Characterization of Polypeptides by Tandem Mass Spectrometry Using Complementary Fragmentation Techniques

In the growing field of proteomics identification of proteins by tandem mass spectrometry (MS/MS) is performed by matching experimental mass spectra against calculated spectra of all possible peptides in a protein database. One problem with this approach is the false-positive identifications. MS-based proteomics experiments are further affected by a rather poor efficiency typical in the range of 10-15%, implicating that only a low percentage of acquired mass spectrometric data is significantly identified and assigned a peptide sequence. In this thesis improvement in spectrum specificity is accomplished by using a combination of high-accuracy mass spectrometry and techniques that will yield complementary sequence information. Performing collision-activated dissociation (CAD) and electron capture dissociation (ECD) upon the same peptide ion will yield such complementary sequence information. Implementing this into a proteomics approach and showing the advantages of using complementary fragmentation techniques for improving peptide identification is shown. Furthermore, a novel database-independent score is introduced (S-score) based upon the maximum length of the peptide sequence tag derived from complementary use of CAD and ECD. The S-score can be used to separate poor quality spectra from good quality spectra. An-other aspect of the S-score is the development of the ‘reliable sequence tag’ which can be used to recover below threshold identifications and for a reliable backbone for de novo sequencing of peptides. A novel proteomics-grade de novo sequencing algorithm has also been developed based upon the RST, which can retrieve peptide identification with the highest reliability (>95%). Furthermore, a novel software tool for unbiased identifications of any post-translational modifications present in a peptide sample is introduced (ModifiComb). Combining all the tools described in this thesis increases the identification specificity (>30 times), recovers false-negative identifications and increases the overall efficiency of proteomics experiements to above 40%. Currently one of the highest achieved in large-scale proteomics.

Development of Electroacoustic Sensors for Biomolecular Interaction Analysis

Biomolecular interaction analysis to determine the kinetics and affinity between interacting partners is important for the fundamental understanding of biology, as well as for the development of new pharmaceutical substances. A quartz crystal microbalance instrument suitable for kinetics and affinity analyses of interaction events was developed. The functionality of the sensor system was demonstrated by development of an assay for relative affinity determination of lectin-carbohydrate interactions. Sensor surfaces allowing for effective immobilization of one interacting partner is a key functionality of a biosensor. Here, three different surfaces and immobilization methods were studied. First, optimized preparation conditions for sensor surfaces based on carboxyl-terminated self assembled monolayers were developed and were demonstrated to provide highly functional biosensor surfaces with low non-specific binding. Second, a method allowing for immobilization of very acidic biomolecules based on the use of an electric field was developed and evaluated. The electric field made it possible to immobilize the highly acidic C-peptide on a carboxylated surface. Third, a method for antibody immobilization on a carboxyl surface was optimized and the influence of immobilization pH on the immobilization level and antigen binding capacity was thoroughly assessed. The method showed high reproducibility for a set of antibodies and allowed for antibody immobilization also at low pH. Three broadly different strategies to increase the sensitivity of electroacoustic sensors were explored. A QCM sensor with small resonator electrodes and reduced flow cell dimensions was demonstrated to improve the mass transport rate to the sensor surface. The use of polymers on QCM sensor surfaces to enhance the sensor response was shown to increase the response of an antibody-antigen model system more than ten-fold. Moreover, the application of high frequency thin film bulk acoustic resonators for biosensing was evaluated with respect to sensing range from the surface. The linear detection range of the thin film resonator was determined to be more than sufficient for biosensor applications involving, for instance, antibody-antigen interactions. Finally, a setup for combined frequency and resistance measurements was developed and was found to provide time resolved data suitable for kinetics determination. / wisenet

