Association of single nucleotide polymorphisms in the leptin gene and segregation by ultrasound backfat at weaning on carcass performance in steers

Marston / One hundred ninety-three crossbred steers from two herds were used to determine the association of leptin gene polymorphisms and effects of feedlot management of lean and fat steers on carcass performance. Steers were sorted into FAT and LEAN groups by ultrasound backfat at weaning and randomly assigned to a finishing phase. Steers were assigned to a backgrounding phase (BACK) and were fed a forage-based diet for 90 days or directly entered a feedlot phase (FEED). Genotypes were determined by IGENITY® (Atlanta, GA) for a panel of nine single nucleotide polymorphisms (SNP) in the leptin gene (UASMS1, UASMS2, C963T, E2FB, A1457G, and A252T), leptin receptor (T945M), growth hormone receptor (G200A), and fat metabolism enzyme (K232A). Initial backfat (BF) means for the FAT and LEAN group were 3.4 mm and 1.8 mm, respectively. Mean on-test weight was heavier for FAT (306.5 kg) than LEAN (292.9 kg). Age-adjusted hot carcass weights (HCWT) were heavier for LEAN/BACK when compared to FAT/FEED and FAT/BACK (P<0.05). Dressing percent for the FAT/FEED group tended to be higher (P<0.10) over all groups except LEAN/BACK. Steers that went directly to the feedlot had higher marbling scores than backgrounded groups. FAT/FEED had higher 12th rib BF than the other contemporaries. None of the SNPs were useful for predicting ultrasound BF at weaning. Some association was detected with UASMS2 and HCWT (P<0.10) resulting in an 11 kg difference between genotype CC and CT (P<0.05). Five of the leptin polymorphisms (UASMS1, UASMS2, A1457G, C963T, and E2FB) were associated with adjusted carcass BF (P=0.01, 0.06, 0.01, 0.01, and 0.01, respectively) and calculated yield grade (P<0.01). A252T was associated with REA, and genotype TT was larger than AA and AT (P<0.05). This study suggests that segregation by initial fatness estimates and feedlot management strategies has the opportunity to increase HCWT by 35 kg. Sorting cattle upon feedlot entry by ultrasound BF and segregation using genetic markers are useful tools that can assist in the estimation of carcass composition in the live animal. With additional research, the possibility exists to incorporate genetic markers into feedlot selection to assist in marketing decisions.

Vliv kondice prasnic na jejich následnou reprodukční užitkovost / Influence of condition sows on their subsequent reproductive performance

The aim was to assess the effect of condition of sows on subsequent reproductive performance. Fitness was assessed based on subjective evaluation using a five-point scale from 1 to 5. Rating condition was performed in a group of around 162 pieces by the current cast piglet always for the reproductive period before birth and before weaning, at intervals during 2009 and 2010. The company has been annually monitored and backfat thickness of sows selected for the period before birth and before weaning. The results of subjective assessment of condition of sows found that sows in the measured density reached an optimal condition in the range of 3,5 to 4 points. The best condition in sows before parturition was on the 3rd, 4th and 7 litter. Before weaning sows were fit from 2,5 to 3,5 points. In the period before weaning reached 450 sows condition 3 with an average of 13,14 pieces of piglets born to 10,21 piglets born alive. In assessing the data on the height of back fat measured now been found that the examined sows ranged average amount of fat in the range of optimum condition for each reproductive period before parturition 19,85 mm, 16,52 mm before weaning, and the difference was 3,33 mm. The statistics were revealed in the amount of fat before birth dependent on the number of piglets (p=0.02), which is statistically significant indicator. At the height of the fat before weaning (p=0.65), the relationship is statistically insignificant. The biggest statistical dependence (p=0.00) difference was found in fat, which tells us that with increasing height of fat before birth increases the amount of fat before weaning. In the last part of the data on the reproductive performance compared to those undertaking trade mark under the key ?reproduction?. It was found that the company has a prejty good level of breeding, despote some fluctuatins in farming losses as a percentage of piglets and sows pregnancy. The company reached the average (between 2008 and 2010) in weaned piglets per sow and year of 21,23 pc, live born piglets per sow and year of 25,37 pc at 2,33 litters per sow per year, but has a large loss of suckling piglets , an average of 10,32 % and 3,78 % rearing, the percentage of sows insemination was 80 % of the company, should be min. 85 %. Due to deteriorating indicators monitored the implementation of an enterprise for restocking.

