a web based participant database system to replace a spreadsheet based one for the project “Vi Ses! Språkvänner i Linköping” and the evaluation of it. The design and implementation was done iteratively using a collection of usability guidelines as well as the results of a set of user tests. User tests were also used for the evaluation of the web database system. The task during the user tests was participant matching and the main measurement taken was task completion time. The project resulted in a web database system that could fully replace the spreadsheet database system. The evaluation tests showed that this system was both faster to match participants in as well as less error prone. / Detta projekt bestod av utvecklingen och evalueringen av ett webbaserat databassystem för deltagare. Den skulle ersätta ett existerande Excel-baserat databassystem för projektet “Vi Ses! Språkvänner i Linköping”. Designen och implementationen gjordes iterativt med hjälp av användbarhetsriktlinjer och resultat från användartester. Användartester användes även för evalueringen av webbdatabassystemet. Uppgiften i användartesterna var deltagarmatchning och det huvudsakliga mätvärdet var hur lång tid uppgiften tog. Projektet resulterade i ett webbaserat databassystem som helt kunde ersätta det Excel-baserade systemet. Evalueringstesterna visade att systemet var både snabbare och mindre felbenäget att matcha deltagare i.