Mass Spectrometric Applications for Diagnosing Metabolic and Endocrine Diseases

Disease-specific compounds (biomarkers) are analyzed in clinical laboratories to assist with diagnosing diseases. This thesis describes development and validation of liquid chromatography tandem mass spectrometry (LC-MS/MS) based tests for diagnosing a diverse group of endocrine and metabolic diseases. The analytical methods used on-line and off-line sample extraction and analytical derivatization as means of enhancing the analytical sensitivity, specificity and clinical utility. All developed methods were extensively validated and reference intervals for the biomarker concentrations were established in blood samples of healthy adults and children. Advantages of the LC-MS/MS as an analytical technique include possibility of simultaneous measurement of multiple analytes and ability of confirming their identity. In this thesis we proposed and evaluated approaches for the assessment of the specificity of analysis in the methods that use tandem mass spectrometry detection. To enhance throughput of the LC-MS/MS tests for the biomarkers that have endogenous or exogenous isomers an approach was developed for quantitation of isomers from unresolved chromatographic peaks. Using methods developed in this thesis we performed a study of the steroidogenesis in ovarian follicles of healthy women and women with polycystic ovary syndrome (PCOS). Obtained data on the steroid concentrations and associations between the steroid metabolites in the pathway would be helpful for better understanding of the ovarian pathophysiology. Potential biomarkers of PCOS were identified in the thesis; further studies will be necessary to confirm their clinical utility.

Database for targeted drug screening with Liquid Chromatography - Time-Of-Flight Mass Spectrometry, (LC-TOFMS)

Today there are no fully general analytical techniques available for detection and confirmation of known and unknown substances in toxicological screening, further tools are therefore needed. The development of mass spectrometry with time-of-flight (TOF) detection is promising but there are still areas to be further developed and evaluated, both instrumentation and applications. During 2009 The National Board of Forensic Medicine-Department of Forensic Genetics and Forensic Toxicology, (RMV) started cooperation with the instrumentation company Waters (Manchester, UK) and the Department of Clinical Pharmacology (KI, Solna) evaluating a new TOF-instrument for toxicological screening. My assignment as a part of this project has been to create a limited and relevant database of drugs and toxics in Excel, including monoisotopic mass, used when screening for pharmaceutical substances and their metabolites most probable to be found in Swedish autopsy material. A limited database has been developed based on information from several sources, it ended up in 875 analytes and metabolites. A limited but complete database is more reliable in practise than a big database, by means of a lower frequency of isobars and more information included (e.g. retention time from liquid chromatography) making analysis faster. Commercial databases are generally theoretical, lacking information about for example retention time that often is an important criterion for identification.

Conditioning of chromatographic systems prior to metabolomic studies : Investigation of the conditioning effect and the possibility to alter it

The conditioning effect in metabolomic studies is the phenomenon of initial variation of analytical results in the first 5-10 injections of a biological sample in chromatographic systems. The deviation manifests itself as a drift in retention time, peak area and in multivariate analysis. It is a major quality assurance problem in the metabolomic field and if not accounted for would result in high analytical variance. The aim of this study was to investigate the conditioning effect and to gain further knowledge about it. The study was carried out on UPLC of hydrophilic liquid chromatography (HILIC) type coupled to quadrupole time of flight (QTOF) MS. A systematic study was designed to investigate the effects of the age of the analytical column. An investigation into certain matrix components as a possible cause of the conditioning effect was made. Different sample preparation methods were investigated. One result showed that no conditioning could be seen and the system appeared stable from the first injection. Differences in sample composition between samples with conditioning effect and samples without conditioning effect were investigated. No correlation between conditioning effect and levels of certain matrix compounds could be found. More studies of correlation between sample composition and the amount of conditioning occurring is needed. Some samples appear to have no retention time drift but have a significant drift in peak area and in multivariate analysis. This is an indication that the conditioning effect should be analysed in more ways than one before determining if a system is stable.