Transcriptoma comparativo e redes de co-expressão gênica associados a características de carcaça de bovinos Nelore / Comparative transcriptome and gene co-expression networks associated with carcass traits of Nellore cattle

O objetivo deste trabalho foi estudar o transcriptoma do músculo Longissimus dorsi (LD) de bovinos da raça Nelore associado as características de área de olho de lombo (AOL) e espessura de gordura subcutânea (EGS), a fim de encontrar genes diferencialmente expressos (GDE), conjuntos de genes co-expressos, vias metabólicas e processos biológicos que regulam essas características. Foram utilizados dados de 385 bovinos Nelore castrados para obtenção das medidas fenotípicas de AOL e EGS, mensuradas entre a 12ª e 13ª costelas do músculo LD. Estes animais foram separados em dois grupos extremos, alto e baixo, contendo seis animais cada, baseados nos valores genômicos estimados (GEBV), para AOL e para EGS. Estes conjuntos de 12 animais foram submetidos à análise de expressão diferencial afim de encontrar GDE entre os grupos. Em seguida, a análise de enriquecimento funcional a partir da lista de GDE foi executada por meio do DAVID (Database for Annotation, Visualization and Integrated Discovery) e da combinação do BiNGO (Biological Networks Gene Ontology) e REVIGO (Reduce + Visualise Gene Ontology). Para AOL, foram identificados 101 GDE entre os grupos de alto e baixo GEBV. Desses, 72 genes apresentaram-se down-regulated e 29 up-regulated no grupo baixo. Para EGS, foram encontrados 18 GDE, dos quais treze apresentaram-se up-regulated e cinco down-regulated no grupo baixo. Dentre as vias metabólicas e processos biológicos encontrados para essas características, destacam-se a via de sinalização MAPK (bta04010) e a via da endocitose (bta04144) - AOL - e, os processos de biossíntese de andrógenos (GO:0006702) e via de sinalização canônica Wnt (GO:0060070) - EGS. Para encontrar conjuntos de genes co-expressos associados aos tratamentos (AOL e EGS), foi utilizado o pacote WGCNA (Weighted Correlation Network Analysis) do R, com dados de contagens de transcritos de 43 animais. Foram identificados 37 módulos, dentre eles, os módulos Azul, Verde-escuro e Salmon apresentaram correlação significativa de 0,3 com a EGS (P&lt;0,10). Os genes destes módulos foram selecionados para análise de enriquecimento funcional quando seus valores de filiação ao módulo foram superiores a 0,7. O módulo azul apresentou o maior número de genes co-expressos, com 953 genes submetidos à análise de enriquecimento funcional. Foram encontradas seis vias metabólicas e 101 termos do Gene Ontology para este módulo, conforme análise do DAVID, além de 108 processos biológicos identificados pelo BiNGO/REVIGO. Os resultados do presente estudo enfatizam a complexidade da regulação gênica do músculo LD de bovinos Nelore associado às características de AOL e EGS. Estes resultados nos auxiliam a compreender melhor os diversos processos moleculares envolvidos na deposição muscular e de gordura de cobertura, características de carcaça economicamente importantes para a produção da carne bovina. / The aim of this work was to study the Longissimus dorsi (LD) muscle transcriptome of Nellore cattle associated with ribeye area (REA) and backfat thickness (BFT) to find differentially expressed genes (DEG), coexpressed gene modules, metabolic pathways, and biological processes that regulate these traits. Data from 385 Nellore steers were used to obtain the phenotypic measures of REA and BFT, measured between the 12th and 13th ribs of the LD muscle. These animals were divided into two extreme groups, high and low, with six animals each, based on the estimated genomic breeding values (GEBV) for REA and for BFT. These sets of 12 animals were submitted to differentially expressed gene analysis to find DEG between the groups. Then, the functional enrichment analysis from the list of DEG was performed by DAVID (Database for Annotation, Visualization and Integrated Discovery) and the combination of BiNGO (Biological Networks Gene Ontology) and REVIGO (Reduce + Visualize Gene Ontology). For REA, 101 DEG were identified between the high and low GEBV groups. Of those, 72 genes were down-regulated and 29 up-regulated in the low group. For BFT, 18 DEG were found, of which thirteen were up-regulated and five were down-regulated in the low group. Among the metabolic pathways and biological processes found for these traits, the MAPK signaling pathway (bta04010) and the endocytosis pathway (bta04144) - for REA -, the androgen biosynthetic processes (GO: 0006702) and the canonical Wnt signaling pathway (GO: 0060070) - for EGS - can be highlighted. To find coexpressed gene modules associated with the traits (REA and BFT), we used the WGCNA (Weighted Correlation Network Analysis) package from R, with data from transcript counts of 43 animals. A total of 37 modules were founded. Among them, the Blue, Dark Green and Salmon modules had a significant correlation of 0.3 with BFT (P&lt;0.10). The genes of these modules were selected for functional enrichment analysis when their module membership values were higher than 0.7. The blue module had the highest number of co-expressed genes, with 953 genes submitted to functional enrichment analysis. Six metabolic pathways and 101 Gene Oontology terms were founded for this module by DAVID analysis, in addition, 108 biological processes were identified by BiNGO/REVIGO. The results of the present study emphasize the complexity of gene regulation in the LD muscle of Nellore cattle associated with REA and BFT. These results can help us to better understand the different molecular processes involved in muscle and fat deposition, which are economically important carcass traits for beef production.

