A phytochemical and pharmacological study of ten Commiphora species indigenous to South Africa

Commiphora species (from which myrrh is obtained) has been a source of several novel and bio-active natural compounds. Traditionally, Commiphora (Burseraceae) is used in southern Africa for the treatment of ulcers, fevers, and as a remedy for snake and scorpion bites. In western Africa, the macerated stem is used in the treatment of rheumatic conditions. The resin of some Commiphora species is applied topically to aid in wound healing. Documented uses include antibacterial and antifungal properties, as well as cytotoxic, cytostatic and anti-oxidant activity. The botanical diversity of this genus in South Africa warrants a study of this plant group, to provide scientific evidence for the traditional use of Commiphora species in African healing rites. Ten Commiphora species were investigated. Fresh plant material of the selected species were identified and collected from natural populations in the Limpopo Province. Active compounds, viz. kaempferol and dihydrokaempferol, in C. glandulosa (stem) were isolated using bioassay-guided fractionation and identified using nuclear magnetic resonance spectroscopy. The stem and leaf extracts of each species were analysed for in vitro anti-oxidant, antimicrobial, anti-inflammatory, anticancer activity, as well as cytotoxicity. The anti-oxidant activity of the extracts was investigated using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and the 2,2’-azino-bis(3-ethyl-benzthiazoline-6-sulfonic acid) (ABTS) assays. Extracts generally exhibited poor anti-oxidant activity in the DPPH assay, with the exception of C. schimperi (stem), C. neglecta (stem), C. tenuipetiolata (stem and leaf), and C. edulis (stem), which possessed IC50 values ranging between 7.31 μg/ml and 10.81 μg/ml. Isolated compounds were subjected to the DPPH assay to determine the anti-oxidant potential of each compound, separately and in combination to establish possible synergistic, antagonistic or additive effects. The flavonol, kaempferol (IC50 = 3.32 μg/ml) showed exceptional radical scavenging activity, in contrast to the low activity displayed by dihydrokaempferol (IC50 = 301.57 μg/ml), their combination being antagonistic. Greater anti-oxidant activity was observed for most species in the ABTS assay when compared to the results obtained in the DPPH assay. The best activity was observed for the stem extracts of C. neglecta (IC50 = 7.28 μg/ml) and C. mollis (IC50 = 8.82 μg/ml). In vitro antimicrobial efficacy was determined against Gram-positive and Gram-negative bacteria as well as yeasts using the MIC microtiter plate assay. A greater selectivity was exhibited by the extracts against the Gram-positive bacteria and yeast than against the Gram-negative bacteria. Using death kinetics studies (time-kill studies), the rate at which the antimicrobial agent kills pathogens over a 24-hour period was determined. The antibacterial activity of Commiphora marlothii (stem) was observed to begin at ca. 30 min of the exposure of S. aureus to the different concentrations of plant extract. All concentrations exhibited antibacterial activity, with a complete bactericidal effect achieved by all test concentrations by the 24th hour. Commiphora pyracanthoides (stem) displayed anti-inflammatory activity through good inhibition of the 5-LOX enzyme (IC50 = 27.86 μg/ml). The ability of extracts and kaempferol to inhibit the in vitro growth of three human cancer cell lines, namely the colon adenocarcinoma (HT-29), breast adenocarcinoma (MCF-7), and the neuronal glioblastoma (SF-268), was evaluated using the sulforhodamine (SRB) antiproliferative assay. The most active Commiphora species against the HT-29 cells were C. glandulosa (leaf and stem) and C. marlothii (leaf). The MCF-7 cell line was the most sensitive to indigenous Commiphora species, with C. edulis (leaf and stem), C. glandulosa (leaf and stem), C. marlothii (leaf), C. pyracanthoides (leaf and stem), C. schimperi (stem), and C. viminea (stem) all possessing an inhibition greater than 80% at 100 μg/ml. Commiphora glandulosa (leaf and stem) and C. pyracanthoides (leaf and stem) were the two most active species against the SF-268 cells, with IC50 values ranging between 68.50 μg/ml and 71.45 μg/ml. The inhibition of the cancer cell proliferation by kaempferol in all three-cancer cell lines was determined, with IC50 values of 9.78 μg/ml in HT-29 cells, 20.21 μg/ml in MCF-7 cells and 43.83 μg/ml in SF-268 cells. The microculture tetrazolium cellular viability (MTT) assay was used to determine the cellular toxicity of the extracts against transformed human kidney epithelium (Graham) cells. Commiphora glandulosa (stem) proved to be most toxic (IC50 = 30.5 μg/ml). The IC50 values for all other extracts were in excess of 95 μg/ml suggesting low in vitro toxicity for the majority of the species. A phytochemical investigation of the non-volatile constituents of the leaf and stems was conducted using high performance liquid chromatography (HPLC). The HPLC profiles and UV spectra of the stem extracts, and the representative flavonoid patterns in the leaf extracts of the species indicate that a similarity exists in their chemical fingerprint.