Electrokinetic devices from polymeric materials

There are multiple applications for polymers: our bodies are built of them, plastic bags and boxes used for storage are composed of them, as are the shells for electronics, TVs, computers, clothes etc. Many polymers are cheap, and easy to manufacture and process which make them suitable for disposable systems. The choice of polymer to construct an object will therefore highly influence the properties of the object itself. The focus of this thesis is the application of commonly used polymers to solve some challenges regarding integration of electrodes in electrokinetic devices and 3D printing. The first part of this thesis regards electrokinetic systems and the electrodes’ impact on the system. Electrokinetic systems require Faradaic (electrochemical) reactions at the electrodes to maintain an electric field in an electrolyte. The electrochemical reactions at the electrodes allow electron-to-ion transduction at the electrode-electrolyte interface, necessary to drive a current at the applied potential through the system, which thereby either cause flow (electroosmosis) or separation (electrophoresis). These electrochemical reactions at the electrodes, such as water electrolysis, are usually problematic in analytical systems and systems applied in biology. One solution to reduce the impact of water electrolysis is by replacing metal electrodes with electrochemically active polymers, e.g. poly(3,4-ethylenedioxythiophene) (PEDOT). Paper 1 demonstrates that PEDOT electrodes can replace platinum electrodes in a gel electrophoretic setup. Paper 2 reports an all-plastic, planar, flexible electroosmotic pump which continuously transports water from one side to the other using potentials as low as 0.3 V. This electroosmotic pump was further developed in paper 3, where it was made into a compact and modular setup, compatible with commercial microfluidic devices. We demonstrated that the pump could maintain an alternating flow for at least 96 h, with a sufficient flow of cell medium to keep cells alive for the same period of time. The second part of the thesis describes the use of 3D printers for manufacturing prototypes and the material requirements for 3D printing. Protruding and over-hanging structures are more challenging to print using a 3D printer and usually require supporting material during the printing process. In paper 4, we showed that polyethylene glycol (PEG), in combination with a carbonate-based plasticizer, functions well as a 3D printable sacrificial template material. PEG2000 with between 20 and 30 wt% dimethyl carbonate or propylene carbonate have good shear-thinning rheology, mechanical and chemical stability, and water solubility, which are advantageous for a supporting material used in 3D printing. The advances presented in this thesis have solved some of the challenges regarding electrokinetic systems and prototype manufacturing. Hopefully this will contribute to the development of robust, disposable, low-cost, and autonomous electrokinetic devices. / Polymera material finns överallt omkring oss; våra kroppar är uppbyggda av dem,plastpåsarna och burkarna vi förvarar vår mat av består av dem, våra kläder och andra tingsom finns i vår vardag är uppbyggda av olika typer av polymerer. Beroende på hur rapporten ser ut såförändras utseendet av mönstret. Hos en polymer påverkar sammansättningen av denrepetitiva gruppen (rapporten) egenskaperna av materialet och polymerer kan vara allt frånhårda och robusta, till flexibla och elektriskt ledande. Arbetet som presenteras i den häravhandlingen berör hur funktionen av olika system påverkas av att man använder sig avpolymerer istället för konventionella material. Första delen av avhandlingen handlar om integrering av elektronik i system som innehållervätska. När vätskor, laddade partiklar, molekyler och joner rör på sig på grund av ett yttreelektriskt fält, så kallas detta för elektrokinetik. Detta kan användas för att pumpa vätska ikanaler som är mindre än 0.2 mm, genom så kallad elektroosmos, samtidigt kommermolekyler med olika laddning att börja separera, så kallad elektrofores. Elektroosmos användsinom t.ex. analytisk kemi för injektion och transport av vätskor. Elektrofores används inombl.a. rättsvetenskap och molekylärbiologi för att separera makromolekyler, så som DNA ochproteiner, med avseende på deras storlek och laddning. I dessa system använder man sig oftastav metallelektroder. När en spänning läggs till ett par metallelektroder som är i kontakt med vatten kommer denhuvudsakliga reaktionen att vara spjälkning av vatten, så kallad vattenelektrolys. Spjälkningav vatten innebär att det bildas vät-och syrgas samt att pH börjar ändras. Gaserna som bildaskan bryta kopplingen mellan elektroderna och därmed stoppar strömmen, så som sker när mandrar ut sladden för t.ex. en elvisp. Förändringar i pH kan t.ex. påverka biologiska provernegativt, så som proteiners funktion och kan leda till celldöd, men kan också minska flödenaen elektroosmotisk pump kan generera. Det finns flera olika sätt hur man kan hanteravattenelektrolys i system med metallelektroder, så som användning av en pH-buffer. Arbetet iden här avhandlingen visar vad som händer om man ersätter metallelektroder med elektrisktledande plastelektroder. I detta fall har metallelektroderna ersatts av den elektriskt ledandepolymeren PEDOT vilket resulterar i att , där man istället för generera gas och pHviförändringar, så förflyttar man joner mellan elektroden och omgivande lösning. Andra delen av avhandlingen handlar om 3D skrivare och hur materialval påverkarutskriften och designen. 3D skrivare är ett bra alternativ för att snabbt och billigt kunnaproducera prototyper och funktionella individanpassade objekt i varierande storlekar.3D skrivare kan beskrivas som en avancerad spritsmaskin där material läggs lager på lager föratt bygga upp det slutgiltiga objektet utifrån en datorgenererade 3D model. Detta förändrarhelt hur man designar objekt och vilka möjliga strukturer och material man kan använda sigav jämfört än då man till exempel använder sig av svarv eller fräs för tillverkning. Det finnsflera olika typer av 3D skrivare, t.ex. smältplastskrivare (den typ som man kan se i flertaletaffärer idag) och den variant som använts i den här avhandlingen, en sprutbaserad. Ensprutbaserad 3D skrivare kan hantera många olika typer av material så länge dessa kan fyllas ien spruta och tryckas ut genom en nål. Det färdiga resultat kan därmed bli mycket olikaberoende på vilka material som använts. Överhängande och utstickande strukturer kan vara komplicerade att skriva ut med en3D skrivare. Utskrift av dessa strukturer kan underlättas genom att man skriver ut en temporärstruktur i ett annat material, ett offermaterial. Offermaterialet fungerar som en mall eller stödtill det slutgiltiga objektet och tas bort (offras) när övriga delar av objektet är klara. I den häravhandlingen beskrivs hur ett offermaterial baserat på polyetylen glykol (PEG, vanligtförekommande i t.ex. schampo och läkemedel) och en mjukgörare kan anpassas för attfungera tillsammans med en sprutbaserad 3D skrivare (se papper 4) för att skriva ut strukturerfrån 0,2 mm och uppåt. Arbetet i den här avhandlingen visar användningen av den ledande polymeren PEDOT i ettelektroforessystem och en elektroosmotisk pump. Detta kan förhoppningsvis underlättautvecklingen av dessa system till att bli mindre, smidigare, snabbare och billigare. Den andradelen presenterar ett vattenlösligt, PEG-baserat material som kan användas som stöd till andramaterial i sprutbaserade 3D utskrifter för att underlätta tillverkningen av 3D utskrivna objekt.