Características de carcaça de bovinos da raça Canchim : estimativas de parâmetros genéticos e associação com marcadores moleculares /

Resumo: A espessura de gordura subcutânea (EGS) e a área de olho de lombo (AOL) são características importantes ligadas à eficiência de produção e à qualidade da carne bovina. Para considerá-las em um programa de avaliação genética e/ou de seleção assistida por marcadores, é necessário quantificar sua variação genética aditiva e validar marcadores moleculares associados a elas. Desta forma, o objetivo neste trabalho foi contribuir para o melhoramento genético da raça Canchim por meio do estudo das características de carcaça EGS e AOL e de peso, obtidas em média aos 18 meses de idade, com abordagens quantitativas e moleculares. Dados de 987 animais da raça Canchim (5/8 Charolês + 3/8 zebu) e MA (filhos de touros Charolês e vacas 1/2 Canchim + 1/2 zebu), machos e fêmeas criados em pastagens, nascidos entre 2003 e 2005, foram analisados pelo método dos quadrados mínimos, cujo modelo estatístico incluiu os efeitos de ano de nascimento, rebanho, sexo, grupo genético e idade à medição (covariável). Foram feitas análises uni e bicaracterísticas utilizando-se um modelo animal que incluiu, além de efeitos fixos, os efeitos aleatórios genético aditivo direto e residual, por meio do método da máxima verossimilhança restrita, para estimar as herdabilidades e as correlações genéticas entre as características. Foram realizadas análises para verificar efeitos de cinco marcadores moleculares (BMS490 e ETH10 do cromossomo 5, INRA133 e ILSTS090 do cromossomo 6 e BMS2142 do cromossomo 19) sobre as características EGS e AOL, com modelo estatístico igual ao anterior mais o efeito do genótipo para o marcador... To consider them as selection criteria in genetic evaluation and marker assisted selection programs, it is necessary to quantify their additive genetic variation and to evaluate the existence of genetic markers associated to them. The objective in this work was to contribute to the Canchim breeding program through the study of BFT, REA and body weight (BW), using quantitative and molecular informations. Data on 987 eighteen months old Canchim (5/8 Charolais + 3/8 zebu) and MA (offspring of Charolais bulls and 1/2 Canchim + 1/2 Zebu cows) bulls and heifers, grown on pasture, born from 2003 to 2005, were analyzed by the least squares method, with a model that included the fixed effects of year of birth, herd, sex, genetic group and age (covariate). One and two-trait analyses with a model that included fixed effects and the additive direct and residual random effects, by the restricted maximum likelihood method, were undertaken to estimate heritabilities and genetic correlations among the traits. Statistical analyses to verify the effect of five genetic markers (BMS490 and ETH10 in chromosome 5, INRA133 and ILSTS090 in chromosome 6 and BMS2142 in chromosome 19) on BFT and REA were also realized with a similar model, but including the genotype of a marker as a fixed effect. Estimativa de características de carcaça e ponto ideal de abate por ultra-sonografia, em bovinos submetidos a diferentes níveis energéticos na ração. / Ultrasonography prediction of carcass characteristics and finishing of cattle fed different energy levels.