Screening of four plants commonly used in ethnoveterinary medicine for antimicrobial, antiprotozoal and anti-oxidant activity

Urginea sanguinea, Aloe marlothii, Elephantorrhiza elephantina and Rhoicissus tridentate are all plants utilized for the management of tick borne diseases in the Madikwe area of North-west province. These plants, in certain concoctions, are believed to be effective against “seme”, “gala” and “Bolwetsi jwa mothlapo o moshibidu” which we have assumed to represent heartwater, gallsickness and redwater from circumstantial epidemiological data available. To obtain a representative extract, which would be indicative of the general activity of the plant, only acetone or methanol extracts were tested for the presence of antimicrobial, antiparasitic or anti-oxidant activity within that specific plant. Activity in all cases made use of either an in vitro biological assay or more specific chemical tests, which were validated in all cases. Ehrlichia ruminantium, Babesia caballi and Theileria equi, all grown in specific cell cultures, were used as a model for evaluating the efficacy against the common protozoan and rickettsial diseases caused by these organisms in livestock. Staphylococcus aureus, Enterococcus faecalis, Pseudomonas aeruginosa and Escherichia coli, four human nosocomial infectious agents, were used as an indicator for the presence of antibacterial activity against these common animal bacterial pathogens. Diphenyl-picrylhydrazyl and the trolox equivalent anti-oxidant chemical assays were used to determine anti-oxidant activity, which although not curative, may aid in the recovery from an infection by stimulating the immune system. The activities demonstrated among the various plants and organisms were not consistent. E. elephantine extracts were the most effective, with activity demonstrable in all biological and chemical screening assays. Although R. tridentate demonstrated poor activity (> 100 ìg/ml) against the tick-borne parasites, the plant extract did demonstrate significant anti-oxidant activity. U. sanguinea extracts showed good activity in both the antibacterial and anti-rickettsial assays (EC50 = 44.49 ng/ml), which may be due to the presence of the toxic bufadienolides present within the plant. A. marlothii possessed significant anti-rickettsial activity (EC50= 111.4 µg/ml) and to a lesser degree antibacterial activity. The results of the study support the use of these plants against heartwater, gallsickness and redwater, which gives credence for the traditional use against “Seme, Gala, and Bolwetsi jwa mothlapo o moshibidu”. Further studies are required to isolate and determine the structure of the active compounds of these plants as well as to confirm the safety and efficacy of the extracts against disease conditions in livestock. E. elephantina ekstrakte was die mees doeltreffende met die biologiese and chemiese bepalings. R. tridentata het sterk anti-oksidantaktiwiteit gehad, maar het lae aktiwiteit (> 100 µg/ml) teen bosluis-oorgedraagde parasiete gehad. U. sanguinea ekstrakte was aktief in beide die antibakteriese en anti-riketsiale bepalings (EC50 = 44.49 ng/ml), wat moontlik toegeskryf kan word aan die giftige bufadienoliede teenwoordig in hierdie species. A. marlothii ekstrakte het betekenisbolle anti-riketsiale aktiwiteit (EC50 = 111.4 µg/ml) maar slegs geringe antibakteriese aktiwiteit gehad. Hierdie resultate bevestig die moontlike waarde van hierdie species teen hartwater, galsiekte and rooiwater, en ondersteun die tradisionele etnoveterinêre gebruik teen “Seme, Gala, and Bolwetsi jwa mothlapo o moshibidu”. INCORPORATION OF BIO-BASED MOLECULES IN SILICONES THROUGH MICHAEL ADDITIONS