Porphyrins and heme in microorganisms : Porphyrin content and its relation to phototherapy and antimicrobial treatments in vivo and in vitro

One of the greatest threats to human health is increasing antimicrobial resistance among pathogens, and finding alternatives for treatment of bacterial infections is of highest importance together with a more controlled use of antibiotics. Porphyrins and heme have both been shown to be a promising class of compounds for inactivation of bacteria; porphyrins by their excellent properties to act as a photosensitizer, and heme by its importance as an iron source during a bacterial infection in vertebrates. This thesis describes the development of analytical methods for the identification and determination of porphyrins and heme using liquid chromatography coupled to tandem mass spectrometry. Subsequently, these developed methods were applied to bacterial samples to investigate different culture conditions and additives effect to the intracellular porphyrin and heme composition. Singlet oxygen production of three naturally occurring porphyrins have been determined together with the photosensitivity for blue light and the porphyrin content in E. coli. Toothbrushes equipped with a LED, emitting light with a wavelength of 450 nm, were used in an eight week randomized clinical trial to investigate any positive periodontal effect of blue light. Porphyrin and heme content in Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis were highly affected by the different cultivation conditions. The culture age of A. actinomycetemcomitans affected the porphyrin profile, while only small changes were observed for P. gingivalis during growth. A large change of the porphyrin profile could be observed when the bacteria were passaged onto a new growth medium. Additional porphyrins were detected and the total porphyrin content increased up to 28 times. These findings highlight the need for more standardized cultivation procedures when performing in vitro experiments. Heme content in Escherichia coli was affected when different additives related to biosynthesis of heme were added to the growth medium. The uptake of heme could be reduced with 52% when a compound that chemically looks similar to heme was added to the growth medium. Since heme acquisition is important for many pathogens, this could be a promising target for antimicrobial drugs. E. coli showed no sensitivity for 405 nm light using light doses up to 172.8 J/cm2 and only low concentrations of porphyrins could be quantified. By adding a porphyrin precursor to E. coli the intracellular concentration of porphyrins increased remarkably and a light dose of 57.6 J/cm2 reduced the bacterial number with &gt; 5 log10 steps. This shows that E. coli can be killed due to their endogenous porphyrins. In the clinical study we could see a weak trend that the 450 nm LED toothbrush possessed a phototherapeutic effect for three clinical indices. All indices were decreased in the intervention group, but there were no statistically significant difference compared to the control group. However, four inflammation markers were significantly decreased in the intervention group while only one decreased significantly in the control group. In conclusion, this thesis has shown that porphyrins and heme are produced endogenously in microorganisms and that the porphyrin profiles vary depending on culture conditions and different additives. Furthermore, porphyrins may be used as endogenous photosensitizers to inactivate bacteria, but more research is necessary to determine if there is a specific porphyrin that contributes more to the photosensitivity. / <p>At the time of the doctoral defense, the following papers were unpublished and had a status as follows: Paper 4: Manuscript. Development of a multi-collector inductively coupled plasma massspectrometry method for measurement of stable sulphur isotope ratios in aerosol sulphate