Este trabalho teve como objetivo avaliar Características de carcaça por ultra-sonografia de 48 animais confinados (24 Brangus e 24 Nelore), com sistema de controle de consumo individual através de portões eletrônicos(Calan Gates), recebendo diferentes níveis de energia na ração (20, 40, 60 e 80%). Foi utilizado um equipamento de ultra-som Piemedical Scanner 200 Vet, com transdutor linear de 18 cm e 3,5 MHz, a cada intervalo de aproximadamente 28 dias, para obter a área de olho de lombo (AOLU), espessura de gordura subcutânea (EGSU) entre a 12ª e a 13ª costelas e a espessura de gordura na picanha (EGPU), sob o terço superior do músculo Bíceps femuris. Após 142 dias de confinamento os animais foram abatidos e obteve-se o peso de carcaça quente (PCQ) e o rendimento de carcaça (RC). Após 24 horas de resfriamento, as carcaças foram cortadas entre a 12ª e a 13ª costelas e obtidas a área de olho de lombo (AOLC) e a espessura de gordura (EGSC) na carcaça. As correlações entre as medidas por ultra-som e na carcaça foram de 0,83 entre AOLU e AOLC e 0,86 entre EGSU e EGSC. Os animais Brangus apresentaram melhores respostas ao tratamento com 60% de concentrado, enquanto que os animais Nelore responderam melhor ao tratamento 40%. Equações de regressão utilizando o peso vivo (PV), AOLU, EGSU e EGPU explicaram 71% da variação do PCQ quando a medida foi realizada no início do experimento e cerca de 97% quando realizada imediatamente antes do abate, nos animais Brangus. Nos animais Nelore as equações explicaram cerca de 50% na medida realizada no início do trabalho e 96% na medida antes do abate. As equações para estimar o RC utilizando as mesmas características explicaram cerca de 28% da variação quando a medida foi realizada 142 dias antes do abate e 25%imediatamente antes do abate, nos animais Brangus. Nos animais Nelore, as equações explicaram cerca de 15% e 21% quando realizadas no início do experimento ou 142 dias depois, respectivamente. A estimativa da espessura de gordura subcutânea ao abate através do PV, AOLU, EGSU e EGPU explicaram cerca de 20% quando utilizados os dados obtidos no início do trabalho até 73% quando os dados utilizados foram os obtidos imediatamente antes do abate. As medidas de ultra-som apresentaram boa acurácia e podem ser utilizadas para estimar diferenças entre grupos de animais, mas as equações para estimar RC e ponto ideal de abate necessitam maiores estudos. / The objective of this experiment was to evaluate trough ultrasonography the carcass characteristics of 48 young bulls (24 Brangus and 24 Nellore), fed diets with four energy levels(20, 40, 60 and 80% of concentrate) in pens with Calan gates. The Piemedical Scanner 200 ultrasound with a linear transductor of 18 cm and 3,5 MHz was utilized, and the measurements taken at 28 days intervals of the rib eye area (AOLU) and subcutaneous fat thickness (EGSU) between the 12 th and 13 th ribs and also over the Biceps femoris muscle (EGPU). After 142 days on feed all the animals were slaughtered and the hot carcass weight (PCQ) was taken and the carcass dressing percentage (RC) calculated. After 24 hours of chilling the carcasses were separated between the 12 th and 13 th ribs and the rib eye area (AOLC) and fat thickness (EGSC) measured directly. The correlations between ultra-sound and carcass measurements were 0,83 for AOLU and AOLC and 0.86 for EGSU and EGSC. The Brangus bulls showed better performance with 60% concentrate diets and the Nellore with 40% concentrate diets. Regression equations using live weight (PV), AOLU, EGSU and EGPU explained 71% of the variation in the PCQ when measured 142 days before slaughter and 97% immediately before slaughter for the Brangus bulls. For the Nellore bulls the equations explained 50% in the first measurement and 96% the day before slaughter. The equations to estimate RC using the same ultrasound measurements explained 50 and 96% and 15 and 21% of the variation when taken 142 days before or in the day before slaughter for the Brangus and Nellore, respectively. To estimate a determined endpoint the PV, AOLU, EGSU and EGPU explained 20% of the variation when the data was taken in the beginning of the feeding period and increased up to 73 when the data were taken just before slaughter. The ultrasound measurements showed good accuracy and could be used to estimate differences among cattle groups, but equations to estimate RC and determined endpoint need further studies.