Silicone stands as an indispensable material for numerous applications; however, its high energy-cost synthesis poses significant environmental challenges. To address these concerns, bio-based silicone has gained considerable attention, showcasing its potential to dilute energy density while offering inherent functional benefits. Despite promising prospects, existing incorporation methods often involve protecting groups, rare metal catalysts, and multistep synthesis, which contradict green chemistry principles. The aza- Michael reaction emerges as a superior choice due to its high atom economy and mild reaction conditions. However, it still suffers from prolonged reaction times, hindering its overall efficiency and sustainability. This thesis utilizes self-activated beta-hydroxy acrylates to greatly enhance aza-Michael kinetics, achieving a 3-fold rate enhancement in solvent-free silicone synthesis. This fast aza-Michael reaction acts as the platform for the incorporation of Vitamin C and amino acids into silicone materials. Vitamin C-modified silicone demonstrates the potential for controlled antioxidant activity release, while amino acid-functionalized silicones are synthesized using choline amino acid ionic liquids, presenting a protecting-group-free and solvent-free synthesis method. Moreover, the synthesized choline amino acid-functional polymers and elastomers are investigated for their dielectric properties revealing promising potential for dielectric elastomer actuator applications. These innovative methods offer green alternatives for incorporating hydrophilic biomolecules into hydrophobic silicone systems, providing new functionalities that address both environmental and functional requirements. / Thesis / Doctor of Science (PhD)

No presente trabalho são relatados os resultados obtidos através de ensaios de caracterização para os diferentes tipos de óleos vegetais estudados, no estado novo e puro, tais como viscosidade, número de acidez, índice de iodo, índice de saponificação, espectroscopia por infravermelho e curvas de resfriamento. Os mesmos ensaios foram realizados com as amostras de óleo de soja com diferentes concentrações de aditivos anti-oxidantes. As diferentes formulações feitas com óleo de soja foram submetidas ao um processo de envelhecimento acelerado, e após esse processo de oxidação, foram realizados novamente os mesmos ensaios. Pode-se então, comparar qual amostra sofreu menor oxidação durante um mesmo tempo de envelhecimento. Assim, pode-se verificar qual aditivo anti-oxidante obteve melhor desempenho, retardando o processo de oxidação no óleo de soja. / In this work has studied different types of vegetable oils such cotton, canola, sunflower, corn and soybean. Characterization in terms of viscosity, number of acidity, index of iodine, index of saponification and by infrared spectroscopy, were performed. Cooling performance was also evaluated. Those tests were performed before and after oxidation. Oxidation of the oils were obtained using a special apparatus, provoking degradation after 72 hours of test. Different formulations using soybean oil and antioxidants additivation were also evaluated. Better performance in terms of oxidation resistance was obtained with formulation 1% Irganox L109.

Research on development of water soluble anti-cancer drugs is one of the great challenges of modern medicinal chemistry. Resveratrol (Res) is one of the many phytoalexins producing stilbenoids present in several medicinal plants, grape skin, peanuts, and red wine. It has been found to exhibit anti-cancer, anti-inflammatory, and anti-oxidant properties. Water solubility and bioavailability are some of the setbacks of this interesting compound. In view of this, effort has been made to synthesize amino acid derivative of resveratrol to improve its bioavailability and solubility in water. Methyl 4-{-[(1E)-2-(3, 5-dihydroxyphenyl)-ethenyl] - phenoxy} butyrate (7), a novel ester intermediate, has been synthesized and could be subjected to further chemical transformations to obtain amino acid derivatives.