Sulphur stable isotope ratios are useful tracers in geological and environmental studies. They can for example be used to trace the origin of atmospheric sulphate aerosols, because anthropogenic sulphate and natural sulphate have distinguishable δ34S-values (δ34S value of approximately +0 to +8 ‰ for anthropogenic and approximately +12 to +19 ‰ for natural). This is useful for climate modelling research, due to the net cooling effect of aerosol sulphate. In the present study a Nu Plasma II (Nu Instruments) multi collector inductively coupled plasma mass spectrometer (MC-ICP-MS) method for measuring stable sulphur isotope ratios in low sulphur content samples, such as sulphate aerosols, was developed. The method was then applied to a sulphate aerosol sample collected in the Maldives.Most of the measurements were performed at high resolution, due to the interferences on 33S. Heated spray chamber coupled to a desolvating membrane, Aridus II (Cetac), increased the sensitivity and reduced interferences notably compared to wet plasma mode. Aridus II gave more stable measurements than DSN-100 (Nu Instruments). Determinations of δ34S for IAEA S1, S3, and S4 were accurate and the determined δ34S-value of the CIT #39 seawater standard (21.05 ± 0.36 ‰, 2SD, n=42) was comparable with published data. In general, Si internal standardization correction increased precision ~2.5 times compared to non-corrected values. The δ34S-value for the sulphate aerosol sample was determined to 3.82 ±0.41 ‰ (2SD, n=40). Repeatability of ~62 nmol introduced sulphur (2 μg/mL) was generally 0.15 ‰ (2SD, n=5) for the SW and 0.19 ‰ (2SD, n=5) for the sulphate aerosol sample. Comparable results for the SW (20.61±0.09 ‰, 2SD, n=4) and sulphate aersosol sample (3.77 ± 0.08 ‰, 2SD, n=8) were obtained with the method applied to Neptune Plus (Thermo Fischer Scientific) MC-ICP-MS in a different laboratory. The determined aerosol sulphate δ34S-value indicated that the sampled sulphate aerosol originated from anthropogenic sources.

Analytical Methods for High Molecular Weight UV Stabilizers / Analytiska metoder för UV-stabilisatorer med hög molekylvikt