Com o objetivo de acasalar novilhas aos 14/15 meses de idade, cem bezerras Hereford foram submetidas a dois sistemas de alimentação (SA) de 20/06 a 20/08: a) PN=50 bezerras em pastagem natural; b) PNs=50 bezerras em pastagem natural suplementadas (14% PB; 72% NDT) a 1%/dia do peso corporal (PC). Em 20/08/2012, em lote único, pastejaram azevém (PA). O período reprodutivo (PR) foi em PN. O escore de trato reprodutivo (ETR-US) e a área pélvica (APR) foram determinados aos 365 dias de idade. De 20/06 a 20/08, o ganho diário médio (GDM) e o peso corporal (PC) das bezerras do PNs foram 67,71% e 9,95% e 6,19% e 15,46% superiores, respectivamente, às do PN. O PNs também apresentou área do músculo longissimus (AOLUS) superior em 8,48%, 22,91% e 21,82%, respectivamente, ao PN. A suplementação proporcionou maior espessura de gordura subcutânea (EGSUS), 95,12% superior em relação ao PN. Em pastagem de azevém, na primeira pesagem, o PNs apresentou 5% de GDM superior às do PN. Bezerras do PNs apresentaram PC 13,36% superior ao PN e 18,20% e 13,33% de AOLUS superiores, respectivamente, aos valores do PN. O PNs obteve maior EGSUS, 1,43±0,67 e 0,89±0,75 mm, superando as do PN. Com o objetivo de acasalar novilhas aos 14/15 meses de idade, cem bezerras Hereford foram submetidas a dois sistemas de alimentação (SA) de 20/06 a 20/08: a) PN=50 bezerras em pastagem natural; b) PNs=50 bezerras em pastagem natural suplementadas (14% PB; 72% NDT) a 1%/dia do peso corporal (PC). Em 20/08/2012, em lote único, pastejaram azevém (PA). O PNs obteve maior EGSUS, 1,43±0,67 e 0,89±0,75 mm, superando as do PN. / Aiming at breeding heifers with 14-15 months of age, 100 Hereford calves were submitted to two feeding systems (FS) between 06/20 and 08/20: a) NP=50 calves grazing on natural pasture and b) NPs=50 calves grazing on natural pasture and fed a supplement (14% CP; 72% TDN) at 1% body weight (BW)/day. On 08/20, heifers were transferred as single group to a ryegrass (RG) pasture. During the last 15 days of ryegrass grazing (10/18) and the first 15 days upon returning to NP (11/03), half of the heifers in each previous FS were fed the supplement, thereby establishing new FS: c) NPs-RGs: previously supplemented on NP and supplemented for 15 days on RG + 15 days on NP; NPs-RG: previously supplemented on NP; e) NP-RGs: supplemented only for 15 days on RG + 15 days on NP; and f) NP-RG: not supplemented in either period. During the breeding season (BS), heifers grazed on NP. Reproductive tract score (TRS) and relative pelvic area (RPA) were determined at 365 days of age. On 07/20 and 08/20, NPs heifers presented 67.71% and 9.95% and 6.19% and 15.46% higher daily weight gain (DWG) and BW than NP heifers, as well as 22.91% and 21.82% larger loin eye area (UREA) and 95.12% average thicker backfat (UFT). At the end of the grazing period, NPs heifers were, on average, 13.36% heavier, and presented higher average UFT than NP (1.43±0.67 vs. 0.89±0.75). Between 10/18 and 11/18, NPs-RGs and NP-RGs heifers presented higher DWG and the highest UFT values were obtained in NPs-RGs heifers. At the start of the BS, NPs-RGs had higher BCS and BW. RPA of NPs-RGs (106.41±11.92 cm²) was only different from NP-RGs (93.74±11.76 cm²). The highest percentage of puberty was determined in NPs-RG (34.3%). The presence of CL was detected in 17.8% of the heifers. Pregnancy rates of 47%, 22%, 18%, and 13% were obtained in NPs-RGs, NPs-RG, NP-RGs, and NP-RG heifers, respectively. Pregnant heifers presented higher DWG between 10/18 and 11/18, and higher RPA, BW, and BCS at the start and end of the BS. Higher pasture DM content and higher supplement levels than those evaluated in the present study are required to achieve better calf development and better reproduction indices when aiming at breeding heifers at 14-15 months of age.