Natural oils are regularly used in cosmetics and as treatment for numeral skin conditions (Nielsen, 2006:575). The natural products industry is a multibillion dollar industry and has grown tremendously over the past few years. Natural oils used in cosmetics contain a range of fatty acids which contribute to several valuable properties in cosmetic- and personal care products. Fatty acids are divided into saturated acids and unsaturated acids (Vermaak et al., 2011:920,922). Because of the popularity and wide diversity of skin care products, it is necessary to create products that will distinguish themselves from the rest of the commercial products. To include natural oils in skin care products is a new way to prevent skin ageing, as well as other dermatological conditions. In this study, a natural oil, namely crocodile oil was used. Crocodile oil is obtained from the fat of the Nile crocodile (Crocodylus niloticus). Crocodile oil has the same composition as human skin oil. It only differs with regard to the percentages of the ingredients present. Crocodile oil contains saturated and unsaturated fatty acids. Because of the similar composition as human skin oil, crocodile oil will rarely be allergenic when applied to human skin and therefore will be a very accepted and harmless product to use (Croc city, 2012). There are many claims of positive results when crocodile oil containing products have been used. It includes fading of freckles, treatment of acne and pimple marks, dark lines, wrinkles and laugh lines. It also includes vanishing of dark shadows, sun spots and other discolorations. It helps prevent discolorations from forming and makes the skin softer, brighter and more attractive. It also controls rashness and dryness (Croc city, 2012). Because of crocodile oil’s anti-ageing, anti-fungal and anti-bacterial effects claimed by crocodile oil suppliers, and due to the fact that little scientific data is available on crocodile oil, it was decided to investigate the claims. In this study, the aims and objectives were to use natural oil, namely crocodile oil, and investigate the fatty acid profile, anti-microbial and anti-fungal activity, anti-oxidant activity, toxicity studies, stability determination of crocodile oil lotion and clinical efficacy testing of the anti-ageing effects. To determine the fatty acid profile of crocodile oil, fatty acid methyl ester (FAME) analysis with gas chromatography were used. Identification of FAME peaks in the samples was made by comparing the relative retention times of FAME peaks from samples to those of reference standards. The composition of fatty acids in crocodile oil compared well to fatty acids found in human skin oil. Anti-microbial and anti-fungal tests were done by Envirocare Laboratories, North-West University, Potchefstroom. Staphylococcus aureus, Esterichia coli, Pseudomanas aeruginosa, Candida albicans, Brasiliensis, Propionibacterium acnes and Trichophyton rubrum cultures were used to determine the anti-microbial and anti-fungal activity of crocodile oil. Unfortunately no activity was observed. The anti-oxidant properties of crocodile oil and crocodile oil lotion were determined by using the most commonly used method for measuring Malondialdehyde (MDA) in biological samples, namely the thiobarbituric acid (TBA) test. This method is based on spectrophotometric quantification of the pink complex formed after reaction of MDA with two molecules of TBA. No anti-oxidant activity was observed in the oil or the lotion. Toxicity studies were performed by Dr. D. Goosen (BVSc Hons. Pret.) from Tswane University of Technology (Pretoria, South Africa). The studies showed that the lotion had no toxicity in the skin sensitisation, acute dermal toxicity and acute dermal irritation studies. To determine the stability of the crocodile oil lotion, the formulated products were store at 25 °C / 60% RH (relative humidity), 30 °C / 60% RH and 40 °C / 75% RH for 6 months in the original packaging as well as a glass container. The stability tests included pH, viscosity, visual appearance assessment, zeta-potential, droplet size and mass loss. The crocodile cream lotion was stable over the 6 months period in both containers. Clinical efficacy testing was performed at the CEL (Clinical Efficacy Laboratory) of the North-West University, Potchefstroom, South Africa. A short-term study over a period of 3 h was performed to investigate the hydrating effects of crocodile oil lotion. A long-term study over a period of 12 weeks was performed to examine the anti-ageing effects of crocodile oil lotion. An erythema study was also conducted to test the anti-erythema properties of crocodile oil lotion. Although the crocodile oil lotion as well as the placebo lotion showed an increase in skin hydration, there was no significant difference between the two treatments.