Utomhusprodukter gjorda i plast är sårbara för nedbrytning på grund av UV-energi från solljus. För att stoppa denna fotonedbrytning används UV-stabilisatorer i plastmaterial; en av huvudkategorierna är hindrade aminljusstabilisatorer (HALS, hindered amine light stabilizers). Detta projekt ämnade att utveckla analytiska metoder passande för polymera HALS-föreningar som används i polyolefin-produkter. Huvudfokuset låg på vätskekromatografi med antingen UV eller masspektrometri (MS) detektion. Identifiering lyckades till viss del, extraktion misslyckades, och det gjordes inga kvantifieringsförsök. Detta examensarbete utfördes som ett samarbete mellan KTH och företaget Norner AS. På grund av detta har detaljerna såsom vilka HALS som användes, hur sammansättningen av elueringsgradienter förändrades och masspektrum från MS analys tagits bort eller ändrats till generiska termer. Högpresterande vätskekromatografi (HPLC, high-performance liquid chromatography) med metoden normalfas (normal phase) ansågs vara ett otillfredsställande alternativ på grund av begränsningen av lämpliga lösningsmedel. Både för HPLC och LC-MS-analys användes omvänd fas (reverse phase) med en C18 50 mm kolonn. Gällande HPLC-analys visade sig en våglängd på 240 nm för UV-detektion vara lämplig. För MS-detektion användes positiv elektrosprayjonisering på grund av föreningarnas basiska pKa-värden. Elueringsgradienten med de mest tillfredsställande resultaten var gradientversion 5, med vatten som lösningsmedel A och metanol som lösningsmedel B. En variant av denna gradient användes för LC-MS; gradientversion 5 mod följer samma mönster men med vatten + Y vol% syra som lösningsmedel A, för att möjliggöra jonisering. Additiv UV 2 har låg löslighet i polära lösningsmedel, men genom förtvålning kunde additiv UV 2 lösas i metanol. Med gradientversion 5 mod har förtvålad additiv UV 2 en retentionstid på 1,2 minuter i både HPLC och LC-MS-analys. För HPLC-analys av additiv UV 3 användes gradientversion 5 som resulterade med en retentionstid på 16,8 minuter, och för LC-MS med gradientversion 5 mod 16 minuter. Analys av additiv UV 5 med HPLC gav en retentionstid på 2 minuter med gradientversion 5, och för LC-MS-analys med gradientversion 5 mod var retentionstiden 1,3 minuter. På grund av inkonsekventa resultat mellan HPLC-analyser eller mellan HPLC och LC-MS-resultat kunde inte retentionstid för identifiering av additiven UV 1, UV 4 och UV 6 bestämmas. Extraktion av polymerprover med olika HALS gjordes i toluen vid 80 °C under en timme. I HPLC var det ingen skillnad i kromatogrammen mellan de olika HALS-produkterna. Resultaten från LC-MS stödde resultaten från HPLC, så slutsatsen att extraktionsmetoden inte var framgångsrik drogs. En anledning till detta kan vara att detektionsgränsen i MS och UV överstiger den extraherade halten av HALS. / Outdoor plastic products are vulnerable to degradation due to UV energy from sunlight. To hinder this photodegradation UV stabilizers are used in plastic materials; one of the main categories is hindered amine light stabilizers (HALS). This project aimed to develop analytical methods suitable for polymeric HALS used in polyolefin products. Liquid chromatography was the primary analytical method, with either UV or mass spectrometry (MS) detection. Identification of HALS was partially successful, the attempted extraction was unsuccessful, and there was no attempt at quantification. This thesis was performed as a collaboration between KTH and the company Norner AS. Due to this, the details such as which HALS were used, how the composition of elution gradients changed, and the mass spectrum from MS analysis have been removed or changed to generic terms. The high-performance liquid chromatography (HPLC) method normal phase was deemed as an unsatisfactory option due to the limitation of appropriate solvents. For HPLC and LC-MS analysis reversed phase with a C18 50 mm column was used. In HPLC analysis, a wavelength of 240 nm for UV detection was found to be suitable. For MS detection, positive electrospray ionization was used due to the compounds' basic pKa values. The elution gradient with the most satisfactory results was gradient version 5, with water as solvent A and methanol as solvent B. A variant of this gradient was used for LC-MS; gradient version 5 mod follows the same pattern but with water + Y vol% acid as solvent A, to enable ionization. UV 2 has low solubility in polar solvents, but through saponification UV 2 could be dissolved in methanol. With gradient version 5 mod, saponified UV 2 has a retention time of 1.2 minutes in both HPLC and LC-MS analysis. For HPLC analysis of additive UV 3, gradient version 5 was used which resulted in a retention time of 16.8 minutes, and for LC-MS with gradient version 5 mod 16 minutes. Analysis of UV 5 with HPLC gave a retention time of 2 minutes with gradient version 5, and for LC-MS analysis with gradient version 5 mod the retention time was 1.3 minutes. Due to inconsistency in results between HPLC analysis or between HPLC and LC-MS results, retention time for identification of additives UV 1, UV 4, and UV 6 could not be stated.  Extraction of polymer samples with different HALS was done in toluene at 80 ॰C for one hour. In HPLC, there was no difference in the chromatograms between the different HALS compounds. The results from LC-MS supported the results from HPLC, thus it was concluded that the extraction method was not successful. A reason for this could be that the detection limit in MS and UV might exceed the extracted amounts of the compounds.