Sows from breeding core of the nucleic stud, sows from nucleic stud as a whole and sows from reproductive stud were integrated in the observation.The Large White breed paternal line focused attention on the reproductive characteristics, the results of their own performance and results of unified testing. The planting of breeding sows focused attention on reproductive characters in the production of stainless crossbreed sow Czech White paternal line x Czech landrase.

Características de carcaça de bovinos da raça Canchim: estimativas de parâmetros genéticos e associação com marcadores moleculares

Para considerá-las em um programa de avaliação genética e/ou de seleção assistida por marcadores, é necessário quantificar sua variação genética aditiva e validar marcadores moleculares associados a elas. Desta forma, o objetivo neste trabalho foi contribuir para o melhoramento genético da raça Canchim por meio do estudo das características de carcaça EGS e AOL e de peso, obtidas em média aos 18 meses de idade, com abordagens quantitativas e moleculares. Dados de 987 animais da raça Canchim (5/8 Charolês + 3/8 zebu) e MA (filhos de touros Charolês e vacas 1/2 Canchim + 1/2 zebu), machos e fêmeas criados em pastagens, nascidos entre 2003 e 2005, foram analisados pelo método dos quadrados mínimos, cujo modelo estatístico incluiu os efeitos de ano de nascimento, rebanho, sexo, grupo genético e idade à medição (covariável). Foram feitas análises uni e bicaracterísticas utilizando-se um modelo animal que incluiu, além de efeitos fixos, os efeitos aleatórios genético aditivo direto e residual, por meio do método da máxima verossimilhança restrita, para estimar as herdabilidades e as correlações genéticas entre as características. Foram realizadas análises para verificar efeitos de cinco marcadores moleculares (BMS490 e ETH10 do cromossomo 5, INRA133 e ILSTS090 do cromossomo 6 e BMS2142 do cromossomo 19) sobre as características EGS e AOL, com modelo estatístico igual ao anterior mais o efeito do genótipo para o marcador... / Carcass traits like back fat thickness (BFT) and rib eye area (REA) are important to determine production efficiency and beef quality. To consider them as selection criteria in genetic evaluation and marker assisted selection programs, it is necessary to quantify their additive genetic variation and to evaluate the existence of genetic markers associated to them. The objective in this work was to contribute to the Canchim breeding program through the study of BFT, REA and body weight (BW), using quantitative and molecular informations. Data on 987 eighteen months old Canchim (5/8 Charolais + 3/8 zebu) and MA (offspring of Charolais bulls and 1/2 Canchim + 1/2 Zebu cows) bulls and heifers, grown on pasture, born from 2003 to 2005, were analyzed by the least squares method, with a model that included the fixed effects of year of birth, herd, sex, genetic group and age (covariate). One and two-trait analyses with a model that included fixed effects and the additive direct and residual random effects, by the restricted maximum likelihood method, were undertaken to estimate heritabilities and genetic correlations among the traits. Statistical analyses to verify the effect of five genetic markers (BMS490 and ETH10 in chromosome 5, INRA133 and ILSTS090 in chromosome 6 and BMS2142 in chromosome 19) on BFT and REA were also realized with a similar model, but including the genotype of a marker as a fixed effect.