The purpose of this thesis work was to develop a headspace solid phase microextraction gas chromatography mass spectrometry (HS-SPME-GC-MS) method to detect volatile organic compounds (VOCs) in board samples and to statistically investigate potential correlation between chromatographic data and flavor data obtained from a trained panel. The developed method would hopefully serve as a complement to the already established routine analyses at Stora Enso and gain an increased understanding of which VOCs in the board influence its flavor properties. The impact of incubation time and adsorption time on the area under curve (AUC) was studied with a Design of Experiment screening using the software MODDE. The screening data showed a correlation between large AUC and low repeatability measured as relative standard deviation (RSD). The data was hard to fit to a model due to the large RSD values for the replicates, AUC for identified compounds as response gave an acceptable fit. The regression coefficients for the model showed that a longer adsorption time gave larger AUC, while incubation time had no significant impact on the response.  Instead of following up the screening with an optimization, the focus was shifted to improving the repeatability of the method, i.e. lowering the RSD. The high RSD was believed to mainly be the result of leakage of analytes and unstable temperature during adsorption, preventing the system from reaching equilibrium. Different heating options and capping options for the vial was tested. Septum in crimp cap ensured a gas tight seal for the vial, giving lower RSD values and larger AUC compared to the other alternatives, showing that there was indeed a leakage. Using oil bath ensured stable temperature during the adsorption and detection of a larger number of VOCs but created a temperature gradient in the vial due to it not being fully submerged in the oil. Oil bath gave larger AUC, but still high RSD due to the temperature gradient making the method sensitive to variance in fiber depth in the vial. The final method was performed with 2 g of board sample in a 20 ml headspace vial sealed with a crimp cap with septa. The incubation and adsorption were performed with the vial immersed in a 90-degree oil bath. 20 min incubation time was chosen based on the time it took to get a stable temperature gradient in the vial, and 20 minutes adsorption time was chosen as a good compromise between large AUC and low RSD. Compared to Stora Ensos routine analysis, the developed SPME method gave chromatograms with an improved signal-to-noise ratio for the base line and several more peaks with larger AUC. For the board sample used during method development, the SPME-method identified 34 VOCs, while the routine analysis only identified 12. The developed method was applied on 11 archived board samples of the same quality that were selected based on their original flavor properties, to get a large diversity of samples. Flavor analysis was performed by letting a trained flavor panel describe the flavor based on intensity and character of the water that had individually been in indirect contact with one of the 11 board sample for 24 h. Potential correlation between chromatographic data obtained with the developed method and the flavor experience described by the flavor panelists was statistically investigated with the multivariate analysis software SIMCA. The correlation study showed that a combination of 12 VOCs with short retention time are most likely the main source of off-flavor which of 5 could only be identified with the developed SPME method. VOCs with long retention time did not contribute to an off-flavor and might have a masking effect on flavor given by other VOCS, however not confirmed in this study. Furthermore, the age of the board samples proved to be a good indicator for prediction of the flavor intensity, whereas the total AUC of the samples was not. Possible correlation between detected VOCs in the samples and flavor character given by the flavor panel were seen, however the variation in the data and the sample set were too small, preventing from making conclusions on individual VOCs impact on the flavor experience. The developed HS-SPME-GC-MS method would serve as a complement to the already established routine analyses at Stora Enso and has slightly increased the understanding of which VOCs in the board influence the flavor properties
